Mandarin fish(Siniperca scherzeri) has high market prices and significant market potential in China because of its highquality meat and high nutritional value. However, due to the limited scale of aquaculture, meeting the market demand is difficult, making the effective development of the aquaculture potential of mandarin fish an important challenge for the industry. In this study, a 30-d breeding experiment was conducted on mandarin fish larvae under three photoperiod conditions: G1 8 h light:16 h dark(8L:16D), G2 12 h light:12 h dark(12L:12D), and G3 16 h light:8 h dark(16L:8D). The results showed that the G2 group exhibited the best growth performance and development status, with final body weights, weight gain rates, and specific growth rates all higher than those of the other two groups(P < 0.05). Observations of sections from each group revealed that the intestinal villi length and muscle thickness of the G2 group were significantly greater than those of the other two groups(P < 0.05). The G2 group inhibited the transcriptional activation of key circadian rhythm genes, including nr1d2a, nr1d1 and per1, while upregulating the expression of BMAL1 in S. scherzeri.The activation of both the insulin signalling pathway and the Fox O signalling pathway enhanced the efficient secretion of insulin, which subsequently played a critical role in regulating fatty acid metabolism. This active fatty acid metabolism provided an optimal energy supply, ensuring that other nutrients were fully utilized during the growth and development process while minimizing unnecessary nutrient loss. Consequently, this mechanism effectively promoted the overall growth and development of S. scherzeri. This study was the first to elucidate the transcriptomic expression patterns of S. scherzeri under varying photoperiod conditions. In response to the cyclic alternation of day and night, S. scherzeri regulated their metabolic levels and the transcriptional activation of downstream target genes via insulin signalling. 展开更多

SiPRR37 exerts dual functions in the regulation of photoperiodic flowering and contributes to the ecological adaption of foxtail millet

Foxtail millet(Setaria italica)is an important crop and an emerging model plant.Photoperiodic flowering is a key determinant of its production and geographic expansion.In this study,we found that SiPRR37 is responsible for the major quantitative trait locus(QTL)Heading date 2(Hd2)identified in 680 foxtail millets using a genome-wide association study.Overexpression of SiPRR37 in foxtail millet significantly delayed the heading date under both natural long-day and short-day conditions.CRISPR/Cas9-induced Siprr37 mutants exhibited earlier flowering in long-day conditions but later flowering in short-day conditions.The critical day length(CDL)for the reversal of Siprr37’s function was around 14.3 h.Haplotype analysis revealed that accessions with the Tc1-Mariner transposon insertion in SiPRR37(Hap 1)flowered significantly earlier at higher latitudes,and later at lower latitudes,indicating that natural variants of SiPRR37 exert dual functions in flowering regulation according to geographic latitude.The gradual,successive decrease in the frequency of Hap 2 from low to high latitudes,with the concurrent increase of Hap 1,demonstrates that these haplotypes have undergone artificial selection.Further FST analysis demonstrated that SiPRR37 has contributed to the ecological adaption of foxtail millet.Additionally,we reveal that OsPRR37 promotes flowering in rice,while GmPRR37 may only inhibit flowering in soybean.Further diurnal expression and transgenic analyses suggest that the dual function of SiPRR37 might depend on SiHd1.Our study uncovered the distinct functional reversal of SiPRR37 and functional diversification of PRR37 homologs in SD crops.These findings not only enrich knowledge about the regulation of photoperiodic flowering,but also contribute to genetic improvement of crops’regional adaptability. 展开更多

The immunohistochemical localization of IAA and the comparison of their relative levels were carried out for the first time in the anthers of Nongken 58S and its wild type Nongken 58 (Oryza sativa subsp. japonica) after long_day and short_day treatments. The distribution of free_IAA in anthers and its dynamic variation could be reflected by this method. The results showed that the IAA level in the anthers of Nongken 58S after long_day treatment was much lower than that in short_day_treated Nongken 58S and those in wild type Nongken 58 in five stages from pistil and stamen primordia formation to late uninucleate stage. The possible reasons for IAA deficiency in Nongken 58S_LD anthers and its relationship with fertility alteration were also discussed. 展开更多

