SlPGR5/SlPGRL1 pathway-dependent cyclic electron transport regulates photoprotection and chloroplast quality in tomato plants

The essential photoprotective role of proton gradient regulation 5(PGR5)-dependent cyclic electron flow(CEF)has been reported in Arabidopsis,rice,and algae.However,its functional assessment has not been performed in tomato yet.In this study,we focused on elucidate the function of SlPGR5 and SlPGR5-like photosynthetic phenotype 1(PGRL1)in tomato.We performed RNA interference and found that SlPGR5/SlPGRL1-suppressed transformants exhibited extremely low CO_(2)assimilation capacity,their photosystem I(PSI)and PSII were severely photoinhibited and chloroplasts were obviously damaged.The SlPGR5/SlPGRL1-suppressed plants almost completely inhibited CEF and Y(ND),and PSII photoinhibition may be directly related to the inability to produce sufficient proton motive force to induce NPQ.The transgenic plants overexpressing SlPGR5 and SlPGRL1 driven by 35S promoter capable alleviate photoinhibition of plants under low night temperature.The transcriptomic and proteomic analyses suggested that the nuclear gene transcription and turnover of chloroplast proteins,including the plastoglobule-related proteins,were closely related to SlPGR5/SlPGRL1 pathway dependent CEF.The bridge relationship between CEF and chloroplast quality maintenance was a novel report to our knowledge.In conclusion,these results revealed the regulatory mechanism of the SlPGR5/SlPGRL1 pathway in photoprotection and maintenance of chloroplast function in tomato,which is crucial for reduce yield loss,especially under adverse environmental conditions. 展开更多

In northern China,light and temperature are major limiting factors for plant growth,particularly during seed production and seedling establishment.While previous studies suggested a possible role for the MYB97 gene in cold-stress,confirmation through documented evidence was lacking.In this study,we transformed the MYB97 gene from Iris laevigata into tobacco,and discovered that the gene boosted photosynthesis,photoprotection and resilience to cold.The transgenic tobacco seeds exhibited enhanced germination and accelerated seedling growth.Moreover,these plants had decreased levels of MDA(Malondialdehyde)and relative conductance,coupled with elevated concentrations of proline and soluble sugars.This response was accompanied by heightened activity of antioxidant enzymes during periods of cold stress(4 and−2℃).Exposure to low temperatures(0–15℃)also reduced heights but accentuated primary root growth in transgenic tobacco plants.Additionally,tobacco leaves showed an increased growth along with higher chlorophyll levels,net photosynthetic rates,stomatal conductance,transpiration rates and non-photochemical quenching coefficient.This study shows that IlMYB97(The MYB97 genes in I.laevigata)improves cold-resistance,and enhances photosynthesis and photoprotective ability,and thus overall growth and development.These findings would offer the genetic resources to further study cold resistance and photosynthesis. 展开更多

Xanthophyll cycle-dependent photo-protection is believed to be the main mechanism for plants to deal with excessive light energy. This review focuses on molecular biological aspects and regulations of violaxanthin de-epoxidase and zeaxanthin epoxidase involved in xanthophyll cycle. We will summarize the functions of xanthophyll cycle, especially recent advances in its thermal dissipation mechanism of photoprotection. In midday ginkgo ( Ginkgo biloba L.) leaves have to bear photon flux density over 1400 μmol·m -2 ·s -1 in combination with high temperatures around 35 ℃ at natural habitat. They show typical midday depression of stomatal conductance and of CO 2 assimilation rate. The zeaxanthin changes with light intensity during the day. The influence of the combination of strong light and temperature on photoinhibition was also examined in the laboratory. A low CO 2 internal conductance (31 mmol·m -2 ·s -1 ) was found in ginkgo leaves, which had been exposed to excessive light at temperature between 15 ℃ and 35 ℃ with reduced CO 2 (80 μL·L -1 ) or oxygen (2%) for 2 h, causing a low CO 2 concentration at the carboxylation site and a high proportion of photorespiration. The ratio of electron transport to CO 2 fixation was rather high in ginkgo (16 e -/CO 2 at 25 ℃) as compared with other plants. It increased with temperature also in 2% O 2 which could not be explained solely as due to change of photorespiration. The reduction of oxygen in 340 or 80 μL·L -1 CO 2 had no effect on the extent of photoinhibition at all temperatures, which indicated that electron flow caused by photorespiration in excess light was negligible in protective effect in ginkgo leaves. However, a decreased CO 2 concentration increased photoinhibition, especially at high temperature. It is concluded that the dissipation of excessive excitation energy in the PSⅡ antennae through the xanthophyll cycle may be the major protective mechanism to preventing from the deteriorated effects of strong light in ginkgo leaves. 展开更多

