Comparison of Growth and Development Traits among Crossbreds of Dorset and Local Sheep Varieties in Gansu Province 被引量:4

In order to improve the meat production performance of local sheep varieties in Gansu Province, Dorset was introduced to crossbreed with the local sheep varieties, including Tan sheep, Small Tail Han sheep and Mongolia sheep. The offspring of different crossbreeding combinations were sampled randomly at different growth stages, and their growth and development traits were measured so as to screen out the best crossbreeding mode. The results showed that under the same crossbreeding mode, the growth rate of F3 was higher than that of F2, and of F2 was higher than that of F1. Among the F3 population, the growth rates of Dorset ×Han and Dorset × Mongolia hybrids were higher. Compared with those of Dorset ×Tan F3 hybrids, the body weights of male and female Dorset × Han and Dorset ×Mongolia F3 hybrids were increased by 5.59%, 4.40%, 5.93% and 3.76%, respectively. Among the F2 population, the growth rates of Dorset × Han and Dorset ×Mongolia hybrids were also higher. The body weights of male and female Dorset ×Han and Dorset × Mongolia F2 hybrids were higher than those of Dorset × Tan ×Han F2 hybrids by 5.99%, 3.67%, 9.80% and 5.00%, respectively. In the F1 population, the growth rates of Dorset × Han and Dorset × Mongolia hybrids were higher.Compared with those of Tan × Han F1 hybrids, the body weights of male and female Dorset × Han and Dorset × Mongolia F1 hybrids were increased by 11.32%,5.22%, 7.60% and 7.20%, respectively. Therefore, in the feeding area of Small Tai Han sheep, Mongolia sheep and Tan sheep, Dorset was the best sire for producing hybrid lambs. The economic benefit of crossbred offspring was obvious. 展开更多

In this study, to understand the variation regulation of physiological and biochemical indexs of White-Tibetan sheep with seasons, the physiological and bio- chemical indexs of White-Tibetan sheep in Hongyuan Areas of Sichuan Province in four seasons were measured. The results showed that the respiratory change range of white-Tibetan sheep in four seasons was from (32.74±21.33) to (49.96±17.62) times/min, and the variation ranges of body temperature and heart rate were from (39.27±1.05) to (39.73±0.31) ℃ and from (88.75±8.32) to (104.63±10.39) times/min, respectively. The indexes of WBC, MCV, MCH and MCHC in autumn and winter were extremely significantly lower than those in spring and summer, while RBC,HCT and RDW-CV in autumn and winter were extremely significantly higher than those in spring and summer, HGB and PLT changed in different seasons with no signifi- cance. The indexes of AST,TP, ALB, GLO and LDH in summer and autumn were relatively higher than those in winter and spring, while the indexes of PCHE, GLU, CHOL and CA in summer and autumn were relatively lower than those in winter and spring, however, the indexes of ALT and ALP were relatively higher in spring and autumn. 展开更多

This applied research seeks to explore feasible plant design for manufacturing insulation materials for construction projects using waste sheep fleece to address environmental issues related to wasted sheep wool and enhance the gross national product. The process starts by collecting low-cost sheep fleece from farms and processed via a production line, including scouring, plucking, carding, thermal bonding, and packing. The design process involves determining an optimal location, infrastructure, staffing, machinery, environmental impact, and utilities. A final economic analysis is undertaken to estimate the product’s cost, selling price, and break-even point based on the anticipated capital and operational costs. The plant is intended to process 6778 tons of sheep wool annually. The study suggests that Mafraq Industrial City is a perfect location for the plant, and purchasing land and structures is the optimal option. The projected capital cost is 1,416,679 USD, while the anticipated operational costs amount to 3,206,275 USD. Insulation material production is estimated to be 114,756 m3 annually. The material may be manufactured into 1 m wide, 0.05 m thick sheets for 2.02 USD per square meter. Thus, for a 10-year plant, a 2.47 USD/m2 selling price breaks even in one year. 展开更多

