红火蚁是近年来侵害我国南方的主要外来入侵物种之一,精确识别红火蚁巢穴是防控红火蚁的关键所在。为解决传统红火蚁防控依赖人工巡视、高危险、效率低的问题,降本增效实现红火蚁巢穴的智能检视,提出了一种基于ResNet34改进的红火蚁巢... 红火蚁是近年来侵害我国南方的主要外来入侵物种之一,精确识别红火蚁巢穴是防控红火蚁的关键所在。为解决传统红火蚁防控依赖人工巡视、高危险、效率低的问题,降本增效实现红火蚁巢穴的智能检视,提出了一种基于ResNet34改进的红火蚁巢穴判别模型。该模型借助采集于不同地貌特征下的红火蚁巢穴图像,结合数据增强技术进行训练,通过在ResNet34的第1层卷积层之后和全连接层之前加入SE注意力机制模块,提升网络的自适应选择和通道权值调整能力,以提取红火蚁巢穴表面局部非线性的纹理特征。经过K折交叉验证试验和超参数探究消融试验,将SE-ResNet34与AlexNet、VGG-16、ResNet18、ResNet34、ResNet50进行对比,分析得出SE-ResNet34的峰值准确率达到了98.76%,比ResNet34的准确率提高了2.17%,较其他测试模型有训练时间短、识别精度高的特点,同时展现出较强的鲁棒性和稳定性。该方法在减少人工成本的同时可降低杀虫剂的使用,为红火蚁巢穴判别提供了一种便捷高效的解决方案。 展开更多

PTH(1-34)诱导骨矿化小鼠实验动物模型的构建及鉴定 被引量:1

目的:探讨骨骼发育及矿化过程,构建骨矿化小鼠实验动物模型。方法:采用甲状旁腺激素(PTH)(1-34)经腹腔注射,诱导6周龄NOD-SCID雌性小鼠成骨细胞分化及骨组织矿化,从而构建骨矿化小鼠实验动物模型。进一步应用Micro-CT检测骨密度,应用钙... 目的:探讨骨骼发育及矿化过程,构建骨矿化小鼠实验动物模型。方法:采用甲状旁腺激素(PTH)(1-34)经腹腔注射,诱导6周龄NOD-SCID雌性小鼠成骨细胞分化及骨组织矿化,从而构建骨矿化小鼠实验动物模型。进一步应用Micro-CT检测骨密度,应用钙黄绿素和盐酸四环素荧光标记检测骨矿化沉积率,应用冯库萨染色鉴定钙盐沉积,并应用马松染色检测骨胶原容积分数,以鉴定小鼠骨组织矿化水平,确定骨矿化小鼠模型是否构建成功。结果:Micro-CT检测发现,PTH(1-34)诱导20 d可以显著增加骨小梁相对体积分数(BV/TV,t=9.473,P<0.001)和骨小梁厚度(Tb.Th,t=18.64,P<0.001),显著减少骨小梁间距(Tb.Sp,t=7.794,P<0.01)。双荧光标记显示,PTH(1-34)处理后骨矿化沉积率(MAR)显著提高(t=29.99,P<0.001)。冯库萨染色显示,PTH(1-34)处理组冯库萨阳性面积(VK.A)显著增加(t=2.914,P<0.05)。马松染色显示,PTH(1-34)处理后胶原容积分数(CVF)显著提升(t=13.94,P<0.001)。结论:PTH(1-34)可有效诱导小鼠骨骼矿化,成功构建骨矿化小鼠实验动物模型。 展开更多