Ammopiptanthus mongolicus (Maxim.) Cheng f. is one of the evergreen shrubs in the desert region of China. In midday its leaves bear photon flux density over 1 500 μmol·m -2 ·s -1 at natural habitat. They show the obvious phenomenon of photoinhibition. For the study of the effects of drought stress on the major protective mechanism against strong light in A. mongolicus leaves, the diurnal variations of photosynthetic rate and chlorophyll fluorescence parameters were investigated under natural conditions with portable photosynthetic measurement system (CIRAS_1) and portable fluorometer (MFMS_2). The experimental results showed that, under normal and drought stress conditions,the net photosynthetic rate ( Pn ), the primary maximum photochemical efficiency of PSⅡ ( Fv/Fm ) and the quantum efficiency of noncyclic electron transport of PSⅡ ( Φ PSⅡ ) decreased obviously at noon (Figs.2,3A,4B). In comparison with plants under normal condition, under drought stress minimal chlorophyll fluorescence ( Fo ) decreased at first and then increased (Fig.3A), non_photochemical quenching ( NPQ ) quickly increased and sustained at a higher level (Fig.4B). This indicated that the major photoprotective mechanism of A. mongolicus leaves was the xanthophyll cycle_dependent thermal energy dissipation under normal condition, while under drought stress, the major photoprotective mechanism was both the xanthophyll cycle_dependent thermal energy dissipation and the reversible inactivation of PSⅡ reaction center. 展开更多

Carotenoids, a class of natural pigments found in all photosynthetic organisms, are involved in a variety of physiological processes, including coloration, photoprotection, biosynthesis of abscisic acid (ABA) and chloroplast biogenesis. Although carotenoid biosynthesis has been well studied biochemically, the genetic basis of the pathway is not well understood. Here, we report the characterization of two allelic Arabidopsis mutants, spontaneous cell death1-1 (spcl-1) and spc1-2. The weak allele spc1-1 mutant showed characteristics of bleached leaves, accumulation of superoxide and mosaic cell death. The strong mutant allele spc1-2 caused a complete arrest of plant growth and development shortly after germination, leading to a seedling-lethal phenotype. Genetic and molecular analyses indicated that SPC1 encodes a putative ζ-carotene desaturase (ZDS) in the carotenoid biosynthesis pathway. Analysis of carotenoids revealed that several major carotenoid compounds downstream of SPC 1/ZDS were substantially reduced in spc1-1, suggesting that SPC 1 is a functional ZDS. Consistent with the downregulated expression of CAO and PORB, the chlorophyll content was decreased in spc1-1 plants. In addition, expression of Lhcb1. 1, Lhcbl. 4 and RbcS was absent in spc1-2, suggesting the possible involvement of carotenoids in the plastid-to-nucleus retrograde signaling. The spc1-1 mutant also displays an ABA-deficient phenotype that can be partially rescued by the externally supplied phytohormone. These results suggest that SPC1/ZDS is essential for biosynthesis of carotenoids and plays a crucial role in plant growth and development. 展开更多