Background Birth weight is a critical economic trait in livestock production.However,its genetic architecture remains poorly understood due to historical limitations in sample size and reliance on low-density SNP arrays.In this study,we utilized low-coverage whole-genome sequencing(lcWGS)to genotype 3,007 Hu sheep,bypassing the cost and resolution constraints of conventional genotyping arrays while achieving scalable genome-wide variant detection.Results LcWGS with high imputation accuracy(97.8%allelic concordance)enabled genome-wide association studies(GWAS)identifying two novel quantitative trait loci(QTLs)on chromosomes 6 and 9.The chromosome 9 QTL encompassed a regulatory region functionally linked to PLAG1 expression through expression quantitative trait locus(eQTL)mapping.Compared with wild-type homozygotes,heterozygous carriers of the lead SNP(chr9:g.35920172A>G)presented a 9.85%increase in birth weight(3.35 kg vs.3.68 kg;Δ=0.33 kg).Notably,the derived allele of this SNP exhibited low frequencies of<0.1 across most global sheep breeds except Dorper,highlighting its potential for selective breeding applications.Leveraging lcWGS data,haplotype-based fine-mapping prioritized three candidate causal variants.A secondary QTL on chromosome 6 colocalized with the FecB mutation,a well-established locus associated with increased litter size.Intriguingly,individuals carrying one FecB allele showed a 6.18%reduction(0.22 kg)in birth weight,which tentatively indicates potential pleiotropic influences on both growth and reproductive traits.Conclusion This study demonstrates the utility of lcWGS as a cost-effective,high-resolution tool for dissecting complex traits in livestock.Our findings not only advance the understanding of birth weight genetics in sheep but also offer a blueprint for accelerating genetic improvement programs in global livestock production through costeffective,genome-wide approaches. 展开更多

Epidemiological characteristics and prevention and control strategies for Eimeria spp.in sheep and goats in China:a systematic review

Sheep and goat coccidiosis has a worldwide distribution and is an important disease on lambing farms.Infection with multiple Eimeria species can lead to severe intestinal damage in sheep/goats and economic losses on farms.Disease is a serious constraint to the healthy development of small ruminant farming.Studies published on PubMed,CNKI,VIP,Wanfang and the resulting references of selected studies were included.Risk factors affecting prevalence were analyzed and stratified by geographic location and climatic variables,age,sex,feeding model,season,sample year,breeds and environment.The total prevalence of coccidia in sheep and goats reached more than 60%in most regions,in which the dominant species in sheep were mainly E.parva,E.ovinoidalis,and E.parva and E.ahsata.East China had the lowest prevalence of coccidia infection in sheep(43.24%),and the dominant species were different from those in the other regions,mainly E.bakuensis and E.gonzalezi.Southwestern China and Central China had slightly less than 40.0%goat coccidia infection,and the dominant species in goats were mainly E.arloingi,E.aligevi,E.hirci and E.ninakohlyakimovae.Sheep/goats of different ages can be infected with coccidiosis,but lambs between 1 and 3 months of age are more susceptible to the disease.When lambs become infected,the pathogen spreads rapidly throughout the herd.Spring,summer and autumn are the seasons with a high incidence of this disease.Environmental pollution may be a significant factor in the development of coccidiosis in sheep raised in large-scale housing.This study provides a comprehensive overview of the species,morphology and geographic distribution of Eimeria species in sheep and goats,summary prevalence in different regions of China,risk factors affecting prevalence,and prevention and control strategies. 展开更多

Background As an indigenous livestock species on the Tibetan Plateau,Tibetan sheep exhibit remarkable adaptability to low temperatures and nutrient-scarce environments.During the cold season,Tibetan sheep are typically managed under two feeding regimes:barn feeding(BF)and traditional grazing(TG).However,the molecular mechanisms underlying their adaptation to these distinct management strategies remain unclear.This study aimed to investigate the adaptive strategies of rumen function in Tibetan sheep to cold-season feeding regimes by integrating analyses of rumen morphology,microbiome,metabolome,and transcriptome.Twelve healthy Tibetan sheep with similar body weights were assigned into two groups(BF vs.TG).At the end of the experiment,rumen tissues were subjected to histological observation.Multi-omics techniques were employed to evaluate the effects of cold-season feeding regimes on rumen function in Tibetan sheep.Results The ruminal papilla height,width,and muscular thickness were significantly higher in BF group.The relative abundances of Actinobacteria and Succiniclasticum were significantly elevated in the rumen of BF group,whereas Rikenellaceae,Gracilibacteria,and Lachnospiraceae showed higher abundances in the TG group.Metabolomic analysis identified 19 differential metabolites between the two groups,including upregulated compounds in BF group(fumaric acid,maltose,L-phenylalanine,and L-alanine)and TG group(e.g.,phenylacetic acid,salicyluric acid and ferulic acid).These metabolites were predominantly enriched in phenylalanine metabolism,alanine,aspartate and glutamate metabolism,and phenylalanine,tyrosine and tryptophan biosynthesis pathways.Additionally,210 differentially expressed genes(DEGs)were identified in rumen epithelium:100 upregulated DEGs in the BF group were enriched in nutrient metabolism-related pathways(e.g.,fatty acid degradation and PPAR signaling pathway),while 110 upregulated DEGs in the TG group were associated with immune-related pathways(e.g.,p53 signaling pathway and glutathione metabolism).Conclusions Among these,we observed distinct rumen functional responses to different cold-season feeding regimes in Tibetan sheep and revealed energy allocation strategies mediated by host-microbe interactions.In the BF group,Tibetan sheep adopted a"metabolic efficiency-priority"strategy,driving rumen microbiota to maximize energy capture from high-nutrient diets to support host growth.In contrast,the TG group exhibited an"environmental adaptation-priority"strategy,where rumen microbiota prioritized cellulose degradation and anti-inflammatory functions,reallocating energy toward homeostasis maintenance at the expense of rumen development and growth performance. 展开更多

