[Objectives] To optimize the extraction process of total flavonoids from Sanguisorbae Radix and carbonized Sanguisorba root,compare quality of different batches of Sanguisorbae Radix,study the effects of processing on the content of flavonoids,and provide scientific basis for reasonable utilization of Sanguisorbae Radix. [Methods] Test samples were prepared by heating,refluxing,and extraction,the extraction process was optimized by orthogonal experiment design,color was developed by NaNO_2-Al( NO_3)3-NaOH,and total flavonoids were measured by UV method at the wavelength of 510 nm. [Results] The linear relationship of rutin was excellent in the concentration range of 0. 1248 mg/mL-0. 5712 mg/mL,R^2= 0. 9997; the average recovery was 99. 67% and the RSD was 0. 70%. The optimum extraction conditions were as follows: the volume fraction of ethanol was 50%,the extraction temperature was 90℃,the extraction time was 90 min,and the solid-to-liquid ratio was 1∶ 20( g/mL). [Conclusions] After optimization of the extraction process,the extraction rate of total flavonoids in samples of Sanguisorbae Radix was significantly increased; there was certain difference in the content of total flavonoids between different batches of Sanguisorbae Radix and processed products; the total flavonoids significantly declines in carbonized sanguisorba root,and the influence of processing on its curative effect was to be further studied. 展开更多

Methods The compounds were isolated and purified by column chromatography and their structures were identified on the basis of physicochemical properties and spectral analysis. Results Five compounds were isolated and identified as 3β-hydroxy-28-norurs-17,19,21-trien (1), 3β-hydroxy-28-norurs-12,17-dien (2), 3β,19α-dihydroxyurs-13(18)- en-28-oic acid (3), 3β-[(α-L-arabin-opyranosyl) oxy]-28-norurs-12,17-dien (4), and pomolic acid (5). Conclusion Compounds 1, 3, and 4 are novel compounds belong to triterpenoids and triterpenoid saponins, named as sanguisorbigenins Z, Y1 , and Y2 , respectively. 展开更多

Objective To explore the protective effects of tannins in Sanguisorba Radix (TSR) on myelosuppression mice induced by cyclophosphamide (CTX). Methods TSR was ig given at the dose of 20 mg/kg for 10 d after ip administration of CTX (200 mg/kg). Results TSR could significantly increase the numbers of white blood ceils, red blood cells, and platelets of myelosuppression in mice. And it could accelerate bone marrow haemopoietic stem/progenitor cells (HSPCs) in myelosuppression mice and enhance cell proliferation by promoting cell cycles from G0/G1 phase to access into S and G2/M phases, then the reduced number of HSPCs induced by CTX was reversed. Moreover, TSR could increase the mRNA and protein expression levels of O(6)-methylguanine- DNA methyltransferase (MGMT) in HSPCs of myelosuppression mice. Concision TSR has a protective function against CTX-induced myelosuppression. The mechanism might be related to protecting hematopoietic stem cells of bone marrow, stimulating hematopoiesis recovery, as well as preventing the apoptosis of hematopoietic stem cells induced by CTX. 展开更多

Objective:To investigate whether Radix Sanguisorbae(RS,Diyu)could restore intestinal barrier function following sepsis using a cecal ligation and puncture(CLP)-induced septic rat model and lipopolysaccharide(LPS)-challenged IEC-6 cell model,respectively.Methods:Totally 224 rats were divided into4 groups including a control,sham,CLP and RS group according to a random number table.The rats in the control group were administrated with Ringer's lactate solution(30 mL/kg)with additional dopamine[10μg/(kg·min)]and given intramuscular injections of cefuroxime sodium(10 mg/kg)12 h following CLP.The rats in the RS group were administrated with RS(10 mg/kg)through tail vein 1 h before CLP and treated with RS(10 mg/kg)12 h following CLP.The rats in the sham group were only performed abdominal surgery without CLP.The rats in the CLP group were performed with CLP without any treatment.The other steps were same as control group.The effects of RS on intestinal barrier function,mesenteric microvessels barrier function,multi-organ function indicators,inflammatory response and 72 h survival window following sepsis were observed.In vitro,the effects of RS on LPS-challenged IEC-6 cell viability,the expressions of zona occludens-1(ZO-1)and ferroptosis index were evaluated by cell counting kit-8,immunofluorescence and Western blot analysis.Bioinformatic tools were applied to investigate the pharmacological network of RS in sepsis to predict the active compounds and potential protein targets and pathways.Results:The sepsis caused severe intestinal barrier dysfunction,multi-organ injury,lipid peroxidation accumulation,and ferroptosis in vivo.RS treatment significantly prolonged the survival time to 56 h and increased 72-h survival rate to 7/16(43.75%).RS also improved intestinal barrier function and relieved intestinal inflammation.Moreover,RS significantly decreased lipid peroxidation and inhibited ferroptosis(P<0.05 or P<0.01).Administration of RS significantly worked better than Ringer's solution used alone.Using network pharmacology prediction,we found that ferroptosis and hypoxia inducible factor-1α(HIF-1α)signaling pathways might be involved in RS effects on sepsis.Subsequent Western blot,ferrous iron measurements,and FerroOrange fluorescence of ferrous iron verified the network pharmacology predictions.Conclusion:RS improved the intestinal barrier function and alleviated intestinal injury by inhibiting ferroptosis,which was related in part to HIF-1α/heme oxygenase-1/Fe^(2+)axis. 展开更多