Physical filters are the safest: they scatter and reflect UV rays, but are difficult to apply on the skin. Chemical filters are capable of absorbing these rays, they are cosmetically pleasant, but recent studies suggest a toxicological nature for some of these agents. Organic filters are the newest advance in sun products. Since 2003 the COLIPA Index [Europe] has regulated sun product availability on the market. In the USA, the FDA establishes the classification and the safety of sun products, which are considered topical medical drugs. Up-Regulated Expression of SOD2 and HPRT1 Following Topical Photoprotection and Photorepair Skincare Formulations in A 3-Dimensional Reconstructed Human Skin Model

Photodamage continues to threaten human skin health despite worldwide sun awareness campaigns and the widespread use of sunscreens. To date, extensive research is lacking into the effects of sun avoidance and solar specific skincare regimens on gene expression changes and DNA repair activity. We have previously reported that photoprotection and photorepair formulations which minimize the harmful effects of ultraviolet, visible light and near-infrared radiation can provide photoprotection, anti-photoaging benefits and rejuvenating effects optically, clinically and genetically. To investigate gene expression changes, specifically antioxidant and DNA repair effects following the use of topical photoprotection and photorepair formulations (The Essential Six, RATIONALE, Victoria, Australia), we used epidermal keratinocytes and dermal fibroblasts derived from a 3-dimensional reconstructed human skin model, and assessed upregulation of SOD2 and HPRT1. Gene expression was assessed via the Genemarkers Standard Skin Panel and quantitative real-time PCR exploration. Tissues were inoculated with solar specific topical formulations, then collected after 24 hours following application of photoprotection formulations and 16 hours following photorepair formulations. The quantitative real-time PCR revealed that, in comparison to the control, the genes encoding SOD2 and HPRT1 have been significantly up-regulated following usage of the photoprotection formulations, 1.86, and 1.41, respectively. SOD2 and HPRT1 were up-regulated following use of the photorepair formulations, 2.15, and 1.28, respectively. We were able to substantiate that the photo protection and photorepair formulations upregulated genes involved in antioxidant and DNA repair mechanisms in a 3-dimensional reconstructed human skin model, suggesting a promising anti-photoaging skin regimen. . 展开更多

When light absorbed by plants exceeds the capacity of photosynthesis, the xanthophyll violaxanthin is reversibly de-epoxidized to zeaxanthin in the so-called xanthophyll cycle. Zeaxanthin plays a key role in the protection of photosynthetic organisms against excess light, by promoting rapidly reversible (qE) and long-term (ql) quenching of excited chlorophylls, and preventing lipid oxidation. The photoprotective role of zeaxanthin, either free or bound to light-harvesting complexes (Lhcs), has been investigated by using mutants lacking Chl b (chl) and/or specific xanthophyll species (npq, lut2). The chl mutation causes (1) the absence of Lhcb proteins; (2) strong reduction of the feedback deexcitation (qE); and (3) accumulation of xanthophylls as free pigments into thylakoids. Chl mutants showed extreme sensitivity to photo-oxidative stress in high light, due to higher singlet oxygen (102) release. The double mutant chlnpql was more sensitive to photo-oxidation than chl, showing that zeaxanthin does protect lipids even when free in the membrane. Nevertheless, lack of zeaxanthin had a much stronger impact on the level of lipid peroxidation in Lhcs-containing plants (WTvs npql) with respect to Lhc-less plants (chl vs chlnpql), implying that its protective effect is enhanced by interaction with antenna proteins. It is proposed that the antioxidant capacity of zeaxanthin is empowered in the presence of PSII- LHCs-Zea complexes, while its effect on enhancement of qE only provides a minor contribution. Comparison of the sensitivity of WT vs npql plants to exogenous 102 suggests that besides the scavenging of 102, at least one additional mechanism is involved in chloroplast photoprotection. 展开更多