Whole‑genome resequencing to investigate the genetic diversity and mechanisms of plateau adaptation in Tibetan sheep

Introduction Tibetan sheep,economically important animals on the Qinghai–Tibet Plateau,have diversified into numerous local breeds with unique characteristics through prolonged environmental adaptation and selective breeding.However,most current research focuses on one or two breeds,and lacks a comprehensive representa-tion of the genetic diversity across multiple Tibetan sheep breeds.This study aims to fill this gap by investigating the genetic structure,diversity and high-altitude adaptation of 6 Tibetan sheep breeds using whole-genome rese-quencing data.Results Six Tibetan sheep breeds were investigated in this study,and whole-genome resequencing data were used to investigate their genetic structure and population diversity.The results showed that the 6 Tibetan sheep breeds exhibited distinct separation in the phylogenetic tree;however,the levels of differentiation among the breeds were minimal,with extensive gene flow observed.Population structure analysis broadly categorized the 6 breeds into 3 distinct ecological types:plateau-type,valley-type and Euler-type.Analysis of unique single-nucleotide polymor-phisms(SNPs)and selective sweeps between Argali and Tibetan sheep revealed that Tibetan sheep domestication was associated primarily with sensory and signal transduction,nutrient absorption and metabolism,and growth and reproductive characteristics.Finally,comprehensive analysis of selective sweep and transcriptome data sug-gested that Tibetan sheep breeds inhabiting different altitudes on the Qinghai–Tibet Plateau adapt by enhancing cardiopulmonary function,regulating body fluid balance through renal reabsorption,and modifying nutrient diges-tion and absorption pathways.Conclusion In this study,we investigated the genetic diversity and population structure of 6 Tibetan sheep breeds in Qinghai Province,China.Additionally,we analyzed the domestication traits and investigated the unique adapta-tion mechanisms residing varying altitudes in the plateau region of Tibetan sheep.This study provides valuable insights into the evolutionary processes of Tibetan sheep in extreme environments.These findings will also contribute to the preservation of genetic diversity and offer a foundation for Tibetan sheep diversity preservation and plateau animal environmental adaptation mechanisms. 展开更多

In this paper,a standardized analysis method is established for identifying meat quality-related genes in Ordos finewool sheep using transcriptome sequencing data.A meticulously standardized approach is utilized to investigate the genetic determinants of meat quality in Ordos fine-wool sheep through transcriptome sequencing analysis.Muscle samples from the longissimus dorsi of one-year-old sheep are collected under controlled conditions,and key texture properties—hardness,elasticity,and chewiness—are measured to categorize samples into high-and low-textural-value groups.Genes significantly associated with meat quality traits are identified through standardized RNA extraction,high-throughput sequencing,and differential gene expression analysis.Functional enrichment analysis reveals their involvement in biological processes such as extracellular matrix organization and metabolic pathways.The findings underscore the pivotal role of standardization in meat quality research,laying a solid scientific foundation for future research on meat quality improvement and molecular breeding. 展开更多

Hu sheep is an indigenous breed from the Taihu Lake Plain in China,known for its high fertility.Although Hu sheep belong to the Mongolian group,their demographic history and genetic architecture remain inconclusive.Here,we analyze 697 sheep genomes from representatives of Mongolian sheep breeds.Our study suggests that the ancestral Hu sheep first separated from the Mongolian group approximately 3000 years ago.As Hu sheep migrated from the north and flourished in the Taihu Lake Plain around 1000 years ago,they developed a unique genetic foundation and phenotypic characteristics,which are evident in the genomic footprints of selective sweeps and structural variation landscape.Genes associated with reproductive traits(BMPR1B and TDRD10)and horn phenotype(RXFP2)exhibit notable selective sweeps in the genome of Hu sheep.A genome-wide association analysis reveals that structural variations at LOC101110773,MAST2,and ZNF385B may significantly impact polledness,teat number,and early growth in Hu sheep,respectively.Our study offers insights into the evolutionary history of Hu sheep and may serve as a valuable genetic resource to enhance the understanding of complex traits in Hu sheep. 展开更多

