Acute ischemic stroke is a clinical emergency and a condition with high morbidity,mortality,and disability.Accurate predictive,diagnostic,and prognostic biomarkers and effective therapeutic targets for acute ischemic stroke remain undetermined.With innovations in high-throughput gene sequencing analysis,many aberrantly expressed non-coding RNAs(ncRNAs)in the brain and peripheral blood after acute ischemic stroke have been found in clinical samples and experimental models.Differentially expressed ncRNAs in the post-stroke brain were demonstrated to play vital roles in pathological processes,leading to neuroprotection or deterioration,thus ncRNAs can serve as therapeutic targets in acute ischemic stroke.Moreover,distinctly expressed ncRNAs in the peripheral blood can be used as biomarkers for acute ischemic stroke prediction,diagnosis,and prognosis.In particular,ncRNAs in peripheral immune cells were recently shown to be involved in the peripheral and brain immune response after acute ischemic stroke.In this review,we consolidate the latest progress of research into the roles of ncRNAs(microRNAs,long ncRNAs,and circular RNAs)in the pathological processes of acute ischemic stroke–induced brain damage,as well as the potential of these ncRNAs to act as biomarkers for acute ischemic stroke prediction,diagnosis,and prognosis.Findings from this review will provide novel ideas for the clinical application of ncRNAs in acute ischemic stroke. 展开更多

Flavonoids,abundant in the fruits,are pivotal to their growth,development,and storage.In addition,they have significant beneficial effects on human health.Consequently,research is increasingly concentrating on the regulatory mechanisms governing flavonoid biosynthesis in fruits.Phytohormones are involved in the regulation of flavonoid biosynthesis.The abscisic acid,ethylene,jasmonic acid,cytokinins,and brassinosteroids promote flavonoid biosynthesis,while auxin negatively regulates flavonoid biosynthesis.Subsequently,transcription factors from the MYB,bHLH,WRKY,NAC,and bZIP families are pivotal in regulating flavonoid biosynthesis.In addition,non-coding RNAs(microRNA and lncRNA)also participate in the regulation of flavonoids biosynthesis.MicroRNAs are generally believed to negatively regulate flavonoid metabolism in fruits,while lncRNAs have the opposite effect.Furthermore,the interactions between plant hormones,transcription factors,and non-coding RNAs in fruit flavonoid biosynthesis were analyzed.Ultimately,a foundational regulatory network for fruit flavonoid biosynthesis was hereby established. 展开更多

Matrix metalloproteinases(MMPs)are essential enzymes involved in extracellular matrix degradation and remodeling.Such processes are integral to normal tissue homeostasis and several pathological conditions such as cancer.Among these MMPs,MMP-13 plays a key role in cancer progression,driving tumor invasion,metastasis,and angiogenesis.Despite significant advancements in understanding its biology,therapeutic targeting of MMP-13 remains challenging owing to its complex and multifaceted regulatory mechanisms.Recent studies have underscored the pivotal role of non-coding RNAs(ncRNAs),including long ncRNAs,microRNAs,and circular RNAs,in modulating MMP-13 expression.This review provides a comprehensive analysis of MMP-13 regulation by several signaling pathways,the influence of ncRNAs on these signaling pathways,and MMP-13 expression during cancer progression and metastasis.Furthermore,we explored the clinical relevance of ncRNA-mediated regulatory networks,highlighting their potential as diagnostic biomarkers and therapeutic targets in various cancers.By unraveling these regulatory mechanisms,this review offers valuable insights into innovative strategies for cancer diagnosis and treatment and emphasizes the translational significance of ncRNA-mediated MMP-13 regulation in oncology. 展开更多

A large body of evidence has highlighted the role of non-coding RNAs in neurodevelopment and neuroinflammation.This evidence has led to increasing speculation that non-coding RNAs may be involved in the pathophysiological mechanisms underlying hydrocephalus,one of the most common neurological conditions worldwide.In this review,we first outline the basic concepts and incidence of hydrocephalus along with the limitations of existing treatments for this condition.Then,we outline the definition,classification,and biological role of non-coding RNAs.Subsequently,we analyze the roles of non-coding RNAs in the formation of hydrocephalus in detail.Specifically,we have focused on the potential significance of non-coding RNAs in the pathophysiology of hydrocephalus,including glymphatic pathways,neuroinflammatory processes,and neurological dysplasia,on the basis of the existing evidence.Lastly,we review the potential of non-coding RNAs as biomarkers of hydrocephalus and for the creation of innovative treatments. 展开更多

