Background:Human skin is affected by ultraviolet rays on a daily basis,and excessive ultraviolet radiation(UVR)can lead to sunburn erythema,tanning,photoaging,and skin tumors.The combination of Astragali Radix(AR)and ...Background:Human skin is affected by ultraviolet rays on a daily basis,and excessive ultraviolet radiation(UVR)can lead to sunburn erythema,tanning,photoaging,and skin tumors.The combination of Astragali Radix(AR)and Anemarrhenae Rhizoma(AAR)is a common pairing in traditional Chinese medicine(TCM).According to earlier studies,they possess properties capable of alleviating the adverse impacts of UVR on the skin.However,the specific actions and underlying mechanisms require further investigation.The study aims to analyze the efficacy of AR-AAR in preventing UVR-induced skin damage and to clarify the associated molecular mechanisms.Methods:Potential signaling pathways by which AR and AAR may protect against UVR-induced skin damage were identified with network pharmacology,molecular docking techniques and molecular dynamics(MD)simulation.Except the normal group,the back skin of SD rats was exposed to 1.1 mW/cm^(2) UVA combined with 0.1 mW/cm^(2) UVB daily,and the UVR skin damage model was established.Morphological features of skin tissues of different groups were discovered through Hematoxylin and Eosin(HE)staining,Masson staining,Weigert staining.ELISA was utilized to measure the levels of reactive oxygen species(ROS),Interleukin 6(IL-6),Interleukin 1β(IL-1β)and Tumor necrosis factos-α(TNF-α)in skin tissues.RT-PCR and Western blot were employed to quantify the mRNA and protein contents of PI3K,AKT,and MMP-9.Results:Network pharmacology analysis predicts that AR-AAR may improve skin damage induced by UVR through the PI3K/AKT signaling pathway.Histological staining shows that AR-AAR can significantly reduce inflammatory infiltration and fibrosis in damaged skin.Treatment with AR-AAR(2:1)significantly reduced the expression levels of IL-1β,IL-6,TNF-αand ROS in UVR-damaged rat skin.After treatment with AR-AAR(2:1),not only did the relative mRNA expression levels of PI3K and AKT and the protein expression levels of PI3K,AKT,P-PI3K,and P-AKT increase,but the mRNA and protein expression levels of MMP-9 decreased.Conclusion:The study indicate that the AR-AAR combination and its active components may mitigate UVR skin damage by modulating the PI3K/AKT signaling pathway.展开更多

Background:Panacis Quinquefolii Radix(PQR)is known for its ability to nourish“Qi”(it serves as the driving force for the functional activities of the body’s organs and meridians,promoting and regulating various phy...Background:Panacis Quinquefolii Radix(PQR)is known for its ability to nourish“Qi”(it serves as the driving force for the functional activities of the body’s organs and meridians,promoting and regulating various physiological functions)and“Yin”(it represents the material foundation of the human body.It plays a role in nourishing,moistening,and cooling the body).Notoginseng Radix et Rhizoma(NRR)is recognized for its properties of resolving blood stasis(it refers to a pathological condition characterized by impaired or stagnant blood circulation within the body).Changes in the compatibility ratio of these herbs often lead to variations in their chemical composition and efficacy.However,the specific alterations in chemical composition and efficacy resulting from compatibility adjustments remain unclear.We aimed to compare the material basis and their effects of different compatibility ratios of PQR and NRR on“Qi”deficiency and blood stasis syndrome(QBS).Methods:This study employed UPLC-Q/TOF-MS to identify effective compounds in the compatibility of PQR and NRR and utilized UPLC-TQ-MS/MS to analyze the dissolution of 16 saponins in PQR and NRR at 9 different ratios.A rat model of QBS was established,and the efficacy of PQR and NRR in treating this syndrome was assessed using hemorheology and coagulation analyses.Results:The study results show that PQR and NRR exhibit significant efficacy,effectively reducing blood viscosity induced by platelet aggregation and lowering inflammatory markers such as IL-6,IL-10,TXB2 and ET associated with vascular injury.Moreover,this combination regulates ATP and ADP levels,enhances energy metabolism,and promotes overall health.A total of 104 compounds in the compatibility of PQR and NRR were identified.The ratios of 1:2 and 1:3 showed the highest total saponin content,but the ratio of 1:1 demonstrated a superior pharmacological effect for the treatment of QBS.Conclusion:In summary,the compatibility of PQR and NRR not only shows the complex interactions between traditional Chinese medicinal materials,but also provides a new idea and method for the treatment of QBS.展开更多

Pinelliae Rhizoma(PR),known as Banxia in Chinese,Hange in Japanese,and Banha in Korean,is a renowned herbal medicine in East Asia derived from the dry tuber of Pinellia ternata(Thunb.)Breit.(PT).It is extensively util...Pinelliae Rhizoma(PR),known as Banxia in Chinese,Hange in Japanese,and Banha in Korean,is a renowned herbal medicine in East Asia derived from the dry tuber of Pinellia ternata(Thunb.)Breit.(PT).It is extensively utilized in dispensing granules,classical prescriptions,and herbal formulas to treat various conditions,including cough,infection,phlegm,nausea,asthma,and inflammation.Despite numerous studies on PR and its classical prescriptions over recent decades,a comprehensive synthesis of available evidence regarding its multifunctional roles and therapeutic potential is lacking.This review aims to address this gap by examining emerging evidence from metabonomics,preclinical studies,and clinical trials,while exploring potential trends and prospects for future research.A systematic literature search was conducted across six electronic databases,including Pub Med,Web of Science,Scopus,Science Direct,Wanfang,and China National Knowledge Infrastructure,to identify relevant articles on PR published until March 2023.PR contains 107 compounds with diverse pharmacological activities,including anti-inflammatory,immune regulatory,anti-viral,anti-cancer,anti-asthma,antitussive and expectorant,antioxidant,anti-obesity,anti-atherosclerosis,antimicrobial,emetic and anti-emetic,anti-convulsant and anti-epileptic,sedative and hypnotic,learning and memory enhancement,and anti-depressant effects.Metabonomic studies suggest that raw PR may exhibit cardiotoxicity and pregnancy toxicity while showing no apparent hepatorenal toxicity.However,limited pharmacokinetic investigations on PR constrain its clinical translation.Furthermore,clinical safety data on PR is scarce,with only four clinical trials assessing its positive effects in pediatric epilepsy,nausea and vomiting,soft tissue injury,and chronic sinus tract.This review aims to enhance understanding of PR and provide valuable information and recommendations for further research and development of herbal medicine.展开更多

