[Objectives]To explore the protective effect of Alpinia zerumbet(Pers.)Burttet Smith ethanol extraction(YSJ)on gastric ulcers.[Methods]The ethanol-induced gastric ulcer model was generated by male Kunming mice.The mice were divided into six groups,including control,model(GU),positive omeprazole enteric-coated capsules(18 mg/kg),YSJ low(1.17 g/kg),medium(2.34 g/kg)and high(4.68 g/kg)dose groups.To observe the state of gastric tissues,hematoxylin and eosin(H&E)staining was applied and immunohistochemistry(IHC)was used to evaluate the expression of nuclear factor kappa-B p65(NF-κB p65),interleukin 6(IL-6),tumor necrosis factor-α(TNF-α),and Cyclooxygenase-2(COX-2)in gastric tissues.The ulcer index,biochemical parameters,and inflammatory proteins were evaluated.In vitro,GES-1 cells were induced by anhydrous ethanol to found gastric ulcer model.The groups were the same as in vivo experiment.Cytotoxicity was tested by MTT and IL-1β,TNF-α,and IL-6 were detected by Elisa assays.[Results]The injuries of gastric tissue in the model group were unambiguously observed and improved after YSJ treatment.The levels of interleukin 1 beta(IL-1β),interleukin 6(IL-6),interleukin 8(IL-8),and TNF-αin serum and tissues were decreased(P<0.05 or 0.01),whereas interleukin 4(IL-4)and prostaglandin E 2(PGE 2)were increased after treatment with YSJ(P<0.05 or 0.01).The levels of IL-1β,IL-6,TNF-α,and IL-8 were decreased in YSJ groups,while IL-4 and PGE 2 presented the opposite trend.The protein expression of p-NF-κB,NF-κB,p-IκBα,IκBα,and TNF-αwas inhibited after treatment with YSJ(P<0.05 or 0.01).[Conclusions]These results demonstrate that YSJ alleviates the occurrence and development of gastric ulcers via inhibiting inflammation. 展开更多

[Objectives]To study the biological characteristics and culture conditions of Hericium coralloides(Scop.)Pers.[Methods]The wild H.coralloides strain of Changbai Mountain was used as the experimental material to conduct a single factor test on its biological characteristics.[Results]The ideal available carbon source and nitrogen source are glucose and ammonium sulfate,respectively,the pH is 3.5-4.0,and the temperature is 20-22℃.Through the orthogonal experiment of 4 factors and 4 levels,it is found that the degree of influence of the 4 factors on H.coralloides is temperature>pH>nitrogen source>carbon source.[Conclusions]The optimum conditions for the mycelium culture of H.coralloides:the carbon source is glucose,and the nitrogen source is yeast extract or peptone,and pH is 3.5-4.0,and temperature is 20-22℃. 展开更多

Objective:To explore the therapeutic effects of the novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan(A.hygrometricus)on human acute lymphoblastic leukemia cells.Methods:Extensive screening of the antiproliferative and chemopreventive potential of different extracts from 5 wild mushrooms,A.hygrometricus,Phallus sp.,Lentinus sp.,Tricholoma sp.,and Serpula sp.was performed against a panel of 6 cancer cell lines and normal cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Apoptosis determination,cell cycle profiling,intracellular reactive oxygen species(ROS)and reactive nitrogen species(RNS),and mitochondrial membrane potential were analyzed by flow cytometry.The activity of caspases was measured colorimetrically,and the expression pattern of mitochondrial proteins was analyzed.Results:The methanol extract of A.hygrometricus and MOLT-4 cells were identified as the most potent extract exhibiting antiproliferative activity and most sensitive cell line,respectively.The mushroom extract induced robust selective apoptosis in MOLT-4 cells and arrested cell cycle progression at the G0/G1 stage.The extract disrupted the mitochondrial membrane potential and enhanced ROS production in MOLT-4 cells.The methanol extract induced apoptosis by downregulating the expression of Bcl-2,increasing the expression of Bax,and activating the caspase cascade.Conclusion:The novel wild edible mushroom is a potential repository of biomolecules for the development of antileukemic drugs. 展开更多

