Understanding how species diverge and adapt is fundamental to unraveling biodiversity.While environmental impacts on species evolution are well-documented,the roles of intrinsic life-history traits remain underexplored.The Qinghai-Xizang Plateau,with its harsh conditions and unique biodiversity,offers a natural laboratory for such investigations.Here,we examine two sympatric small mammals—the solitary,low-dispersal plateau zokor(Eospalax baileyi)and the social,high-dispersal plateau pika(Ochotona curzoniae)—to elucidate how life-history traits shape population structures and adaptive strategies.Through whole-genome sequencing and cardiac-blood phenotype analyses,we reveal striking differences in their evolutionary trajectories.Despite enduring similar environmental pressures,plateau zokor populations exhibit pronounced genetic subdivisions,high inbreeding,and distinct local adaptations.In contrast,plateau pika populations display genetic panmixia,widespread diversity,and adaptive uniformity.Demographic inference highlights that plateau zokors experienced severe population bottlenecks and restricted gene flow during glacial periods,underscoring the impact of dispersal capacity on evolutionary outcomes.Our findings demonstrate that intrinsic biological traits,particularly dispersal ability,fundamentally influence genetic architecture,population connectivity,and local adaptation.This study not only provides empirical evidence of how life-history traits shape evolutionary dynamics but also offers a framework for integrating intrinsic and extrinsic factors in understanding biodiversity formation. 展开更多

This paper evaluated the impacts of mounds created by the plateau pika (Ochotona curzoniae) on the vegetation composition, structure, and species diversity of an alpine Kobresia steppe meadow in Nagqu County, Tibet Autonomous Region, China. Based on mound height or the depth of erosion pit, we defined five stages of erosion and compared the floristic features of communities at these stages with those in undisturbed sites. In the study area, the mounds and pits covered up to 7% of the total area. Lancea tibetica, Lamiophlomis rotata, and Potentilla biflarca were the dominant species in erosion pits, and Kobresia pygmaea, the dominant species in undisturbed sites, became a companion species in eroded areas. In the process of erosion, the original vegetation was covered by soil ejected by the pika, then the mounds were gradually eroded by wind and rain, and finally erosion pits formed. The vegetation coverage increased with increasing erosion stages but remained significantly lower than that in undisturbed sites. Improved coverage eventually reduced soil erosion, and pit depth eventually stabilized at around 20cm. Aboveground biomass increased with increasing erosion stage, but the proportion of low-quality forage reached more than 94%. The richness index and Shannon-Wiener index increased significantly with increasing erosion stage, but the richness index in mound and pit areas was significantly lower than that in undisturbed sites. 展开更多

However, safe and potent adjuvan... Severe Acute Respiratory Syndrome (SARS) is a deadly infectious disease caused by SARS Coronavirus (SARS-CoV). Inactivated SARS-CoV has been explored as a vaccine against SARS-CoV. However, safe and potent adjuvants, especially with more efficient and economical needle-free vaccination are alw needed more urgently in a pandemic. The development of a safe and effective mucosal adjuvant and vaccine ays for prevention of emergent infectious diseases such as SARS will be an important advancement. PIKA, a stabilized derivative of Poly (I:C), was previously reported to be safe and potent as adjuvant in mouse models. In the present study, we demonstrated that the intraperitoneal and intranasal co-administration of inactivated SARS-CoV vaccine together with this improved Poly (I:C) derivative induced strong anti-SARS-CoV mucosal and systemic humoral immune responses with neutralizing activity against pseudotyped virus. Although intraperitoneal immunization of inactivated SARS-CoV vaccine alone could induce a certain level of neutralizing activity in serum as well as in mucosal sites, co-administration of inactivated SARS-CoV vaccine with PIKA as adjuvant could induce a much higher neutralizing activity. When intranasal immunization was used, PIKA was obligatorily for inducing neutralizing activity in serum as well as in mucosal sites and was correlated with both mucosal IgA and mucosal IgG response. Overall, PIKA could be a good mucosal adjuvant candidate for inactivated SARS-CoV vaccine for use in possible future pandemic. 展开更多