This study aimed to assess the therapeutic potential of Anemarrhenae Rhizoma(AR)in osteoporotic rats and to elucidate the metabolic pathways involved in AR’s role in alleviating osteoporosis(OP).OP was induced in rat...This study aimed to assess the therapeutic potential of Anemarrhenae Rhizoma(AR)in osteoporotic rats and to elucidate the metabolic pathways involved in AR’s role in alleviating osteoporosis(OP).OP was induced in rats through ovariectomy(OVX),followed by oral administration of either high or low doses of AR,as well as estradiol valerate,over a 14-week period.Micro-computed tomography(Micro-CT)was employed to examine the femur tissue morphology,while enzyme-linked immunosorbent assay(ELISA)was adopted to measure serum levels of PINP and CTX-I to evaluate AR’s efficacy in treating OP.Additionally,metabolomic profiling of femur tissues was conducted using gas chromatography-mass spectrometry(GC-MS).The bioactive components of AR,along with its therapeutic targets for OP,were identified through UPLC-MS/MS and online database searches,and metabolic networks were established by integrating differential metabolites and potential targets.Furthermore,Western blotting analysis confirmed key molecular targets.The findings revealed that AR treatment significantly mitigated OVX-induced OP in rats.Metabolomic analysis indicated that AR exerted its effects by modulating the levels of 10 key metabolites(such as linoleic acid and inositol)and influencing five crucial metabolic pathways,including linoleic acid metabolism and the phosphoinositide signaling system.Among these,the linoleic acid metabolic pathway emerged as a pivotal focus for further investigation based on the constructed interaction network of differential metabolites and targets.Western blotting analysis demonstrated that AR reversed the up-regulation of CYP1A2 and CYP2C9,two targets associated with the linoleic acid metabolic pathwa y,in OP rats.In conclusion,AR appeared to ameliorate OP by modulating metabolite levels in OVX rats,with its mechanism of action likely centered on regulating the linoleic acid metabolic pathway.展开更多

This study aimed to evaluate the antidepressant effects of the combined administration of total saikosaponins(SSA)and volatile oil of Cyperi Rhizoma(VO)using a mouse depression model induced by chronic restraint plus ...This study aimed to evaluate the antidepressant effects of the combined administration of total saikosaponins(SSA)and volatile oil of Cyperi Rhizoma(VO)using a mouse depression model induced by chronic restraint plus mild stress(CRMS),and to compare the effects with the traditional antidepressant fluoxetine.Male Kunming mice were subjected to 14-day CRMS modeling and then randomly divided into four groups:the combined treatment group(intraperitoneal injection of SSA 3.5 mg·kg^(-1)+VO 35 mg·kg^(-1)),the fluoxetine treatment group(20 mg·kg^(-1)),the normal saline treatment group,and the non-model group.Drugs were administered continuously for 14 days.Depressive-like behaviors were assessed using the Forced Swimming Test(FST),Tail Suspension Test(TST),and Open-Field Test(OFT).The results showed that the absolute immobility time of mice in the CRMS model group was significantly prolonged in FST and TST.Combined administration of SSA and VO significantly improved depressive-like behaviors,restoring the absolute immobility time in FST and TST to levels close to the control group,with efficacy comparable to fluoxetine.This study confirms that the combination of SSA and VO exhibits antidepressant effects equivalent to fluoxetine in the CRMS model,providing experimental evidence for the further clinical development of this traditional Chinese medicine(TCM)compatibility.展开更多
OBJECTIVE:To evaluate the effects of Huangqi(Radix Astragali Mongolici)-Ezhu(Rhizoma Curcumae Phaeocaulis)(HQEZ)on colorectal cancer therapies and to elucidate the potential mechanisms of HQEZ,especially in combinatio...OBJECTIVE:To evaluate the effects of Huangqi(Radix Astragali Mongolici)-Ezhu(Rhizoma Curcumae Phaeocaulis)(HQEZ)on colorectal cancer therapies and to elucidate the potential mechanisms of HQEZ,especially in combination with 5-Fluorouracil(5-FU).METHODS:The anti-tumor effects of HQEZ were evaluated in colorectal cancer models both in vivo and in vitro.The network pharmacological assay was used to investigate potential mechanisms of HQEZ.Potential target genes were selected by Gene Ontology(GO)enrichment analysis,Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis,protein-protein interaction network(PPI)and molecular docking.Within key targets,potential targets related to drug sensitivity,especially the sensitivity to 5-FU,were evaluated in HCT116 in vitro by immunofluorescence,quantitative real-time polymerase chain reaction(qPCR)and Western-blot.Then,changes in potential targets were assessed in tumors from tumor-bearing mice and the expression of these targets was also evaluated in colorectal cancer(COAD)patients from the Cancer Genome Atlas Program(TCGA)database.RESULTS:HQEZ significantly enhanced the anti-tumor activity of 5-FU in vivo and inhibit the growth of HCT116 in vitro.By network pharmacological analysis,key targets,such as protein kinase B(AKT1),epidermal growth factor receptor(EGFR),adenosine triphosphate(ATP)binding cassette subfamily B member 1(ABCB1,also named multidrug resistance protein 1,MDR1),ATP binding cassette subfamily G member 2(ABCG2),thymidylate synthetase(TYMS,also named TS),prostaglandinendoperoxide synthase 2(PTGS2),matrix metallopeptidase 2(MMP2),MMP9,toll like receptor 4(TLR4),TLR9 and dihydropyrimidine dehydrogenase(DPYD),were identified.Additionally,4 potential core active ingredients(Folate,Curcumin,quercetin and kaempferol)were identified to be important for the treatment of colorectal cancer with HQEZ.In key targets,chemoresistance related targets were validated to be affected by HQEZ.Furthermore,5-FU sensitivity related targets,including MDR1,TS,EGFR,ribonucleotide reductase catalytic subunit M1,Breast and Ovarian Cancer Susceptibility Protein 1(BRCA1)and mutl homolog 1 were also significantly reduced by HQEZ both in vitro and in vivo.Finally,these validated key targets and 5-FU sensitivity related targets were demonstrated to be up-regulated in COAD patients based on TCGA database.CONCLUSION:HQEZ has synergistic effects on the antitumor activity of 5-FU in the treatment of colorectal cancer both in vivo and in vitro.The beneficial effect of HQEZ results from the inhibition of the drug sensitivity targets associated with 5-FU.The combination therapy of HQEZ with 5-FU or other chemotherapeutic drugs will also improve the anti-tumor efficacy of chemotherapy.展开更多

