Background: Some depressed patients receive acupuncture as an adjunct to their conventional medications.Objective: This review aims to provide evidence on whether acupuncture can enhance the therapeutic effectiveness ...Background: Some depressed patients receive acupuncture as an adjunct to their conventional medications.Objective: This review aims to provide evidence on whether acupuncture can enhance the therapeutic effectiveness of antidepressants for treating depression, and explore whether acupuncture can reduce the adverse reactions associated with antidepressants.Search strategy: English and Chinese databases were searched for randomized controlled trials(RCTs)published until December 1, 2021.Inclusion criteria: RCTs with a modified Jadad scale score ≥ 4 were included if they compared a group of participants with depression that received acupuncture combined with antidepressants with a control group that received antidepressants alone.Data extraction and analysis: Meta-analysis was performed, and statistical heterogeneity was assessed based on Cochran’s Q statistic and its related P-value. Primary outcomes were the reduction in the severity of depression and adverse reactions associated with antidepressants, while secondary outcomes included remission rate, treatment response, social functioning, and change in antidepressant dose.The Grading of Recommendations Assessment, Development and Evaluation(GRADE) framework was used to evaluate the overall quality of evidence in the included studies.Results: This review included 16 studies(with a total of 1958 participants). Most studies were at high risk of performance bias and at low or unclear risk of selection bias, detection bias, attrition bias, reporting bias, and other bias. Analysis of the 16 RCTs showed that, compared with antidepressants alone, acupuncture along with antidepressants reduced the Hamilton Depression Rating Scale-17(HAMD-17) scores(standard mean difference [SMD]-0.44, 95% confidence interval [CI]-0.55 to-0.33, P < 0.01;I^(2)= 14%), Self-rating Depression Scale(SDS) scores(SMD-0.53, 95% CI-0.84 to-0.23, P < 0.01;I^(2)= 79%), and the Side Effect Rating Scale(SERS) scores(SMD-1.11, 95% CI-1.56 to-0.66, P < 0.01;I^(2)= 89%). Compared with antidepressants alone, acupuncture along with antidepressants improved World Health Organization Quality of Life-BREF scores(SMD 0.31, 95% CI 0.18 to 0.44, P < 0.01;I^(2)= 15%), decreased the number of participants who increased their antidepressant dosages(relative risk[RR] 0.32, 95% CI 0.22 to 0.48, P < 0.01;I^(2)= 0%), and resulted in significantly higher remission rates(RR1.52, 95% CI 1.26 to 1.83, P < 0.01;I^(2)= 0%) and treatment responses(RR 1.35, 95% CI 1.24 to 1.47, P < 0.01;I^(2)= 19%) in terms of HAMD-17 scores. The HAMD-17, SDS and SERS scores were assessed as low quality by GRADE and the other indices as being of moderate quality.Conclusion: Acupuncture as an adjunct to antidepressants may enhance the therapeutic effectiveness and reduce the adverse drug reactions in patients receiving antidepressants. Ginsenosides Rg 1,Rb 1,R 1,Rd,and Re are major constituents of Panax notoginseng,a famous traditional Chinese medicinal herb,which has both stimulative and inhibitory effects on the central nervous system (CNS).The mo...Ginsenosides Rg 1,Rb 1,R 1,Rd,and Re are major constituents of Panax notoginseng,a famous traditional Chinese medicinal herb,which has both stimulative and inhibitory effects on the central nervous system (CNS).The monoamine hypothesis proposes that depression is a result of the depletion of 5-hydroxytryptamine (5-HT),norepinephrine (NE) and dopamine (DA) in addition to the activation of monoamine oxidase in the CNS.The purpose of this study was to determine whether P.notoginseng Saponin (PNS) has an antidepressant activity.We investigated the antidepressant-like activities of Rg 1,Rb 1,R 1,Rd,and Re in mice,using two animal models of depression.In addition,we analyzed the neurochemicals by the chronic unpredictable mild stress test.Our results showed that Rb 1,Rd,and Re treatment at 10 mg kg-1 significantly reduced the duration of immobility in both the tail suspension and forced swimming tests.Rb 1,Rd,and Re increases in 5HT and NE levels at 10 mg kg-1 in both the frontal cortex and hippocampus.Dopamine levels increased in the hippocampus and the striatum.Moreover,5-hydroxyindoleacetic acid (5-HIAA) levels were found increased in the hippocampus.These findings suggest that the antidepressant effects of Rb 1,Rd,and Re may be related to the increase in 5-HT and NE in the CNS,and through the alterations in the synthesis or metabolism of dopamine.展开更多
The aim of the present work is to evaluate the putative antidepressant-like effects of pomegranate fruit extract including seeds (PFE) on the performance of male mice in the forced swimming test (FST), after acute adm...The aim of the present work is to evaluate the putative antidepressant-like effects of pomegranate fruit extract including seeds (PFE) on the performance of male mice in the forced swimming test (FST), after acute administration, after short-term treatment (7 days) and, after repeated administration in a 24-h period (24, 12 and 1 h before swimming test). A single dose (20 ml/kg p.o.) of PFE, in male mice provoked a significant reduction of the immobility time. Such effect was also observed with short-term treatment (7 days) with doses of 1 and 10 ml/kg/day of PFE. Moreover, it was noted that there were important differences in the onset of the antidepressant-like effect in the FST, depending on the modality of treatment with PFE. Both efficacy and potency were higher when repeated administration of PFE was used, and surprisingly the dose of 10 ml/kg (24, 12 and 1 h before swimming test) was as effective as Fluoxetine. In the same way, the short term administration (7 days) improved significantly efficacy and potency of the PFE in comparison to a single dose treatment. These results indicate an antidepressant-like profile of action for PFE which deserves further research.展开更多
BACKGROUND Recently,hepatic arterial infusion chemotherapy(HAIC)plus lenvatinib has been frequently used to treat unresectable hepatocellular carcinoma(uHCC)in China.In the clinic,the hepatic arteries of some patients...BACKGROUND Recently,hepatic arterial infusion chemotherapy(HAIC)plus lenvatinib has been frequently used to treat unresectable hepatocellular carcinoma(uHCC)in China.In the clinic,the hepatic arteries of some patients shrink significantly during this treatment,leading to improved short-term efficacy.AIM To investigate the relationship between the shrinkage of hepatic arteries and the short-term effect of HAIC plus lenvatinib treatment.METHODS Sixty-seven participants with uHCC were enrolled in this retrospective study.The patients received HAIC every 3 wk,followed by oral lenvatinib after the first HAIC course.Hepatic artery diameters were measured on CT before treatment and after 1 and 2 mo of treatment.Meanwhile,the changes in tumor capillaries were also examined on pathological specimens before and after 1 mo of treatment.The antitumor response after 1,3,and 6 mo of treatment was assessed using the modified Response Evaluation Criteria in Solid Tumors(mRECIST).The relationship between the changes in vessel diameters and the short-term effect of the combination treatment was evaluated by receiver-operating characteristic and logistic regression analyses.RESULTS The hepatic artery diameters were all significantly decreased after 1 and 2 mo of treatment(P<0.001),but there was no difference in the vessel diameters between 1 and 2 mo(P>0.05).The microvessel density in the tumor lesions decreased significantly after 1 mo of combination treatment(P<0.001).According to mRECIST,46,41,and 24 patients had complete or partial responses after 1,3,and 6 mo of treatment,respectively,whereas 21,21,and 32 patients had a stable or progressive disease at these times,respectively.Shrinkage of the tumor-feeding artery was significantly associated with the tumor response after 1,3,and 6 mo of treatment(P<0.001,P=0.004,and P=0.023,respectively);however,changes in other hepatic arteries were not significantly associated with the tumor response.Furthermore,shrinkage of the tumor-feeding artery was an independent factor for treatment efficacy(P=0.001,P=0.001,and P=0.002 and 1,3,and 6 mo,respectively).CONCLUSION The hepatic arteries shrank rapidly after treatment with HAIC plus lenvatinib,and shrinkage of the tumor-feeding artery diameter was closely related to improved short-term efficacy.展开更多