Changes in the pattern of organization of microtubules in the meiotic stages of development of pollen (i.e. from pre-meiotic interphase to more or less metaphase I) of a normal (IR36) and a temperature/photoperiod sensitive male sterile line (Peiai 64S) of rice were studied using immunofluorescence confocal microscopy. In IR36, from pre-meiotic interphase to metaphase I, the pattern of microtubule distribution in the meiocytes underwent a series of changes. Some new organizational patterns of microtubules (that have not been described before) were observed during microsporogenesis, including the existence of a broad band of perinuclear microtubules at the diakinesis stage of development. The pattern of microtubule distribution in the meiocytes of the male sterile line, Peiai 64S, was quite different front that seen in IR36. In Peiai 64S, the microtubules showed abnormal patterns of distribution from pre-meiotic interphase to metaphase I. For example the broad band of perinuclear microtubules seen at diakinesis in IR36 was much disorganized and loosened in Peiai 64S. The spindles formed were also very abnormal and different from the normal spindle. The appearance of abnormal microtubule distribution in the early stages of microsporogenesis may contribute to the malformation and ultimate abortion of pollen in Peiai 64S. 展开更多

[ Method ] The sowing-date tests and designed conditions were conducted during 2003 -2005. [ Result] Fertility of Vtai911289a, could alter under specific photoperiod and temperature conditions. Temperature is one of the main factors influencing male fertility of the male sterile lines. Vtai911289a, showed stable sterility under the condition of the mean of daily temperature at fertility sensitive stage lower than 19℃ and presented partial fertility when the mean of daily temperature at fertility sensitive stage lower than 20 - 22℃. Photoperiod to some extent affects the male fertility of Vtai911289a, long-day condition is helpful for the male fertility of the sterile line. Short_day (SD) induced the plant senescence in pumpkin ( Cucurbita pepo Linn.) strain 185. Structural assay, gene expression and a series of biochemical analyses were performed to analyze the senescence mechanism in pumpkin strain 185 exposed to SD. Two aspects of important changes initiated in SD exposure contributed to the senescence process. SD functionally led to the initiation of the apical transformation from vegetative to reproductive growth, and then programmed cell death (PCD) in the apical meristem, causing the loss of vigorous growth activity. Moreover, SD treatment resulted in the formation of a great number of dying cells in mesophyll tissue later in the development compared with the phenotype of plants under long_day (LD) conditions. During the senescence process, high expression of nuclease is an important molecular event. These results indicate that the initiation of senescence process in pumpkin stain 185 plants is closely related to the death of cells in apical meristem and mesophyll. 展开更多

QTL Identification of the Insensitive Response to Photoperiod and Temperature in Soybean by Association Mapping 被引量:6

The insensitive response to photoperiod and temperature is an important quantitative trait for soybean in wide adaptation breeding. The natural variation in response to photoperiod and temperature was detected using 275 accessions of soybean [Glycine max (L.) Merrill] from China. Genome-wide association mapping, based on population structure analysis, was carried out using 118 SSR markers by the TASSEL GLM (general linear model) program. Nine SSR markers (P〈0.01) were associated with the value of the response to photoperiod and temperature (VRPT) caused by days to flowering (DF), among which, Satt308 (LG M), Sattl50 (LG M) and Satt440 (LG l), were identified in both 2006 and 2007. Twelve SSR markers (P〈0.01) were associated with VRPT caused by days to maturity (DM), among which three markers, Satt387 (LG N), Satt307 (LG C2) and AW310961 (LG J), were detected in both 2006 and 2007. In addition, a total of 20 elite alleles were screened out over 2006 and 2007 for being associated with an insensitive response to photoperiod and temperature (IRPT) caused by DF and a total of seven different elite alleles were screened out for being associated with IRPT caused by DM. Among these elite alleles, five alleles, Satt150-244, Satt308-164, Satt308-206, Satt440-176, and Satt440-206, were associated with IRPT caused by DF and were identified in both years, but only one allele, Satt307-170, was identified as being associated with an IRPT caused by DM. Based on these elite alleles, a set of typical accessions were screened out. The result about the genetic basis of IRPT is meaningful for soybean wide adaption breeding. 展开更多

Photoperiod sensitivity in maize plays an essential role in utilizing tropic and sub-tropic germplasm to temperate areas. This study aims to identify and map the QTLs responsible for the characteristics measuring photoperiod sensitivity, days from planting to silking (SD), photoperiod response coefficient of silking (PRC), and anthesis-silking interval (ASI). Using the population derived from Zheng 58, photoperiod-insensitive parent, and Ya 8701, photoperiod-sensitive parent, a linkage map was constructed with 93 single sequence repeat (SSR) markers. Phenotyping of 296 F2-3 families of the population in replicated-field test was conducted in both long-day (Beijing, China) and short-day (Sichuan, China) conditions. Ten QTLs were identified to be associated with the SD and ASI on chromosomes 3, 4, 6, 8, and 10 in the longday conditions, and 11 QTLs were detected to be related to the SD and ASI on chromosomes 2, 3, 4, 5, 6, 8, and 10 in the short-day conditions, respectively. A QTL associated with the PRC as a major effect in the long-day conditions located in the same position as the QTL related to the SD and ASI in the map, and was on chromosome 10 linked with marker bnlg1655. Using these QTLs in the marker-assisted selection, the photoperiod sensibility could be reduced by selection of the alleles responsible for the SD, PRC, and ASI in breeding programs. 展开更多