However, the efficacy of the AR-NR medicine pair(AN) in improving cardiac remodeling and its underlying mechanism remains unclear. This study aimed to evaluate AN's cardioprotective effect and potential mechanism on cardiac remodeling using transverse aortic constriction(TAC) in mice and angiotensin II(Ang II)-induced neonatal rat cardiomyocytes(NRCMs) and fibroblasts in vitro. High-performance liquid chromatography-quadrupole-time of flight tandem mass spectrometry(HPLC-Q-TOF-MS/MS) characterized 23 main components of AN. AN significantly improved cardiac function in the TAC-induced mice. Furthermore, AN considerably reduced the serum levels of N-terminal pro-B-type natriuretic peptide(NT-pro BNP), cardiac troponin T(CTn-T), and interleukin-6(IL-6) and mitigated inflammatory cell infiltration. Post-AN treatment, TAC-induced heart size approached normal. AN decreased cardiomyocyte cross-sectional area and attenuated the upregulation of cardiac hypertrophy marker genes(ANP, BNP, and MYH7) in vivo and in vitro.Concurrently, AN alleviated collagen deposition in TAC-induced mice. AN also reduced the expression of fibrosis-related indicators(COL1A1 and COL3A1) and inhibited the activation of the transforming growth factor-β1(TGF-β1)/mothers against decapentaplegic homolog 3(Smad3) pathway. Thus, AN improved TAC-induced cardiac remodeling. Moreover, AN downregulated p-dynamin-related protein(Drp1)(Ser616) expression and upregulated mitogen 2(MFN-2) and optic atrophy 1(OPA1) expression in vivo and in vitro, thereby restoring mitochondrial fusion and fission balance. In conclusion, AN improves cardiac remodeling by regulating mitochondrial dynamic balance, providing experimental data for the rational application of Chinese medicine prescriptions with AN as the main component in clinical practice.展开更多
Background:Sophorae Tonkinensis Radix et Rhizoma(ST,also known as ShanDouGen)pertains to the Sophora genus,which is generally distributed in Southwest China.As a celebrated folk medicine,it has heat-clearing and detox...Background:Sophorae Tonkinensis Radix et Rhizoma(ST,also known as ShanDouGen)pertains to the Sophora genus,which is generally distributed in Southwest China.As a celebrated folk medicine,it has heat-clearing and detoxifying,reducing swelling and soothing pharynx.In recent years,there has been a notable rise in adverse events,including hepatotoxicity,associated with the use of ST,however,the mechanism behind ST-induced hepatotoxicity is unclear.Methods:The effects of ST on liver injury were investigated in vivo.Then,serum and liver samples were then analyzed using high-throughput metabolomics techniques.Furthermore,the application of network pharmacology and multivariate statistical analysis has been instrumental in identifying and predicting biomarkers and targets linked to liver toxicity.Results:In this study,we found that ST extract has a certain degree of damage to the rat liver.Using UPLC-MS/MS techniques,463 compounds were identified from ST,of which 73 compounds were absorbed in the blood of the STH group.Metabolomics results showed that amino acid metabolic biomarkers were associated with liver toxicity induced by ST.Notably,we identified 22 core ingredient as toxic effects Q-markers of ST through correlation analysis between biomarkers and absorbed components.On this basis,it is concluded by network pharmacology that ST may cause liver toxicity through PI3K-Akt signaling pathway,NF-kappa B signaling pathway,MAPK signaling pathway,and other pathways.Conclusion:Our findings also demonstrated that the“chemical composition-blood migration component-liver metabolism”stactics has an enormous potential to discern biomarkers and ingredients,and to elaborate the complexity toxicity mechanism of ST.展开更多