目的:通过分析行经阴道McDonald子宫颈环扎术的单胎妊娠孕妇术后孕34周前分娩的影响因素,建立风险预测模型。方法:回顾性分析2019年1月1日至2024年2月29日于中国人民解放军北部战区总医院和平院区妇产科行McDonald子宫颈环扎术的500例... 目的:通过分析行经阴道McDonald子宫颈环扎术的单胎妊娠孕妇术后孕34周前分娩的影响因素,建立风险预测模型。方法:回顾性分析2019年1月1日至2024年2月29日于中国人民解放军北部战区总医院和平院区妇产科行McDonald子宫颈环扎术的500例单胎妊娠孕妇的临床资料,按7∶3随机划分为训练集350例与验证集150例。对训练集的资料利用单因素联合多因素Logistic回归筛选出影响子宫颈环扎术后孕34周前分娩的预测变量,建立预测模型和评估,并在验证集中进行内部验证。结果:多因素Logistic回归示,术前子宫颈长度(CL)<20 mm、有羊水沉积物(AFS)、阴道乳杆菌缺失、子宫颈扩张>3 cm为子宫颈环扎术后孕34周前分娩的独立危险因素(OR>1,P<0.05),据此构建的Nomogram模型的曲线下面积(AUC)为0.887,模型的一致性良好(H-L检验:P=0.905,Brier得分为0.095)。临床决策曲线示,当阈值在0.10~0.92时,该模型的临床决策价值较高。内部验证后AUC为0.812,预测结果与实际情况之间具有较高的一致性(H-L检验:P=0.859)。结论:基于影响因素构建的经阴道McDonald子宫颈环扎术后孕34周前分娩的列线图模型具有良好的临床应用价值,对筛选出高危孕妇并给予个体化干预具有指导作用。 展开更多

采用吸附法对模型油中二氯乙烷的脱除进行研究。以SAPO-34分子筛为载体,以Ni2+、Cu2+、Mg2+、Zn2+为活性组分,利用等体积浸渍法制得不同金属离子改性的SAPO-34分子筛吸附剂。采用低温氮气吸附-脱附(BET)、X射线衍射(XRD)和氨吸附及程序... 采用吸附法对模型油中二氯乙烷的脱除进行研究。以SAPO-34分子筛为载体,以Ni2+、Cu2+、Mg2+、Zn2+为活性组分,利用等体积浸渍法制得不同金属离子改性的SAPO-34分子筛吸附剂。采用低温氮气吸附-脱附(BET)、X射线衍射(XRD)和氨吸附及程序升温脱附(NH3-TPD)对吸附剂进行了表征,考察了5种吸附剂对二氯乙烷的吸附脱氯效果及金属负载量对脱氯效果的影响,得到了最优吸附剂,同时考察了脱氯实验的吸附条件对脱氯效果的影响,得到了最佳吸附条件,最后研究了吸附剂的再生能力。结果表明:Ni/SAPO-34分子筛的吸附脱氯效果较好,并且有很好的再生能力,其吸附效果的最佳条件为镍金属离子的负载量为4%,吸附温度为20℃,剂油比为1∶30,吸附时间为50min。这一研究为真实油中有机氯化物的脱除提供了经验和依据。 展开更多