Pikas(Lagomorpha: Ochotonidae) are small mouselike lagomorphs. To investigate their adaptation to different ecological environments during their dispersal from the Qinghai-Xizang(Tibet) Plateau(QTP), we collected 226 pikas and measured 20 morphological characteristics and recorded habitat information. We also sequenced the genome of 81specimens, representing 27 putative pika species.The genome-wide tree based on 4?090 coding genes identified five subgenera, i.e., Alienauroa, Conothoa,Lagotona, Ochotona, and Pika, consistent with morphometric data. Morphologically, Alienauroa and Ochotona had similar traits, including smaller size and earlier divergence time compared to other pikas.Consistently, the habitats of Alienauroa and Ochotona differed from those of the remaining subgenera. Phylogenetic signal analysis detected 83 genes significantly related to morphological characteristics, including several visual and hearingrelated genes. Analysis of shared amino acid substitutions and positively selected genes(PSGs) in Alienauroa and Ochotona identified two genes, i.e.,mitochondrial function-related TSFM(p.Q155E) and low-light visual sensitivity-related PROM1(p.H419Y).Functional experiments demonstrated that TSFM-155E significantly enhanced mitochondrial function compared to TSFM-155Q in other pikas, and PROM1-419Y decreased the modeling of dynamic intracellular chloride efflux upon calcium uptake. Alienauroa and Ochotona individuals mostly inhabit different environments(e.g., subtropical forests) than other pikas, suggesting that a shift from the larger ancestral type and changes in sensory acuity and energy enhancement may have been required in their new environments. Human disturbance,particularly road traffic,is one of the greatest threats to wildlife.Considering the association between alerting behavior and the survival of animals,it is important to study the effects of road traffic on alerting behavior of wildlife.Previous studies assessing the shortterm impact of road traffic on alerting behavior of wildlife have focused on vigilance distances.However,studies on the use of alarm calls are scarce,and it is unclear whether such behavioral responses change after repeated exposure to road traffic.We assessed the alerting behavior of plateau pikas(Ochotona curzoniae)who were near or far from roads when facing a potential predator.We found that pikas near roads exhibited shorter vigilance and tolerance distances,and produced fewer alarm calls than those relatively far away from roads.Furthermore,both vigilance and tolerance distances of plateau pikas were significantly positively correlated with the distance from the burrow to the road.Road traffic reduced antipredator responses and shaped alerting behavior;that is,pikas near roads were bolder and more silent compared to those far away from roads.Our findings suggest that increasing urbanization will have corresponding effects on animal behavior,which may have significant fitness effects in the future. 展开更多

Influences of pika and simulated grazing disturbances on bare patches of alpine meadow in the Yellow River Source Zone

Bare patches in alpine meadow are the main manifestation of its degradation.The change of bare patches in an alpine meadow in the Yellow River Source Zone during 2018-2019 was studied in relation to the disturbances caused by plateau pika(Ochotona curzoniae)population and simulated grazing via artificial mowing both independently and interactively.The disturbance was set at three levels of high,medium and no disturbance(control group).Bare patches were mapped by from unmanned aerial vehicle(UAV)images with fine resolution of 1 cm obtained in August 2018 and August 2019 in ArcGIS.The results showed that the total area of bare patches decreased by 112.05 m2 in sub-plots devoid of pika disturbance but increased by 126.37 m2 in other subplots.The highest rate of increase is 89.02%.The individual effect of pika exceeds the joint effect of pika and mowing disturbances.The sole effect of mowing is lower than the joint effect of pika disturbance and intensive mowing,but higher than the joint influence of pika disturbance and moderate mowing.Strong pika disturbance(14 per sub-plot)caused the influence of mowing from moderate to intensive to increase by five-fold.The area of bare patches treated with moderate mowing and no pika disturbance decreased at the highest pace(-37.22%).Intensive mowing and medium density pikas(100 pikas/ha)are considered the thresholds at which the bare patches start to expand.Even if the meadow is mowed at the medium and high intensity,the area of bare patches can be significantly reduced if plateau pika population is controlled to a low level.ANOVA analysis and longterm macro-scale satellite-derived results reveal that pika disturbance is more important in causing the bare patches to change than simulated grazing.Therefore,it is more important to control the number of pikas than to reduce grazing intensity to prevent the expansion of bare patches in the degraded alpine meadow in the study area. 展开更多

Emerging and re-emerging viruses from wild animals have seriously threatened the health of humans and domesticated animals in recent years.Herein,we isolated a new mammalian orthoreovirus(MRV),Pika/MRV/GCCDC7/2019(PMRV-GCCDC7),in the Qinghai-Tibet Plateau wild pika(Ochotona curzoniae).Though the PMRVGCCDC7 shows features of a typical reovirus with ten gene segments arranged in 3:3:4 in length,the virus belongs to an independent evolutionary branch compared to other MRVs based on phylogenetic tree analysis.The results of cellular susceptibility,species tropism,and replication kinetics of PMRV-GCCDC7 indicated the virus could infect four human cell lines(A549,Huh7,HCT,and LoVo)and six non-human cell lines,including Vero-E6,LLCMK2,BHK-21,N2a,MDCK,and RfKT cell,derived from diverse mammals,i.e.monkey,mice,canine and bat,which revealed the potential of PMRV-GCCDC7 to infect a variety of hosts.Infection of BALB/c mice with PMRVGCCDC7 via intranasal inoculation led to relative weight loss,lung tissue damage and inflammation with the increase of virus titer,but no serious respiratory symptoms and death occurred.The characterization of the new reovirus from a plateau-based wild animal has expanded our knowledge of the host range of MRV and provided insight into its risk of trans-species transmission and zoonotic diseases. 展开更多