The mechanism by which the mitochondrial alternative oxidase (AOX) pathway contributes to photosystem II (PSII) photoprotection is in dispute. It was generally thought that the AOX pathway protects photosystems by dissipating excess reducing equivalents exported from chloroplasts through the malate/oxaloacetate (Mal/OAA) shuttle and thus preventing the over-reduction of chloroplasts. In this study, using the aoxla Arabidopsis mutant and nine other C3 and C4 plant species, we revealed an additional action model of the AOX pathway in PSII photoprotection. Although the AOX pathway contributes to PSII photoprotection in C3 leaves treated with high light, this contribution was observed to disappear when photorespiration was suppressed. Disruption or inhibition of the AOX pathway significantly decreased the photorespiration in C3 leaves. Moreover, the AOX pathway did not respond to high light and contributed little to PSII photoprotection in C4 leaves possessing a highly active Mal/OAA shuttle but with little photorespiration. These results demonstrate that the AOX pathway contributes to PSII photoprotection in C3 plants by maintaining photo- respiration to detoxify glycolate and via the indirect export of excess reducing equivalents from chloro-plasts by the MaI/OAA shuttle. This new action model explains why the AOX pathway does not contribute to PSII photoprotection in C4 plants. 展开更多

Major light-harvesting complex of photosystemⅡ(LHCⅡ)plays a dual role in light-harvesting and excited energy dissipation to protect photodamage from excess energy.The regulatory switch is induced by increased acidity,temperature or both.However,the molecular origin of the protein dynamics at the atomic level is still unknown.We carried out temperature-jump time-resolved infrared spectroscopy and molecular dynamics simulations to determine the energy quenching dynamics and conformational changes of LHCⅡtrimers.We found that the spontaneous formation of a pair of localα-helices from the 310-helix E/loop and the C-terminal coil of the neighboring monomer,in response to the increased environmental temperature and/or acidity,induces a scissoring motion of transmembrane helices A and B,shifting the conformational equilibrium to a more open state,with an increased angle between the associated carotenoids.The dynamical and allosteric conformation change leads to close contacts between carotenoid lutein 1 and chlorophyll pigment 612,facilitating the fluorescence quenching.Based on these results,we suggest a unified mechanism by which the LHCⅡtrimer controls the dissipation of excess excited energy in response to increased temperature and acidity,as an intrinsic result of intense sun light in plant photosynthesis. 展开更多

Objective:Awareness regarding the negative effects of ultraviolet radiation are commonly neglected in Asian countries.This study sought to evaluate the ultraviolet radiation knowledge and photoprotective behaviors of university students in Indonesia.Methods:This observational cross-sectional was conducted at Pelita Harapan University,Indonesia.Results:A total of 89(45.4%)medical students and 107(54.6%)nonmedical students participated in this study.Their mean age was 21.03±2.03 years,and 46.4%of them had Fitzpatrick type III skin.Medical students as well as older students exhibited a higher level of knowledge regarding photoprotection(P≤0.05).Factors related to good photoprotective behaviors were sex,older age,and average expenditure on skincare.Further analysis revealed that a high level of knowledge regarding photoprotection was related to acceptable photoprotective behaviors(P=0.01;odds ratio=1.97;95%confidence interval=1.13-3.54).Conclusions:Knowledge regarding sun-related damage and practice of photoprotective behaviors were low among the participants.The results indicate that formal education and academic subjects should incorporate the topic of sun-related damage and promote the practice of photoprotective behaviors early on to increase awareness among populations. 展开更多

Purpose: To investigate the efficacy of a solar repair Formulation following ultraviolet and environmental exposure in order to improve overall skin health and appearance through three hypotheses: The Formulation increases expression of DNA repair mechanisms markers;The Formulation enhances overall skin appearance through reducing signs of inflammation, elevating hydration, reinforcing skin firmness and amplifying radiance;In-Vivo efficacy test results are aligned with measured gene expression changes. Methods: The Formulation (#6NIC1.V1.1-1) was tested for: In-vitro LDH cytotoxicity activity, In-vitro qPCR gene expression with and without ultraviolet exposure on a reconstructed 3-dimensional skin model, and In-Vivo efficacy study on a panel of 22 participants objectively and subjectively. Results: Skin radiance, firmness, hydration, redness, and inflammation are significantly improved after In-Vivo skin exposure to the Formulation and environmental challenges such as ultraviolet radiation. These outcomes were confirmed by in-vitro genetic testing on a reconstructed human skin model. Conclusion: The studies allowed us to identify and group results in four main skin functions that were significantly enhanced following the application of the Formulation: firmness, hydration, radiance and soothing. 展开更多