目的通过建立肺癌骨转移小鼠模型,探讨甲状旁腺激素(PTH)(1-34)对小鼠肺癌骨转移瘤生长的影响。方法将30只4~6周龄BALB/c雌性小鼠,用人肺癌A549细胞悬液骨内注射的方法制作胫骨近端肺癌骨转移模型,7 d后X线确定模型建立,随机分成3组... 目的通过建立肺癌骨转移小鼠模型,探讨甲状旁腺激素(PTH)(1-34)对小鼠肺癌骨转移瘤生长的影响。方法将30只4~6周龄BALB/c雌性小鼠,用人肺癌A549细胞悬液骨内注射的方法制作胫骨近端肺癌骨转移模型,7 d后X线确定模型建立,随机分成3组,一组注射甲状旁腺素(PTH)(1-34)(40 mg/kg)作为观察组,一组注射等量溶解剂作为空白对照组,一组注射环磷酰胺作为阳性对照组,每2 d测体质量一次及观察右后肢瘤体生长情况,给药28 d后处死小鼠,所有小鼠行X线及显微CT检查观察肿瘤形态、大小,CT检测胫骨局部骨密度及肿瘤体积,HE染色及免疫组织化学法观察肿瘤形态及性质,检测血清碱性磷酸酶浓度了解成骨情况。所有数据采用SPSS 13.0统计软件进行分析,体质量以均数±标准差表示,ALP、肿瘤体积及CT数据分析采用单因素方差分析做整体检验,两两比较采LSD-t检验,P〈0.05被认为差异有统计学意义。结果观察组小鼠体质量变化曲线与对照组组间差异无显著性(P〉0.05),X线及显微CT可见观察组及对照组均出现明显的骨破坏缺损,环磷酰胺组骨皮质相对完整,轻度骨破坏,CT数据分析观察组及对照组肿瘤体积大小组间差异无显著性(P〉0.05),环磷酰胺组肿瘤体积明显小于前两组,组间有显著性(P〈0.05),而观察组骨量大于对照组,低于环磷酰胺组,3组组间差异有显著性(P〈0.05),病理学显示溶骨性病变为主的混合性骨质破坏,观察组及对照组骨结构破坏严重,肿瘤细胞填充明显,免疫组化显示腺瘤,两组差异相似,观察组ALP浓度大于对照组及环磷酰胺组,观察组与对照组及环磷酰胺组相比,组间差异有显著性(P〈0.05)。结论间歇性小剂量注射甲状旁腺激PTH(1-34)不促进小鼠肺癌骨转移瘤的生长,但增加骨转移瘤灶周围的骨量。 展开更多

目的探讨血清白介素34(IL-34)与T2DM及肥胖的相关性。方法选取研究对象99例,按照1998年WHO肥胖诊断标准分为正常体重组(NW组,n=32)和超重/肥胖组(OW组,n=67)。NW组按照1999年WHO糖尿病诊断标准分为无T2DM组(N-Con组,n=17)和T2DM组(N-T2D... 目的探讨血清白介素34(IL-34)与T2DM及肥胖的相关性。方法选取研究对象99例,按照1998年WHO肥胖诊断标准分为正常体重组(NW组,n=32)和超重/肥胖组(OW组,n=67)。NW组按照1999年WHO糖尿病诊断标准分为无T2DM组(N-Con组,n=17)和T2DM组(N-T2DM组,n=15),OW组分为无T2DM组(O-Con组,n=33)及T2DM组(O-T2DM组,n=34)。检测各组血清IL-34、高敏C反应蛋白(hsC-RP)、FIns、FPG、TC、TG、LDL-C、HDL-C、HbA1c,测量SBP、DBP、WC、臀围(HC),记录年龄、性别,计算胰岛素抵抗指数(HOMA-IR)、BMI。对OW组中8例伴有糖尿病和5例不伴糖尿病的患者行腹腔镜Roux-en-Y胃旁路术(RYGB),术后1个月再次随访以上指标。结果各组血清IL-34浓度分别为(167.1±39.38)vs(170.71±35.68)vs(244.48±42.20)vs(208.67±46.50)ng/L。O-Con组高于N-Con、N-T2DM、O-T2DM组,同时O-T2DM组高于N-Con及N-T2DM组,N-Con组与N-T2DM组比较,差异无统计学意义(P>0.05)。Pearson相关分析显示,IL-34与hsC-RP、FIns、HOMA-IR、WC、HC、BMI呈正相关(r=0.672,0.442,0.566,0.678,0.476,0.651,P<0.01)。多元逐步回归分析显示,WC、BMI是IL-34的独立影响因素(β=0.434、0.321,P<0.01)。OW组8例代谢手术后随访1个月,血清IL-34水平、hsC-RP、HOMA-IR、BMI、WC较手术前下降,但只有HOMA-IR差异有统计学意义(P<0.01)。结论血清IL-34水平在肥胖人群中升高,可能参与IR,与机体炎症反应相关,和T2DM关系尚不明确。 展开更多