Gastric cancer(GC)has high morbidity and mortality worldwide.Due to the absence of noticeable symptoms,diagnosing GC at an early stage is very difficult,which consequently leads to advanced GC and poor prognosis.Effective biomarkers are essential for prolonging patients’survival.Helicobacter pylori(H.pylori)infection represents the most significant risk factor for GC,with nearly all cases linked to this infection.Many non-coding RNAs(ncRNAs)are dysregulated in H.pylori-infected GC,indicating that ncRNAs may serve as biomarkers of early-stage GC.In this editorial,we discuss the study by Chen et al.Although previous studies have identified roles for miR-136 in gastric cancer proliferation,apoptosis,and invasion,none have specifically explored its relationship with H.pylori-associated gastric carcinogenesis. 展开更多

Anther is a key male reproductive organ that is essential for the plant life cycle,from the sporophyte to the gametophyte generation.To explore the isoform-level transcriptional landscape of developing anthers in maize(Zea mays L.),we analyzed Iso-Seq data from anthers collected at 10 developmental stages,together with strand-specific RNA-seq,CAGE-seq,and PAS-seq data.Of the 152,026 high-confidence full-length isoforms identified,68.8%have not been described;these include 22,365 isoforms that originate from previously unannotated loci and 82,167 novel isoforms that originate from annotated protein-coding genes.Using our newly developed strategy to detect dynamic expression patterns of isoforms,we identify 13,899 differentially variable regions(DVRs);surprisingly,1275 genes contain more than two DVRs,revealing highly efficient utilization of limited genic regions.We identify 7876 long non-coding RNAs(lncRNAs)from 4098 loci,most of which were preferentially expressed during cell differentiation and meiosis.We also detected 371 long-range interactions involving intergenic lncRNAs(lincRNAs);interestingly,243 were lincRNA-gene ones,and the interacting genes were highly expressed in anthers,suggesting that many potential lncRNA regulators of key genes are required for anther development.This study provides valuable resources and fundamental information for studying the essential transcripts of key genes during anther development. 展开更多

Hepatocellular carcinoma(HCC)remains one of the most prevalent and lethal malignancies worldwide.Long non-coding RNAs(lncRNAs)have emerged as crucial regulators of gene expression and cancer progression,yet the functional diversity of RP11-derived lncRNAs—originally mapped to bacterial artificial chromosome(BAC)clones from the Roswell Park Cancer Institute—has only recently begun to be appreciated.This mini-review aims to systematically synthesize current findings on RP11-derived lncRNAs in HCC,outlining their genomic origins,molecular mechanisms,and biological significance.We highlight their roles in metabolic reprogramming,microRNA network modulation,and tumor progression,as well as their diagnostic and prognostic value in tissue and serum-based analyses.Finally,we discuss therapeutic opportunities and propose future directions to translate RP11-derived lncRNAs into clinically actionable biomarkers and targets for precision liver cancer therapy. 展开更多

目的基于高通量测序技术系统筛选结直肠癌患者血清差异表达微小RNA(microRNA,miRNA),构建多miRNA组合诊断标志物,为结直肠癌早期无创诊断提供新的分子标志物。方法采用病例对照研究设计,纳入结直肠癌患者30例和健康对照者20例,运用Illum... 目的基于高通量测序技术系统筛选结直肠癌患者血清差异表达微小RNA(microRNA,miRNA),构建多miRNA组合诊断标志物,为结直肠癌早期无创诊断提供新的分子标志物。方法采用病例对照研究设计,纳入结直肠癌患者30例和健康对照者20例,运用Illumina HiSeq 2500平台进行高通量小RNA测序,筛选差异表达miRNA(log2 Fold Change,|log2FC|≥1,P<0.05)。通过miRanda和RNAhybrid算法预测靶基因,采用DAVID平台进行基因本体(gene ontology,GO)功能富集和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。运用受试者工作特征曲线(receiver operating characteristic curve,ROC)分析和逻辑回归方法构建最优miRNA组合诊断模型。结果高通量测序共鉴定出205个差异表达miRNA,其中上调98个,下调107个。功能富集分析显示,差异miRNA主要富集于Notch信号通路(P<0.01)、自噬相关通路(P<0.05)、跨膜受体蛋白酪氨酸激酶信号通路(P<0.001)等肿瘤相关生物学过程。成功构建包含miR-21-5p、miR-92a-3p、miR-155-5p、miR-143-3p和miR-145-5p的5-miRNA组合诊断标志物,其ROC曲线下面积达0.924(95%CI:0.867~0.981),诊断敏感性90.00%,特异性85.00%,显著优于传统血清肿瘤标志物癌胚抗原(carcinoembryonic antigen,CEA)(AUC=0.663)和糖类抗原19-9(carbohydrate antigen 19-9,CA19-9)。该标志物对早期结直肠癌(Ⅰ~Ⅱ期)同样具有良好诊断效能(AUC=0.895)。结论基于高通量测序筛选的5-miRNA血清组合标志物对结直肠癌具有优异的诊断效能,特别适用于早期无创筛查,为结直肠癌的精准诊断和临床决策提供了新的分子工具。 展开更多

