Reader proteins that bind specific methyllysine are important to biological functions of lysine methylation,but readers of many methyllysine sites are still unknown.Therefore,development of covalent probes is importan...Reader proteins that bind specific methyllysine are important to biological functions of lysine methylation,but readers of many methyllysine sites are still unknown.Therefore,development of covalent probes is important to identify readers from cell samples so as to understand biological roles of lysine methylation.Generally,readers bind methyllysine via aromatic cages that contain tryptophan,tyrosine and phenylalanine,that offer a unique motif for selective crosslinking.We recently reported a site-selective tryptophan crosslinking strategy based on dimethylsulfonium that mimics dimethyllysine to crosslink tryptophan in aromatic cages of readers.Since tyrosine is a key residue for binding affinity to methyllysine,especially some readers that do not contain tryptophan residues in the binding pocket.Here we developed strategies of site-selective crosslinking to tyrosine.Ultraviolet(UV)source was applied to excite tyrosine at neutral pH or phenoxide at basic p H,and subsequent single-electron transfer(SET)from Tyr*to sulfonium inside the binding pocket enables selective crosslinking.In consequence,methyllysine readers with tyrosine-containing aromatic cages could be selectively crosslinked by site-specific sulfonium peptide probes.In addition,we expanded substrates from aromatic cages to tyrosine residues of proximate contact with sulfonium probes.The pair of LgBiT and SmBiT exhibited orthogonal crosslinking in complicated cell samples.As a result,we may expand sulfonium tools to target local tyrosine in future investigations.展开更多
In the context of the continuous deepening of the“Double Reduction”policy and the growing demand for quality education,leveled mathematics readers,as an emerging form of publishing that integrates subject education ...In the context of the continuous deepening of the“Double Reduction”policy and the growing demand for quality education,leveled mathematics readers,as an emerging form of publishing that integrates subject education and reading experience,face challenges such as unclear leveling logic,insufficient functional support,and weak user engagement.This paper introduces the 4V marketing theory and constructs an analytical framework from four dimensions:differentiation,functionality,added value,and resonance.Two representative products,“Climbing Mathematics”and“Spark Mathematics,”are selected for a typical case comparison to identify their strengths and weaknesses in content design,service systems,and brand operation,and to extract transferable strategic elements.The study finds that the user-value-oriented strategy based on the 4V model can effectively address the core issues in the market promotion and user relationship building of leveled mathematics readers,providing practical paths and theoretical support for educational publishing institutions to achieve product innovation and brand upgrading in this niche field.展开更多

The analysis indicates it has better performance than a conventional Gilbert mixer pair in commutating mode. A quadrature down-conversion mixer based on this topology is designed and optimized for an ultra high frequency RFID reader. Operating in the 915MHz ISM band, the presented quadrature mixer measures a conversion gain of 12.5dB,an IIP3 of 10dBm, an IIP2 of 58dBm, and an SSB noise figure of 17.6riB. METHODS: Ten capsule endoscopy cases of small bowel lesions were administered to the reade...AIM: To determine the performance of novice readers (4^th year medical students) for detecting capsule endoscopy findings. METHODS: Ten capsule endoscopy cases of small bowel lesions were administered to the readers. Gold standard findings were pre-defined by gastroenterologists. Ten gold standard "targets" were identified among the 10 cases. Readers were given a 30-min overview of Rapid Reader software and instructed to mark any potential areas of abnormalities. A software program was developed using SAS to analyze the thumbnailed findings. RESULTS: The overall sensitivity for detecting the gold standard findings was 80%. As a group, at least 5 out of 10 readers detected each gold standard finding per recording. All the gold standard targets were identified when the readers' results were combined. Incidental finding/false positive rate ranged between 8.2-59.8 per reader. CONCLUSION: A panel of medical students with minimal endoscopic experience can achieve high sensitivity in detecting lesions on capsule endoscopy. A group of novice readers can pre-screen recordings to thumbnail potential areas of small bowel lesions for further review. These thumbnails must be reviewed to determine the clinical relevance. Further studies are ongoing to assess other cohorts.展开更多