G蛋白偶联受体34(GPR34)在肥大细胞、肿瘤组织和人体免疫器官中高度选择性表达,但因属于膜蛋白,其三维结构难以测定,不能从分子水平上分析其与药物作用情况。本文选用PDB数据库中与GPR34一级序列同源性最高的4NTJ为模版蛋白进行同源建模,运用拉氏图和Profile-3D进行评估,通过加膜限制和loop区能量优化等方法最终得到合理并且可信度高的3D结构模型。最优模型Verify Score为124.97(期望Verify Score值区间为69.071 1~153.491),预测分析得到14个可能的活性位点。通过最陡下降法和共轭梯度法构建GPR34激动剂分子的最低能量结构,将激动剂分子与预测的蛋白3D结构模型上的活性位点进行分子模拟对接,根据对接模型比较不同激动剂分子与不同蛋白活性位点的相互作用。实验结果对设计GPR34激动剂分子和进一步研究GPR34的结构和功能具有理论指导意义。 展开更多

AIM: To study the effect of mobilized peripheral blood autologous CD34 positive(CD34+) cell infusion in patients with non-viral decompensated cirrhosis.METHODS: Cirrhotic patients of non-viral etiology were divided into 2 groups based on their willingness to be listed for deceased donor liver transplant(DDLT)(control, n = 23) or to receive autologous CD34+ cell infusion through the hepatic artery(study group, n= 22). Patients in the study group were admitted to hospital and received granulocyte colony stimulating factor injections 520 μg/d for 3 consecutive days to mobilize CD34+ cells from the bone marrow. On day 4,leukapheresis was done and CD34+ cells were isolated using CliniMAC magnetic cell sorter. The isolated CD34+ cells were infused into the hepatic artery under radiological guidance. The patients were discharged within 48 h. The control group received standard of care treatment for liver cirrhosis and were worked up for DDLT as per protocol of the institute. Both groups were followed up every week for 4 wk and then every month for 3 mo.RESULTS: In the control and the study group, the cause of cirrhosis was cryptogenic in 18(78.2%) and16(72.72%) and alcohol related in 5(21.7%) and6(27.27%), respectively. The mean day 3 cell count(cells/μL) was 27.00 ± 20.43 with a viability of 81.84± 11.99%. and purity of 80%-90%. Primary end point analysis revealed that at 4 wk, the mean serum albumin in the study group increased significantly(2.83± 0.36 vs 2.43 ± 0.42, P = 0.001) when compared with controls. This improvement in albumin was,however, not sustained at 3 mo. However, at the end of3 mo there was a statistically significant improvement in serum creatinine in the study group(0.96 ± 0.33 vs 1.42 ± 0.70, P = 0.01) which translated into a significant improvement in the Model for End-Stage Liver Disease score(15.75 ± 5.13 vs 19.94 ± 6.68,P = 0.04). On statistical analysis of secondary end points, the transplant free survival at the end of 1 mo and 3 mo did not show any significant difference(P =0.60) when compared to the control group. There was no improvement in aspartate transaminase, alanine transaminase, and bilirubin at any point in the study population. There was no mortality benefit in the study group. The procedure was safe with no procedural or treatment related complications.CONCLUSION: Autologous CD 34+ cell infusion is safe and effectively improves liver function in the short term and may serve as a bridge to liver transplantation. 展开更多

BACKGROUND Although expression of interleukin(IL)-34 is upregulated in active ulcerative colitis(UC),the molecular function and underlying mechanism are largely unclear.AIM To investigate the function of IL-34 in acute colitis,in a wound healing model and in colitis-associated cancer in IL-34-deficient mice.METHODS Colitis was induced by administration of dextran sodium sulfate(DSS),and carcinogenesis was induced by azoxymethane(AOM).Whether the impact of IL-34 on colitis was dependent on macrophages was validated by depletion of macrophages in a murine model.The association between IL-34 expression and epithelial proliferation was studied in patients with active UC.RESULTS IL-34 deficiency aggravated murine colitis in acute colitis and in wound healing phase.The effect of IL-34 on experimental colitis was not dependent on macrophage differentiation and polarization.IL-34-deficient mice developed more tumors than wild-type mice following administration of AOM and DSS.No significant difference was shown in degree of cellular differentiation in tumors between wild-type and IL-34-deficient mice.IL-34 was dramatically increased in the active UC patients as previously reported.More importantly,expression of IL-34 was positively correlated with epithelial cell proliferation in patients with UC.CONCLUSION IL-34 deficiency exacerbates colonic inflammation and accelerates colitis-associated carcinogenesis in mice.It might be served as a potential therapeutic target in UC. 展开更多