Results A qualitative TLC method and a quantitative HPLC method with piceid as the reference substance were established, respectively. With piceid as the reference substance and ethyl acetate-methanol-formic acid-water ( 19:3:0.5:1) as the mobile phase, a TLC method for the identification of Huzhang from the commonly used crude drugs of the same family was also set up. Conclusion The established TLC method can reasonably appraise the quality of the drug and easily distinguish Huzhang from the other commonly used crude drugs of the same family. The HPLC method for determining piceid is simple, reproducible, accurate, and feasible.展开更多
In this study, we developed a novel and simple HPLC-DAD method for simultaneous qualitative and quantitative determination of five major components of Glycyrrhizae Radix et Rhizoma (GRR) in Xiaoer Zhike Tangjiang (...In this study, we developed a novel and simple HPLC-DAD method for simultaneous qualitative and quantitative determination of five major components of Glycyrrhizae Radix et Rhizoma (GRR) in Xiaoer Zhike Tangjiang (XEZKTJ) with standardized reference extract (SRE). The five analytes (liquiritin apioside, liquiritin, isoliquiritin apioside, liquirigenin and glycyrrhizic acid) were well separated with good linearity, precision, stability and repeatability. The recovery rates ranged from 95.69% to 100.80%. The content of the five compounds in 34 batches of commercial XEZKTJ products was determined using standardized GRR extract (SRE method) and individual chemical reference standards (CRS method). Highly similar results were obtained from the two methods, demonstrating the feasibility of the proposed SRE method. Taken together, we proposed an efficient and low-cost way to perform multi-component quality control of XEZKTJ in this study.展开更多

In order to evaluate the crude and pr...The crude and processed Paeoniae Radix Alba-Atractylodis Macrocephalae Rhizoma herbal pairs, originated from Bai-zhu-shao-yao-san, are used to treat different diseases clinically. In order to evaluate the crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs, a simple, easy, and sensitive high-performance liquid chromatography coupled with diode array detectors was developed for simultaneous determination of nine bioactive components in the herbal pairs. The calibration curve exhibited good linearity(r2≥0.9992). The LODs and LOQs were ≤7.30 and 11.53 μg/m L, respectively. The intra-, inter-day and repeatability RSD values of the nine compounds were less than 3.86%, 2.71%, and 4.29%, respectively. The RSD stability values were less than 3.64%. The recovery of the method was in the range of 96.70%–102.10%, with RSD values less than 3.52%. The developed method can be applied to the intrinsic quality control of crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs.展开更多

文摘Background:Human skin is affected by ultraviolet rays on a daily basis,and excessive ultraviolet radiation(UVR)can lead to sunburn erythema,tanning,photoaging,and skin tumors.The combination of Astragali Radix(AR)and Anemarrhenae Rhizoma(AAR)is a common pairing in traditional Chinese medicine(TCM).According to earlier studies,they possess properties capable of alleviating the adverse impacts of UVR on the skin.However,the specific actions and underlying mechanisms require further investigation.The study aims to analyze the efficacy of AR-AAR in preventing UVR-induced skin damage and to clarify the associated molecular mechanisms.Methods:Potential signaling pathways by which AR and AAR may protect against UVR-induced skin damage were identified with network pharmacology,molecular docking techniques and molecular dynamics(MD)simulation.Except the normal group,the back skin of SD rats was exposed to 1.1 mW/cm^(2) UVA combined with 0.1 mW/cm^(2) UVB daily,and the UVR skin damage model was established.Morphological features of skin tissues of different groups were discovered through Hematoxylin and Eosin(HE)staining,Masson staining,Weigert staining.ELISA was utilized to measure the levels of reactive oxygen species(ROS),Interleukin 6(IL-6),Interleukin 1β(IL-1β)and Tumor necrosis factos-α(TNF-α)in skin tissues.RT-PCR and Western blot were employed to quantify the mRNA and protein contents of PI3K,AKT,and MMP-9.Results:Network pharmacology analysis predicts that AR-AAR may improve skin damage induced by UVR through the PI3K/AKT signaling pathway.Histological staining shows that AR-AAR can significantly reduce inflammatory infiltration and fibrosis in damaged skin.Treatment with AR-AAR(2:1)significantly reduced the expression levels of IL-1β,IL-6,TNF-αand ROS in UVR-damaged rat skin.After treatment with AR-AAR(2:1),not only did the relative mRNA expression levels of PI3K and AKT and the protein expression levels of PI3K,AKT,P-PI3K,and P-AKT increase,but the mRNA and protein expression levels of MMP-9 decreased.Conclusion:The study indicate that the AR-AAR combination and its active components may mitigate UVR skin damage by modulating the PI3K/AKT signaling pathway.

文摘Background:Cyperi Rhizoma,derived from Cyperus rotundus L.,is a widely used medicinal herb in traditional Chinese medicine(TCM),with Shandong Province recognized as its geo-authentic habitat.However,the quality of Cyperi Rhizoma varies significantly across different regions,potentially influencing its therapeutic efficacy.This study investigates the influence of geographic origin on the chemical composition and overall quality of Cyperi Rhizoma.Methods:A comprehensive approach,including traditional quality assessment,GC-MS(g as c hromatography-m ass s pectrometry),RP-HPLC(r everse p hase h igh-p erformance l iquid c hromatography),and FT-IR(f ourier t ransform i nfrared s pectroscopy)techniques,was employed to analyze Cyperi Rhizoma samples from Shandong Province.These methods examined the physical appearance,chemical profile,and content variations,particularly focusing onα-cyperone.Results:Traditional quality assessments revealed noticeable differences in the external characteristics of the samples.GC-MS analysis identified a variety of unique chemical constituents,while RP-HPLC and FT-IR showed significant variations inα-cyperone content,with higher levels found in Shandong samples.Conclusion:These results demonstrate that geographic origin is a critical determinant of Cyperi Rhizoma quality,with Shandong specimens exhibiting superiorα-cyperone levels and characteristic phytochemical profiles.This validates the geo-authenticity concept in TCM and provides actionable data for developing evidence-based quality standards,suggesting that provenance should be prioritized in medicinal material selection and pharmacopeial specifications.