Subsequently,an index of short-term cumulative effect is established with air diffusive equation,and then the macro-scale meteorological situation and micro-scale factors of forming the short-term cumulative effect are discussed with the observation data. The macro-scale meteorological variables contain upper-level weather situations and surface weather situations. The micro-scale factors mainly include the boundary-layer height and boundary-layer stability. The analyses show that boundary-layer factors and weather variables have a significant influence on the short-term cumulative effect. The notion of short-term cumulative effect will play an important role in interpreting the severe pollution weather.展开更多

Other changcs in the respiratory epithelium are contraction of epithelium, formation of interlamellar bridge and diminishing number of microridges. Massive destruction of the respiratory epithelium by deep lesions and sloughing of epithelial layer in 0.5 and 1.0 ppm solutions after 6-12 h of exposure apparently causes death to the fish. OBJECTIVE To find that the extracellular cap of a K2P channel can act as a new allosteric site and may serve as a direct drug target.METHODS Molecular biology and cell transfection,electrophysiology,molecular docking,...OBJECTIVE To find that the extracellular cap of a K2P channel can act as a new allosteric site and may serve as a direct drug target.METHODS Molecular biology and cell transfection,electrophysiology,molecular docking,molecular dynamics simulations,virtual screening for TREK1,and depressive-related behavior tests.RESULTS Extracellular domain of TREK1 channel existed a dynamic cavity in the extracellular domain by the method of computations,mutagenesis and electrophysiology.Molecular dynamics simulations suggested that ligand-induced allosteric conformational transitions lead to blockage of the ion conductive pathway.Using virtual screening approach,we identified other inhibitors targeting the extracellular allosteric ligand-binding site of these channels.Overall,our results suggested that the allosteric site at the extracellular cap of the K2P channels might be a promising drug target for these membrane proteins.The TREK1 inhibitor TKDC had significantly faster onset than that of fluoxetine in chronic administration trials,and the study confirms that TREK1 was an important target for the development of rapid antidepressants.CONCLUSION The study is a significant step forward for understanding the function of TREK and for identifying specific inhibitors,which should be of interest to others in the field.展开更多

