In Tessala Mountain (Sidi Bel Abbes Country, North-Western of Algeria), the Ruta genus commonly known by "fidjel" is represented by two species: Ruta Chalepensis L. and Ruta Montana (L.) L. frequently used by local population in a traditional treatment. The water-distilled essential oils from this species yielded 7.23% and 6.104% for Ruta chalepensis L. and Ruta montana (L.) L. respectively. The essential oil of Ruta montana (L.) L. showed a strong antibacterial activity against all bacterial strains tested (Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, Mycobacterium kansasii ATCC ! 2478 and Mycobacterium vaccae ATCC 1548314) compared to Ruta chalepensis L. oils which have a moderate effect only on Staphylococcus aureus ATCC 25923 and Pseudomonas aeruginosa ATCC 27853. Therefor, the antibacterial properties of the essential oils of Ruta Chalepensis L. and Ruta Montana (L.) L. are now well established through this study and therefore could justify their future uses in the treatment of nosocomial infections.展开更多

In t...OBJECTIVE: Chemopreventive approach with natural products, particularly plants and plant-derived ones, is receiving increasing attention for their effective role against cancer without any palpable side effects. In this study, efficacy of ethanolic extract of Ruta graveolens (RG) on skin melanoma cells (A375) in vitro and on 7,12-dimethylbenz(a)anthracene (DMBA)-induced skin cancer in vivo has been tested in Swiss albino mice. METHODS: Studies on cell viability, apoptosis and autophagy induction were conducted in vitro. To check apoptosis, assays like alteration in mitochondrial membrane potential, annexin V-fluorescein isothiocyanate/propidium iodide assay and immunoblot were performed. Fluorescence microscopic and immunoblot assays were performed to confirm autophagy induction. The effects of RG were determined by evaluating body weight, tumor incidence, tumor volume and tumor burden in mice. Enzymatic and non-enzymatic antioxidant status was assessed. The role of some relevant signaling proteins was also analyzed. RESULTS: RG caused death ofA375 cells through induction of caspase 3-mediated apoptosis and Beclin-l-associated autophagy. Moreover, RG administration (75 mg/kg body weight) which showed no acute or chronic toxicity, showed significant reduction in the skin tumor burden of DMBA-painted mice. RG also demonstrated potent anti-lipid peroxidative and antioxidant functions during the course of skin cancer induction by DMBA. CONCLUSION: Chemopreventive potential of RG was demonstrated from overall results of this study indicating its possible use in therapeutic formulation of an effective drug to treat skin cancer.展开更多

The Ruta graveolens (Ruta) is a medicinal plant used in different parts of the world with different therapeutics properties like dermatologic as far as anti-helmintic properties. We analyzed the vascular action measuring the tension to identify vasodilator effect of Ruta extract in norm-tense rat’s aorta incubated and measured in isolated organ bath, and evaluating the toxicity effect in CRL-1730 cell line, through enzymatic assay (MTT), confocal microscopy (propidium iodide stain) and flow cytometry (TUNEL assay), including extracellular reactive oxygen species (ROS) production through luminescence assay. The results show with DE50 29 ± 0.1 μg/mL evidenced vasodilatation, partially endothelium-depend. The cytotoxicity showed with DE50 304.6 ± 2 μg/mL in enzymatic assay (MTT) while evidenced membrane permeability in high concentrations (1500 μg/mL), DNA fragmentation in absence of oxidative stress in only observed when high concentrations of Ruta are used over the cell culture. The vasodilatation activity is executed in subtoxic concentration and partially endothelium-depend without permeability effect in the membrane and deterioration of the cells viability suggesting a complex effect of Ruta preparation in the regulation of vascular tone.展开更多

Usually the cells have several mechanisms for scavenging those free radicals, but, in several pathologic processes, such as cancer, those free radicals increase their production, making it impossible to sustain the system stable, generating the condition called oxidative stress. Ruta graveolens L. (Rue) is a plant commonly used in traditional medicine, mainly as antiinflamatory; this has been related to some organic components, such as Rutin, but there hasn't been any lectin studies in Rue stem. Lectins are glycoproteins of non-enzymatic and no immune origin, able to bind to simple carbohydrates, which lets them bind selectively to malignant cells against normal cells, killing them via apoptosis and reducing the free radicals level. In this study we intended to characterize rue stem lectins as those weren't reported yet. Also, the anticancer and antioxidant activity of these lectins was evaluated, Rue stem lectins were extracted using a saline solution and semipurified to obtain an enriched extract and administrated to nickel oxide treated mice. Oxidative stress was cuantified using the tiobarbituric acid (TBARS) method to quantify Malondialdehyde (MDA), the Griess method to cuantify Nitrites and enzymatic activity of catalase were cuantified in liver. In this study was found that rue stem lectins are useful as a therapeutic auxiliar, considering that its ratio of antioxidant activity is limited, being a prooxidant agent at high concentrations.展开更多

METHOD: For anti-infla...AIM: In the present study, the anti-inflammatory and antioxidant activities of the methanol extract of Ruta graveolens leaves (RG-M) were evaluated using various in vivo and in vitro models. METHOD: For anti-inflammatory activity, RG-M was administered by the oral route (p.o.) in a carrageenan-induced paw edema model, and by the intraperitoneal route (i.p.) in an exudative inflammation model. In vitro inhibition of cyelooxygenase and li- poxygenase enzymes was evaluated. In vitro antioxidant activity was also examined. Endogenous antioxidant status was fiarther evaluated by ferric reducing ability of plasma model. RESULTS: RG-M showed maximum inhibition of carrageenan-induced edema (100 mg·kg ^-1- 33.36%; 200 mg·kg ^-1 _ 45.32% and 400 mg·kg ^-1 56.28%). In the exudative inflammation model, a significant reduction in leukocyte migration (200 mg·kg ^-1_ 54.75% and 400 mg·kg ^-1 - 77.97%) and protein exudation (200 mg·kg ^-131.14% and 400 mg·kg ^-1 - 49.91%) were observed. RG-M also exhibited inhibition of COX-1 (ICs0 182.27 μg·mL ^-1) and COX-2 (IC50 190.16 μg·mL ^-1) as well as 5-LOX (IC50 215.71μg·mL ^-1). Antioxidant activity was significant with improved endogenous antioxidant status. CONCLUSION: The results demonstrated the anti-inflammatory and antioxidant activity of RG-M with potent inhibitory effects on the arachidonic acid pathways.展开更多

The effects of heat treatment...The basic martensitic transformation (MT) properties of Ru50Ta50 alloy, i.e. MT temperature (MTT), temperature hysteresis (△T), and MT heat (△H) were investigated in this paper. The effects of heat treatment and thermal cycling on MT behavior of Ru50Ta50 alloy, the deformation and microstructure characteristics of Ru50Ta50 alloy were also studied for its engineering application as high temperature actuator/sensor materials by means of differential scanning calorimetry, X-ray diffraction, transmission electron microscope, optical microscope, and hardness test. The results showed that a two-stage reversible MT takes place in Ru50Ta50 alloy. The two-stage MT starting temperatures (Ms^1, Ms^2) and the temperature hysteresis (△T1, △T2) are 1047, 784 and 11, 14℃, respectively. No significant effect of aging treatment and thermal cycling on MTT and AT of Ru50Ta50 alloy were observed, but △H decreases slowly with increasing thermal cycles. The hardness and brittleness of Ru50Ta50 alloy are high. The deformation mode of RuTa alloys is twinning.展开更多