Atherosclerosis,characterized by the formation of fibrofatty lesions in the arterial wall,remains a leading cause of global morbidity and mortality.Emerging evidence highlights the critical regulatory roles of long non-coding RNAs(lncRNAs)and microRNAs(miRNAs)in atherogenesis.LncRNAs can function as competing endogenous RNAs(ceRNAs)by sponging miRNAs,thereby modulating the expression of downstream target mRNAs.This review summarizes current knowledge on lncRNA-miRNA-mRNA regulatory networks and their functional roles in the three major cell types involved in atherosclerotic plaque development:endothelial cells(ECs),vascular smooth muscle cells(VSMCs),and macrophages.In ECs,these networks are implicated in inflammation,apoptosis,proliferation,angiogenesis,pyroptosis,and autophagy.In VSMCs,they regulate proliferation,apoptosis,and migration.In macrophages,they influence lipid metabolism,inflammatory responses,oxidative stress,and autophagy.Although the ceRNA mechanism is predominant,some lncRNAs also act as primary transcripts for miRNAs.Additionally,exosome-mediated non-coding RNA delivery mediates intercellular crosstalk,further expanding the complexity of RNA-based regulation in atherosclerosis.Despite significant progress,challenges remain due to the complexity and context-specificity of these networks.Further research is essential to elucidate these mechanisms and explore their potential as therapeutic targets for atherosclerosis. 展开更多

目的:探讨过表达环状RNA(circRNAs)修饰牙髓干细胞(DPSCs)来源外泌体(Exo)促进人脐静脉内皮细胞(HUVECs)血管生成在牙髓血管再生中的作用,并阐明相关分子机制。方法:分离乳齿源-、成人智齿源-和老年人恒牙源-原代DPSCs。流式细胞术检测... 目的:探讨过表达环状RNA(circRNAs)修饰牙髓干细胞(DPSCs)来源外泌体(Exo)促进人脐静脉内皮细胞(HUVECs)血管生成在牙髓血管再生中的作用,并阐明相关分子机制。方法:分离乳齿源-、成人智齿源-和老年人恒牙源-原代DPSCs。流式细胞术检测3种原代DPSCs表面标志物蛋白阳性表达情况。使用3种来源的DPSCs与HUVECs建立共培养体系,将细胞分为乳牙源-原代DPSCs组、成人智齿源-原代DPSCs组和老年人恒牙源-原代DPSCs组。另将细胞分为Exo-OE vector组、Exo-circ_0026827 OE组和Exo-circRNA 124534 OE组,分别转染腺病毒介导的OE vector、circ_0026827 OE和circRNA124534 OE后,分离其细胞培养上清液Exo。细胞计数试剂盒8(CCK-8)法检测各组HUVECs增殖活性,实时荧光定量PCR(RT-qPCR)法检测各组Exo中血管生成相关circRNAs表达水平,Western blotting法检测共培养细胞上清中Exo标志物相关蛋白表达水平。验证各组细胞Exo摄取情况,血管生成-诱导实验检测细胞上清液中Exo分离情况。结果:大部分细胞表面标志物表达情况为白细胞分化抗原44(CD44)(+)、白细胞分化抗原34(CD34)(-)和基质细胞抗原1(Stro-1)(+),鉴定为原代DPSCs。CCK-8法检测,与乳牙源-原代DPSCs组比较,成人智齿源-原代DPSCs组和老年人恒牙源-原代DPSCs组共培养的HUVECs增殖活性明显降低(P<0.01);与成人智齿源-原代DPSCs组比较,老年人恒牙源-原代DPSCs组共培养的HUVECs增殖活性明显降低(P<0.01)。3种共培养体系细胞培养上清液Exo中白细胞分化抗原9(CD9)、热激蛋白70(HSP70)和肿瘤易感基因101(TSG101)均表达阳性。3种共培养体系细胞上清液中Exo的粒径为50~110 nm。RT-qPCR法检测,与乳牙源-原代DPSCs组比较,成人智齿源-原代DPSCs组和老年人恒牙源-原代DPSCs组Exo中circRNA124534和circ_0026827 mRNA表达水平均明显降低(P<0.01);与成人智齿源-原代DPSCs组比较,老年人恒牙源-原代DPSCs组Exo中circRNA124534和circ_0026827 mRNA表达水平均明显降低(P<0.01);3组Exo中circ_信号诱导增殖相关基因1(SIPA1L1) mRNA表达水平比较差异无统计学意义(P>0.05)。Western blotting法检测,与Exo-OE vector组比较,Exo-circ_0026827 OE组HUVECs中磷酸化p38丝裂原活化蛋白激酶(p-p38 MAPK)、血管内皮生长因子A(VEGF-A)、血管内皮生长因子受体2(VEGFR2)、血管发生素1(Ang-1)、基质细胞衍生因子1(SDF-1)和基质金属蛋白酶9(MMP-9)蛋白表达水平均明显升高(P<0.01);与Exo-OE vector组比较,Exo-circRNA124534 OE组HUVECs细胞中p-p38MAPK、VEGF-A、VEGFR2、Ang-1、SDF-1和MMP-9蛋白表达水平均明显升高(P<0.01)。各组HUVECs均成功摄取Exo。Exo-OE vector组HUVECs呈现较为平铺的生长状态;与Exo-OE vector组比较,Exo-circ_0026827 OE组和Exo-circRNA124534 OE组HUVECs呈现网状排列的生长状态,并有排列成管状的趋势。结论:过表达circ_0026827和circRNA124534修饰乳牙DPSCs来源的Exo对HUVECs血管生成有一定促进作用。 展开更多