In case of binary data, with observing YES/NO on each subject in certain period of time, it is the proportion of subjects who respond in treatment group minus the proportion of responders in placebo group (for example, 50% vs. 30%). However, a greater difference was proposed by Rihmer et al. (2011) [1] with their supporting arguments, in that antidepressant response and placebo response had different mechanisms and there were equal chances for antidepressant responder to be responding to placebo and not responding to placebo at all. Therefore, the authors proposed 50% - 30% * 50% when the response rate in the treatment group and the placebo group are 50% and 30% respectively, resulting in higher drug-placebo difference than traditional understanding of 50% - 30%. In this article, we tried to explain why the authors misunderstood the drug-placebo concept for evaluating drug superiority, their misunderstanding of assumptions of traditional calculation, as well as their wrong reasoning on their proposed approach. All in all, we conclude the traditional approach of 50% - 30% is the right way of evaluating drug-placebo difference and the possible methods to control impact of placebo effect are briefly discussed at the end of this article.展开更多

Previous studies have reported tha...As serotoninergic transmission plays a crucial role in higher brain function in mammals, the disturbance of this system will likely have significant effects on emotion and cognition. Previous studies have reported that chronic treatment with Specific Serotonin Reuptake Inhibitors (SSRIs) during both late pregnancy and lactation was associated with abnormal behavior in adult rats. These data imply that disturbances in serotoninergic transmission during neurodevelopment may have negative effects on both the structure and function of the resultant adult brain. Therefore, the effect of a single exposure to an SSRI or a tricyclic antidepressant that preferentially inhibits serotonin reuptake during the pre-weaning period was examined in adult mice. An oral infusion of paroxetine (70 mg/kg), fluvoxamine (250 mg/kg), clomipramine (180 mg/kg), or saline was administered on postnatal day 14. Starting at 11 weeks of age, mice were assessed using a comprehensive behavioral test battery. Mice treated with paroxetine demonstrated altered behavior on the open field and hole-board tasks;those treated with fluvoxamine had behavioral changes on the light-dark box, hole-board, and sucrose preference tasks, while alteration in forced swimming and cued fear behavior were noted in mice treated with clomipramine. These results suggest that even a single administration of an antidepressant could have profound effects on behavior in adulthood, although the effects might differ dependent on the specific drug that was administered.展开更多

[Method] Taking a heavy rainstorm covering most parts of Shaanxi Province in late July 2010 as an example, data of five Doppler weather radars in Shaanxi Province were employed for a detailed analysis of the evolution of the heavy rainstorm pro- cess. [Result] Besides the good large-scale weather background conditions, the de- velopment and evolution of some mesoscale and small-scale weather systems direct- ly led to short-term heavy precipitations during the heavy rainstorm process, involv- ing the intrusion of moderate IS-scale weak cold air and presence of small-scale wind shear, convergence and adverse wind area. In addition, small-scale convection echoes were arranged in lines and formed a "train effect", which would also con- tribute to the generation of short-term heavy precipitation. Objective:Zhi-zi-chi Decoction(ZZCD)is a classical antidepressant traditional Chinese medicine(TCM)formula.However,the microbiological basis of its antidepressant effects remains unclear.The objective of this study wa...Objective:Zhi-zi-chi Decoction(ZZCD)is a classical antidepressant traditional Chinese medicine(TCM)formula.However,the microbiological basis of its antidepressant effects remains unclear.The objective of this study was to investigate the microbial basis of ZZCD.Methods:The bacteria that exhibit enhanced growth in ZZCD extracts were identified through in vitro culture.A depressed rat model was subsequently established to assess the antidepressant effects of the bacteria.A GC-MS-based metabolomics approach and molecular docking simulation were employed to investigate the potential mechanism of the antidepressant effects of the bacteria from the perspective of changes in the metabolic profile of the gut microbiota.Finally,the antioxidant effects of gut microbial metabolites following Lactobacillus rhamnosus treatment were validated through cellular experimentation.Results:ZZCD was able to promote the growth of L.rhamnosus.Furthermore,L.rhamnosus was demonstrated to increase the percentage of sucrose preference,reduce the immobility time in tail suspension and forced swimming tests,and alleviate depressive symptoms in rats.Additionally,L.rhamnosus was shown to alter the metabolic profile of gut microbiota.A total of six metabolites(ethanamin,butyric acid,2-methylbutanoic acid,behenic acid,3-methylvaleric acid and 1-hexadecanol)associated with depressive behavior in rats were identified.All six metabolites demonstrated the capacity to bind to kelchlike ECH-associated protein 1(Keap 1)in molecular docking simulations.Gut microbial metabolites after L.rhamnosus treatment increased nuclear factor erythroid 2-related factor 2(Nrf2)levels in HT22 cells.Conclusion:L.rhamnosus may be one of the microbial bases for the antidepressant effects of ZZCD.Its effects may be related to the regulation of gut microbiota metabolism and anti-oxidative stress.展开更多
Cemented paste backfill(CPB) is largely used in underground mines worldwide.A key issue associated with application of CPB is to estimate the stresses in backfilled stopes and on barricades.Recent numerical and experi...Cemented paste backfill(CPB) is largely used in underground mines worldwide.A key issue associated with application of CPB is to estimate the stresses in backfilled stopes and on barricades.Recent numerical and experimental results show that arching effect is absent shortly after the placement of CPB in stopes.However,stress decreases in barricade drift with increasing distance between the measurement points and drawpoint have also been observed,demonstrating arching effect shortly after the pouring of CPB.To explain these paradoxes,CPB is considered as Bingham fluid having a yield shear stress.Three dimensional analytical solutions are proposed to evaluate the short-term total stresses in backfilled stopes and on barricades,accounting for the CPB's yield shear stress-induced arching effect.Stress diminution due to such arching effect in the backfilled stopes and on barricades is indeed obtained.But the reduction becomes insignificant using typical yield shear stress and stope geometry.More analyses indicate that the typical yield shear stress values do not fully correspond to field conditions where the yield shear stress would increase exponentially due to apparent consolidation(loss of water by drainage,a phenomenon similar to the desiccation of overly saturated fine-grained materials).展开更多

