RCN2和PEAK1在结直肠癌中的表达与临床病理特征及预后的关系 被引量:2

目的:检测网织钙结合蛋白2(RCN2)和伪足富集的非典型激酶1(PEAK1)蛋白在结直肠癌组织中的表达情况,分析RCN2和PEAK1表达与患者临床病理特征和预后的关系。方法:免疫组织化学法检测90例结直肠癌组织及其癌旁正常组织中RCN2和PEAK1蛋白表... 目的:检测网织钙结合蛋白2(RCN2)和伪足富集的非典型激酶1(PEAK1)蛋白在结直肠癌组织中的表达情况,分析RCN2和PEAK1表达与患者临床病理特征和预后的关系。方法:免疫组织化学法检测90例结直肠癌组织及其癌旁正常组织中RCN2和PEAK1蛋白表达情况,分析结直肠癌组织RCN2和PEAK1表达与患者临床病理特征的关系,Kaplan-Meier生存曲线分析RCN2和PEAK1表达对患者预后的影响,Spearman等级相关检验结直肠癌组织RCN2和PEAK1表达的相关性。结果:RCN2和PEAK1蛋白在结直肠癌组织中的阳性表达率均明显高于癌旁正常组织(P<0.05)。结直肠癌组织RCN2表达与肿瘤直径、浸润深度和TNM分期均有关(P<0.05),PEAK1表达与肿瘤浸润深度、淋巴结转移和TNM分期均有关(P<0.05)。Log Rank检验结果显示,RCN2阳性表达组和PEAK1阳性表达组患者的术后5年总生存率均分别低于RCN2阴性表达组和PEAK1阴性表达组患者(P<0.05)。结直肠癌组织RCN2和PEAK1表达呈正相关性(r=0.586,P=0.000)。结论:RCN2和PEAK1蛋白在结直肠癌组织中呈高表达,且均与肿瘤恶性进展和不良预后关系密切。RCN2和PEAK1可作为结直肠癌治疗靶标的候选分子。 展开更多

目的研究miR-378c对宫颈癌细胞增殖、迁移和愈合的影响及其对靶基因的调控作用。方法合成miR-378c的类似物(miR-378c mimics)和抑制物(miR-378c inhibitors)转染宫颈癌细胞SiHa,无miRNA转染细胞做阴性对照(negative control,NC),CCK-8... 目的研究miR-378c对宫颈癌细胞增殖、迁移和愈合的影响及其对靶基因的调控作用。方法合成miR-378c的类似物(miR-378c mimics)和抑制物(miR-378c inhibitors)转染宫颈癌细胞SiHa,无miRNA转染细胞做阴性对照(negative control,NC),CCK-8检测细胞增殖能力,Transwell和划痕实验检测细胞迁移和愈合情况,实时荧光定量PCR(qRT-PCR)检测Peak1和JAK2在各组细胞中的表达情况。结果划痕实验显示,与NC组比较,miR-378c mimics组SiHa细胞划痕愈合率明显增高(P<0.05),而miR-378c inhibitor组SiHa细胞划痕愈合率明显降低(P<0.05)。CCK-8实验显示,与NC组比较,miR-378c mimics组和miR-378c inhibitor组细胞增殖能力均没有明显改变(均P>0.05)。Transwell迁移实验显示,与NC组相比,miR-378c mimics组SiHa细胞迁移能力显著增强,而miR-378c inhibitor组细胞迁移明显抑制(均P<0.05)。qRT-PCR实验显示,与NC组相比,miR-378c mimics组Peak1基因表达显著降低(P<0.05),而JAK2基因不受影响(P>0.05);而miR-378c inhibitor组Peak1基因表达显著上升(P<0.05),JAK2基因表达量变化无统计学意义(P>0.05)。结论miR-378c通过靶向Peak1基因,促进宫颈癌细胞的迁移和愈合,而对宫颈癌细胞增殖影响不显著。 展开更多

A novel method based on the extended infomax approach of independent component analysis (ICA) is proposed for the single trial estimation of multi channel pattern reversal visual evoked potential (PVEP). The clinical applications illustrate the encouraging performance of the method. The number of trials needed is reduced to 3, but the estimated VEP complex is clearer than that obtained by conventional ensemble averaging with 50 trials. By analyzing the time course and spatial pattern of the independent components (ICs) of the multi channel PVEP, one component is found to be reasonably related to the P100 peak. 展开更多

It is verified that the phonon scattering process and the residual linewidthare the dominant factors of the linewidth of 2210 cm^(-1) IR absorption peak except the anomalous linewidth at 200 K. By investigating the anomalities of the peak shape and thelinewidth of the peak at 200 K, we put forward a mechanism that the T_d symmetry of defect-complex corresponding to the 2210 cm^(-1) peak can he transferred into the D_(2d) symmetry as temperature rises to above 200 K. The quantitative analysis shows that the V+4H-model is indeed of two states: The T_d configuration is stable at temperature lower than 200 K, while the D_(2d) one is stable at temperature higher than 200 K. We can draw the conclusion that the V +4H-model corresponds to the 2210 cm^(-1) IR absorption peak from the symmetric breaking mechanism, which can quantitatively fit the experimental results. 展开更多
