耐甲氧西林金黄色葡萄球菌基因组测序及putP基因克隆

[目的]对耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)ATCC43300进行基因组测序,克隆钠/脯氨酸同向转运蛋白(Sodium/proline symporter,PutP)基因,构建细菌双杂交诱饵重组质粒pBT-PutP,对PutP蛋白进行... [目的]对耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)ATCC43300进行基因组测序,克隆钠/脯氨酸同向转运蛋白(Sodium/proline symporter,PutP)基因,构建细菌双杂交诱饵重组质粒pBT-PutP,对PutP蛋白进行生物信息学分析。[方法]采用CTAB法提取基因组DNA,PacBio SequelⅡ进行全基因组测序,使用SMRTLINK软件进行数据处理,flye软件进行组装,并使用Circos软件绘制MRSA基因组圈图。利用各种数据库及软件进行基因组结构注释、基因组基本功能数据库注释和基因组特定功能注释。PCR扩增putP基因,构建pBT-PutP诱饵重组质粒,对PutP蛋白利用生物信息方法进行初步分析。[结果]耐甲氧西林金黄色葡萄球菌ATCC43300基因组全长2946753 bp,基因数量2750个,基因平均长度892.01 bp,基因区域GC%含量33.39%,含反义RNA(antisense RNA)2个,RNA抗毒素4个,小RNA(small non-coding RNA,sRNA)89个,19个CRISPR序列,12个基因岛,以及2个前噬菌体结构。成功构建了pBT-PutP重组质粒。[结论]成功对耐甲氧西林金黄色葡萄球菌ATCC43300进行了全基因组测序,并系统性地注释了基因组序列,成功构建细菌双杂交诱饵重组质粒pBT-PutP,为后续开发靶向PutP的特异肽适体奠定前期基础。 展开更多

基于微阵列数据预测乳腺癌生物标志物及其靶向中药

[目的]基于微阵列数据挖掘乳腺癌的生物标志物,筛选其靶向中药。[方法]下载乳腺癌的临床微阵列数据以筛选差异表达基因,构建基因共表达网络,并基于12种算法筛选核心基因。通过卡普兰迈尔(Kaplan-Meier)和比例风险回归模型(Cox proportional hazards model)筛选预后基因,构建预后模型并挖掘相关微RNA(miRNAs)。最后,预测其靶向中药及小分子,并通过分子建模和分子对接验证其结合力。[结果]共获得464个差异表达基因,包括50个核心基因。其中,酰基辅酶A合成酶长链家族成员1(Acyl-CoA Synthetase Long Chain Family Member 1,ACSL1),CD24分子(CD24 Molecule,CD24),谷氨酰氧乙酸转氨酶2(Glutamic-Oxaloacetic Transaminase 2,GOT2)和角蛋白(Keratin 14,KRT14)可被has-miR-373等20个microRNAs调控,是乳腺癌的预后标志物。大青叶、麻黄、雷公藤等35种中药所包含的汉防己甲素等13种成分是上述标志物的靶向药物,且它们的结合能均小于-4.0 kcal/mol。[结论]ACSL1[log-rank P=0.039,风险比=1.4(95%CI,1.02-1.94)],CD24[log-rank P=0,风险比=1.98(95%CI,1.42-2.76)],GOT2[log-rank P=0.046,风险比=1.38(95%CI,1.01-1.91)]和KRT14[log-rank P=0.003,风险比=0.612(95%CI,0.444-0.843)]是乳腺癌的潜在生物标志物,且汉防己甲素等13种成分是其潜在的靶向药物(结合能均小于-4.0 kcal/mol),这为乳腺癌的诊疗提供了新思路。 展开更多

Several low and high affinity osmolyte transport systems have been identified in various bacterial species. A lot of research has centered on characterizing the osmolyte transport systems of Gram-negative bacteria, but less has been done to characterize the same transport systems in Gram-positive bacteria. This review will focus on the previous work that has been done to understand the osmolyte transport systems in the species Staphylococcus aureus and how these transporters may serve dual functions in allowing the bacteria to survive and grow in a variety of environments, including on the surface or within humans or other animals. 展开更多

Variable structure control for descriptor Markovian jump systems subject to partially unknown transition probabilities

The descriptor Markovian jump systems( DMJSs)with partially unknown transition probabilities( PUTPs) are studied by means of variable structure control. First,by virtue of the strictly linear matrix inequality( LMI) technique,a sufficient condition is presented, under which the DMJSs subject to PUTPs are stochastically admissible. Secondly,a novel sliding surface function based on the system state and input is constructed for DMJSs subject to PUTPs; and a dynamic sliding mode controller is synthesized, which guarantees that state trajectories will reach the pre-specified sliding surface in finite time despite uncertainties and disturbances. The results indicate that by checking the feasibility of a series of LMIs,the stochastic admissibility of the overall closed loop system is determined. Finally,the validity of the theoretical results is illustrated with the example of the direct-current motor. Furthermore,compared with the existing literature,the state convergence rate,buffeting reduction and overshoot reduction are obviously optimized. 展开更多
