The capsid protein(CP) gene of the major outer capsid protein of RRSV was expressed in Escherichia coli BL21(DE3) using the pMAL-C2 X expression vector. The recombinant protein was used as the immunogen to immunize BALB/c mice. A hybridoma cell line 8A12 secreting monoclonal antibody(MAb) against RRSV was obtained by fusing mouse myeloma cells(Sp 2/0) with spleen cells from the immunized BALB/c mice. Western blot analysis showed that the MAb 8A12 can specifically react with RRSV CP. Using the MAb, an antigen-coated-plate enzyme-linked immunosorbent assay(ACP-ELISA), a dot enzyme-linked immunosorbent assay(dot-ELISA), and immunocapture-RT-PCR(IC-RT-PCR) assay were developed to detect RRSV. The established ACP-ELISA, dot-blot ELISA and IC-RT-PCR methods could detect RRSV in infected rice tissue crude extracts with dilutions of 1:40 960, 1:1 280 and 1:655 360(w/v, g mL-1), respectively. The ACP-ELISA and dot-blot ELISA methods could detect RRSV in infected insect vector crude extracts with dilutions of 1:12 800 and 1:1 600(an individual planthopper μL-1), respectively. The field survey revealed that Rice ragged stunt disease occurs on rice in Hainan, Yunnan, Guangxi, Sichuan, Guizhou, Fujian, Hunan, Jiangxi and Zhejiang in China.展开更多
Given its direct borrowings from and intertextual relations to other similar texts,“A Tale of the Ragged Mountains”has often been accused of flagrant plagiarism or lack of originality.Such a simple assertion ignores...Given its direct borrowings from and intertextual relations to other similar texts,“A Tale of the Ragged Mountains”has often been accused of flagrant plagiarism or lack of originality.Such a simple assertion ignores not only Poe’s deviation from these sources but also his rationale for such initiatives.While duly recognizing Poe’s indebtedness to his predecessors and contemporaries,this paper proposes to examine Poe’s literary re-creation and parody by concentrating on his subtle treatment of these borrowed texts,and in so doing,look also at his subversive approach to colonialist discourses.It suggests that in recounting Bedloe’s encounters with“Indians”in America and Asia,the tale actually creates a world of doubles to connect the present with the past,the imperial with the exotic,and the self with the other.In this regard,Dr.Templeton’s mesmeric treatment of Bedloe is both a reenactment and purgation of haunting colonial memories.The so-called medical accident at the end of the tale,therefore,registers Poe’s reflection on and warning against the imperial mindset.展开更多

T-to-C transition at nucl...Background: Myoclonic epilepsy with ragged red fibers (MERRF) syndrome is characterized by myoclonus, generalized epilepsy, cerebellar ataxia, and ragged red fibers (RRFs) in the muscle. T-to-C transition at nucleotide position 14709 in the mitochondrial tRNA glutamic acid (tRNA^Gla) gene has previously been associated with maternally inherited diabetes and deathess. However, the association between MERRF and mitochondrial T14709C mutation (m.TI4709C) has never been reported before. Methods: Clinical information of a 17-year-old patient was collected; muscle biopsy and next-generation sequencing (NGS) of whole mitochondrial and neuromuscular disease panel were then conducted. Finally, sanger sequencing was carried out to confirm the mutations. Results: The patient presented a typical MERRF phenotype with muscle weakness, epileptic seizure, clonic episodes, cerebellar ataxia, and spinal scoliosis. Muscle biopsy showed RRFs which indicated abnormal mitochondrial functions. NGS of whole mitochondrial gene revealed m.TI4709C mutation, confirmed by Sanger sequencing. Conclusion: We present a sporadic patient with typical MERRF presentation carrying the mutation ofm.T14709C, which expanded the spectrum of re.T14709C.展开更多
