It was found to be dysregulated in diabetic rats (Psammomys obesus, Israeli sand rat) and its homologue for humans is SelS/AD-015. The present study analyzed mRNA expression of SelS in omental adipose tissue biopsies from patients with type 2 diabetes mellitus (T2DM), and age- and weight-matched nondiabetic patients, the relationship of SelS mRNA with Homa-IR and serum SAA level. Methods Human omental adipose tissues from ten cases of type 2 diabetic patients and twelve cases of nondiabetic individuals were analyzed for the expression level of SelS mRNA by semiquantitative polymerase chain reaction (PCR), Homa-IR estimated by standard formula and SAA level by enzyme-linked immunosorbent assay (ELISA). Results SelS mRNA expression, Homa-IR and serum SAA were higher in T2DM sufferers than in nondiabetic control group. SelS mRNA level was positively correlated with Homa-IR and SAA level in each group. Conclusions SelS protein may be involved in insulin resistance in Chinese with T2DM by acting as the SAA receptor, thus playing an important role in the development of T2DM and atherosclerosis.展开更多

Social affective ability learning is a comprehensive ability gradually formed by individuals through formal and informal learning, which is related to emotion management, communication and cooperation, effective cognition and achievement of goals, etc. It is manifested in the continuous development of ideas, emotions and behavior patterns. Studies have shown that social-emotional ability not only has an important impact on individual’s academic and career development and physical and mental health, but also an important factor to reduce anti-social behavior, promote social harmony and enhance happiness. The stage of junior high school students is the key stage of the development of social-emotional ability. Based on the theoretical framework of social-emotional learning (SEL), this paper explores the cultivation of junior high school students’ social-emotional ability in school education, and takes psychological classroom as an example, in order to provide a theoretical basis for quality education and the cultivation of students’ non-cognitive ability.展开更多
BACKGROUND Proteins play a central role in regulating biological functions,and various pathways regulate their synthesis and secretion.Endoplasmic reticulum-associated protein degradation(ERAD)is crucial for monitorin...BACKGROUND Proteins play a central role in regulating biological functions,and various pathways regulate their synthesis and secretion.Endoplasmic reticulum-associated protein degradation(ERAD)is crucial for monitoring protein synthesis and processing unfolded or misfolded proteins in actively growing tumor cells.However,the role of the multiple ERAD complexes in liver cancer remains unclear.AIM To elucidate the effects of SEL1L-mediated ERAD on Huh7 and explore the underlying mechanisms in vivo and in vitro.METHODS Huh7 cells were treated with ERAD inhibitor to identify ERAD’s role.Cell counting kit-8,5-ethynyl-2’-deoxyuridine and colony formation experiments were performed.Apoptosis level and migration ability were assessed using fluorescence activated cell sorting and Transwell assay,respectively.Huh7 SEL1L knockout cell line was established via clustered regularly interspaced short palindromic repeats,proliferation,apoptosis,and migration were assessed through previous experiments.The role of SEL1L in vivo and the downstream target of SEL1L were identified using Xenograft and mass spectrometry,respectively.RESULTS The ERAD inhibitor suppressed cell proliferation and migration and promoted apoptosis.SEL1L-HRD1 significantly influenced Huh7 cell growth.SEL1L knockout suppressed tumor cell proliferation and migration and enhanced apoptosis.Mass spectrometry revealed EXT2 is a primary substrate of ERAD.SEL1L knockout significantly increased the protein expression of EXT2.Furthermore,EXT2 knockdown partially restored the effect of SEL1L knockout.CONCLUSION ERAD inhibition suppressed the proliferation and migration of Huh7 and promoted its apoptosis.EXT2 plays an important role and ERAD might be a potential treatment for Huh7 hepatocellular carcinoma.展开更多