The Versatile Role of Period Circadian Regulators(PERs)in Oral Squamous Cell Carcinoma

This review explores the pivotal role of circadian rhythm regulators,particularly the PER genes,in Oral Squamous Cell Carcinoma(OSCC).As key constituents of the biological clock,PERs exhibit a downregulated expression pattern in OSCC,and the expression levels of PERs in OSCC patients are correlated with a favorable prognosis.PERs impact the occurrence and development of OSCC through multiple pathways.In the regulation of cell proliferation,they can function not only through cell cycle regultion but also via metabolic pathways.For example,PER1 can interact with receptors for activated C kinase 1(RACK1)and phosphatidylinositol 3-kinase(PI3K)through its PAS domain to inhibit glycolysis and thereby reduce cell proliferation.Regarding the regulation of cell death,PERs mediate various types of cell death in OSCC cells,such as p53-dependent apoptosis,protein kinase B(AKT)/mammalian target of rapamycin(mTOR)dependent autophagy,or hypoxia-inducible factor l-alpha(HIF-1a)mediated ferroptosis.In regulating epithelia-mesenchymal transition(EMT),PERs can lead to the downregulation of EMT related genes,such as zinc finger E-box binding homeobox 1/2(ZEBI/2),twist family BHLH transcription factor 1/2(TWIST1/2),and Vimentin,thereby influencing the migration and invasion capabilities of OSCC cells.In tumor angiogenesis,PERs exert regulatory effects on related factors,such as methionyl aminopeptidase 2(MetAP2)and vascular endothelial growth factor(VEGF).In the tumor immune microenvironment,PERs can inhibit the inhibitor of kappa B kinase(IKK)/nuclear factor kappa B(NF-kB)pathway and programmed cell death ligand 1(PD-L1)expression,thereby enhancing the cytotoxic effect of CD8+T cells on OSCC cells.In-depth studies focusing on elucidating the precise regulatory mechanisms of PERs can facilitate the development of therapeutic strategies targeting PERs,including restoration of PERs expression/activity,targeting PERs-regulated pathways,combination therapies,and chronotherapy.These furnish a theoretical foundation for formulating individualized treatment plans to achieve precise treatment for patients with OSCC. 展开更多

Hydroponic experiments were conducted in a growth chamber and changes in the hydraulic conductivity of sorghum (Sorghum vulgare Pers.) roots (Lpr) at the three-leaf stage were measured using the pressure chamber method. Water deficiency was imposed with polyethylene glycol (PEG) 6000 and the phosphorus (P) levels were controlled by complete Hoagland solution with and without P nutrient. The objective of this study was to investigate the effect of P nutrition on root Lpr under water deficiency. The results showed that the Lpr in P deficiency treatments decreased markedly, but the Lpr recovered to the same value as that of control when sufficient P was supplied for 4-24 h. Water deficiency decreased Lpr, but the hydraulic conductivity of the roots with sufficient P supply was still higher than that of plants without P supply. When resuming water supply, the Lpr of the water-deficient plants under P supply recovered faster than that of plants without P supply, which indicates that plants with sufficient P nutrient are more drought tolerant and have a greater ability to recover after drought. The treatment of HgCl2 indicated that P nutrient could regulate the Lpr by affecting the activity and the expression levels of aquaporins. 展开更多

