Objectives:To observe the clinical effect on opioid-induced constipation of liver stagnation and spleen deficiency treated with the ultrasonic penetration of the No.1 constipation herbal formula at Tiānshū(天枢ST25)...Objectives:To observe the clinical effect on opioid-induced constipation of liver stagnation and spleen deficiency treated with the ultrasonic penetration of the No.1 constipation herbal formula at Tiānshū(天枢ST25).Methods:A total of 126 patients that were in compliance with the inclusion criteria were randomized into an acupoint group(41 cases),a Chinese medication group(42 cases)and a western medication group(43 cases).In the acupoint group,the ultrasonic penetration of the No.1 constipation herbal formula at bilateral Tiānshū(天枢ST25)were given.In the Chinese herbal medication group,the No.1 constipation herbal formula was prescribed for oral administration.In the western medication group,polyethylene glycol electrolyte powder was prescribed for oral administration.The treatment was given once a day,for 14 days totally.Before and after treatment,the constipation symptom score,the score of the patient assessment of constipation quality of life(PAC-QOL),the situation of nitric oxide synthetase(NOS)in serum were observed in each group.The safety indicators in the observation included diarrhea,allergic reaction,liver dysfunction,skin lesions,etc.Results:A total of 122 cases accomplished the trial.The total effective rate was 82.5%in the acupoint group,80.48%in the Chinese medication group and 78.04%in the western medication group.The constipation symptoms were all relieved in the three groups,indicating the statistical significance as compared with those before treatment(P<0.01,P<0.05).In the acupoint group and the western medication group,the improvements in the three symptom items,i.e.distention or scurrying pain in the chest,hypochondriac region or the bilateral sides of the abdomen,depression or irritability and poor appetite,were better than the western medication group(P<0.01,P<0.05).The improvements in all of the symptoms in the acupoint group were better than the Chinese medication group(all P<0.05).The PACQOL score was improved in each group after treatment and it was different significantly in statistics as compared with that before treatment in each group.After treatment,the improvement of PAC-QOL score in the acupoint group and the Chinese medication group was better than that in the western medication group(both P<0.05),and the improvement in the acupoint group was better than the Chinese medication group(P<0.05).As to serum NOS,after treatment,it was different significantly as compared with that before treatment in each group(all P<0.05),the improvement in the acupoint group was better than that of either the Chinese medication group or the western medication group,indicating the statistical significance(both P<0.05).The incidence of adverse reaction in the acupoint group was less and different significantly as compared with the western medication group(P<0.01),and it was different statistically as compared with the Chinese medication group(P<0.05).Conclusion:The ultrasonic penetration of the No.l constipation herbal formula at Tiānshū(天枢ST25)is effective on opioid-induced constipation of liver stagnation and spleen deficiency.This therapy achieves the significant therapeutic effect and presents the good compliance in treatment.Hence,it deserves to be further promoted in clinical application.展开更多

METHODS: A total of 53 male Spragu...AIM: To investigate the effects of moxibustion on the morphology and function of mast cells (MC) at Tianshu (ST25) in rats with trinitro-benzene-sulfonic acid (TNBS)-induced colitis. METHODS: A total of 53 male Sprague-Dawley rats were randomly divided into a normal group and experimental group. In the experimental group, a rat model of TNBS-induced colitis was established, and the rats were then randomly divided into a model group, moxi-bustion group, moxibustion plus disodium cromoglycate (M + DC) group and moxibustion plus normal saline (M+ NS) group. Rats in the moxibustion group received suspended moxibustion at bilateral ST25 for 10 min, once a day for 7 d. Rats in the M + DC and M + NS groups were pretreated with disodium cromoglycate and normal saline at bilateral ST25, respectively, and were then concurrently subjected to the same treatment as rats in the moxibustion group. The hematoxy- lin-eosin staining method was used to observe histology of the colon and the toluidine blue-improved method was used to observe mast cells at ST25 acupoint areas. RESULTS: An improvement in colonic injury in the moxibustion group was observed and the degranulation ratio of MC at ST25 acupoint was markedly higher in the moxibustion group than in the model group (45.91 ± 11.41 vs 32.58 ± 8.28, P < 0.05). After inhibition of degranulation of MC at ST25 by disodium cromoglycate, no improvement in colon tissue injury was observed. CONCLUSION: Moxibustion exerted its effect on healing impaired colonic mucosa in rats with TNBS-induced colitis by increasing the degranulation ratio of local MC, but had little effect on the morphology of MC at ST25 acupoint.展开更多
