Rauvolfia serpentina(L.)Benth.Ex Kurz is a greatly appreciated medicinal plant,well-known for its therapeutic benefits in traditional medicine,particularly in Ayurveda,where the roots and whole plant are used to treat...Rauvolfia serpentina(L.)Benth.Ex Kurz is a greatly appreciated medicinal plant,well-known for its therapeutic benefits in traditional medicine,particularly in Ayurveda,where the roots and whole plant are used to treat a variety of ailments.However,studies focusing on R.serpentina seeds are relatively scarce.Hence,the present study provides a novel approach by analysing the seed oil of R.serpentina extracted using the supercritical-carbon dioxide-fluid-extraction(SCFE)technique.The research employed advanced analytical methods including gas-chromatography with flame ionization detector(GC-FID),gas-chromatography-tandem mass spectrometry(GC-MS/MS),and high performance thin layer chromatography(HPTLC)to characterise the chemical composition of the extracted oil.Functional moieties were evaluated by Fourier transform infrared spectroscopy(FT-IR),while proton nuclear-magnetic-resonance(1H NMR)spectroscopy was utilised to identify the phytometabolites as well as to assess the physico-chemical parameters.The anti-microbial potential of the supercritically extracted oil was demonstrated through its activity against Klebsiella pneumoniae.The inhibitory effects on K.pneumoniae were quantified using the broth microdilution method,showing activity at both minimum inhibitory concentrations(MIC50 and MIC90).Furthermore,the oil was found to be non-genotoxic,as demonstrated by the Ames assay,which showed no mutagenic effects against S.typhimurium and E.coli WP2 uvrA.Since previous reports on R.serpentina seeds and their novel contribution in the field of pharmaceutics are rather limited,the present study is of utmost importance.The study may pave the way for future investigations into the therapeutic potentials of R.serpentina seeds.展开更多

Objective:To assess the in-vitro antihacterial activity and anti-inflammatory activity of orally administered different extracts(Hydro-alcoholic,methanolic,ethyl acetate and hexane)of Rauvolfia tetraphylla(R.tetraphyl...Objective:To assess the in-vitro antihacterial activity and anti-inflammatory activity of orally administered different extracts(Hydro-alcoholic,methanolic,ethyl acetate and hexane)of Rauvolfia tetraphylla(R.tetraphylla)root bark in Carrageetiaii induced acute inflammation in rats.Methods:In-vitro antibacterial activity was evaluated for extracts against four Gram positive and four Gram negative bacteria by using cylinder plate assay.Hydro-alcoholic extract(70%v/v ethanol)at 200,400 and 800 mg/kg doses and methanolic,ethyl acetate and hexane extracts at doses 100,200 and 400 mg/kg were tested for anti-inflammatory activity in Carrageenan induced rat paw oedema model and paw thickness was measured every one hour up to 6 hrs.Results:All extracts of R.tetraphylla root bark showed good zone of inhibition against tested bacterial strains.In Carrageenan induced inflammation model,hydro-alcoholic and methanolic extract of R.tetraphylla root bark at three different doses produced significant(P<0.00l)reduction when compared to vehicle treated control group and hexane,ethyl acetate extracts.Conclusions:In the present study extracts of R.tetraphylla root bark shows good in-vitro antibacterial activity and in-vivo anti-inflammatory activity in rats.展开更多
Copper oxide nanoparticles (CuO Nps) were successfully synthesized by solution combustion method using aqueous leaf extract of Rauvolfia serpentina as a fuel. The structure and morphology of the CuO nanoparticles (...Copper oxide nanoparticles (CuO Nps) were successfully synthesized by solution combustion method using aqueous leaf extract of Rauvolfia serpentina as a fuel. The structure and morphology of the CuO nanoparticles (Nps) were characterized by powder X-ray diffraction (PXRD), UV-visible spectroscopy (UV-visible), scanning electron microscopy (SEM), transmission electron microscopy (TEM), etc. The PXRD patterns reveal the formation of monoclinic phase with crystallite structure. SEM images indicate that the particles have sponge-like structure being highly porous and agglom- erated with large surface area. The average crystallite sizes were found to be in the range of 10-20 nm by Scherrer's method. The CuO Nps size was further confirmed by TEM. Further, CuO Nps exhibit good photocatalytic activity for the photodegradation of trypan blue dye, indicating that it acts as a promising semiconducting material. The antibacterial properties of CuO nanoparticles were investigated against pathogenic bacterial strains, namely Gram -ve Escherichia coli (NCIM-5051) and Pseudomonas desmolyticum (NCIM-2028) and Gram +ve bacteria Staphylococcus aureus (NCIM- 5022) using the agar well diffusion method.展开更多

