The test were conducted from May 15 to December 24 in 2013 using six-year-old ponkan (C. blanco cv. Ponkan) trees with 40% soil water conditions by taken regular watering as control. The content of acids in fruit were determined by HPLC, and relative expression of related genes of citric acid metabolic were determined by relative fluorescence quantitative PCR. The results showed that the content of citric acid, malic acid, quinic acid and total organic acids per gram sarcocarp were extremely increased by 285.2%, 320%, 480% and 299.1%, and the content of per-fruit organic acid were 77.39%, 89.64%, 117.24% and 75.9% respectively compared to those control in the fruit mature stage. Relative expression of CitCS1, CitCS2 were higher than control, and relative expression of CitAco1, CitAco2, CitAco3 had a certain increase in the late fruit development, were lower in mature stage. Three relative expression of CitIDH gene were higher than control in mature stage. Low CitGAD4 relative expression and undetectable in mature stage, the relative expression of CitGAD5 gene had a role in promoting under water stress. Furthermore, the relative expression of CitCS1, CitCS2, CitACO1, CitACO3, CitIDH1, CitIDH2, CitIDH3, CitGAD4 and CitGAD5 were influenced by water stress through the correlation analysis. Water stress caused the accumulation of citric acid, declined fruit quality, leaded to change of the genetic rela- tive expression about citric acid synthesis and degradation. The down-regulation of CitACO1, CitGAD4 and up-regulation of CitCS1, CitCS2 might be one of the reasons that promoted to the accumulation of citric acid.展开更多
Ponkan(Citrus reticulate Blanco)is one of the most popular citrus fruits for daily consumption and processing in China.In this study,limonene was removed from ponkan essential oil(PEO)by using an effective methodology...Ponkan(Citrus reticulate Blanco)is one of the most popular citrus fruits for daily consumption and processing in China.In this study,limonene was removed from ponkan essential oil(PEO)by using an effective methodology to estimate the optimum levels of three independent variables of molecular distillation(MD)in order to minimize the limonene content and(b)maximize the removal rate.A three-factor Box-Behnken design(BBD)was employed to evaluate the effect of distilling temperature(45.00~55.00℃),wiper rolling speed(150.00~250.00 r/min)and distilling pressure(25.00~35.00 Pa).The optimum parameters within the experimental range of the variables were extrapolated to be:distilling temperature 47.00℃,wiper rolling speed 203.00 r/min and distilling pressure 30.00 Pa.Optimal values extrapolated by response surface methodology(RSM)for the limonene content and removal rate of the desirable PEO were 47.82%and 84.67%,respectively.展开更多

Oral ulcers,a common mucosal affliction,severely diminish patients’quality of life due to pain and discomfort.Despite existing treatments,the frequent recurrence of oral ulcers underscores the need for more effective...Oral ulcers,a common mucosal affliction,severely diminish patients’quality of life due to pain and discomfort.Despite existing treatments,the frequent recurrence of oral ulcers underscores the need for more effective therapeutic options.This study aimed to investigate the inhibitory effects of Ponkan(Citrus reticulata‘Ponkan’)extracts and their flavonoid components on oral ulcers,exploring their potential as a natural therapy.Twenty different Ponkan cultivars were selected for flavonoid extraction and analysis.Ultra-performance liquid chromatography(UPLC)and high-resolution mass spectrometry(HRMS)were utilized for substance identification,leading to the discovery of 16 flavonoid compounds.Using an oral ulcer model established in Wistar rats,Ponkan extracts were found to effectively reduce ulcer area and pain sensitivity.The study revealed that four polymethoxyflavones(PMFs)were particularly effective in treating oral ulcers,while hesperidin did not show significant improvement.Mechanistic studies indicated that PMFs could inhibit the phosphorylation of Jak2 and Stat3,whereas hesperidin lacked this effect.This research supports the potential use of Ponkan and its flavonoids as a natural and effective alternative therapy for oral ulcers,with a focus on the role of PMFs in modulating the Jak2-Stat3 pathway,highlighting the possibilities for targeted natural treatments.展开更多