文摘Background:Panacis Quinquefolii Radix(PQR)is known for its ability to nourish“Qi”(it serves as the driving force for the functional activities of the body’s organs and meridians,promoting and regulating various physiological functions)and“Yin”(it represents the material foundation of the human body.It plays a role in nourishing,moistening,and cooling the body).Notoginseng Radix et Rhizoma(NRR)is recognized for its properties of resolving blood stasis(it refers to a pathological condition characterized by impaired or stagnant blood circulation within the body).Changes in the compatibility ratio of these herbs often lead to variations in their chemical composition and efficacy.However,the specific alterations in chemical composition and efficacy resulting from compatibility adjustments remain unclear.We aimed to compare the material basis and their effects of different compatibility ratios of PQR and NRR on“Qi”deficiency and blood stasis syndrome(QBS).Methods:This study employed UPLC-Q/TOF-MS to identify effective compounds in the compatibility of PQR and NRR and utilized UPLC-TQ-MS/MS to analyze the dissolution of 16 saponins in PQR and NRR at 9 different ratios.A rat model of QBS was established,and the efficacy of PQR and NRR in treating this syndrome was assessed using hemorheology and coagulation analyses.Results:The study results show that PQR and NRR exhibit significant efficacy,effectively reducing blood viscosity induced by platelet aggregation and lowering inflammatory markers such as IL-6,IL-10,TXB2 and ET associated with vascular injury.Moreover,this combination regulates ATP and ADP levels,enhances energy metabolism,and promotes overall health.A total of 104 compounds in the compatibility of PQR and NRR were identified.The ratios of 1:2 and 1:3 showed the highest total saponin content,but the ratio of 1:1 demonstrated a superior pharmacological effect for the treatment of QBS.Conclusion:In summary,the compatibility of PQR and NRR not only shows the complex interactions between traditional Chinese medicinal materials,but also provides a new idea and method for the treatment of QBS.

文摘Thrombosis is a leading cause of mortality and morbidity in patients suffering from polycythemia vera(PV).Drug pair Salviae Miltiorrhizae Radix et Rhizoma(Danshen,DS)and Notoginseng Radix et Rhizoma(Sanqi,SQ)is common traditional Chinese medicine(TCM)used in clinical practice to promote blood circulation and eliminate blood stasis.In this study,network pharmacology and molecular docking were used to analyze the potentially active ingredients and underlying mechanisms of drug pair DS-SQ against thrombosis after PV.These results show that 54 targets are related to both disease and the drug pair.Nineteen core targets,including IL-6 and AKT1,were screened.Luteolin and tanshinone IIa from DS as well as quercetin from SQ might be the major substances in the treatment of thrombosis after PV.KEGG enrichment analysis demonstrated that the lipid and atherosclerosis signaling pathway might play a significant role.These results provide valuable insights and a reference for the use of drug pair DS-SQ in management of thrombosis after PV and lay a foundation for further exploration of pharmacological effects.

文摘Pinelliae Rhizoma(PR),known as Banxia in Chinese,Hange in Japanese,and Banha in Korean,is a renowned herbal medicine in East Asia derived from the dry tuber of Pinellia ternata(Thunb.)Breit.(PT).It is extensively utilized in dispensing granules,classical prescriptions,and herbal formulas to treat various conditions,including cough,infection,phlegm,nausea,asthma,and inflammation.Despite numerous studies on PR and its classical prescriptions over recent decades,a comprehensive synthesis of available evidence regarding its multifunctional roles and therapeutic potential is lacking.This review aims to address this gap by examining emerging evidence from metabonomics,preclinical studies,and clinical trials,while exploring potential trends and prospects for future research.A systematic literature search was conducted across six electronic databases,including Pub Med,Web of Science,Scopus,Science Direct,Wanfang,and China National Knowledge Infrastructure,to identify relevant articles on PR published until March 2023.PR contains 107 compounds with diverse pharmacological activities,including anti-inflammatory,immune regulatory,anti-viral,anti-cancer,anti-asthma,antitussive and expectorant,antioxidant,anti-obesity,anti-atherosclerosis,antimicrobial,emetic and anti-emetic,anti-convulsant and anti-epileptic,sedative and hypnotic,learning and memory enhancement,and anti-depressant effects.Metabonomic studies suggest that raw PR may exhibit cardiotoxicity and pregnancy toxicity while showing no apparent hepatorenal toxicity.However,limited pharmacokinetic investigations on PR constrain its clinical translation.Furthermore,clinical safety data on PR is scarce,with only four clinical trials assessing its positive effects in pediatric epilepsy,nausea and vomiting,soft tissue injury,and chronic sinus tract.This review aims to enhance understanding of PR and provide valuable information and recommendations for further research and development of herbal medicine.
文摘Based on multidisciplinary research methods,this study analyzes the mechanisms and potential of Gastrodiae Rhizoma(Tianma)and Uncariae Ramulus Cum Uncis compatibility in treating hypertension.The research confirms that components including rhynchophylline and gastrodin exert antihypertensive effects by inhibiting vascular smooth muscle proliferation and modulating calcium signaling pathways.The combination of these two herbs enhances the bioavailability of core components,synergistically regulates targets such as PTGS2 and NOS3,activates the PPAR signaling pathway,and strengthens vasodilation and antioxidant capacity.Clinical studies demonstrate that Tianma Gouteng Decoction exhibits superior efficacy in improving mitochondrial function and delaying vascular aging compared to single components.This investigation provides scientific evidence for the multi-target antihypertensive effects of traditional Chinese medicine and promotes its modern application in cardiovascular disease treatment.