Objective:To investigate the anti-inflammatory effects of Sanguisorbae Radix on contact dermatitis(CD).Methods:Mice were sensitized by painting 30μL of 1-fluoro-2,4-dinitrofluorobenzene(DNFB)onto each ear for 3 days.Four days later,mice were challenged by painting with 50μL of DNFB onto the shaved dorsum every 2 days.Sanguisorbae Radix methanol extract(MESR)was applied onto the shaved dorsum every 2 days.The effects of MESR on skin thickness,skin weights,histopathological changes,skin lesions and cytokine production in DNFB-induced CD mice were investigated,as well as its effects on body weights and spleen/body weight ratio.Results:Topical application of MESR effectively inhibited enlargement of skin thickness and weight(P<0.05).MESR treatment also inhibited hyperplasia,spongiosis and immune cell infiltration induced by DNFB in inflamed tissues and improved lesions on dorsum skin in CD mice.Moreover,treatment with MESR suppressed the increase in the levels of tumor necrosis factor α(TNF-α,P<0.01)and interferonγ(IFN-γ,P<0.05),respectively.Finally,MESR had no effect on body weight gain or spleen/body weight ratio.Conclusion:These data suggest that MESR acts as an anti-inflammatory agent that decreases the production of TNF-α and IFN-γ,resulting in reductions of skin lesions and histopathological changes in inflamed skin tissues. 展开更多

响应面法对地榆总黄酮超声提取工艺的优化

以地榆(Sanguisorba officinalis L.)根为材料,对其所含黄酮类物质进行了提取工艺研究。通过单因素试验考察了乙醇体积分数、原料粒度、液料比、超声功率、超声时间、超声次数对地榆总黄酮提取率的影响,在单因素基础上,利用响应面法设... 以地榆(Sanguisorba officinalis L.)根为材料,对其所含黄酮类物质进行了提取工艺研究。通过单因素试验考察了乙醇体积分数、原料粒度、液料比、超声功率、超声时间、超声次数对地榆总黄酮提取率的影响,在单因素基础上,利用响应面法设计,优化了各项工艺参数。结果表明,超声提取地榆根总黄酮的最佳工艺条件为乙醇体积分数47.78%、原料粒度40-60目、液料比20∶1(mL∶g)、超声功率374.25 W、超声时间33.70 min。此工艺条件下,地榆根中总黄酮提取率为10.565%。按照响应面优化的最佳提取工艺条件,对40-60目地榆粉末超声提取3次,测量总黄酮提取率为11.035%。研究结果表明,超声辅助提取的黄酮得率要高于一些传统方法,适合对地榆总黄酮进行提取加工。 展开更多