小核仁RNAs(small nucleolarRNAs,snoRNAs)是一类发现较早且位于核仁内的小非编码RNAs,在核糖体RNAs(ribosomalRNAs,rRNAs)、信使RNAs(messengerRNAs,mRNAs)、小核RNAs(small nuclearRNAs,snRNAs)的成熟及修饰中均发挥重要作用。snoRNA... 小核仁RNAs(small nucleolarRNAs,snoRNAs)是一类发现较早且位于核仁内的小非编码RNAs,在核糖体RNAs(ribosomalRNAs,rRNAs)、信使RNAs(messengerRNAs,mRNAs)、小核RNAs(small nuclearRNAs,snRNAs)的成熟及修饰中均发挥重要作用。snoRNAs的功能及其作用途径一直以来均是学术界的研究热点。目前,snoRNAs对rRNAs的化学修饰作用已得到广泛认可。另外,有研究表明snoRNAs与一些遗传疾病以及肿瘤性疾病的发生发展存在密切关联。近年来研究发现,部分snoRNAs经切割可生成更小的、有功能的RNAs,即小核仁RNAs衍生RNAs(snoRNAs derivedRNAs,sdRNAs),这些sdRNAs中部分具有微小RNAs(microRNAs,miRNAs)的特征,可发挥类似miRNA的作用,这一发现极大地拓展了snoRNAs的作用机制方式。结合国内外研究现状,在总结snoRNAs的结构和基本功能的基础上对sdRNAs与miRNAs之间的相关性进行了综述,以期为后续的相关研究提供参考。 展开更多

背景:尽管目前针对脊髓损伤复杂生理病理机制开展了众多研究,但仍缺乏令人满意的治疗方法。研究表明,在脊髓损伤后与氧化应激、炎症、细胞自噬和凋亡等继发性损伤相关的非编码RNAs均有差异表达。因此,非编码RNAs具有作为脊髓损伤后治疗... 背景:尽管目前针对脊髓损伤复杂生理病理机制开展了众多研究,但仍缺乏令人满意的治疗方法。研究表明,在脊髓损伤后与氧化应激、炎症、细胞自噬和凋亡等继发性损伤相关的非编码RNAs均有差异表达。因此,非编码RNAs具有作为脊髓损伤后治疗靶点的潜力。目的:综述非编码RNAs在脊髓损伤后的差异表达、功能作用和调控机制,以期为脊髓损伤的临床治疗提供新思路。方法:以“spinal cord injury”AND“non-coding RNA”OR“microRNA”OR“long non-coding RNA”OR“circRNA”AND“oxidative stress”OR“inflammation”OR“autophagy”OR“apoptosis”为关键词,检索PubMed、Web of Science数据库;以“脊髓损伤”和“非编码RNA”或“微小RNA”或“长链非编码RNA”或“环状RNA”和“氧化应激”或“炎症”或“自噬”或“凋亡”为关键词检索CNKI、万方数据库。依照纳入和排除标准进行筛选,最终对75篇文献进行综述。结果与结论:脊髓损伤后,与氧化应激、炎症、自噬和细胞凋亡等继发性损伤相关的非编码RNAs出现显著差异表达,导致靶基因表达差异和细胞功能发生变化。通过调节非编码RNAs拮抗氧化应激,干预小胶质细胞活化、极化和炎症通路以减轻炎症反应,维持适度水平细胞自噬和保持自噬通量通畅,以及抑制细胞凋亡,可能成为促进脊髓损伤修复的突破点。 展开更多