The Influence of Fence on Body Mass and Population Dynamics of Plateau Pika

To determine the influence of the fence on plateau pika (Ochotona curzoniae), we measured the body mass, breeding status, population densities, using live-trapping and line-transect sampling in April and September 2006. In April, body mass of both male and female plateau pika inside of fence were significant higher than that outside of fence (♀: F = 6.583, df = 1.20, p = 0.019;♂: F = 6.49, df = 1.17, p = 0.021). The ratio of spermary drop of an adult male was also higher (92.31% vs 57.14%). In September, body mass of adult male in both study sites showed no significant difference (F = 0.001, df = 1.23, p = 0.975), but were all significant higher than that of April (F = 121.713, df = 1.22, p < 0.001;F = 105.819, df = 1.20, p < 0.001). In April, the population densities of both study sites showed on significant difference (F = 2.388, df = 1.38, p = 0.131), but the population density inside of fence was much higher than that outside of fence (F = 7.534, df = 1.38, p = 0.009) in September. Our results show that the changes of quality and quantity of food that available to plateau pika in winter, can depress the range of reducing body mass of plateau pika, make breeding season ahead, and then promote the increase of population. 展开更多

With the overuse of antibiotics in health care and animal husbandry,antibiotic resistance becomes a serious threat to public health.Antibiotic residues from veterinarymedicine have increased the dissemination of antibiotic resistance genes(ARGs)by horizontal gene transfer globally,leading to the enrichment of ARGs in wildlife.Plateau pika(Ochotona curzoniae)is a small herbivore endemic to the Qinghai–Tibetan Plateau.Previous studies reveal that pika evolves a coprophagy behavior toward cohabitated yak,which makes the pika population a potential reservoir of ARGs.Yet,little is known about the resistome of pika under different grazing intensities.Here,we sampled the cecum content of pika from three different grazing intensity areas in the Qinghai–Tibetan Plateau to evaluate the effect of grazing on its gut microbiota and resistome.By using the 16S full-length amplicon and metagenomic sequencing,our study revealed that livestock grazing significantly altered the gut microbial community of plateau pika as compared to prohibited grazing areas.We found bacterial lineage Prevotellaceae,Lachnospirales,and RF39 increased in grazing areas.Analysis of the resistome revealed that pika from continuous grazing areas enriched a higher abundance of colistin(MCR)and streptogramin(vat)resistance genes.Moreover,we observed significant correlations between the gut microbial community,ARGs,and mobile genetic element profiles,hinting that pika gut microbiota was an important shaping force of the resistome.In future studies,the continuous monitoring of wildlife gut resistome and environmental antibiotic residues is imperative for a better understanding and for tackling the horizontal gene transfer of ARGs across the wildlife–livestock interface. 展开更多

德格县草地资源较为丰富,长期以来放牧利用较为粗放,“生态、生产、生计”功能低下。以往对全县草地资源类型、分布、利用以及对草地主要生物灾害-鼠害的发生、分布、数量、防控尚不明确。本研究通过文献检索,并结合草地基况调查、草地... 德格县草地资源较为丰富,长期以来放牧利用较为粗放,“生态、生产、生计”功能低下。以往对全县草地资源类型、分布、利用以及对草地主要生物灾害-鼠害的发生、分布、数量、防控尚不明确。本研究通过文献检索,并结合草地基况调查、草地有害生物普查、草原鼠害防控示范区建设的数据和成果,结合德格县气候、土壤、水文及社会经济等自然和人文要素,对德格县草地类型、地域分布、利用特征、害鼠类型及危害进行分析,首次对草原鼠害进行定义,明确了草地类型及分布,剖析了全县草地利用及管理存在的问题,并提出对策和建议,以期为川西高原>4000m高海拔区同类草地合理利用和高质量发展提出参考和科学依据。 展开更多

