目的:报道表现为泛发性毛囊角化性丘疹伴慢性瘙痒的特应性皮炎患者一例,明确其可能的致病基因。方法:收集临床资料,提取先证者及其父母外周血DNA,进行目标基因外显子组测序,并通过Sanger测序验证,结合变异类型、人群数据库、有害性预测... 目的:报道表现为泛发性毛囊角化性丘疹伴慢性瘙痒的特应性皮炎患者一例,明确其可能的致病基因。方法:收集临床资料,提取先证者及其父母外周血DNA,进行目标基因外显子组测序,并通过Sanger测序验证,结合变异类型、人群数据库、有害性预测结果和既往文献评估其致病性。结果:先证者,男,13岁,全身泛发性毛囊角化性丘疹、干燥伴瘙痒10年。患者外周嗜酸性粒细胞及IgE水平升高,外院筛查过敏原示麦麸过敏,有过敏性鼻炎、面食过敏史。家族中无类似疾病。检测到患儿CARD11基因存在c.89G>A(p.Arg30Gln)杂合变异,经Sanger测序验证共分离后提示为自发变异。患儿父母未检测到CARD11基因突变。结论:本研究报道了一例以泛发性毛囊丘疹为临床特征的特应性皮炎患者,丰富了CARD11变异的临床表型谱。 展开更多

Microglia are the first immune cells that are activated in the brain following ischemic stroke.Mitochondrial dysfunction exacerbates microglia-mediated neuroinflammation post-stroke.Caspase activation and recruitment domain 19(CARD19)is involved in innate immune response and inflammatory response,which are also important functions of microglia.However,the role of CARD19 in microglial biology and ischemic stroke remains unknown.Here,we observed that CARD19 expression was significantly elevated in microglia in the penumbra after ischemic stroke via analyzing the spatial transcriptomic sequencing data of ischemic brain tissue,as well as in an in vitro model of microglial activation.Remarkably,conditional knockdown of Card19 in microglia promoted post-stroke neuroinflammation and worsened neurological outcomes in a mouse model of ischemic stroke.Mechanistically,we found that CARD19 localized to mitochondria and promoted the assembly of mitochondrial intermembrane bridge components,while CARD19 deficiency in microglia caused ultrastructural and functional damage to the mitochondrial cristae,leading to an exaggerated pro-inflammatory response.Thus,our findings suggest that preserving mitochondrial cristae,by targeting CARD19 could be a novel therapeutic strategy for ameliorating neuroinflammation post-stroke and decreasing the volume of the ischemic penumbra. 展开更多

Data analytics to prevent retail credit card fraud: empirical evidence from Latin America

Reducing the risk of fraud in credit card transactions is crucial for the competitiveness of companies,especially in Latin American countries.This study aims to establish measures for preventing and detecting fraud in the use of credit cards in shops through analytical methods(data mining,machine learning and artificial intelligence).To achieve this objective,the study employs a predictive methodology using descriptive and exploratory statistics and frequency,frequency&monetary(RFM)classification techniques,differentiating between SMEs and large businesses via cluster analysis and supervised models.A dataset of 221,292 card records from a Latin American merchant payment gateway for the year 2022 is used.For fraud alerts,the classification model has been selected for small and medium–sized merchants,and the multilayer perceptron(MLP)neural network has been selected for large merchants.Random forest or Gini decision tree models have been selected as backup models for retraining.For the detection of punctual fraud patterns,the K-means and partitioning around medoids(PAM)models have been selected,depending on the type of trade.The results revealed that the application of the identified models would have prevented between 48 and 85%of fraud transactions,depending on the trade size.Despite the promising results,continuous updating is recommended,as fraudsters frequently implement new fraud techniques. 展开更多

Credit Card Fraud Detection Method Based on RF-WGAN-TCN

Credit card fraud is one of the primary sources of operational risk in banks,and accurate prediction of fraudulent credit card transactions is essential to minimize banks’economic losses.Two key issues are faced in credit card fraud detection research,i.e.,data category imbalance and data drift.However,the oversampling algorithm used in current research suffers from excessive noise,and the Long Short-Term Memory Network(LSTM)based temporal model suffers from gradient dispersion,which can lead to loss of model performance.To address the above problems,a credit card fraud detection method based on Random Forest-Wasserstein Generative Adversarial NetworkTemporal Convolutional Network(RF-WGAN-TCN)is proposed.First,the credit card data is preprocessed,the feature importance scores are calculated by Random Forest(RF),the features with lower importance are eliminated,and then the remaining features are standardized.Second,the Wasserstein Distance Improvement Generative Adversarial Network(GAN)is introduced to construct the Wasserstein Generative Adversarial Network(WGAN),the preprocessed data is input into the WGAN,and under the mutual game training of generator and discriminator,the fraud samples that meet the target distribution are obtained.Finally,the temporal convolutional network(TCN)is utilized to extract the long-time dependencies,and the classification results are output through the Softmax layer.Experimental results on the European cardholder dataset show that the method proposed in the paper achieves 91.96%,98.22%,and 81.95%in F1-Score,Area Under Curve(AUC),and Area Under the Precision-Recall Curve(AUPRC)metrics,respectively,and has higher prediction accuracy and classification performance compared with existing mainstream methods. 展开更多