[ Objective ] The study systematically studied the biological characteristics of ginseng Botrytis cinerea Pers. [ Method ] The pure pathogenic fungus was isolated from ginseng B. cinerea collected in the field by tissue segregation and purification cultivation. Subsequently, using PDA medium plate culture method, the effect of various culture conditions on mycelium growth and sporulation of ginseng B. cinerea was detected. [ Result] The optimum temperature for mycelium growth and sporulation of B. cinerea was 25 ℃. The appropriate temperature for conidia germination ranged from 20 to 25 ℃. And the optimum pH value for mycelium growth and conidia germination was 6.0. The optimum carbon source was sucrose, followed by glucose and fructose. The optimum nitrogen source was peptone, fol- lowed by beef extract, yeast extract, alanine and ammonium nitrate. Among the media, the growth of mycelium cultured on PDA medium was the fastest with the production of gray mycelium and dense colonies. Lethal temperatures for sclerotia, mycelium and conidia were 60, 55 and 50 ℃, respectively. [ Conclusion] The study provided the scientific basis for the research on the incidence law of B. cinerea and its control. 展开更多

On the MS medium supplemented with 2 ppm 2,4-D,calli were induced after 4-6weeks from the petioles of an American celery plant (Apium graveolens var.Dulce pers.cv.Florida).Suspension culture was started from the calli in a hormone-free liquid MS medium on agyratory shaker at 110 rpm,and kept at 26℃.To stimulate cell division and dedifferentiation,thesubcultures were conducted for 7 days each on the same medium.The liquid suspension containingsingle cells,cell aggregates,and somatic embryos in different stages were screened 2-3 weekslater and 1.0-1.5mm somatic embryos were obtained.These embryos were encapsulated withsodium alginate by dropping-bead method and solidified with 0.1mol CaCl<sub>2</sub>,.These synthetic seedsgerminated and developed well into seedlings in the sterilized vermiculite substrate. 展开更多

为了探究不同施肥处理对羊肚菌[Morchella esculenta(L.)Pers.]产量及经济效益的影响,设置不施肥为对照组,以黄腐酸生物有机肥(10 000 kg/hm^(2))、有机酵素肥(10 000 kg/hm^(2))、草炭(5 000 kg/hm^(2))、草木灰(5 000 kg/hm^(2))为试... 为了探究不同施肥处理对羊肚菌[Morchella esculenta(L.)Pers.]产量及经济效益的影响,设置不施肥为对照组,以黄腐酸生物有机肥(10 000 kg/hm^(2))、有机酵素肥(10 000 kg/hm^(2))、草炭(5 000 kg/hm^(2))、草木灰(5 000 kg/hm^(2))为试验组,对羊肚菌子实体产量、形态特征进行测定,并计算其经济效益及产投比。结果表明,不同施肥处理中羊肚菌子实体产量(干重)从高到低依次为草炭(1 335.38 kg/hm^(2))、草木灰(1 170.54 kg/hm^(2))、对照(1 096.03 kg/hm^(2))、有机酵素肥(881.62 kg/hm^(2))、黄腐酸生物有机肥(855.49 kg/hm^(2))。草炭和草木灰处理中羊肚菌产量明显提高,且草炭处理中羊肚菌的整体表现要优于草木灰处理,而有机酵素肥、黄腐酸生物有机肥处理中羊肚产量不增反降。与对照处理相比,草炭处理羊肚菌产量(干重)增加21.84%,总产值增加21.84%,经济效益增加24.29%,产投比增加5.85%。施用草炭有利于提高羊肚菌产量和经济效益。 展开更多

