Pik2-H4是从水稻高抗稻瘟病株系H4中克隆的广谱型抗稻瘟病基因。Pik2-H4基因的CDS全长序列3066 bp,与日本晴中的等位基因相比,其序列存在较大差异。Pik2-H4属于NBS-LRR类基因,其蛋白序列含有NBARC结构域及富亮氨酸重复序列。采用酵母双杂交系统,以Pik2-H4蛋白为诱饵从水稻cDNA表达文库中钓取互作蛋白质,并成功筛选到3个与Pik2-H4有相互作用的蛋白质(LOC-Os08g39300、LOC-Os03g25960、LOC-Os09g29130)。本研究结果有助于进一步了解由Pik2-H4介导的水稻抗稻瘟病反应机理。 展开更多

Abstract The broad-spectrum rice blast resistance gene Pik2-H4 was cloned from the highly resistant line H4. The full-length sequence of Pik2-H4 was 3 066 bp, which was different from the corresponding allele sequence of Nipponbare. Pik2-H4 belonged to NBS-LRR genes and coded a protein containing NB-ARC domain and leueine-rieh repeats. To find the interaction proteins with Pik2-H4 from the rice, the yeast two-hybrid system was used and three important proteins ( LOC- Os08939300, LOCOs03g25960 and LOC-Os09929130)were identified. The results could provide some new information for the mechanism of rice blast resistance mediated by Pik2-H4. 展开更多