The aim of this study was to confirm the antibacterial activity of an extract of Berberis heter...The dried roots of Berberis heteropoda Schrenk have traditionally been used to treat acute gastroenteritis and dysentery. The aim of this study was to confirm the antibacterial activity of an extract of Berberis heteropoda Schrenk root in vitro and its therapeutic effects on rats with diarrhea-predominant irritable bowel syndrome(D-IBS) in vivo, as well as to identify the related signaling pathways. A water extract of Berberis heteropoda Schrenk root(BHS) inhibited the growth of S. aureus, E. coli, P. aeruginosa and S. faecalis. BHS potentially damaged the structure of the bacterial cell membrane and decreased the activity of some membranous enzymes, eventually killing the S. aureus, E. coli, P. aeruginosa and S. faecalis bacteria. Oral administration of BHS(low, middle and high dose group, L, M and H) significantly alleviated the abdominal pain, diarrhea, and depression-like symptoms of D-IBS rats, and the efficacy index ranged from 30% to 60%, indicating that the BHS treatment was effective. BHS(L, M and H) alleviated the abnormal pathological changes in the brain, as evidenced by HE staining. The expression of CHAT, 5-HT, C-FOS and CGRP was reduced by the BHS treatment(L, M and H). Our findings provide novel insights into the use of the natural product BHS to inhibit pathogenic bacteria by destroying the bacterial structure, indicating that BHS possesses certain biological activities. Furthermore, BHS has the potential to alleviate diarrhea, abdominal pain and depression-like behaviors in D-IBS rats by regulating the brain-gut peptide levels.展开更多
Short-term temperature fluctuations(STFs),including amplitude and frequency fluctuations,are one of the main features of weather and play vital roles in determining the type of ecosystem present.Although temperature f...Short-term temperature fluctuations(STFs),including amplitude and frequency fluctuations,are one of the main features of weather and play vital roles in determining the type of ecosystem present.Although temperature fluctuations at different time scales have been extensively discussed,the research on week-scale STFs is lacking.In this study,we developed a method,that can quantify the amplitude and frequency of STFs by the thresholds from all years.We used this method to quantify the amplitude and frequency of the 7-d STFs from 1951 to 2019 across China.Our results indicate that the amplitude of the STF was much higher in the eastern part of China than in the western part,while the frequency of the STF was higher in the middle part than in the southern and northern parts;further-more,the STF was highly dependent on internal factors such as topography.The long-term STF mainly showed a decreasing trend before 1990,which implies that temperature became increasingly stable from the 1950s to the 1990s.The main influencing factors were related to topography since the trends were relatively consistent in space.A case study in Taihu Lake showed that an unstable STF in winter and summer resulted in a smaller bloom area in the following spring and autumn.Our method could eliminate seasonal effects and is capable of analyzing STFs at scales ranging from days to years.Quantifications of the amplitude and frequency also make the STF indicators more comprehensive.Furthermore,the STF increased significantly across most of China after 1990,which implies that temperature is becoming increasingly unstable.The drivers of these STFs are related to human impacts since the trends are different in space.展开更多