In nature, Apostichopus japonicus exhibits a behavioral characteristic of emerging at night and sheltering during the day. Hence, it was commonly believed that longer and darker light conditions are better for a population of A. japonicus. In aquaculture situations therefore, animals have commonly been cultured without lighting, especially during juvenile-hatching culture. However, how the length of darkness each day affects .4.japonicus and what the mechanism is of behavioral response of A. japonicus to different photoperiods remain unclear, and are specifically addressed in this study. We applied nine photoperiod treatments (light:dark (LD) cycles at 0:24, 3:21, 6:18, 9:15, 12:12, 15:9, 18:6, 21:3, and 24:0 under 500Ix) to 60 individuals (29.73±0.23 g) per treatment (with 3 replicates). The enviroramental conditions were set as: water temperature = 16.9±0.3℃, dissolved oxygen〉6.0 rag/L, ammonia〈0.3 mg/L, pH=7.8-8.2, and salinity=30-32. Under experimental conditions of continuous darkness as well as continuous light, cyclic nocturnal activity patterns of A. japonicus (viz. the animals emerged and fed at night and sheltered during the day) were observed. However, they spent more and less time, respectively, moving and feeding under continuous darkness and continuous light, than those under a natural light cycle. Under photoperiods with 6-12 h of light, the animals showed one sheltering behavior transition and two emerging behavior transitions. These behavior transitions appeared to be governed by some internal physiological factors and induced by daily light variation. The behavior of the animals was significantly affected by different photoperiods, and the distribution rate (DR) of the animals emerging at "daytime" increased with lengthening "light time" (except for the LD 3:21 cycle). We also found that there was no significant difference in mean DR per day under LD cycles of 6:18, 9:15, 15:9, and 12:12. Mean DRs per day were significantly lower for these photoperiods, than for those under LD cycles of 0:24 and 3:21, and significantly higher than for those under LD cycles of 18:6, 21:3 and 24:0. These results imply that 6-15 h light per day were the similar photoperiods to natural sunlight. From the viewpoint of activity and feeding behaviors, these might be the optimal photoperiods for rearing A. japonicus. 展开更多

Pei'ai64S, an indica sterile variety with photoperiod and thermo-sensitive genic male sterile (PTGMS) genes, has been widely exploited for commercial seed production for "two-line" hybrid rice in China. One PTGMS gene from Pei'ai64S, pmsl(t), was mapped by a strategy of bulked-extreme and recessive-class approach with simple sequence repeat (SSR) and insert and deletion (In-Del) markers. Using linkage analysis for the F2 mapping population consisting of 320 completely male sterile individuals derived from a cross between Pei'ai64S and 93-11 (indica restorer) lines, the pmsl(t) gene was delimited to the region between the RM21242 (0.2 cM) and YF11 (0.2 cM) markers on the short arm of chromosome 7. The interval containing the pmsl(t) locus, which was co-segregated with RM6776, is a 101.1 kb region based on the Nipponbare rice genome. Fourteen predicted loci were found in this region by the Institute for Genomic Research (TIGR) Genomic Annotation. Based on the function of the locus LOC_Os07g12130 by bioinformatics analysis, it is predicted to encode a protein containing a Myb-like DNA-binding domain, and may process the transcript with thermosensory response. The reverse transcription-polymerase chain reaction (RT-PCR) results revealed that the mRNA levels of LOC_Os07g12130 were altered in different photoperiod and temperature treatments. Thus, the LOC_Os07g12130 locus is the most likely candidate gene for pmsl(t). These results may facilitate not only using the molecular marker assisted selection of PTGMS genes, but also cloning of the pmsl(t) gene itself. 展开更多