为改善竹荪单作产量逐年降低问题,以长裙竹荪[Dictyophora indusiata(Vent.Pers.)Fisch.]和羊肚菌(Morchella vulgaris)为材料,设计长裙竹荪单作、羊肚菌单作、长裙竹荪-羊肚菌轮作、长裙竹荪-圆白菜轮作、羊肚菌-辣椒轮作对比试验,连续... 为改善竹荪单作产量逐年降低问题,以长裙竹荪[Dictyophora indusiata(Vent.Pers.)Fisch.]和羊肚菌(Morchella vulgaris)为材料,设计长裙竹荪单作、羊肚菌单作、长裙竹荪-羊肚菌轮作、长裙竹荪-圆白菜轮作、羊肚菌-辣椒轮作对比试验,连续3年测定单位面积长裙竹荪和羊肚菌产量和部分品质指标。结果表明,与单作相比,轮作能够一定程度上提高长裙竹荪和羊肚菌产量及品质,其中羊肚菌-长裙竹荪轮作下二者的综合表现最好。羊肚菌-长裙竹荪轮作中长裙竹荪产量、单位面积子实体平均产出数量、去盖单株鲜重在栽培第2年和第3年均较单作显著增加,菌盖鲜重、菌高、菌柄直径和伞裙长度均显著增大,灰分、粗蛋白、粗脂肪和粗纤维含量较单作也略有增加,但二者间差异不显著;在栽培第2年和第3年,羊肚菌-长裙竹荪轮作下羊肚菌产量、子实体平均单株鲜重、单位面积子实体平均产出数量均较单作显著增加,羊肚菌灰分、粗脂肪和粗纤维含量均较单作显著增高,羊肚菌伞裙长度和菌柄直径在栽植第3年也较单作有所增大,菌盖与菌柄比大幅提高。羊肚菌-长裙竹荪轮作下羊肚菌形态、质量均显著提升。 展开更多

羊肚菌[Morchella esculenta(L.)Pers.]在我国分布极其广泛,有关其种类分布、生态环境、资源调查与开发利用等方面均有研究报道。2013—2016年,采用区域调查和定点调查的研究方法,对甘肃甘南藏族自治州境内野生羊肚菌的种类、自然分布... 羊肚菌[Morchella esculenta(L.)Pers.]在我国分布极其广泛,有关其种类分布、生态环境、资源调查与开发利用等方面均有研究报道。2013—2016年,采用区域调查和定点调查的研究方法,对甘肃甘南藏族自治州境内野生羊肚菌的种类、自然分布和生态环境等资源学方面进行了调查研究。调查结果显示:(1)该地区有7种野生羊肚菌,分别为羊肚菌(Morchella esculenta)、粗柄羊肚菌(Morchella crassipes)、尖顶羊肚菌(Morchella conica)、黑脉羊肚菌(Morchella angusticeps)、小羊肚菌(Morchella deliciosa)、紫褐羊肚菌(Morchella purpurascens)和半开羊肚菌(Morchella semilibera),其中紫褐羊肚菌(Morchella purpurascens)和半开羊肚菌(Morchella semilibera)为甘肃省新纪录种。(2)该地区野生羊肚菌发生季节为每年的4—5月,春夏之交的雨后广泛生于桦、栎等阔叶林中及林缘空旷草丛中,并且每年的10—11月也偶有发生。(3)该地区野生羊肚菌大多发生在壤质潮土、粘质潮土和褐壤腐殖质较多的土壤中,土壤中富含微量元素镁、铁、锰、铜、锌、硼、钼等。(4)该地区野生羊肚菌发生温度一般在4.4~16.8℃,相对湿度在75%左右,若阴、晴天交替和有足够的新鲜空气,则非常有利于羊肚菌子实体的生长。 展开更多

Pers.]进行筛选敏感度和抗真菌能力的比较。测试了狗芽根种子在温度、培养基量、生长时期3种不同条件下对标准化合物的敏感度。结果表明,狗芽根比早熟禾对除草剂标准化合物更敏感,且有更强的抗真菌污染能力。狗芽根种子在25℃的条件下,对除草剂标准化合物的敏感度最高;减少水琼脂用量,即提高样品的终浓度,等同于提高狗芽根筛选的敏感度;狗芽根种子培养48 h后,对标准化合物敏感度最高。对1 880个微生物源提取物进行筛选,表明狗芽根作为单子叶杂草,用于先导化合物筛选具有稳定、高效、敏感度高等突出优点;狗芽根是目前单子叶除草剂先导化合物筛选的首选靶标。 展开更多
