Objective:To explore the mechanism of the Peiyuan Jieyu formula in treating depression by assessing its impact on a lipopolysaccharide-induced(LPS-induced)depression mouse model.Methods:We created a mouse model of dep...Objective:To explore the mechanism of the Peiyuan Jieyu formula in treating depression by assessing its impact on a lipopolysaccharide-induced(LPS-induced)depression mouse model.Methods:We created a mouse model of depression by exposing mice that had previously received chronic stress to intraperitoneal LPS injections.The mice were divided into the following groups:control,model,fluoxetine,Tiansi Yin,Sini powder,and low-,medium-,and high-dose Peiyuan Jieyu formula groups.Forced swim and tail suspension tests were used to assess the efficacy of the depression(despair)model,and weight gain rates were also measured.Furthermore,serum levels of various depression and inflammation-associated molecules,including tumor necrosis factor-a(TNF-a),interferon-γ(IFN-γ),tryptophan,5-hydroxytryptamine,kynurenine(KYN),and kynurenic acid(KA)were assessed.Furthermore,the expression levels of ionic calcium-binding adaptor molecule-1(IBA-1)and indoleamine 2,3-dioxygenase(IDO)mRNA in hippocampal microglia were measured.Results:The model group displayed greater despair-associated immobility,which was shortened in response to various doses of Peiyuan Jieyu formula.Furthermore,formula administration significantly reduced serum TNF-a levels and hippocampal IDO mRNA expression.The high formula dose also reduced IFN-γand IBA-1 levels,the latter was also decreased in response to the medium formula dose.However,the low formula dose reduced serum KYN level and KYN/tryptophan(TRP)and KYN/KA ratios.Conclusion:The Peiyuan Jieyu formula holds immense potential in treating depression in a mouse model,potentially inhibiting inflammation and improving TRP-KYN metabolic disorders.展开更多
Objective:To study the intervention effect of Peiyuan Huayu Decoction on the neuron damage in model rats with acute subdural hematoma (ASDH).Methods: 160 SD rats were randomly divided into four groups, and the ASDH mo...Objective:To study the intervention effect of Peiyuan Huayu Decoction on the neuron damage in model rats with acute subdural hematoma (ASDH).Methods: 160 SD rats were randomly divided into four groups, and the ASDH model rats were made by stereotactic autoblood injection, and sham operation group received craniotomy without blood injection. Sham operation group and model group were normally bred after model establishment, and 6 h after model establishment, the treatment group received intragastric administration of Peiyuan Huayu Decoction, and control group received intragastric administration of Piracetam Tablets, 1 time a day. On the 1d, 3d, 5d and 7d after model establishment, the general conditions of rats (activity, food intake and mental state) were observed, blood was collected via auricula dextra, ELISA method was used to determine peripheral plasma NSE and S100β protein contents, routine HE staining was conducted after perfusion fixation, the neurons in blood injection side of brain tissue were counted, and the neuron damage was observed.Results: 26 rats were dead in the experiment. The general conditions of sham operation group were significantly better than those of other groups, treatment group was significantly better than model group and control group on the 5d group (P<0.05), and there was no significant difference on the 1d, 3d and 7d (P>0.05);neuron count of sham operation group was basically stable, treatment group was not different from model group and control group on the 1d (P>0.05), treatment group was better than model group (P<0.05), and not different from control group (P>0.05) on the 3d, and treatment group was better than model group and control group on the 5d and 7d (P<0.05);peripheral plasma S100β protein and NSE contents of sham operation group were at lower levels, treatment group was not significantly different from model group and control group on the 1d (P>0.05), S100β protein and NSE contents decreased significantly on the 3d, and treatment group was significantly different from model group and control group (P<0.05), S100β protein and NSE contents increased on the 5d and 7d, the increase in treatment group was slower than that in model group and control group, and there was significant difference (P<0.05).Conclusion:Peiyuan Huayu Decoction has obvious protective effect on the neurons in ASDH model rats, and this effect may be based on the inhibition of secondary neuron damage.展开更多
Objective:To study the effective compound action target signal pathway of Peiyuan bushenan taifang(PYBSATF)has achieved good clinical efficacy in the treatment of recurrent spontaneous abortion(RSA),but its mechanism ...Objective:To study the effective compound action target signal pathway of Peiyuan bushenan taifang(PYBSATF)has achieved good clinical efficacy in the treatment of recurrent spontaneous abortion(RSA),but its mechanism has not been clarified because of its complex components.In this study,network pharmacology is applied to study the effective compounds,targets and signal pathways of PYBSATF in the treatment of RSA,so as to reveal its pharmacological mechanism of action in the treatment of recurrent spontaneous abortion.Methods:The Traditional Chinese Medicine Systems Pharmacology database(TCMSP)and CNKI are used to obtain the main compounds and drug action targets of PYBSATF.Genecards and the Online Mendelian Inheritance of Man databases(OMIM)are searched to collect the known genes related to RSA,so as to construct compound-target network and screen out the common target proteins and main active compounds.We also use string database to construct a visual protein-protein interaction network(PPI).Cluster Profiler and R software were used to analyze the common targets of drugs and diseases for GO function analysis and KEGG pathway enrichment analysis.Finally,the compound and the protein