The chrysanthemum genome harbors three FT-like genes:CmFTL1 and CmFTL3 are thought to act as regulators of floral induction under long-day(LD)and short-day(SD)conditions,respectively,whereas the function of CmFTL2 is currently unclear.The objective of the present research was to explore the function of CmFTL2 in the determination of flowering time of the photo-insensitive chrysanthemum cultivar‘Floral Yuuka’,both in response to variation in the photoperiod and to the exogenous provision of sucrose.Spraying leaves of‘Floral Yuuka’plants with 50 mM sucrose accelerated flowering and increased the level of CmFTL2 transcription in the leaf more strongly than either CmFTL1 or FTL3 under both long and SD conditions.Transcription profiling indicated that all three CmFTL genes were upregulated during floral induction.The relationship of the CmFTL2 sequence with that of other members of the PEBP family suggested that its product contributes to the florigen rather than to the anti-florigen complex.The heterologous expression of CmFTL2 in the Arabidopsis thaliana ft-10 mutant rescued the mutant phenotype,showing that CmFTL2 could compensate for the absence of FT.These results suggest that CmFTL2 acts as a regulator of floral transition and responds to both the photoperiod and sucrose. 展开更多

It is vital to determine the effective photoperiods of maize for making full use of tropical germplasm, which is the foundation for determining the effect of latitude and planting date on the development of photoperiod-sensitive maize cultivars. The objective of this study is to determine the photoperiod-sensitive inductive phase using reciprocal transfer between long- day (LD) (15 h d^-1) and short-day conditions (SD) (9 h d^-1). For Huangzao 4 and CML288, days to tassel and pollen shedding were recorded, and stem apical meristems (SAM) were observed by a laser scanning confocal microscope. The results show that the seedlings are insensitive to photoperiod when they are very young (juvenile). However, after this period, LD delays flowering and increases the leaf numbers below the inflorescence, and the length of the interval of the photoperiod-sensitive inductive phase is longer under LD conditions than under SD conditions. Transferred from SD to LD, plants show a sudden decrease in leaf numbers once sufficient SD has been received for flower commitment. While transferred from LD to SD, plants have a continuous increase in leaf numbers during the photoperiod sensitive inductive phase under LD conditions. At the same time, when plants are competent to flowers, the obvious morphology is the elongation of maize SAM. There is an obvious variance of the photoperiod sensitive phase under LD and SD conditions in different maize. 展开更多

The responses of dormancy induction to illumination and the characteristics of respiratory rate were studied with the nectarine peach bud in this article. The trial was conducted with nectarine (Prunus persica var. nectariana cv. Shuguang) and involved three treatments: a short day treatment (8 h), a long day treatment (16 h), and the normal condition as the control. The dormancy status was determined with the growth of shoot and the sprouting ability, and the respiratory rate was mensurated with oxygen electrode. Short day treatment could induce the growth stopping of peach shoots ahead, promote the development of dormancy, and induce buds into dormancy with 21 d previous to control. Long day treatment postponed the growth stopping and the induction and development of dormancy. The respiratory rate decreased according to the development of dormancy induction. The minimum respiratory rate appeared about 7 days after the start of dormancy induction. Bud respiratory rate increased during this period and then declined and remained at low level during dormancy period. Long day reduced buds respiratory rate slightly. Short day could induce dormancy obviously, and long day postponed dormancy induction. The changes of respiratory rate were correlated with the development of dormancy induction, and the bud respiratory rate was also affected by photoperiod. 展开更多

To understand the male sterility mechanism of photoperiod/thermo-sensitive genic male sterile [P(T)GMS] lines in rice, the research progress on genetics of photoperiod and/or temperature sensitive genic male sterility in rice was reviewed. A new idea was proposed to explain the sterility mechanism of P(T)GMS rice. The fertility transition from sterile to fertile is the result of cooperative regulation of major-effect sterile genes with photoperiod and/or temperature sensitive genes, but not the so-called pgms gene in P(T)GMS rice. The minor-effect genes, which exhibit accumulative effect on sterility, are the important factors for the critical temperature of sterility transition. The more minor-effect genes the sterile line holds, the lower the critical temperature of sterility transition is. The critical temperature of sterility transition will be invariable if all the minor-effect genes are homozygous. The strategies for breeding P(T)GMS rice were also proposed. The selective indices of critical photoperiod and temperature for sterility transition should be set according to varietal type and ecological region. Imposing selection pressure is a key technology for breeding P(T)GMS rice with lower critical temperature for sterility, and improving the comprehensive performance of the whole traits and combining ability is vital for breeding P(T)GMS rice lines. 展开更多