文摘Background: Some depressed patients receive acupuncture as an adjunct to their conventional medications.Objective: This review aims to provide evidence on whether acupuncture can enhance the therapeutic effectiveness of antidepressants for treating depression, and explore whether acupuncture can reduce the adverse reactions associated with antidepressants.Search strategy: English and Chinese databases were searched for randomized controlled trials(RCTs)published until December 1, 2021.Inclusion criteria: RCTs with a modified Jadad scale score ≥ 4 were included if they compared a group of participants with depression that received acupuncture combined with antidepressants with a control group that received antidepressants alone.Data extraction and analysis: Meta-analysis was performed, and statistical heterogeneity was assessed based on Cochran’s Q statistic and its related P-value. Primary outcomes were the reduction in the severity of depression and adverse reactions associated with antidepressants, while secondary outcomes included remission rate, treatment response, social functioning, and change in antidepressant dose.The Grading of Recommendations Assessment, Development and Evaluation(GRADE) framework was used to evaluate the overall quality of evidence in the included studies.Results: This review included 16 studies(with a total of 1958 participants). Most studies were at high risk of performance bias and at low or unclear risk of selection bias, detection bias, attrition bias, reporting bias, and other bias. Analysis of the 16 RCTs showed that, compared with antidepressants alone, acupuncture along with antidepressants reduced the Hamilton Depression Rating Scale-17(HAMD-17) scores(standard mean difference [SMD]-0.44, 95% confidence interval [CI]-0.55 to-0.33, P < 0.01;I^(2)= 14%), Self-rating Depression Scale(SDS) scores(SMD-0.53, 95% CI-0.84 to-0.23, P < 0.01;I^(2)= 79%), and the Side Effect Rating Scale(SERS) scores(SMD-1.11, 95% CI-1.56 to-0.66, P < 0.01;I^(2)= 89%). Compared with antidepressants alone, acupuncture along with antidepressants improved World Health Organization Quality of Life-BREF scores(SMD 0.31, 95% CI 0.18 to 0.44, P < 0.01;I^(2)= 15%), decreased the number of participants who increased their antidepressant dosages(relative risk[RR] 0.32, 95% CI 0.22 to 0.48, P < 0.01;I^(2)= 0%), and resulted in significantly higher remission rates(RR1.52, 95% CI 1.26 to 1.83, P < 0.01;I^(2)= 0%) and treatment responses(RR 1.35, 95% CI 1.24 to 1.47, P < 0.01;I^(2)= 19%) in terms of HAMD-17 scores. The HAMD-17, SDS and SERS scores were assessed as low quality by GRADE and the other indices as being of moderate quality.Conclusion: Acupuncture as an adjunct to antidepressants may enhance the therapeutic effectiveness and reduce the adverse drug reactions in patients receiving antidepressants. These findings must be interpreted with caution, as the evidence was of low or moderate quality and there was a lack of comparative data with a placebo control.Systematic review registration: INPLASY202150008.

文摘Ginsenosides Rg 1,Rb 1,R 1,Rd,and Re are major constituents of Panax notoginseng,a famous traditional Chinese medicinal herb,which has both stimulative and inhibitory effects on the central nervous system (CNS).The monoamine hypothesis proposes that depression is a result of the depletion of 5-hydroxytryptamine (5-HT),norepinephrine (NE) and dopamine (DA) in addition to the activation of monoamine oxidase in the CNS.The purpose of this study was to determine whether P.notoginseng Saponin (PNS) has an antidepressant activity.We investigated the antidepressant-like activities of Rg 1,Rb 1,R 1,Rd,and Re in mice,using two animal models of depression.In addition,we analyzed the neurochemicals by the chronic unpredictable mild stress test.Our results showed that Rb 1,Rd,and Re treatment at 10 mg kg-1 significantly reduced the duration of immobility in both the tail suspension and forced swimming tests.Rb 1,Rd,and Re increases in 5HT and NE levels at 10 mg kg-1 in both the frontal cortex and hippocampus.Dopamine levels increased in the hippocampus and the striatum.Moreover,5-hydroxyindoleacetic acid (5-HIAA) levels were found increased in the hippocampus.These findings suggest that the antidepressant effects of Rb 1,Rd,and Re may be related to the increase in 5-HT and NE in the CNS,and through the alterations in the synthesis or metabolism of dopamine.
文摘The aim of the present work is to evaluate the putative antidepressant-like effects of pomegranate fruit extract including seeds (PFE) on the performance of male mice in the forced swimming test (FST), after acute administration, after short-term treatment (7 days) and, after repeated administration in a 24-h period (24, 12 and 1 h before swimming test). A single dose (20 ml/kg p.o.) of PFE, in male mice provoked a significant reduction of the immobility time. Such effect was also observed with short-term treatment (7 days) with doses of 1 and 10 ml/kg/day of PFE. Moreover, it was noted that there were important differences in the onset of the antidepressant-like effect in the FST, depending on the modality of treatment with PFE. Both efficacy and potency were higher when repeated administration of PFE was used, and surprisingly the dose of 10 ml/kg (24, 12 and 1 h before swimming test) was as effective as Fluoxetine. In the same way, the short term administration (7 days) improved significantly efficacy and potency of the PFE in comparison to a single dose treatment. These results indicate an antidepressant-like profile of action for PFE which deserves further research.
文摘BACKGROUND Recently,hepatic arterial infusion chemotherapy(HAIC)plus lenvatinib has been frequently used to treat unresectable hepatocellular carcinoma(uHCC)in China.In the clinic,the hepatic arteries of some patients shrink significantly during this treatment,leading to improved short-term efficacy.AIM To investigate the relationship between the shrinkage of hepatic arteries and the short-term effect of HAIC plus lenvatinib treatment.METHODS Sixty-seven participants with uHCC were enrolled in this retrospective study.The patients received HAIC every 3 wk,followed by oral lenvatinib after the first HAIC course.Hepatic artery diameters were measured on CT before treatment and after 1 and 2 mo of treatment.Meanwhile,the changes in tumor capillaries were also examined on pathological specimens before and after 1 mo of treatment.The antitumor response after 1,3,and 6 mo of treatment was assessed using the modified Response Evaluation Criteria in Solid Tumors(mRECIST).The relationship between the changes in vessel diameters and the short-term effect of the combination treatment was evaluated by receiver-operating characteristic and logistic regression analyses.RESULTS The hepatic artery diameters were all significantly decreased after 1 and 2 mo of treatment(P<0.001),but there was no difference in the vessel diameters between 1 and 2 mo(P>0.05).The microvessel density in the tumor lesions decreased significantly after 1 mo of combination treatment(P<0.001).According to mRECIST,46,41,and 24 patients had complete or partial responses after 1,3,and 6 mo of treatment,respectively,whereas 21,21,and 32 patients had a stable or progressive disease at these times,respectively.Shrinkage of the tumor-feeding artery was significantly associated with the tumor response after 1,3,and 6 mo of treatment(P<0.001,P=0.004,and P=0.023,respectively);however,changes in other hepatic arteries were not significantly associated with the tumor response.Furthermore,shrinkage of the tumor-feeding artery was an independent factor for treatment efficacy(P=0.001,P=0.001,and P=0.002 and 1,3,and 6 mo,respectively).CONCLUSION The hepatic arteries shrank rapidly after treatment with HAIC plus lenvatinib,and shrinkage of the tumor-feeding artery diameter was closely related to improved short-term efficacy.