高原鼠兔(Ochotonacurzoniae)是青藏高原优势草食小型哺乳动物,通过采食、挖掘及排泄等行为活动影响草地生态系统功能。艾美耳球虫(Eimeriaspp.)是高原鼠兔的主要肠道寄生物,具有较高的种属特异性,是一种潜在的高原鼠兔种群防控新模式... 高原鼠兔(Ochotonacurzoniae)是青藏高原优势草食小型哺乳动物,通过采食、挖掘及排泄等行为活动影响草地生态系统功能。艾美耳球虫(Eimeriaspp.)是高原鼠兔的主要肠道寄生物,具有较高的种属特异性,是一种潜在的高原鼠兔种群防控新模式。然而,艾美耳球虫防控高原鼠兔后高寒草地植被的变化情况尚不清楚。该研究选取艾美耳球虫防控高原鼠兔后的草地和对照草地,通过分析不同处理后高寒草地植被多样性、共现网络和指示种等来探究艾美耳球虫防控高原鼠兔后对草地植被群落结构的影响。结果表明:在野外投放艾美耳球虫后,高原鼠兔的有效洞口数显著下降,控鼠效果良好。艾美耳球虫投放后草地总盖度、Shannon-Wiener指数、Simpson指数和β多样性指数等均显著增高;混合效应模型的结果表明,艾美耳球虫投放对草地植被Simpson指数和Pielou均匀度指数的影响更大。艾美耳球虫防控高原鼠兔后植被群落共现网络分析中的群落间关联性和平均连通度增高,草地关键物种由杂类草变成了禾本科和莎草科物种。研究结果为青藏高原高寒草地生物多样性保护、高寒草地啮齿动物科学防控以及生态系统适应性管理提供新的见解。 展开更多

目的:基于超高效液相色谱—四极杆飞行时间质谱联用(ultra high performance liquid chromatography quadrupole timeof-flight mass spectrometry,UHPLC-QTOF-MS)研究不同海拔下高原鼠兔棕色脂肪组织(brown adipose tissue,BAT)中的内... 目的:基于超高效液相色谱—四极杆飞行时间质谱联用(ultra high performance liquid chromatography quadrupole timeof-flight mass spectrometry,UHPLC-QTOF-MS)研究不同海拔下高原鼠兔棕色脂肪组织(brown adipose tissue,BAT)中的内源性差异代谢物及代谢通路,探讨BAT适应高原极端环境的代谢分子机制。方法:分别捕捉青海省果洛地区(4360 m)(MD组)和海北州门源地区(2900 m)(MY组)高原鼠兔各8只,利用UHPLC-QTOF-MS检测2组高原鼠兔BAT样本代谢产物,采用Simca软件对BAT代谢轮廓特征进行主成分分析(principal component analysis,PCA)和正交偏最小二乘判别分析(orthogonal projections to latent structures-discriminant analysis,OPLS-DA),结合t检验、火山图分析差异代谢物,并对差异代谢物进行KEGG代谢通路分析,构建气泡图。结果:PCA和OPLS-DA模式结果显示,2组BAT代谢组均表现出明显聚类型分布,这些代谢物参与体内糖代谢、脂代谢、氨基酸代谢、核苷酸代谢、胆汁酸代谢等多种生命活动。其中,葡萄糖-6-磷酸、胆酸、ADP-核糖、腺苷、脱氧胞苷、L-苯丙氨酸等代谢物水平在MD组高原鼠兔BAT中明显上调(P<0.05);去甲肾上腺素、硬脂酸、尿素、鸟氨酸、丙硫氧嘧啶等代谢物水平明显下调(P<0.05)。富集通路分析发现BAT代谢与苯丙氨酸、磷酸戊糖途径、酪氨酸、色氨酸等代谢通路相关。结论:高原极端环境影响机体的能量相关代谢通路有效激活BAT非颤抖产热,促进机体脂肪组织分化与白色脂肪褐变,增加BAT比重,以此适应高原极端环境。 展开更多

[目的]探究高寒草甸退化秃斑地土壤风蚀规律,为退化草甸区的植被恢复和生态环境保护提供科学依据。[方法]选取青海省河南县因高原鼠兔活动而退化形成的秃斑地作为研究对象,在9,12,15 m/s这3种风速条件下,对不同植被覆盖度(0%,20%,40%,60%,80%)的退化秃斑地进行原位模拟风力侵蚀试验,以分析植被盖度在不同风速下对土壤风蚀的影响。[结果]①退化秃斑地的土壤风蚀速率随时间推移呈现先急剧下降后趋于平缓的变化趋势(p<0.05)。在总时长为60 min的风蚀试验下,前10 min是土壤风蚀速率变化的敏感期。②3种风速下,植被盖度与总输沙量之间均呈显著负相关的指数函数关系(R^(2)=0.98),其指数系数绝对值越大,表明植被覆盖度对风蚀的抑制作用随风速增加而减弱。③随着植被盖度的增加,0~5 cm高度范围内输沙量显著降低,且各植被盖度的输沙量均随高度增加而逐渐减少。[结论]增加植被盖度能够有效降低风蚀速率和输沙高度,但在较高风速下,植被盖度对风蚀的抑制效果显著减弱,且植被盖度抑制土壤风蚀的关键阈值在40%左右,并随着植被盖度的持续增大,风蚀减少的边际效益逐渐降低。 展开更多