The effects of light intensities and photoperiods on proteolytic activity in Afifella marina strain ME (KC205142) were investigated. Secretion of proteolytic enzymes in Afifella marina was preliminarily assessed by skim milk agarose media. Subsequently, light intensities, such as, dark, 1000, 1500, 2000, 2500, 3000, 3500, 4000, 4500 and 5000 lux were used to evaluate the effects on proteolytic activity in Afifella marina strain ME under anaerobic condition. After that, the effect of photoperiods on proteolytic activity was monitored under anaerobic light condition (3000 lux) at 0 h (0L/24D), 6 h (6L/18D), 12 h (12L/12D), 18 h (18L/6D) and 24 h (24L/0D) of photoperiod. The highest proteolytic activity of 74.67 U was recorded at 3000 lux illumination light intensity. The proteolytic activity in bacterium Afifella marina strain ME was positively associated with the dry cell weight. The proteolytic activity of 72.67 U in bacterium Afifella marina strain ME at 18 h (18L/6D) photoperiod is not significantly different (p > 0.05) from proteolytic activity of 74.67 U recorded at continuous light (24L/0D) condition. Light intensity of 3000 lux, culture period of 48 h and a photoperiod of 18 h (18L/ 6D) were the optimum parameters for proteolytic activity in bacterium Afifella marina strain ME. 展开更多

A prominent example of seasonal phenotypic flexibility is the winter increase in thermogenic capacity (=summit metabolism, Msurn) in small birds, which is often accompanied by increases in pectoralis muscle mass and lipid catabolic capacity. Temperature or photoperiod may be drivers of the winter phenotype, but their relative impacts on muscle remodeling or lipid transport pathways are little known. We examined photoperiod and temperature effects on pectoralis muscle expres- sion of myostatin, a muscle growth inhibitor, and its tolloid-like protein activators (TLL-1 and TLL- 2), and sarcolemmal and intracellular lipid transporters in dark-eyed juncos Junco hyemalis. We acclimated winter juncos to four temperature (3~C or 24~C) and photoperiod [short-day (SD) = 8L:16D; long-day (LD) = 16L:8D] treatments. We found that myostatin, TLL-I, TLL-2, and lipid transporter mRNA expression and myostatin protein expression did not differ among treatments, but treatments interacted to influence lipid transporter proteinexpression. Fatty acid translocase (FAT/CD36) levels were higher for cold SD than for other treatments. Membrane-bound fatty acid binding protein (FABPpm) levels, however, were higher for the cold LD treatment than for cold SD and warm LD treatments. Cytosolic fatty acid binding protein (FABPc) levels were higher on LD than on SD at 3℃, but higher on SD than on LD at 24℃. Cold temperature groups showed upregulation of these lipid transporters, which could contribute to elevated Msum compared to warm groups on the same photoperiod. However, interactions of temperature or photoperiod effects on muscle remodeling and lipid transport pathways suggest that these effects are context-dependent. 展开更多

Shuguang) nectarine was used to study effects of photoperiod on key-enzyme activities of respiration during dormancy induction. The dormancy status was determined with sprouting ability. Spectrophotometry was used to investigate activities of phosphohexose isomerase (PGI), malic dehydrogenase (MDH), and glucose-6-phosphate dehydrogenase (G6PDH). The results revealed that short day (SD) treatment promoted dormancy induction while long day (LD) treatment postponed the process. During dormancy induction, PGI activities declined, MDH activities changed little, and G6PDH activities increased both in flower buds and leaf buds. PGI activities and MDH activities in SD treatment were lower than control, and G6PDH activities were higher, which was opposite with LD treatment. The changes of respiratory key-enzyme activities were adjusted by photoperiod and correlated with the development of dormancy induction. 展开更多

To understand the genetic characteristics of a new photoperiod-sensitive genic male sterile line Mian 9S, some reciprocal crosses were made between Mian 9S and six indica rice materials, Yangdao 6, Luhui 602, Shuihui 527, Mianhui 725, Fuhui 838 and Yixiang 1B. Genetic analysis results suggested that the photoperiod-sensitive genic male sterility (PGMS) of Mian 9S was controlled by a single recessive nuclear gene. Thus, the F2 population derived from the cross of Yangdao 6/Mian 9S was used to map the PGMS gene in Mian 9S. By using SSR markers, the PGMS gene of Mian 9S was mapped on one side of the markers, RM6659 and RM1305, on rice chromosome 4, with the genetic distances of 3.0 cM and 3.5 cM, respectively. The gene was a novel PGMS gene and designated tentatively as pms4. In addition, the application of the pms4 gene was discussed. 展开更多