The task of detecting fraud in credit card transactions is crucial to ensure the security and stability of a financial system,as well as to enforce customer confidence in digital payment systems.Historically,credit card companies have used rulebased approaches to detect fraudulent transactions,but these have proven inadequate due to the complexity of fraud strategies and have been replaced by much more powerful solutions based on machine learning or deep learning algorithms.Despite significant progress,the current approaches to fraud detection suffer from a number of limitations:for example,it is unclear whether some transaction features are more effective than others in discriminating fraudulent transactions,and they often neglect possible correlations among transactions,even though they could reveal illicit behaviour.In this paper,we propose a novel credit card fraud detection(CCFD)method based on a transaction behaviour-based hierarchical gated network.First,we introduce a feature-oriented extraction module capable of identifying key features from original transactions,and such analysis is effective in revealing the behavioural characteristics of fraudsters.Second,we design a transaction-oriented extraction module capable of capturing the correlation between users’historical and current transactional behaviour.Such information is crucial for revealing users’sequential behaviour patterns.Our approach,called transactional-behaviour-based hierarchical gated network model(TbHGN),extracts two types of new transactional features,which are then combined in a feature interaction module to learn the final transactional representations used for CCFD.We have conducted extensive experiments on a real-world credit card transaction dataset with an increase in average F1 between 1.42%and 6.53%and an improvement in average AUC between 0.63%and 2.78%over the state of the art. 展开更多

目的:探讨秋水仙碱对动脉粥样硬化(AS)小鼠主动脉斑块的干预作用,并分析其对炎症反应、核苷酸结合寡聚化结构域样受体3(NLRP3)/含CARD结构域的凋亡相关颗粒样蛋白(ASC)/含半胱氨酸的天冬氨酸蛋白酶1(Caspase-1)信号通路的影响。方法:12... 目的:探讨秋水仙碱对动脉粥样硬化(AS)小鼠主动脉斑块的干预作用,并分析其对炎症反应、核苷酸结合寡聚化结构域样受体3(NLRP3)/含CARD结构域的凋亡相关颗粒样蛋白(ASC)/含半胱氨酸的天冬氨酸蛋白酶1(Caspase-1)信号通路的影响。方法:12只正常小鼠作为空白对照组,48只同品系小鼠随机分为模型组、秋水仙碱-L组、秋水仙碱-M组、秋水仙碱-H组,每组各12只。比较各组主动脉斑块病理改变、主动脉斑块病变程度、非酒精性脂肪性肝病活动度评分(NAS)、附睾脂肪横截面面积及血脂、血糖、动脉粥样硬化指数(AI)、炎症指标、氧化应激指标、NLRP3/ASC/Caspase-1信号通路(NLRP3 mRNA、ASC mRNA、Caspase-1 mRNA及蛋白)水平。结果:与模型组比较,秋水仙碱-L组、秋水仙碱-M组、秋水仙碱-H组主动脉油红面积、主动脉窦病变面积、主动脉斑块面积占比、主动脉红染脂质面积占比、肝脏NAS评分及附睾脂肪横截面面积降低,且呈剂量依赖性(均P<0.05);与模型组比较,秋水仙碱-L组、秋水仙碱-M组、秋水仙碱-H组血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、空腹血糖(FPG)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、单核细胞趋化蛋白-1(MCP-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)、高敏C反应蛋白(hs-CRP)、丙二醛(MDA)、氧化修饰低密度脂蛋白(ox-LDL)水平及胰岛素抵抗指数(HOMA-IR)、AI降低,血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-px)水平升高,且呈剂量依赖性(均P<0.05);与模型组比较,秋水仙碱-L组、秋水仙碱-M组、秋水仙碱-H组NLRP3 mRNA、ASC mRNA、Caspase-1 mRNA及蛋白水平降低,且呈剂量依赖性(均P<0.05)。结论:秋水仙碱对AS小鼠主动脉斑块具有良好的干预作用,可改善AS病变程度,降低血脂、血糖水平,减轻血管炎症反应,缓解氧化应激反应,其作用机制可能与抑制NLRP3/ASC/Caspase-1信号通路活化有关。 展开更多