Photoinhibition in Shaded Cotton Leaves After Exposing to High Light and the Time Course of Its Restoration 被引量:26

Chlorophyll fluorescence emission, pigment composition and photosynthetic rate of shade-grown cotton ( Gossypium hirsutum L.) plants were measured immediately after suddenly exposing to full sunlight and at regular intervals there after within 15 d. Photoinhibition occurred in shade-grown cotton leaves immediately after exposed to full sunlight. The chlorophyll fluorescence parameter F-v/F-m and PhiPS II, which reflect the efficiency of PS II,obviously decreased in shade-grown leaves, much lower than that of the full sunlight-grown leaves. On the contrary, F-o value was sharply increased. Neither of these parameters could completely recover till next morning. The photoinhibition was chronic and continued for about 4 d, while the F-v/F-m and the net photosynthetic rate ( P-n) continued to decline, then began to increase gradually 6 d later and turned stable after 10 - 12 d, appearing as an acclimation phenomenon. However, the final value of F-v/F-m and P-n did not reach the level as in those leaves grown in the full sunlight ever before. The final P-n was higher by 60% than that before exposure, but lower for more than 40% than that of the full sunlight-grown leaves. The most notable response of chloroplast pigment composition was a pronounced increase in the pool size of carotenoids in xanthophyll cycle over a period of 3 d. The results indicated that when shade-grown cotton seedlings were suddenly transferred to the full sunlight, the decline of F-v/F-m and P-n might associate with the damage of the PS II reaction center. During the light acclimation, photoprotective mechanisms such as the xanthophyll cycle-dependent energy dissipation were increased, so that photodamage in leaves transferred from low to high light might be reduced. 展开更多

A wheat (Triticum aestivum L cv. Xiaoyan 54) VDE cDNA was obtained using RT-PCR method. Its deduced protein sequence shares high identity with that of Arabidopsis and rice. Southern blot revealed that there are three copies of VDE gene per haploid genome of wheat. VDE transcript levels were higher in green leaf than in root, seed and etiolated leaf. Northern blotting analysis indicated that VDE mRNA level is induced during greening process of etiolated wheat seedling and increased by intense light illumination. 展开更多

Ultraviolet(UV)-induced photoaging skin has become an urgent issue.The functional foods and cosmetics aiming to improve skin photoaging are developing rapidly,and the demand is gradually increasing year by year.Collagen peptides have been proven to display diverse physiological activities,such as excellent moisture retention activity,hygroscopicity,tyrosinase inhibitory activity and antioxidant activity,which indicates that they have great potential in amelioration of UV-induced photoaging.The main objective of this article is to recap the main mechanisms to improve photoaging skin by collagen peptides and their physiological activities in photo-protection.Furthermore,the extraction and structural characteristics of collagen peptides are overviewed.More importantly,some clinical trials on the beneficial effect on skin of collagen peptides are also discussed.In addition,prospects and challenges of collagen peptides are emphatically elucidated in this review.This article implies that collagen peptides have great potential as an effective ingredient in food and cosmetics industry with a wide application prospect. 展开更多