Currently, aluminium (Alum, a generic ter... An adjuvant is usually used to enhance the immune response induced by vaccines. The choice of adjuvant or immune enhancer determines the effectiveness of the immune response. Currently, aluminium (Alum, a generic term for salts of aluminium) is the only FDA-approved adjuvant. Alum predominantly induces the differentiation of Th2 cells and thus mediates an antibody immune response. Therefore, there is an urgent need for new adjuvants that enhance not only humoral but also cellular immune responses. In the present study, we demonstrates that PIKA (a stabilized dsRNA) as an adjuvant directly induces the activation and the proliferation of both B and NK cells in vitro. Injection of PIKA into mice results in the production of cytokines in vivo. In addition, the study demonstrates that PIKA promotes the maturation of bone marrow-derived dendritic cells (BMDCs) including up-regulation of the co-stimulatory molecules CD80, CD86 and CD40, and the induction of cytokines such as IL-12p70, IL-12p40 and IL-6. Importantly, after immunization of mice with HBsAg plus PIKA, the presence of PIKA enhances the titers of HBsAg-specific IgG and HBsAg-specific IFN-γ production. These results demonstrate that PIKA as an adjuvant can promote both humoral and cellular immune responses. These might have an implication in applying PIKA as an adjuvant to be used in the design and development of both therapeutic and preventive vaccines, and used in the clinical study. 展开更多

精准识别高原鼠兔危害地秃斑,是科学评估其危害程度的前提与基础。传统的秃斑识别与面积测算方法存在计算繁琐、效率低下等问题。本研究基于深度学习的无人机影像分割方法,提出了小波增强U型卷积神经网络(W-UNet)分割方法。该方法以U型... 精准识别高原鼠兔危害地秃斑,是科学评估其危害程度的前提与基础。传统的秃斑识别与面积测算方法存在计算繁琐、效率低下等问题。本研究基于深度学习的无人机影像分割方法,提出了小波增强U型卷积神经网络(W-UNet)分割方法。该方法以U型卷积神经网络(UNet)为基础架构,采用视觉几何组16层网络(VGG16)作为主干网络。在跳跃连接部分引入坐标注意力机制(CA)提升目标区域的空间定位能力。在编码阶段引入小波变换卷积(WTConv),增强高频信息提取与细粒度特征的恢复能力。此外,采用Focal Loss与Dice Loss组合构建损失函数,有效解决类别不平衡问题。结果表明:本研究提出的无人机影像分割方法在平均交并比(MIoU)、类别平均像素准确率(MPA)和整体准确率(ACC)方面分别达到81.2%、89.4%和95.8%,显著优于传统的UNet-Vgg模型。该研究为高原鼠兔鼠害地秃斑的高效精准监测提供了有力的技术支撑。 展开更多

Seasonal variations in gut microbiota of small mammals and how they are influenced by environmental variables are relatively poorly understood.We sampled 162 wild plateau pikas(Ochotona curzoniae)in 4 seasons over 2 and a half years and recorded the air temperature,precipitation,and nutrient content in edible vegetation at the sampling site.After conducting 16S rRNA and shotgun metagenomic sequencing,we found that the highest alpha diversity,the relative abundance of Firmicutes,and the simplest co-occurrence network occurred in winter,whereas the highest relative abundance of Proteobacteria and the most complex network structure were observed in spring.The highest relative abundance of Verrucomicrobiota and Spirochaetota was seen in summer and autumn,respectively.Air temperature,precipitation,and the contents of crude protein,crude fiber,and polysaccharide in vegetation had significant effects on the seasonal changes in gut microbiota.Diet contributed more to microbial variation than climatic factors.Metagenomic analysis revealed that the amino acid metabolism pathway and axillary activity enzymes were most abundant in summer,while abundance of carbohydrate-binding modules and carbohydrate esterases was highest in spring.These microbial variations were related to the changes in dietary nutrition,indicating that gut microbiota of plateau pika contribute to the efficient use of food resources.This study provides new evidence of how external environmental factors affect the intestinal environment of small mammals. 展开更多