Objective:Sheep are commonly used as large animal pre-clinical models for inves-tigating cardiovascular therapies,interventions,anatomy and physiology.Further,novel small diameter vascular grafts are frequently tested via implantation into sheep carotid arteries(CAs).This is because,unlike humans,acute occlusion of one or both sheep CAs is not associated with morbidity or mortality and thus provides safer experimental testing,with reduced ethical constraints,animal numbers and costs.However,to date there has been no evidence regarding sheep tolerance of femoral artery(FA)occlusion.Methods:In this study,seven sheep underwent CA graft surgery,with digital subtrac-tion angiography(DSA)of the CAs performed every 2 months via femoral access,for a total of 8 months.Four months into the study,the left FA of two sheep became inac-cessible due to a suspected FA occlusion.Thus,femoral angiography was performed,followed by FA dissection,FA histology and retrospective analysis of both veterinar-ian animal monitoring and pain scores.Results:FA angiography and histology confirmed complete left FA occlusion in two sheep.Retrospective animal monitoring demonstrated sheep with occluded FAs did not display increased pain scores or deleterious effects on their gait or wellbeing.Conclusion:Our data shows that sheep tolerate FA occlusion with no symptoms,similar to their cerebral circulation,making them an appropriate model for assessing small diameter femoral graft interposition studies and testing other cardiovascular interventions. 展开更多

The results indicated that,for the same crossbreeding mode,the growth rate of progeny was in order F3】 F2】 F1; for the F3 progeny,the combinations Dorset- Borderdale- Small tail Han sheep and Dorset- Borderdale- Mongolia sheep gave a higher growth rate,with a body weight of 1. 57%,3. 17%,8. 23%,1. 15% higher in male and female individuals than the counterparts of Dorset and Tan sheep and Small tail Han sheep; for the F2 progeny,the combinations Dorset- Borderdale- Small tail Han sheep and Dorset- Borderdale- Mongolia sheep also gave a higher growth rate,with a body weight of 2. 15%,4. 53%,9. 21% and 2. 75% higher in male and female individuals than the counterparts of Dorset and Tan sheep and Small tail Han sheep; for the F1 progeny,the combination Borderdale and Small tail Han sheep assumed a higher growth rate,with a body weight of 3. 23%,6. 07%,7. 42% and 8. 66% higher in male and female individuals than the counterparts of Borderdale- Mongolia sheep and Tan sheep- Small tail Han sheep,respectively. Therefore,in the Small-tail Han sheep and Mongolia sheep producing regions,the F2 or F3progeny bred by using Dorset or Borderdale sheep as male parent to cross with local breeds,or the hybrid lambs of Small-tail Han sheep and Borderdale sheep as highly qualified commodity,would produce significant economic benefit. To study the physiological and biochemical indexes of White-tibetan sheep in plateau environment, we measured the physiological and biochemical inde- xes of White-tibetan sheep at 3, 6, 9, 12, 15, 18,21 and 24 months of age. The results showed that the body temperature, respiration and heart rate of White-ti- betan sheep at 3 - 24 months of age were (38.89 ±0. 56) - (39.83 ± 0.32) ℃, (26.00±6.67 ) - (58.64 ± 12.67 ) times/rain and ( 83.90 ± 7.64) - (111.36 ±7.46) times/rain, respectively. The blood physiological indexes of WBC, HCT and RDW-CV at 3, 6, 15 and 18 months of age were lower than those at 9, 12, 21 and 24 months of age. On the contrary, the indexes of RBC, MCV, MCH and MCHC at 3, 6, 15 and 18 months of age were higher than those at 9, 12, 21 and24 months of age, while HGB and PLT did not vary with months of age. For blood biochemical indexes, ALT, AST, TP, ALB, LDH and CHOL reached the maximums at 21 months of age, while ALP, PCHE and GLU reached the maximums at 3 months of age. Besides, the GLO and GA reached the maxi- mums at 18 and 24 months of age, respectively. The physiological and biochemical indexes of White-tibetan sheep followed certain rules in growth and development process, and might be affected by plateau climate and season. 展开更多