文摘This study aimed to assess the therapeutic potential of Anemarrhenae Rhizoma(AR)in osteoporotic rats and to elucidate the metabolic pathways involved in AR’s role in alleviating osteoporosis(OP).OP was induced in rats through ovariectomy(OVX),followed by oral administration of either high or low doses of AR,as well as estradiol valerate,over a 14-week period.Micro-computed tomography(Micro-CT)was employed to examine the femur tissue morphology,while enzyme-linked immunosorbent assay(ELISA)was adopted to measure serum levels of PINP and CTX-I to evaluate AR’s efficacy in treating OP.Additionally,metabolomic profiling of femur tissues was conducted using gas chromatography-mass spectrometry(GC-MS).The bioactive components of AR,along with its therapeutic targets for OP,were identified through UPLC-MS/MS and online database searches,and metabolic networks were established by integrating differential metabolites and potential targets.Furthermore,Western blotting analysis confirmed key molecular targets.The findings revealed that AR treatment significantly mitigated OVX-induced OP in rats.Metabolomic analysis indicated that AR exerted its effects by modulating the levels of 10 key metabolites(such as linoleic acid and inositol)and influencing five crucial metabolic pathways,including linoleic acid metabolism and the phosphoinositide signaling system.Among these,the linoleic acid metabolic pathway emerged as a pivotal focus for further investigation based on the constructed interaction network of differential metabolites and targets.Western blotting analysis demonstrated that AR reversed the up-regulation of CYP1A2 and CYP2C9,two targets associated with the linoleic acid metabolic pathwa y,in OP rats.In conclusion,AR appeared to ameliorate OP by modulating metabolite levels in OVX rats,with its mechanism of action likely centered on regulating the linoleic acid metabolic pathway.

文摘[Objectives]This study was conducted to perform visual analysis of the research hotspots and development trends of Cyperi Rhizoma in the field of antidepressant based on CiteSpace,so as to explore the application and development direction of Cyperi Rhizoma in the field of antidepressant.[Methods]Highly-relevant literatures were selected from the core database of China National Knowledge Infrastructure(CNKI),and CiteSpace and WPS office software were employed to visually analyze relevant contents such as publishing institutions,scholars,keywords,publishing time,and citation frequency.[Results]A total of 297 domestic relevant literatures were selected.Most of the publications,institutions and authors were concentrated in universities,affiliated institutions and scientific research institutes of traditional Chinese medicine in China,and no relatively novel applied research direction has emerged.At present,the hot spots and frontiers of application were mostly concentrated in its role in treating depression,anxiety,gynecological diseases and other disorders.[Conclusions]The research on Cyperi Rhizoma for its antidepressant effects in China originated in the late 20 th century.From 2004 to 2024,studies have primarily focused on its pharmacological principles,mechanisms of action,and classification,while the exploration of its application in specific depressive disorders was limited.Overall,research progress has been relatively slow.Currently,further efforts are needed to optimize the active antidepressant components of Cyperi Rhizoma and clarify its mechanisms of action,which will facilitate its broader application in treating various stages of depressive disorders.
文摘As a diuretic and dampness-eliminating medicinal agent in traditional Chinese medicine(TCM),Zexie(Alismatis Rhizoma)is commonly used for symptoms such as dysuria,edema and abdominal distension,diarrhea with oliguria,and dizziness due to phlegm–fluid retention.Systematic phytochemical studies have revealed that the active components of Zexie(Alismatis Rhizoma)are primarily triterpenoids,along with sesquiterpenes,polysaccharides,sterols,alkaloids,phenolic acids,and lignans.These components form the material basis for its pharmacological activities.Recent breakthroughs in pharmacological research have expanded beyond its traditionally recognized diuretic and anti-inflammatory effects:Its antiurolithiasis effect has been clearly linked to inhibiting the crystallization of stone components and promoting stone expulsion;its renal protective effect can ameliorate kidney injury by reducing oxidative stress and suppressing inflammatory responses in renal tissues;its lipid-lowering mechanism involves regulating lipid metabolism pathways and reducing lipid deposition;in terms of anticancer activity,it exhibits proliferation inhibition and apoptosis induction in various tumor cells such as liver,lung,and colon cancers;additionally,it shows significant antibacterial activity against pathogens,including Escherichia coli and Staphylococcus aureus.

文摘Ultra-performance liquid chromatography-mass spectrometry(UPLC-MS)technology was employed to analyze the chemical composition of three Clematidis Radix et Rhizoma sourced from different origins.Common characteristic components were identified through comprehensive literature review and the integration of fragment ion data.A rheumatoid arthritis(AA)model was induced in Sprague-Dawley(SD)rats using Freund’s complete adjuvant,and simultaneous administration of extracts from the three original plants of Clematidis Radix et Rhizoma,along with different doses of magnoflorine,was conducted to evaluate differences in therapeutic efficacy.A common characteristic component was identified,magnoflorine,present in Clematis chinensis Osbeck.Clematis hexapetala Pall.,and Clematis manshurica Rupr.Extracts from all three original plants,as well as each dose group of the Rhizomas of Clematis chinensis Osbeck.Clematis hexapetala Pall.,Clematis manshurica Rupr.,and magnoflorine,demonstrated efficacy in alleviating rheumatoid arthritis symptoms by enhancing cellular infiltration,reducing vascular opacification,and lowering serum levels of inflammatory factors.Magnoflorine has the potential to be used as a quality marker for the three original plants of Clematidis Radix et Rhizoma.Additionally,both the three original plants and magnoflorine exhibit therapeutic potential for rheumatoid arthritis,which provides a solid theoretical and experimental basis for the establishment of more scientifically grounded quality standards of Clematidis Radix et Rhizoma and promotes the rational utilization of medicinal resources.