文摘In order to reveal the rapid increase mechanism of particulate concentration in short time,a notion of short-term cumulative effect of air particulate is defined as the significant increase of pollutant concentration in a short time under the condition of breeze,stable weather and constant emission caused by human being's activities. Subsequently,an index of short-term cumulative effect is established with air diffusive equation,and then the macro-scale meteorological situation and micro-scale factors of forming the short-term cumulative effect are discussed with the observation data. The macro-scale meteorological variables contain upper-level weather situations and surface weather situations. The micro-scale factors mainly include the boundary-layer height and boundary-layer stability. The analyses show that boundary-layer factors and weather variables have a significant influence on the short-term cumulative effect. The notion of short-term cumulative effect will play an important role in interpreting the severe pollution weather.
文摘Alterations in the respiratory epithelium of gills of a catfish, Heteropneustes fossilis by the sublethal and lethal concentrations of HgCl2 have been observcd using scanning electron microscope. Fish exposed to 0.1 ppm shows a thin coat ot mucus, but the continuous layer of mucus is not retained in 0.3 ppm after 24 h of exposure. Epithelial cells of the gill filament are hyperplastic. Other changcs in the respiratory epithelium are contraction of epithelium, formation of interlamellar bridge and diminishing number of microridges. Massive destruction of the respiratory epithelium by deep lesions and sloughing of epithelial layer in 0.5 and 1.0 ppm solutions after 6-12 h of exposure apparently causes death to the fish. SEM observations are well in conformity with the findings made at light microscopic level

文摘OBJECTIVE To find that the extracellular cap of a K2P channel can act as a new allosteric site and may serve as a direct drug target.METHODS Molecular biology and cell transfection,electrophysiology,molecular docking,molecular dynamics simulations,virtual screening for TREK1,and depressive-related behavior tests.RESULTS Extracellular domain of TREK1 channel existed a dynamic cavity in the extracellular domain by the method of computations,mutagenesis and electrophysiology.Molecular dynamics simulations suggested that ligand-induced allosteric conformational transitions lead to blockage of the ion conductive pathway.Using virtual screening approach,we identified other inhibitors targeting the extracellular allosteric ligand-binding site of these channels.Overall,our results suggested that the allosteric site at the extracellular cap of the K2P channels might be a promising drug target for these membrane proteins.The TREK1 inhibitor TKDC had significantly faster onset than that of fluoxetine in chronic administration trials,and the study confirms that TREK1 was an important target for the development of rapid antidepressants.CONCLUSION The study is a significant step forward for understanding the function of TREK and for identifying specific inhibitors,which should be of interest to others in the field.
文摘In clinical trials, drug effect is measured by a difference between subjects who are treated by experimental drug against placebo-treated subjects. In case of binary data, with observing YES/NO on each subject in certain period of time, it is the proportion of subjects who respond in treatment group minus the proportion of responders in placebo group (for example, 50% vs. 30%). However, a greater difference was proposed by Rihmer et al. (2011) [1] with their supporting arguments, in that antidepressant response and placebo response had different mechanisms and there were equal chances for antidepressant responder to be responding to placebo and not responding to placebo at all. Therefore, the authors proposed 50% - 30% * 50% when the response rate in the treatment group and the placebo group are 50% and 30% respectively, resulting in higher drug-placebo difference than traditional understanding of 50% - 30%. In this article, we tried to explain why the authors misunderstood the drug-placebo concept for evaluating drug superiority, their misunderstanding of assumptions of traditional calculation, as well as their wrong reasoning on their proposed approach. All in all, we conclude the traditional approach of 50% - 30% is the right way of evaluating drug-placebo difference and the possible methods to control impact of placebo effect are briefly discussed at the end of this article.