[Objective] The study aimed to provide references for the time of oocyte maturation in vitro and enucleation in the course of sheep nuclear transfer(NT).[Method] Compared the effects of different maturation time of oocytes on enucleation efficiency and reconstructed embryo development by means of blind enucleation and fluorescence microscopy.[Result] Treatment of IVM(in vitro maturation)19-21 h was significantly higher than IVM 16-18 h treatment in oocyte maturation rate(P<0.05)and was significantly higher than IVM 22-24 h treatment in enucleation rate(P<0.05).Three treatments had no significant difference in cleavage rate and blastocyst rate(P>0.05),but IVM 19-21 h treatment was significantly higher than the other 2 treatments in average cell number of blastocysts(P<0.05).[Conclusion] The appropriate in vitro maturation time of oocytes was 19-21 h for sheep nuclear transfer,which could significantly improve the quality of blastocysts according to the cell number per blastocyst(P<0.05). 展开更多

[ Result] Four microsatellite loci detected were high in heterozygosity, more in effective alleles number and rich in polymorphic information, all the three indices passed through the high polymorphic level (PIC 〉0.5). [Method] Fifteen sheep about one-year old as experimental material were divided into three groups (group A, group B end group C), into whose basal feed 0, 4 and 10 g h/sine were respectively added. After 28 d of feeding, the experimental sheep were all slaughtered for sampling; then total RNAs were extrac- ted from the samples and used to clone GHR and GAPDH genes via retrotrenscription for analyzing the expression abundance of GHR mRNA in Iongissimus dorsi muscle from different treatments. [Result] The expression of GHR mRNA in treatment B was significantly higher than that in treatment A( P 〈0.01 ), and significantly higher then that in treatment C ( P 〈0.05) ; while that in treatment C and treatment A was insignificantly different( P 〉 0.05). [ Conclusion] Addition of lysine into basal feed of sheep could dose-independently improve the expression of GHR gene in Iongissimus dorsi muscle. 展开更多

In the present study with Tan sheep, small-tailed Han sheep, Hu sheep, Tong sheep, and Wadi sheep, we detected the distribution of gene frequency of several microsatellite sites in different chromosomes, the result showed that: 1) Hu sheep was in the status of Hardy-Weinberg extreme disequilibrium (P 〈 0.01), while populations including Tong sheep, small-tailed Han sheep, Tan sheep, and Wadi sheep were in Hardy-Weinberg equilibrium (P 〉 0.05). 2) Variance analysis of the heterozygosity and poly- morphic information content at rnicrosatellite makers showed that there were not significant differences among populations as to heterozygosity and PIC (P 〉 0.05), as to effective number of alleles there were not significant differences both among Tan sheep, Hu sheep, Tong sheep, and Wadi sheep, and between Wadi sheep and small-tailed sheep (P 〉 0.05), but between the former three populations and the latter two populations, there were significant differences (0.01〈 P 〈0.05). The variation levels of small-tailed Han sheep was the highest in the five populations based on microsatellite maker data, subsequently followed by Wadi sheep, Tong sheep, Tan sheep, and then Hu sheep. 3) The phylogenetic relationships of the five sheep populations in this study did not meet the mechanism of isolation by distance, and the genetic differentiation relationships among five sheep populations were not closely linearly correlative with their geography distribution. Our findings supported related records in literature: The five populations originated on different time stage from the primogenitor population and communicated genetically with each other thereafter in the process of natural and artificial selection and on different ecological environment. 展开更多

Carnoy′s fluid and neutral buffered formalin(NBF)have been proved to be good fixatives for preservation of mast cells in pig,cattle and sheep except NBF blocked staining of most porcine mast cells,especially those located in intestinal mucosa(MMC)and in thymus medulla(TMMC). Both toluidine blue and Alcian blue were the excellent stains generally,but Alcian blue stained more porcine mast cells than did toluidine blue( P <0 01). Staining with toluidine blue of a wide pH range(from 0 1 to 7 0)showed that porcine mast cells were not very pH dependent,but the dye at pH 0 5 seemed to have the strongest affinity for all mast cells in pigs and it was also suitable for bovine and ovine mast cell staining. In the three species,unlike in rodents,the Alcian blue method did not distinguish between mast cells in the intestinal mucosa(MMC)and those in the connective tissue of the intestinal submucosa,tongue and skin(CTMC). Porcine CTMC,but not MMC,fluoresced strongly when stained with berberine sulphate or with a mixture of berberine sulphate and acridine orange. It suggested that porcine CTMC contained heparin proteoglycan. 展开更多