文摘Polygonati Rhizoma,a functional food and a traditional Chinese medicine broadly used in China and several Southeast Asia countries,possesses effective health-promoting activities.Prepared from 3 plants in Polygonatum genus(Polygonatum kingianum,Polygonatum sibiricum,and Polygonatum cyrtonema),Polygonati Rhizoma has drawn increasing attention due to its remarkable immune-enhancing and metabolic regulatory activities in recent years.In this review,we summarized the updated research of chemical constituents and biological activities of Polygonati Rhizoma,especially the metabolic regulation,immunomodulatory effects,and anti-fatigue activities,aiming to provide a comprehensive understanding,broaden the usage and promote more in-depth exploration of Polygonati Rhizoma as a functional food.

文摘This study aimed to evaluate the antidepressant effects of the combined administration of total saikosaponins(SSA)and volatile oil of Cyperi Rhizoma(VO)using a mouse depression model induced by chronic restraint plus mild stress(CRMS),and to compare the effects with the traditional antidepressant fluoxetine.Male Kunming mice were subjected to 14-day CRMS modeling and then randomly divided into four groups:the combined treatment group(intraperitoneal injection of SSA 3.5 mg·kg^(-1)+VO 35 mg·kg^(-1)),the fluoxetine treatment group(20 mg·kg^(-1)),the normal saline treatment group,and the non-model group.Drugs were administered continuously for 14 days.Depressive-like behaviors were assessed using the Forced Swimming Test(FST),Tail Suspension Test(TST),and Open-Field Test(OFT).The results showed that the absolute immobility time of mice in the CRMS model group was significantly prolonged in FST and TST.Combined administration of SSA and VO significantly improved depressive-like behaviors,restoring the absolute immobility time in FST and TST to levels close to the control group,with efficacy comparable to fluoxetine.This study confirms that the combination of SSA and VO exhibits antidepressant effects equivalent to fluoxetine in the CRMS model,providing experimental evidence for the further clinical development of this traditional Chinese medicine(TCM)compatibility.

文摘OBJECTIVE:To evaluate the effects of Huangqi(Radix Astragali Mongolici)-Ezhu(Rhizoma Curcumae Phaeocaulis)(HQEZ)on colorectal cancer therapies and to elucidate the potential mechanisms of HQEZ,especially in combination with 5-Fluorouracil(5-FU).METHODS:The anti-tumor effects of HQEZ were evaluated in colorectal cancer models both in vivo and in vitro.The network pharmacological assay was used to investigate potential mechanisms of HQEZ.Potential target genes were selected by Gene Ontology(GO)enrichment analysis,Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis,protein-protein interaction network(PPI)and molecular docking.Within key targets,potential targets related to drug sensitivity,especially the sensitivity to 5-FU,were evaluated in HCT116 in vitro by immunofluorescence,quantitative real-time polymerase chain reaction(qPCR)and Western-blot.Then,changes in potential targets were assessed in tumors from tumor-bearing mice and the expression of these targets was also evaluated in colorectal cancer(COAD)patients from the Cancer Genome Atlas Program(TCGA)database.RESULTS:HQEZ significantly enhanced the anti-tumor activity of 5-FU in vivo and inhibit the growth of HCT116 in vitro.By network pharmacological analysis,key targets,such as protein kinase B(AKT1),epidermal growth factor receptor(EGFR),adenosine triphosphate(ATP)binding cassette subfamily B member 1(ABCB1,also named multidrug resistance protein 1,MDR1),ATP binding cassette subfamily G member 2(ABCG2),thymidylate synthetase(TYMS,also named TS),prostaglandinendoperoxide synthase 2(PTGS2),matrix metallopeptidase 2(MMP2),MMP9,toll like receptor 4(TLR4),TLR9 and dihydropyrimidine dehydrogenase(DPYD),were identified.Additionally,4 potential core active ingredients(Folate,Curcumin,quercetin and kaempferol)were identified to be important for the treatment of colorectal cancer with HQEZ.In key targets,chemoresistance related targets were validated to be affected by HQEZ.Furthermore,5-FU sensitivity related targets,including MDR1,TS,EGFR,ribonucleotide reductase catalytic subunit M1,Breast and Ovarian Cancer Susceptibility Protein 1(BRCA1)and mutl homolog 1 were also significantly reduced by HQEZ both in vitro and in vivo.Finally,these validated key targets and 5-FU sensitivity related targets were demonstrated to be up-regulated in COAD patients based on TCGA database.CONCLUSION:HQEZ has synergistic effects on the antitumor activity of 5-FU in the treatment of colorectal cancer both in vivo and in vitro.The beneficial effect of HQEZ results from the inhibition of the drug sensitivity targets associated with 5-FU.The combination therapy of HQEZ with 5-FU or other chemotherapeutic drugs will also improve the anti-tumor efficacy of chemotherapy.