In this study, we used a classic rat model of traumatic brain injury to test the hypothesis that cold water swimming preconditioning improved the recovery of cognitive functions and explored the mechanisms. Results showed that after traumatic brain injury, pre-conditioned rats(cold water swimming for 3 minutes at 4℃) spent a significantly higher percent of times in the goal quadrant of cold water swim, and escape latencies were shorter than for non-pretreated rats. The number of circulating endothelial progenitor cells was significantly higher in pre-conditioned rats than those without pretreatment at 0, 3, 6 and 24 hours after traumatic brain injury. Immunohistochemical staining and Von Willebrand factor staining demonstrated that the number of CD34~+ stem cells and new blood vessels in the injured hippocampus tissue increased significantly in pre-conditioned rats. These data suggest that pretreatment with cold water swimming could promote the proliferation of endothelial progenitor cells and angiogenesis in the peripheral blood and hippocampus. It also ameliorated cognitive deficits caused by experimental traumatic brain injury. 展开更多

The tumor suppressor p53 is at the hub of the cellular DNA damage response network.P53-dependent cell fate decision is inseparable from p53 dynamics.A type of non-coding microRNA miR-34 has the function of enhancing p53 content.An intriguing question arises:How does miR-34 affect p53 kinetics?To address this question,we reconstruct a p53 signal transduction network model containing miR-34.Some experimental phenomena of p53 pulses are reproduced to explain the rationality of the model.The method of numerical bifurcation is used to investigate the effect of miR-34 on p53 kinetics.We point out that appropriate or higher miR-34 transcription rates can prevent DNA-damaged cell proliferation by causing p53 oscillation or high steady-state kinetic behavior,respectively.However,the lack of miR-34 synthesis ability will induce p53 to remain at a low level,and cells cannot respond correctly to DNA damage.These results are well in line with the anti-cancer role of miR-34.Our work sheds light on how miR-34 carries out its tumor-suppressive function from tuning p53 dynamic aspect. 展开更多

目的:通过独活凝胶贴膏穴位贴敷的药效学与其有效成分蛇床子素、二氢欧山芹醇当归酸酯的药动学和药效学研究,揭示药物在体内的药动学与药效学动态变化规律,验证穴位贴敷的滞后性及应答性。方法:采用微透析采样法对类风湿性关节炎(RA)家... 目的:通过独活凝胶贴膏穴位贴敷的药效学与其有效成分蛇床子素、二氢欧山芹醇当归酸酯的药动学和药效学研究,揭示药物在体内的药动学与药效学动态变化规律,验证穴位贴敷的滞后性及应答性。方法:采用微透析采样法对类风湿性关节炎(RA)家兔关节腔组织液进行采样,并用UPLC-MS/MS法测定独活有效成分蛇床子素、二氢欧山芹醇当归酸酯的含量建立药物动力学模型(PK);用ELISA法检测药效学指标IL-1β、TNF-α和IL-6的抑制率(PD),以效应-时间-药物浓度绘制曲线并进行相关分析;采用Win Nonlin Examples Guide 5.2.1软件对药物效应-血药浓度进行PK-PD模型拟合。结果:在药动学研究中,阳陵泉经皮给药途径符合血管外给药的一级药物动力学模型并得到两组药动学参数:蛇床子素药动学参数:k_(01)=0.15(1/h),k_(10)=0.17(1/h),AUC=3597.07(h·ng/mL),T_(max)=6.00 h,C_(max)=373.18 ng/mL;二氢欧山芹醇当归酸酯药动学数:k_(01)=0.12(1/h),k_(10)=0.18(1/h),AUC=1197.80(h·ng/mL),T_(max)=6.90 h,C_(max)=125.65 ng/mL。在PK-PD模型研究中,通过药效学拟合得到阳陵泉给药途径下IL-1β、TNF-α和IL-6的PK-PD参数以及拟合方程,分别为E=95.72·Ce/568.08+Ce;E=46.32·Ce/618.27+Ce;E=48.32·Ce/118.47+Ce。结论:基于微透析技术和ELISA法的独活凝胶贴膏PK-PD研究方法可行、数据直观、结论理想,说明经络穴位对药物的治疗作用具有放大效应,穴位贴敷对经皮给药途径具有生物促进作用。 展开更多

