Quince(Cydonia oblonga)leaves are traditionally reported to alleviate the risk of cardiovascular disease.The study envisaged to explore the in vitro and in vivo phytochemical and cardio protective potential of quince extract respectively.Different solvent extracts(Aqueous,Ethanolic,Hydroethanolic,Methanolic)of quince leaf obtained via two different extraction methods(Ultrasonication and Reflux)were evaluated for the phytochemical and antioxidant analysis.Among all the extracts,quince ethanolic extract,extracted via sonication was found to have the maximum phenolics(39.89±0.67 mg GAE/g DW),flavonoids(21.51±1.0 mgRE/g DW)and an anti-oxidant potential 2,2-diphenyl-1-picryl-hydrazyl radical(DPPH)with IC_(50) value of 168.88±2.24μg/ml.The High-performance thin layer chromatograph(HPTLC)and ultrahigh performance liquid chromatography mass spectrometry(UPLC-MS)analysis confirmed the presence of bio active compounds.Furthermore,the in vivo activity was ascertained against Doxorubicin(DOX)induced cardio toxicity in rats fed orally with the extract at 160 and 320 mg/kg body weight.The pre-treated groups significantly assuaged the changes induced by DOX in the electrocardiogram(ECG).Additionally,it mitigated the elevation in the blood serum parameters;aspartate transaminase(AST),lactate dehydrogenase(LDH),creatinine kinase-MB(CK-MB)and also repressed glutathione(GSH)depletion and rise in malondialdehyde(MDA)level induced by DOX.Furthermore,improvement in histopathological changes of the heart tissue concomitant to DOX induced toxicity was also observed such as,reduced degeneration,necrosis,inflammation and apoptosis.Thus,it could be concluded that cardioprotective activity of quince extract accredited to the occurrence of phytoconstituents prevented the DOX induced cardiotoxicity and helped to reinstate the cardiac damage in rats. 展开更多

Inhibitory effect of a Chinese quince seed peptide on protein glycation:A mechanism study

Non-enzymatic glycation can cause the formation and accumulation of advanced glycation end products(AGEs),and it poses great threats to human health.It is urgent to search for safe and efficient inhibitors to prevent reducing sugar induced protein glycation.In this study,we inves-tigated the anti-glycation activity and mechanism of an identified peptide,Asparagine-Tyrosine-Arginine-Arginine-Glutamic acid(NYRRE)from Chinese quince seed protein hydrolysate,by mul-tispectroscopy,confocal imaging,and computational molecular simulation.Firstly,it was found that NYRRE could scavenge hydroxyl radicals and chelate Fe 2+.Besides,the NYRRE was effective in every stage of fructose induced bovine serum albumin(BSA)glycation.The NYRRE could re-duce the formation of fructosamine,carbonyl compounds,glycoxidation products and𝛽-amyloid structure.Meanwhile,NYRRE could protect thiol groups and stabilize the spatial conformation of BSA.The NYRRE presented strong inhibition in fluorescent AGEs,and 68.19%of total AGEs formation was prevented with NYRRE at 15 mmol/L.The results of molecular simulation indi-cated that NYRRE could insert into the hydrophobic pocket of BSA and interact with hot spots,including arginine and lysine residues.The mechanism of NYRRE inhibiting protein glycation could be due to its antioxidant activity,BSA structure stabilizing ability,and specific bond with glycation sites of BSA.These results provided a valuable reference for developing NYRRE as an efficient antiglycation agent in preventing glycation-mediated diseases. 展开更多

Edible coatings offer many advantages in cheese coating for prolonging the shelf life and improving quality properties.Mould spoilage that occurs during ripening is one of the most important issues in Kas¸ar cheese.The current study focused on the effects of an edible coating solution based on quince seed mucilage(QSM)containing thyme oil(TO)in different ratios,on the surface moulding of Kas¸ar cheese and evaluated the physicochemical,microbiological,textural,and proteolysis characteristics of Kas¸ar cheese throughout the 120 days of ripening.Also,the effect of edible coating on the in-vitro Angiotensin-converting enzyme(ACE)-inhibitory activities of pH 4.6-soluble extracts of Kas¸ar cheeses was investigated.Results showed that the coating of Kas¸ar cheese with QSM and the increased levels of thyme oil significantly suppress mould growth throughout the ripening.Until 120^(th) days of ripening,counts of yeasts and moulds were lower<2 log cfu g^(-1)in coated cheese sample with QSM containing 2%TO.Coating with QSM caused an increase in moisture loss compared to vacuum-packed cheeses,however,didn’t change the normal ripening conditions such as microbiological,chemical,textural,and proteolysis properties of Kas¸ar cheese.As the ripening progressed,ACE-inhibitory activity increased in all samples,with the highest inhibition in vacuum-packed and coated cheeses.Overall,it was determined that the QSM coating showed good potential as a coating material for the protection of cheese shelf life and quality as well as for enhancing ACE-inhibitory activity. 展开更多

In this study,phytochemically characterized extracts from the fruits of Chaenomeles japonica(CJ),Cornus mas(CM),and Hippophaёrhamnoides(HR)were investigated using an interdisciplinary approach for their effects on human gut microbiome(GM)composition and selected health parameters in a preclinical model organism,Caenorhabditis elegans.Initially,due to the availability of proteomics data of the probiotic bacterium Bacillus subtilis,analysis was conducted in silico to investigate the impact of phytochemicals from CJ,CM,and HR on its nucleases.Next,the extracts were incubated with human fecal slurry(FS)for 24 and 72 h,and the isolated genomic DNA was analyzed using next-generation sequencing(NGS)of bacterial 16S rRNA genes.These data revealed no significant alterations in GM between the different extracts.In the GM analysis,B.subtilis was not detected,which may be related to the interaction of its enzymes with the phytochemicals.Principal coordinate analysis showed that the extract type influences community composition,particularly after prolonged time.A further focus was on survival,fat accumulation,and Aβproteotoxicity in extract-treated nematodes.All extracts were well tolerated up to the highest tested concentration(200μg/mL).Notably,the CM extract significantly increased nematode survival by 23%at 200μg/mL,while HR demonstrated the most pronounced effect on fat reduction,markedly decreasing lipid accumulation in genetically modified SS104 worms at the same concen-tration.These findings suggest that while the extracts do not substantially modulate GM composition in vitro,they exert distinct beneficial effects on metabolic and longevity-associated parameters in C.elegans. 展开更多