深度学习法在游梁式抽油机示功图诊断中发挥着重要作用,但实际应用中,不同故障类型的示功图样本在数量、多样性和平衡性方面存在不足,影响现有诊断模型的性能。为此,提出了一种基于卷积变分自编码器(CVAE)的示功图样本扩展方法。将CVAE... 深度学习法在游梁式抽油机示功图诊断中发挥着重要作用,但实际应用中,不同故障类型的示功图样本在数量、多样性和平衡性方面存在不足,影响现有诊断模型的性能。为此,提出了一种基于卷积变分自编码器(CVAE)的示功图样本扩展方法。将CVAE模型编码器和解码器设计为浅层卷积神经网络,作为示功图样本生成模型;原始示功图样本归一化和二值化处理后,提取图像的像素值作为模型输入。由CVAE模型单独学习每类示功图的形状特征,以生成与原始数据集相似的新样本;将生成的新样本与原始数据集结合,构建扩展数据集;利用诊断模型对扩展数据集进行训练,以验证生成样本的有效性,并分析样本扩展后改善类不平衡性对模型诊断性能的影响。研究结果表明:采用该方法扩展后的数据集显著提升了诊断模型的性能,其宏平均分数(F_(macro))达到98.66%,优于随机复制和全连接变分自编码器扩展数据集的结果。该方法为示功图故障诊断提供了样本数量、样本多样性和平衡性兼备的深度学习数据,显著提升了判断的准确性,可为智慧油田建设提供技术支撑。 展开更多

目的比较紫外线辐射剂量指示卡与照度计在评价紫外线杀灭医疗环境物体表面微生物效果方面的一致性,为医疗环境紫外线消毒效果快速评价提供依据。方法选择病房内高频接触物体表面试验点,分别放置照度计、剂量指示卡和大肠埃希菌、金黄色... 目的比较紫外线辐射剂量指示卡与照度计在评价紫外线杀灭医疗环境物体表面微生物效果方面的一致性,为医疗环境紫外线消毒效果快速评价提供依据。方法选择病房内高频接触物体表面试验点,分别放置照度计、剂量指示卡和大肠埃希菌、金黄色葡萄球菌、枯草杆菌黑色变种芽孢3种菌片。开启紫外线消毒灯作用30 min,记录照度计读数和剂量指示卡颜色变化,并收集菌片进行菌落培养计数。结果60个紫外线辐射剂量指示卡全部变色合格。剂量指示卡与照度计显示的紫外线累计辐射剂量具有一致性,且主要数据与3种指示微生物平均杀灭对数值均呈显著正相关性(P<0.001)。结论紫外线辐射剂量指示卡评估医疗环境条件下的紫外线消毒效果,使用方便且结果较可靠。 展开更多

目的应用生物学信息分析软件预测结核分枝杆菌CarD蛋白的结构和功能。方法运用NCBI中Blast工具,将结核分枝杆菌H37Rv carD基因序列与GenBank中相似序列进行核苷酸比对;利用MEGA 6.06软件,采用邻位相连法构建N-J进化树;运用ClustalX 2.1... 目的应用生物学信息分析软件预测结核分枝杆菌CarD蛋白的结构和功能。方法运用NCBI中Blast工具,将结核分枝杆菌H37Rv carD基因序列与GenBank中相似序列进行核苷酸比对;利用MEGA 6.06软件,采用邻位相连法构建N-J进化树;运用ClustalX 2.1软件,将结核分枝杆菌H37Rv CarD及与其同源性较高的分枝杆菌CarD氨基酸序列进行多重比对分析;登录ExPASy网站,运用ProtParam工具分析CarD蛋白的理化性质,运用ProtScale进行蛋白质疏水性分析,运用SignalP 4.1Server预测蛋白质信号肽,运用TMHMM Server v.2.0以及TMPred预测蛋白质跨膜螺旋区,运用SOPMA预测蛋白质二级结构,运用SWISS-MODEL进行蛋白质三级结构同源建模;运用NetPhos 3.1Server预测蛋白质磷酸化位点;运用NCBI中CDD工具预测CarD蛋白保守结构域。结果分枝杆菌carD基因序列相似度为100%,进化关系较近。H37Rv与田鼠分枝杆菌起源于同一物种,同源性较高。CarD分子在进化过程中高度保守,为稳定、亲水性蛋白,无跨膜区、无信号肽,蛋白序列中存在4个丝氨酸磷酸化位点,二级结构中以α-螺旋为主,三级结构同源建模成功。CarD蛋白是一种结合RNA聚合酶的转录调控因子,属于CarD/CdnL/TRCF家族。结论 CarD蛋白为RNA聚合酶结合蛋白,调控转录和控制结核分枝杆菌的生长。该蛋白序列保守稳定,是治疗结核病潜在的新靶标。 展开更多