sequences were conducted according to the node parameters,so that the core protein and core compounds are used to perform molecular docking.Results:186 potential active components and 65 predicted action targets of PYBSATF were screened out.At the same time,1658 genes were also screened out to be closely related to RSA,among which 65genes overlaped with PYBSATF targets and were considered to be related to way of treatment.PPI network showed that VEGFA,IL6,EGFR,MAPK8 and ESR1were the core targets of PYBSATF for the treatment of RSA.GO and KEGG enrichment analysis obtained 93 biological processes of cells(P<0.01)and 87 signaling pathways(P<0.01).PYBSATF played a pharmacological role through a variety of pathways including anti-inflammatory,anti-apoptosis,promoting proliferation and angiogenesis.Molecular docking showed that most active components and key targets of PYBSATF had strong efficiency.Conclusion:Through the study of network pharmacology,it predicted that PYBSATF might treat RSA through multiple targets and multiple signal pathways.Significantly,the predictive targets may be potential targets for treatment of RSA.展开更多

Objective To observe the therapeutic effect of Piwei Peiyuan Decoction(PWPYD)in treating chronic atrophic gastritis patients with Pi-Wei weakness type accompanied with intestinal metaplasia.Methods A total of 100 chro...Objective To observe the therapeutic effect of Piwei Peiyuan Decoction(PWPYD)in treating chronic atrophic gastritis patients with Pi-Wei weakness type accompanied with intestinal metaplasia.Methods A total of 100 chronic atrophic gastritis patients with Pi-Wei weakness type accompanied with intestinal metaplasia were included in this study from June 2022 to December 2023 at the Second Affiliated Hospital of Anhui University of Chinese Medicine.Totally 96 patients met the inclusion criteria and were randomly assigned to the control group and the treatment group,48 cases in each group.The patients in the treatment group were treated with PWPYD,and those in the control group were treated with folic acid tablets.The treatment course lasted for 24 weeks.The changes of pathological scores,Chinese medicine(CM)symptom scores,and gastroscopic scores,serum pepsinogen I(PG I),pepsinogen II(PG II),gastrin-17(G-17),interleukin-6(IL-6),IL-10,and tumor Necrosis factor-Alpha(TNF-α)levels were observed before and after treatment between the two groups.Meanwhile,Western Blot and RT-qPCR were used to detect the levels of cystine glutamate reverse transporter(xCT),glutathione eroxidase 4(GPX4),and peroxisome proliferator activated receptorα(PPARα)expressions.Results Compared with baseline,pathological scores,CM symptom scores,and gastroscopic scores decreased in the two group after treatment(P<0.05).The serum PG I,PG II,G-17,and IL-10 expression levels increased(P<0.05),and the serum IL-6 and TNF-αexpression levels decreased(P<0.05).The protein and mRNA levels of xCT,GPX4,and PPARαin the focal tissue decreased(P<0.05).Compared with the control group after treatment,pathological scores and gastroscopic scores of mucous membrane color,mucous membrane texture decreased(P<0.05),CM symptom scores of gastric distension and fullness,gastric pain,low food intake and nausea,fatigue and weakness,and shortness of breath and laziness decreased in the treatment group(P<0.05).The expression levels of serum PG I,G-17,IL-6,IL-10,and TNF-αin the treatment group were better than in the control group(P<0.05).The protein and mRNA levels of xCT,GPX4,and PPARαin lesion tissues in the treatment group were lower than those in the control group(P<0.05).Conclusion By regulating the expression levels of serum inflammatory factors and the protein and mRNA expression levels of xCT,GPX4,and PPARαin the lesion tissue,PWPYD can improve the clinical symptoms and pathological changes of the gastric mucosa of the patients,and improve their quality of life.展开更多

Materials and Methods:...Objective: To explore the effects of Peiyuan Tongnao(PYTN) Capsule on working memory, the content of glutamic acid, and the expression of NMDA receptor 2 B(NR2 B) in rats with cerebral ischemia. Materials and Methods: 52 Sprague-Dawley rats were randomly divided into sham group, model group, nimodipine group, and PYTN group. The bilateral common carotid artery occlusion(BCAO) was performed to establish rat model with cerebral ischemia in the model, nimodipine, and PYTN groups. Gastric lavage with some drug based on body weight conversion was performed daily for 4 weeks in the nimodipine and PYTN groups. The working memory of the rats was tested by Morris water maze. The expression of Nissl body in hippocampus tissue was observed by Nissl staining. The determination of Glu content in hippocampus was detected by high-performance liquid chromatography. The expression level of NR2 B in hippocampus area was determined using Western blot. Results: Morris water maze test on working memory escape latency. In the model group, day 1 versus day 4, there was a statistical difference(P < 0.05). In the nimodipine group, day 1 vs. day 4 displayed a statistical difference(P < 0.05).In the PYTN group, day 1 versus day 3 and day 1 versus day 4 were in significant differences(P < 0.05). The Glu content in hippocampus of the sham group was significantly different from that of model group(P < 0.05). The Glu content in the PYTN group was significantly different from that of the model group(P < 0.05). With regard to the expression of NMDA 2 B in hippocampus between the sham group and the model, nimodipine, and PYTN groups, all were displayed statistical significance(P < 0.01). As the same, the expression of NMDA2 B in the model group was significant from that of the nimodipine group(P < 0.05) and PYTN group(P < 0.01). Conclusion: PYTN capsule was beneficial for improving working memory and protect neural cells in rats of cerebral ischemia, which may be associated with upregulation of the expression of Glu and NMDAR2 B in hippocampus.展开更多