This was a multi-center clinical study, and a total of 138 patients include</span><span style="font-family:Verdana;">d</span><span style="font-family:;" "=""><span style="font-family:Verdana;"> 96 males and 42 females, with the age from 60 to 76 years. </span><span style="font-family:Verdana;">Ai-Jing has obtained the Chinese invention patent since 2008</span><span style="font-family:Verdana;"> (ZL991-03528.3). A novel technique acupoint injection method was adopted. Except for lung cancer, patients with other malignant tumors were injected once every other day, 2</span></span><span style="font-family:;" "=""> </span><span style="font-family:Verdana;">ml each time. A course of treatment needs 6 Ai-Jing injections, lung cancer patients need 10</span><span style="font-family:;" "=""> </span><span style="font-family:Verdana;">-</span><span style="font-family:;" "=""> </span><span style="font-family:Verdana;">20 Ai-Jing injections. At the same time, patients were treated with traditional medicine decoction. Due to the limited number of cases, cure rate, recovery rate and remission rate were calculated as a whole, respectively</span><span style="font-family:;" "=""> </span><span style="font-family:;" "=""><span style="font-family:Verdana;">18.12%, 44.20% and 32.15%. All the invalid patients were lung cancer, accounting for 16.67% of the patients treated with lung cancer, and the total treatment inefficiency was 6.52%. 文摘As serotoninergic transmission plays a crucial role in higher brain function in mammals, the disturbance of this system will likely have significant effects on emotion and cognition. Previous studies have reported that chronic treatment with Specific Serotonin Reuptake Inhibitors (SSRIs) during both late pregnancy and lactation was associated with abnormal behavior in adult rats. These data imply that disturbances in serotoninergic transmission during neurodevelopment may have negative effects on both the structure and function of the resultant adult brain. Therefore, the effect of a single exposure to an SSRI or a tricyclic antidepressant that preferentially inhibits serotonin reuptake during the pre-weaning period was examined in adult mice. An oral infusion of paroxetine (70 mg/kg), fluvoxamine (250 mg/kg), clomipramine (180 mg/kg), or saline was administered on postnatal day 14. Starting at 11 weeks of age, mice were assessed using a comprehensive behavioral test battery. Mice treated with paroxetine demonstrated altered behavior on the open field and hole-board tasks;those treated with fluvoxamine had behavioral changes on the light-dark box, hole-board, and sucrose preference tasks, while alteration in forced swimming and cued fear behavior were noted in mice treated with clomipramine. These results suggest that even a single administration of an antidepressant could have profound effects on behavior in adulthood, although the effects might differ dependent on the specific drug that was administered.
文摘Histopathological effects of sublethal (0.1, 0.2, 0.3 ppm) and lethal (0.5, 1.0 ppm)solutions of mercuric chloride (HgCl2) have been studied on the gills of the air breathing catfish, Heteropneustes fossilis at the intervals ranging from 6h to 15 days. Thickening of the epithelium, swelling and hyperplasia of the mucous cells, fusion of secondary lamellae, formation of interlamellar bridge and deposition of mucous over the entire surface are some noteworthy features of mercury poisoning in sublethal concentrations. Acute pathological manifestations are formation of subepithelial space, sloughing of the epithelial layer, hemorrhage and hypertrophy of the mucous cells. 文摘[Objective] This study aimed to analyze the cause of the generation of short-term heavy precipitations in a regional heavy rainstorm in Shannxi Province. [Method] Taking a heavy rainstorm covering most parts of Shaanxi Province in late July 2010 as an example, data of five Doppler weather radars in Shaanxi Province were employed for a detailed analysis of the evolution of the heavy rainstorm pro- cess. [Result] Besides the good large-scale weather background conditions, the de- velopment and evolution of some mesoscale and small-scale weather systems direct- ly led to short-term heavy precipitations during the heavy rainstorm process, involv- ing the intrusion of moderate IS-scale weak cold air and presence of small-scale wind shear, convergence and adverse wind area. In addition, small-scale convection echoes were arranged in lines and formed a "train effect", which would also con- tribute to the generation of short-term heavy precipitation. [Conclusion] This study provided basic information for more clear and in-depth analysis of the formation mechanism of short-term heavy precipitations.