Their slructures were characterized by spectroscopic analysis and chemical method, including 1D NMR, 2D NMR, and HR-ESI-MS. BACKGROUND Many natural products confer health benefits against diverse diseases through their antioxidant activities.Carbon tetrachloride(CCl4)is often used in animal experiments to study the effects of substances on liver injury and the related mechanisms of action,among which oxidative stress is a major pathogenic factor.AIM To compare antioxidant and hepatoprotective activities of ten herbs and identify and quantify phytochemicals for the one with strongest hepatoprotection.METHODS The antioxidant activity of ten medicinal herbs was determined by both ferricreducing antioxidant power and Trolox equivalent antioxidant capacity assays.The total phenolic and flavonoid contents were determined by Folin–Ciocalteu method and aluminum chloride colorimetry,respectively.Their effects on CCl4-induced oxidative liver injury were evaluated and compared in a mouse model by administrating each water extract(0.15 g/mL,10 mL/kg)once per day for seven consecutive days and a dose of CCl4 solution in olive oil(8%,v/v,10 mL/kg).The herb with the strongest hepatoprotective performance was analyzed for the detailed bioactive components by using high-performance liquid chromatography-electrospray ionization source-ion trap tandem mass spectrometry.RESULTS The results revealed that all tested herbs attenuated CCl4-induced oxidative liver injury;each resulted in significant decreases in levels of serum alanine transaminase,aspartate transaminase,alkaline phosphatase,and triacylglycerols.In addition,most herbs restored hepatic superoxide dismutase and catalase activities,glutathione levels,and reduced malondialdehyde levels.Sanguisorba officinalis(S.officinalis)L.,Coptis chinensis Franch.,and Pueraria lobata(Willd.)Ohwi root were the three most effective herbs,and S.officinalis L.exhibited the strongest hepatoprotective effect.Nine active components were identified in S.officinalis L.Gallic acid and(+)-catechin were quantified(7.86±0.45 mg/g and 8.19±0.57 mg/g dried weight,respectively).Furthermore,the tested herbs displayed a range of in vitro antioxidant activities proportional to their phenolic content;the strongest activities were also found for S.officinalis L.CONCLUSION This study is of value to assist the selection of more effective natural products for direct consumption and the development of nutraceuticals or therapeutics to manage oxidative stress-related diseases. 展开更多

OBJECTIVE To investigate the pharmacological effect and mechanism of Sanguisorba officinalis L.(SOL)in non-small cell lung cancer(NSCLC)in vitro and in vivo based on network pharmacology.METHODS Network pharmacology was used to analyze the improving effect of SOL on NSCLC and possible targets.Cell counting kit 8(CCK-8)and 5-ethynyl-2′-deoxyuridine(EdU)staining,Western blotting,flow cytometry of AnnexinⅤ/PI,Hoechst 33342/PI staining detection and immunofluorescence were utilized in vitro.H&E staining,immunohistochemistry staining and Western blotting were performed in vivo.RESULTS Based on network prediction,we analyzed the 208 common targets of SOL and NSCLC.36 core targets in 208 common targets were obtained through cytoscape analysis.And the top 10 core targets included Akt,mTOR,EGFR,etc..KEGG analysis showed that PI3K-Akt signaling pathway was the most likely pathway.Furthermore,the mechanism study found that SOL could activate the PI3K/Akt/mTOR signaling pathway in vitro and in vivo.The anti-proliferative effect of SOL in A549 and H1299 cells was measured and validated by CCK-8 and EdU assay.Immunohistochemical results of Ki67 showed that SOL effectively inhibited tumor growth in vivo.SOL also significantly inhibited the migration and invasion of A549 and H1299 cells.SOL significantly increased the percentage of cells with PI signal in A549 and H1299,and the process of cell death of A549 cells indicated that SOL induced apoptosis.The PARP-1 and caspase-3 in A549 and H1299 were found to be activated in a dose manner.The results in vivo were consistent with those in vitro.CONCLUSION SOL-induced,caspase-3-mediated apoptosis via the induction of the PI3K/Akt/mTOR signaling pathway in NSCLC,which further clarified the mechanism of SOL in the inhibition of NSCLC,and thereby provided a possibility for SOL to serve as a novel therapeutic agent for NSCLC in the future. 展开更多

It is known that high-fructose intake leads to obesity, diabetes, dyslipidemia in rodents. The aim of our study was to evaluate the effect of Blood bumet's extract on body weight, visceral fat, blood glucose, plasma insulin, lipid profiles, lipid peroxidation and enzymatic antioxidants (superoxide dismutase, catalase, glutathione reductase) in fructose-induced type-2 diabetic rats. Our results showed that in high-fructose-induced diabetic rats were observed abdominal obesity, hyperglycemia, hyperinsulinemia, increased lipid profiles and atherogenic index, decreased insulin sensitivity, activation of lipid peroxidation and reduced activities of enzymatic antioxidants. Blood bumet's extract was able to improve insulin resistance, glucose toleration, rectifies dyslipidemia and abolish the damage in antioxidant system in terms of high-fructose feeding in rats. 展开更多

[Objectives] To establish a method for the determination of tannin in Sanguisorba officinalis L.,and provide guarantee for further control over quality of semi-finished tannin products from Sanguisorba officinalis L. [Methods] With gallic acid as reference substance,the UV method was used to determine the content of tannin in Sanguisorba officinalis L.,and the methodological examination was done. [Results]With gallic acid as standard,when the concentration of tannin in Sanguisorba officinalis L. was 1. 02-10. 14 μg·m L^(-1),there was a good linear relationship with absorbance,the precision,reproducibility and recovery were in line with the requirements,and the absorbance value was stable within 40 min after coloration.