文摘Astragali Radix(AR) and Notoginseng Radix et Rhizoma(NR) are frequently employed in cardiovascular disease treatment. However, the efficacy of the AR-NR medicine pair(AN) in improving cardiac remodeling and its underlying mechanism remains unclear. This study aimed to evaluate AN's cardioprotective effect and potential mechanism on cardiac remodeling using transverse aortic constriction(TAC) in mice and angiotensin II(Ang II)-induced neonatal rat cardiomyocytes(NRCMs) and fibroblasts in vitro. High-performance liquid chromatography-quadrupole-time of flight tandem mass spectrometry(HPLC-Q-TOF-MS/MS) characterized 23 main components of AN. AN significantly improved cardiac function in the TAC-induced mice. Furthermore, AN considerably reduced the serum levels of N-terminal pro-B-type natriuretic peptide(NT-pro BNP), cardiac troponin T(CTn-T), and interleukin-6(IL-6) and mitigated inflammatory cell infiltration. Post-AN treatment, TAC-induced heart size approached normal. AN decreased cardiomyocyte cross-sectional area and attenuated the upregulation of cardiac hypertrophy marker genes(ANP, BNP, and MYH7) in vivo and in vitro.Concurrently, AN alleviated collagen deposition in TAC-induced mice. AN also reduced the expression of fibrosis-related indicators(COL1A1 and COL3A1) and inhibited the activation of the transforming growth factor-β1(TGF-β1)/mothers against decapentaplegic homolog 3(Smad3) pathway. Thus, AN improved TAC-induced cardiac remodeling. Moreover, AN downregulated p-dynamin-related protein(Drp1)(Ser616) expression and upregulated mitogen 2(MFN-2) and optic atrophy 1(OPA1) expression in vivo and in vitro, thereby restoring mitochondrial fusion and fission balance. In conclusion, AN improves cardiac remodeling by regulating mitochondrial dynamic balance, providing experimental data for the rational application of Chinese medicine prescriptions with AN as the main component in clinical practice.

文摘Background:Sophorae Tonkinensis Radix et Rhizoma(ST,also known as ShanDouGen)pertains to the Sophora genus,which is generally distributed in Southwest China.As a celebrated folk medicine,it has heat-clearing and detoxifying,reducing swelling and soothing pharynx.In recent years,there has been a notable rise in adverse events,including hepatotoxicity,associated with the use of ST,however,the mechanism behind ST-induced hepatotoxicity is unclear.Methods:The effects of ST on liver injury were investigated in vivo.Then,serum and liver samples were then analyzed using high-throughput metabolomics techniques.Furthermore,the application of network pharmacology and multivariate statistical analysis has been instrumental in identifying and predicting biomarkers and targets linked to liver toxicity.Results:In this study,we found that ST extract has a certain degree of damage to the rat liver.Using UPLC-MS/MS techniques,463 compounds were identified from ST,of which 73 compounds were absorbed in the blood of the STH group.Metabolomics results showed that amino acid metabolic biomarkers were associated with liver toxicity induced by ST.Notably,we identified 22 core ingredient as toxic effects Q-markers of ST through correlation analysis between biomarkers and absorbed components.On this basis,it is concluded by network pharmacology that ST may cause liver toxicity through PI3K-Akt signaling pathway,NF-kappa B signaling pathway,MAPK signaling pathway,and other pathways.Conclusion:Our findings also demonstrated that the“chemical composition-blood migration component-liver metabolism”stactics has an enormous potential to discern biomarkers and ingredients,and to elaborate the complexity toxicity mechanism of ST.

文摘Aim To establish reliable methods for evaluating the quality of rhizoma of Polygonum cuspidatum( Huzhang in Chinese). Methods TLC and HPLC were employed for the chemical identification and content determination,respectively. Results A qualitative TLC method and a quantitative HPLC method with piceid as the reference substance were established, respectively. With piceid as the reference substance and ethyl acetate-methanol-formic acid-water ( 19:3:0.5:1) as the mobile phase, a TLC method for the identification of Huzhang from the commonly used crude drugs of the same family was also set up. Conclusion The established TLC method can reasonably appraise the quality of the drug and easily distinguish Huzhang from the other commonly used crude drugs of the same family. The HPLC method for determining piceid is simple, reproducible, accurate, and feasible.

文摘In this study, we developed a novel and simple HPLC-DAD method for simultaneous qualitative and quantitative determination of five major components of Glycyrrhizae Radix et Rhizoma (GRR) in Xiaoer Zhike Tangjiang (XEZKTJ) with standardized reference extract (SRE). The five analytes (liquiritin apioside, liquiritin, isoliquiritin apioside, liquirigenin and glycyrrhizic acid) were well separated with good linearity, precision, stability and repeatability. The recovery rates ranged from 95.69% to 100.80%. The content of the five compounds in 34 batches of commercial XEZKTJ products was determined using standardized GRR extract (SRE method) and individual chemical reference standards (CRS method). Highly similar results were obtained from the two methods, demonstrating the feasibility of the proposed SRE method. Taken together, we proposed an efficient and low-cost way to perform multi-component quality control of XEZKTJ in this study.
文摘Coptidis Rhizoma, a well known heat-clearing Chinese Materia Medica, exhibited multi-target anti-Alzheimer effectsin vitro through inhibition of AI342 aggregation, ACHE, and BACE1. Six berberine-like alkaloids were found to be the majoractive constituents of Coptidis Rhizoma by HPLC analysis along with activity assays. A standard combination of the six majoralkaloids was demonstrated to fully reproduce the in vitro effects of Coptidis Rhizoma. Molecular docking studies revealed thatberberine-like alkaloids could directly bind at the active sites of AI342, ACHE, and BACE1 through hydrophobic interactions,aromatic stacking, and electrostatic interactions.

文摘The crude and processed Paeoniae Radix Alba-Atractylodis Macrocephalae Rhizoma herbal pairs, originated from Bai-zhu-shao-yao-san, are used to treat different diseases clinically. In order to evaluate the crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs, a simple, easy, and sensitive high-performance liquid chromatography coupled with diode array detectors was developed for simultaneous determination of nine bioactive components in the herbal pairs. The calibration curve exhibited good linearity(r2≥0.9992). The LODs and LOQs were ≤7.30 and 11.53 μg/m L, respectively. The intra-, inter-day and repeatability RSD values of the nine compounds were less than 3.86%, 2.71%, and 4.29%, respectively. The RSD stability values were less than 3.64%. The recovery of the method was in the range of 96.70%–102.10%, with RSD values less than 3.52%. The developed method can be applied to the intrinsic quality control of crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs.