Objective: To investigate damage effects of ultraviolet irradiation on eternal keratinocyte-HaCaT cells and to evaluate photo-protective efficiency of hydroxychloroqine and Traditional Chinese Medicines(epigallocatechingallate[EGCG], baikal skullcap root and szechwan lovge rhizome) on HaCaT cells damaged by middle wave ultraviolet(UVB) irradiation. Methods: Subconfluent HaCaT cells were sham or UVB irradiated and treated with above TCM agents. The damage degree of HaCaT cells was observed by a light microscop. Cell growth was recorded by cell count and cellular activity was detected by MTT method. The secretion amount of IL-6 and TNF-α was measured by ELISA. Results: The irradiation damage of HaCaT cells was depended on the irradiated dosages and cellular activity was reduced by 36%-80%, with a maximum decrease over 90% after 72 h. The intervention of the above drugs may increase the cellular activity by 10%-72%. The photo-protective efficiency was more apparent in EGCG (from 1.19±0.07 to 1.28±0.06, P<0.01) than that in hydroxychloroqine (from 0.43±0.04 to 0.96±0.04, P<0.05). The other two tested drugs also showed photo-protective effect(from 0.44±0.07 to 1.21±0.02, P<0.05). As to cytokine secretion, EGCG could decline the secretion amount of IL-6 and TNF-α apparently. Hydroxychloroqine and baikal skullcap root baikal skullcap root could only reduce the secretion of IL-6. The secretion of IL-6 and TNF-α could not be inhibited by szechwan lovge rhizome. Conclusion: The injury effect of UVB irradiation on cultured keratinocytes is dose-dependent and the tested drugs have photo-protective potency. Inhibition of cytokine secretion may be one of the mechanisms related to reducing the damage effect of UVB irradiation. 展开更多

The effects of light intensity on photosynthesis and photoprotective mechanisms under progressive drought were studied on apple trees (Malus domestica Borkh.) Fuji. The potted trees were exposed to drought stress for 12 days and different light conditions (100, 60 and 25%sunlight). During the progressive drought, the relative water content (RWC) in leaf declined and was faster in ful light than in 60 and 25%sunlight. However, the decrease in the net photosynthetic rate (Pn), stomatal conductance (Gs) and Rubisco activity were slower under 100%sunlight condition than other light conditions. After the 6 days of drought, the maximum PSII quantum yield (Fv/Fm), the capacity of electrons move beyond QA?(1–VJ) and electron move from intersystem to PSI acceptor side (1–VI)/(1–VJ) decreased, with greater decline extent in brighter light. While RWCs were>75%, the variations in different light intensities of Gs and Rubisco activity at identical RWC, suggested the direct effects of light. While the little difference in the state of photosynthetic electron transport chain among tested light intensities indicates the results of faster water loss rate of light. Our results also demonstrated that the enhancement the de-epoxidations of xanthophyl cycle, activities of ascorbate peroxidase (APX) and catalase (CAT) were directly regulated by light intensity. While the higher photorespiration rate (Pr) under stronger light condition was mainly caused by faster water loss rate of light. 展开更多

Polypeptide from Chlamys farreri (PCF) , a topical polypeptide isolated from Chlamys farreri , was used in this experiment aimed to investigate the photoprotective effect of PCF against chronic skin damage induced by ultraviolet A (UVA) and ultraviolet B (UVB) radiation. The chronic ultraviolet irradiated guinea pig model was established, and visible changes in the skin including wrinkling, sagging and erythema were observed. Malondialdehyde (MDA) and antioxidant enzymes including superoxide dismutase (SOD) and glutathione peroxidase (GSH px) in the dorsal skin were determined using biochemical methods. The results showed:(1)PCF (5 % and 20%) could greatly protect the dorsal skin of guinea pig against wrinkling, sagging and erythema induced by UV radiation in a concentration dependent manner.(2)PCF could reduce MDA formation in the dorsal skin caused by UV irradiation, while increasing the activities of SOD and GSH px.(3)The differences among the PCF groups and UV model group were significant ( P <0.05, P <0.01). These results indicated that topical application of PCF provided broad solar UV spectrum photoprotection; and that the antioxidant property of PCF might play a role in photoprotection. 展开更多