Objective:To explore the therapeutic mechanism of moxibustion in Crohn disease(CD)-associated intestinal fibrosis by observing its effects on the angiotensin-converting enzyme(ACE)/angiotensinⅡ(AngⅡ)/angiotensinⅡty...Objective:To explore the therapeutic mechanism of moxibustion in Crohn disease(CD)-associated intestinal fibrosis by observing its effects on the angiotensin-converting enzyme(ACE)/angiotensinⅡ(AngⅡ)/angiotensinⅡtype 1 receptor(AT1R)axis in CD mouse models.Methods:Six randomly selected male C57BL/6 mice were assigned to a normal group,while the remaining mice were administered 0.1 mL of 2,4,6-trinitrobenzene sulfonic acid via enema to establish a CD intestinal fibrosis model.After successful modeling,the mice were randomly divided into a model group,a moxibustion group,and a Western medication group,with 6 rats in each group.The normal group and the model group only received grabbing without intervention.In the moxibustion group,mild moxibustion was applied to Qihai(CV6)and bilateral Tianshu(ST25)once a day for 10 min each time over 7 consecutive days.The Western medication group was administered mesalazine suspension via oral gavage once a day for 7 consecutive days.At the end of the intervention,the general condition,disease activity index(DAI)score,and gross colon score of mice in each group were evaluated.Hematoxylin-eosin staining was used to observe and score the histological changes in the colon tissue in each group.Masson staining was used to observe colonic fibrosis and the ratio of collagen-positive areas was analyzed;the expression of AngⅡin the colon tissue was detected by the enzyme-linked immunosorbent assay;immunohistochemistry and real-time quantitative reverse transcription polymerase chain reaction were used to detect the protein and mRNA expression of ACE and AT1R in the colon tissue,respectively;Western blotting was used to detect the expression of transforming growth factor(TGF)-β1 and connective tissue growth factor(CTGF)in the colon tissue.Results:Compared to the normal group,the DAI score,gross colon score,colonic histological score,collagen-positive area ratio,ACE protein and mRNA,AngⅡprotein,AT1R protein and mRNA,TGF-β1 protein,and CTGF protein in the colon tissue in the model group increased significantly(P<0.01).In contrast,the above indicators in both the moxibustion group and the Western medication group reduced significantly compared to the model group(P<0.01 or P<0.05).There was no statistical difference in these indicators between the moxibustion group and the Western medication group(P>0.05).Conclusion:Moxibustion can alleviate intestinal fibrosis in CD mice,and its therapeutic mechanism may be associated with the regulation of colonic ACE/AngⅡ/AT1R axis.展开更多
Objective:To observe the effect of acupuncture on local skin tissue metabolites of Tianshu(ST25)in rats with Crohn's disease(CD)using metabolomics and to explore the mechanism of acupuncture treatment metabolites ...Objective:To observe the effect of acupuncture on local skin tissue metabolites of Tianshu(ST25)in rats with Crohn's disease(CD)using metabolomics and to explore the mechanism of acupuncture treatment metabolites and pathway on local skin tissue of Tianshu(ST25).Materials and Methods:Thirty-eight Sprague-Dawley rats were randomly assigned to a normal group(n=14)and a CD modeling group(n=24).Rats in the CD modeling group were administered with 2,4,6-trinitrobenzene sulfonic acid and ethanol enema,in addition to the normal group.Four normal rats and four CD modeling rats were selected at random after model identification was established.Furthermore,CD modeling group rats were randomly assigned to two groups of 10 rats:the model group and acupuncture group.Rats in the acupuncture group underwent acupuncture of Tianshu(ST25)and Shangjuxu(ST37)for 7 days.After the intervention,the colon tissue was collected from each group of rats and the pathological changes were observed through hematoxylin and eosin(HE)staining.The local skin tissues at Tianshu(ST25)of the rats in each group were taken,and the proton nuclear magnetic resonance technique was used to detect the metabonomics of the local skin tissues of Tianshu(ST25)in rats.Results:Compared with the normal group,the following were observed in the model group after HE staining:irregular colon morphology and other pathological changes such as intestinal mucosal hyperemia,edema,ulcers,polyps,and inflammatory cell infiltration.In the acupuncture group,colon tissue structure was relatively complete and layered,the colon gland structure was restored,and inflammatory cell infiltration was significantly improved.The choline,glycerin,glycine,guanidoacetic acid,and proline levels were significantly higher in the model group than in the normal group(P<0.05 or P<0.01).Compared with the model group,the acupuncture group had significantly increased contents of alanine,leucine,L-phenylalanine,and tyrosine in the skin(P<0.05 or P<0.01).Valine,leucine,and L-isoleucine biosynthesis and L-phenylalanine metabolism were the main metabolic pathways involved in the changes in the local skin tissues of the rats in each group,biosynthesis of tyrosine,and tryptophan.Conclusion:The change in the metabolites in the local area of Tianshu(ST25)may be related to intestinal disease.The acupuncture of Tianshu(ST25)can improve intestinal inflammatory reaction in rats with CD,and this finding may be related to the regulation of amino acid metabolites and their pathways in the local acupoint tissues of Tianshu(ST25)by acupuncture.展开更多