[Method] The full-length cDNA of SGD was cloned from R. verticillata with RA...[Objective] This study aimed to clone Scrictosidine-β-D-glucosidase (SGD) gene from Rauvolfia verticillata and analyze its characteristics. [Method] The full-length cDNA of SGD was cloned from R. verticillata with RACE technique. Then the expression levels in different tissues were analyzed with quantitative RT-PCR and the bioinformatic characteristics were also predicted. [Result] The full-length cDNA of RvSGD was 1 856 bp, containing a 1 608 bp CDS that encoded 536 amino acids with a calculated molecular mass of 61.0 kDa and an isoelectric point of 6.16. Bioinformatic analysis revealed that RvSGD shared high similarity with SGDs from Cantharanthus roseus and Rauvolfia serpentina at the amino acids; three conserved catalytic sites His-161, Glu-207 and Glu-419 were also presented in RvSGD. Quantitative RT-PCR showed that expression level of RvSGD was the highest in barks, followed by old leaves, roots, tender leaves and tender stems. [Conclusion] The present study helps to understand more about the functions of the SGD gene at the level of molecular genetics, and provides new targets for molecular regulation of TIAs biosynthesis.展开更多

Baill. var. hainanensis Tsiang extract on mice with colitis induced by Dextran sulphate sodium (DSS). Methods: 80 BALB/C mice were randomly divided into (1) normal control group, (2) DSS group, (3) DSS + SASP treatment group, (4) DSS + PP treatment group. After the experiment, the mice in each group were observed daily general conditions, Immunohistochemistry was used to detect the expression of ERK, p38 MAPK and JNK protein in colonic mucosa of mice in each group. The expression of TNF-_and IL-6 cytokines in colonic tissue of mice was detected by ELASA. Results: The general situation of SASP group and PP group was better than DSS group;Compared with DSS group, the expression of ERK, p38 MAPK and JNK protein was significantly decreased. The levels of TNF-a and IL-6 in colon tissue of SASP group and PP group were significantly decreased. Conclusion: The results suggest that pectin polysaccharide from Rauvolfia hainanensis extract has a significant therapeutic effect on colitis in DSS mice, which may via regulating the MAPK signal pathway pathways.展开更多

文摘目的:探究萝芙木在溃疡性结肠炎中的作用及潜在分子机制。方法:利用HERB数据库和相关文献对萝芙木的活性成分进行筛选,并确定其潜在靶点。从GeneCards数据库获得与溃疡性结肠炎相关的靶点。随后构建成分-靶点网络,并通过Gene Ontology(GO)和Kyoto Encyclopedia of Genes and Genomes(KEGG)通路分析探究萝芙木治疗溃疡性结肠炎的潜在机制。此外,利用葡聚糖硫酸钠(dextran sulfate sodium,DSS)诱导的溃疡性结肠炎小鼠模型评估萝芙木治疗溃疡性结肠炎的疗效和机制。结果:本研究共筛选出26个萝芙木治疗溃疡性结肠炎的潜在靶点。成分-靶点网络分析表明,果胶可能是治疗溃疡性结肠炎的关键成分。功能富集分析显示萝芙木治疗溃疡性结肠炎与JAK激酶(Janus kinase,JAK)-信号传导及转录激活蛋白(signal transducer and activator of transcription,STAT)信号通路密切相关。体内实验结果表明,萝芙木果胶可以减轻DSS诱导的溃疡性结肠炎小鼠的临床症状(如疾病活动指数、结肠长度和病理变化),并降低肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素1β(interleukin-1β,IL-1β)和白细胞介素6(interleukin-6,IL-6)的表达水平(P<0.05)。此外,萝芙木果胶可以使JAK-STAT信号通路中的JAK2和STAT3表达下调(P<0.05)。结论:萝芙木尤其是其果胶成分可能通过调节JAK-STAT信号通路对溃疡性结肠炎产生治疗作用,本研究旨在为萝芙木在溃疡性结肠炎中的应用提供理论基础。
文摘采用固相萃取-电导抑制离子色谱法测定了萝芙木中常见有机酸和无机阴离子。以Ionpac AS11为分离柱,Ionpac AG11为保护柱,萝芙木样品过IC SPE-C18前处理柱去除杂质和有机物,在确定的色谱条件下,柱温30℃,淋洗液为7.5 mmol/L Na OH溶液,流速为0.7 m L/min,电导检测器-离子色谱法直接测定样品,方法的相关系数在0.9987~0.9999之间,样品回收率在83.0%~100.8%之间,RSD〈5%。方法适用于中草药中常见有机酸和无机阴离子含量的测定。