文摘Objective:Zhi-zi-chi Decoction(ZZCD)is a classical antidepressant traditional Chinese medicine(TCM)formula.However,the microbiological basis of its antidepressant effects remains unclear.The objective of this study was to investigate the microbial basis of ZZCD.Methods:The bacteria that exhibit enhanced growth in ZZCD extracts were identified through in vitro culture.A depressed rat model was subsequently established to assess the antidepressant effects of the bacteria.A GC-MS-based metabolomics approach and molecular docking simulation were employed to investigate the potential mechanism of the antidepressant effects of the bacteria from the perspective of changes in the metabolic profile of the gut microbiota.Finally,the antioxidant effects of gut microbial metabolites following Lactobacillus rhamnosus treatment were validated through cellular experimentation.Results:ZZCD was able to promote the growth of L.rhamnosus.Furthermore,L.rhamnosus was demonstrated to increase the percentage of sucrose preference,reduce the immobility time in tail suspension and forced swimming tests,and alleviate depressive symptoms in rats.Additionally,L.rhamnosus was shown to alter the metabolic profile of gut microbiota.A total of six metabolites(ethanamin,butyric acid,2-methylbutanoic acid,behenic acid,3-methylvaleric acid and 1-hexadecanol)associated with depressive behavior in rats were identified.All six metabolites demonstrated the capacity to bind to kelchlike ECH-associated protein 1(Keap 1)in molecular docking simulations.Gut microbial metabolites after L.rhamnosus treatment increased nuclear factor erythroid 2-related factor 2(Nrf2)levels in HT22 cells.Conclusion:L.rhamnosus may be one of the microbial bases for the antidepressant effects of ZZCD.Its effects may be related to the regulation of gut microbiota metabolism and anti-oxidative stress.

文摘Cemented paste backfill(CPB) is largely used in underground mines worldwide.A key issue associated with application of CPB is to estimate the stresses in backfilled stopes and on barricades.Recent numerical and experimental results show that arching effect is absent shortly after the placement of CPB in stopes.However,stress decreases in barricade drift with increasing distance between the measurement points and drawpoint have also been observed,demonstrating arching effect shortly after the pouring of CPB.To explain these paradoxes,CPB is considered as Bingham fluid having a yield shear stress.Three dimensional analytical solutions are proposed to evaluate the short-term total stresses in backfilled stopes and on barricades,accounting for the CPB's yield shear stress-induced arching effect.Stress diminution due to such arching effect in the backfilled stopes and on barricades is indeed obtained.But the reduction becomes insignificant using typical yield shear stress and stope geometry.More analyses indicate that the typical yield shear stress values do not fully correspond to field conditions where the yield shear stress would increase exponentially due to apparent consolidation(loss of water by drainage,a phenomenon similar to the desiccation of overly saturated fine-grained materials).

文摘The dried roots of Berberis heteropoda Schrenk have traditionally been used to treat acute gastroenteritis and dysentery. The aim of this study was to confirm the antibacterial activity of an extract of Berberis heteropoda Schrenk root in vitro and its therapeutic effects on rats with diarrhea-predominant irritable bowel syndrome(D-IBS) in vivo, as well as to identify the related signaling pathways. A water extract of Berberis heteropoda Schrenk root(BHS) inhibited the growth of S. aureus, E. coli, P. aeruginosa and S. faecalis. BHS potentially damaged the structure of the bacterial cell membrane and decreased the activity of some membranous enzymes, eventually killing the S. aureus, E. coli, P. aeruginosa and S. faecalis bacteria. Oral administration of BHS(low, middle and high dose group, L, M and H) significantly alleviated the abdominal pain, diarrhea, and depression-like symptoms of D-IBS rats, and the efficacy index ranged from 30% to 60%, indicating that the BHS treatment was effective. BHS(L, M and H) alleviated the abnormal pathological changes in the brain, as evidenced by HE staining. The expression of CHAT, 5-HT, C-FOS and CGRP was reduced by the BHS treatment(L, M and H). Our findings provide novel insights into the use of the natural product BHS to inhibit pathogenic bacteria by destroying the bacterial structure, indicating that BHS possesses certain biological activities. Furthermore, BHS has the potential to alleviate diarrhea, abdominal pain and depression-like behaviors in D-IBS rats by regulating the brain-gut peptide levels.

文摘Short-term temperature fluctuations(STFs),including amplitude and frequency fluctuations,are one of the main features of weather and play vital roles in determining the type of ecosystem present.Although temperature fluctuations at different time scales have been extensively discussed,the research on week-scale STFs is lacking.In this study,we developed a method,that can quantify the amplitude and frequency of STFs by the thresholds from all years.We used this method to quantify the amplitude and frequency of the 7-d STFs from 1951 to 2019 across China.Our results indicate that the amplitude of the STF was much higher in the eastern part of China than in the western part,while the frequency of the STF was higher in the middle part than in the southern and northern parts;further-more,the STF was highly dependent on internal factors such as topography.The long-term STF mainly showed a decreasing trend before 1990,which implies that temperature became increasingly stable from the 1950s to the 1990s.The main influencing factors were related to topography since the trends were relatively consistent in space.A case study in Taihu Lake showed that an unstable STF in winter and summer resulted in a smaller bloom area in the following spring and autumn.Our method could eliminate seasonal effects and is capable of analyzing STFs at scales ranging from days to years.Quantifications of the amplitude and frequency also make the STF indicators more comprehensive.Furthermore,the STF increased significantly across most of China after 1990,which implies that temperature is becoming increasingly unstable.The drivers of these STFs are related to human impacts since the trends are different in space.