目的建立H1N1流感病毒感染树鼩模型,探讨流感病毒动力学变化和在呼吸系统组织中的分布特征。方法选择3~3.5周岁成年树鼩(Tupaia Belangeri Chinensis)24只,雌雄各半,随机分为空白组(B组)和模型组(M组)各12只,使用H1N1流感病毒每只600μL... 目的建立H1N1流感病毒感染树鼩模型,探讨流感病毒动力学变化和在呼吸系统组织中的分布特征。方法选择3~3.5周岁成年树鼩(Tupaia Belangeri Chinensis)24只,雌雄各半,随机分为空白组(B组)和模型组(M组)各12只,使用H1N1流感病毒每只600μL(1×10^(6.8)TCID50/0.1 mL)经鼻腔感染M组树鼩,在-3~10 d期间每天早晨测量肛温,同时采集咽拭子、鼻拭子和血液样本测定病毒载量。并测定2、4、7 d血液中抗流感的中和抗体。2、3、5、10 d随机处死3只树鼩,采集鼻甲、气管、咽部和肺组织进行病毒载量检测,取3、5、10 d组织进行病理学检测。结果M组的树鼩表现为被毛蓬乱、食欲不振、运动迟缓、体温升高和鼻咽分泌物增加等临床症状;感染后1 d开始可以在呼吸道中检测到病毒载量,3 d血液中出现病毒载量;病理结果显示鼻甲、咽、气管和肺组织中均有一定程度的病理改变。结论树鼩感染流感病毒后出现的临床症状与人类极为相似,所建立的流感树鼩模型对研究流感的发病机制与抗流感药物评价提供了有重要价值的工具。 展开更多

Thermax 700 thermo gravimetric analysis (TGA) instrument is introduced for the investigation of the reaction and deactivation kinetics of Methanol-to-Olefins (MTO) process with SAPO-34 catalyst.By the use of a special sample basket,the TGA instrument can be viewed as a plug flow fixed-bed reactor,while the weight change of SAPO-34 during reaction can be recorded online.Kinetic data are acquired in the temperature range of 648.2?748.2 K and space velocities of 7.08?35.91 h^-1 (WHSV).Catalyst activity is expressed with average coke content,and selectivity for different products is related as a function of coke content and temperature.Methane is also introduced into the lumping kinetic model,and power exponent function with first-order reaction is adopted for model deduction.Exponential function is tested to give the best fit for catalyst activity and product selectivity with the highest correlation coefficient.The nicely agreed results between experimental and calculated data suggest that the overall kinetic model would be meaningful in both product distribution prediction and reactor simulation. 展开更多

In this paper, a characteristic model based longitudinal control design for the trans-aerosphere vehicle X-34 in its transonic and hypersonic climbing phase is proposed. The design is based on the dynamic characteristics of the vehicle and the curves it is to track in this climbing phase. Through a detailed analysis of the aerodynamics and vehicle dynamics during this climbing phase, an explicit description of the tracking curve for the flight path angle is derived. On the basis of this tracking curve, the tracking curves for the two short-period variables, the angle of attack and the pitch rate, are designed. An all-coefficient adaptive controller is then designed, based on the characteristic modeling, to cause these two short-period variables to follow their respective tracking curves. The proposed design does not require multiple working points, making the design procedure simple. Numerical simulation is performed to validate the performance of the controller. The simulation results indicate that the resulting control law ensures that the vehicle climbs up successfully under the restrictions on the pitch angle and overloading. 展开更多
