Peaches (Prunus persica L.) pulp mitigate type 2 diabetic mice by modulating of glucose metabolism and gut microbiota

Peach are a fruit with high nutritional and economic value,but their safety and suitability for diabetic patients have been questioned.This study investigated the effects and potential mechanisms of peach pulp(PP)on type 2 diabetes mellitus(T2DM)in mice induced by a high-fat diet(HFD)combined with streptozotocin(STZ).The results showed that PP alleviated hyperglycemia,hyperlipidemia,hyperuricemia,and tissue dysfunction in T2DM mice through the synergistic effect of nutrients and non-nutrient compounds.Analysis of mRNA expression levels revealed that PP improved glucose metabolism in T2DM mice by promoting glycogen synthesis and inhibiting gluconeogenesis.Furthermore,elevated levels of PP resulted in an increase in acetic acid content following a 4 weeks intervention period.Additionally,it led to the restoration of gut microbiota balance by decreasing the Firmicutes/Bacteroidota(F/B)ratio and enhancing the presence of Romboutsia,Allobaculum,Alloprevotella,and Bacteroides after an 8 weeks intervention.Ultimately,our results suggest that PP may offer advantages for individuals with diabetes. 展开更多

[Objectives]To clarify the effects of varying potassium application rates on leaf growth and soil mineral nutrients of greenhouse peaches,and to identify the optimal potassium fertilizer dosage.[Methods]Using the greenhouse peach superior line C26-7-17 as the test material and maintaining a fixed ratio of nitrogen and phosphorus fertilizers,five potassium fertilizer treatment groups were established.The application rates of potassium sulfate were 0,67,135,202,and 269 g/plant,respectively.The regulatory effects of varying potassium application rates were analyzed by measuring leaf area,chlorophyll content,and soil mineral element content.[Results]Applying nitrogen,phosphorus,and potassium fertilizers in specific proportions increased the leaf area of greenhouse peaches,enhanced chlorophyll content,and significantly improved the soil s nutritional status.Considering the overall optimization of soil nutrients,the recommended potassium fertilizer dosage under the experimental conditions was 202 g/plant.[Conclusions]This study offers theoretical insights that may enhance the quality and efficiency of greenhouse peaches,as well as inform nutrient management strategies. 展开更多

Late-mature peaches (Prunus persica L. Batsch, cv. Dongxuemi) were stored in air, modification atmosphere (MA) and controlled atmospheres (CA) at 1℃ to investigate enzymatic activities, flesh browning, quality attributes and fruit storability during storage periods. Peaches stored in CA conditions showed significant lower activities of polyphenol oxidase (PPO) and peroxidase (POD) compared to that kept in MA and in air during the early period of storage. CA treatments indicated a better result in maintaining fruit firmness, color and titratable acidity than MA and control treatments. But there was no significant difference in vitamin C and total soluble solids of peaches stored in CA, MA and air. Increasing PPO activity obviously stimulated flesh browning. Peach fruits stored in CA conditions showed a lower activity of POD and higher firmness in the early period of storage. The peaches stored in 5%CO2 + 5%O2 and 10%CO2 + 5%O2 for 92 days had a good quality without any off-flavor. The results indicated CA conditions obviously inhibited the enzymatic browning, delayed senescence and maintained quality of late-mature peaches. 展开更多

Zhaohui peaches(Prunus persica Batsch)were treated with 0(CK), 1, 10 or 100 μmol L-1 methyl jasmonate(MeJA)vapor at 20℃ for 24 h before stored at 0℃ for 35 d. The untreated fruits showed chilling injury(CD symptoms after 4 wk of storage, as indicated by increased fruit firmness and reduced ex-tractable juice, which is referred as leatheriness. Treatment with 1 and 10 μmol L-1 MeJA promoted normal ripening and softening, maintained higher levels of extractable juice, titratable acidity, pectinesterase(PE)and polygalacturonase(PC)activities, inhibited increases in fruit fresh weight loss, decay incidence, electrolyte leakage and MDA content, and improved color development, thereby preventing chilling injury symptoms development and maintaining edible quality. MeJA treatment also delayed the climacteric rise in respiratory rate, but promoted ethylene production during the later period of cold storage, suggesting that ethylene may involve in CI development of peaches. These results indicate that 1 and 10 μmol L-1 MeJA treatments could be used to reduce CI development and decay incidence in peaches. 展开更多

The use of near infrared (NIR) spectroscopy was proved to be a useful tool for quality analysis of fruits. A bifurcated fiber type NIR spectrometer, with a detection range of 800-2500 nm by InGaAs detector, was used to evaluate the firmness of peaches. Anisotropy of NIR spectra and firmness of peaches in relation to detecting positions of different parts (including three latitudes and three longitudes) were investigated. Both spectra absorbency and firmness of peach were influenced by longitudes (i, ii, iii) and latitudes (A, B, C). For modeling, two thirds of the samples were used as the calibration set and the remaining one third were used as the validation or prediction set. Partial least square regression (PLSR) models for different longitude and latitude spectra and for the whole fruit show that collecting several NIR spectra from different longitudes and latitudes of a fruit for NIR calibration modeling can improve the modeling performance. In addition, proper spectra pretreatments like scattering correction or derivative also can enhance the modeling performance. The best results obtained in this study were from the holistic model with multiplicative scattering correction (MSC) pretreatment, with correlation coefficient of cross-validation γcv=0.864, root mean square error of cross-validation RMSECV=6.71 N, correlation coefficient of calibration r=0.948, root mean square error of calibration RMSEC=4.21 N and root mean square error of prediction RMSEP=5.42 N. The results of this study are useful for further research and application that when applying NIR spectroscopy for objectives with anisotropic differences, spectra and quality indices are necessarily measured from several parts of each object to improve the modeling performance. 展开更多

Peach, an economically important model plant of the Rosaceae family, has been domesticated and cultivated in China for approximately5 000 years. The Hexi Corridor, an important corridor connecting east-central China and the Tarim Basin, is the starting point of the Silk Road that links China and the Eurasian region. As a globally distributed fruit tree, the spread of peach was accomplished through historical trade routes in the Hexi Corridor and the Tarim Basin. However, knowledge of peach genetic diversity in these regions remains limited. In this study,we examined the relationships and the spread history of domesticated peaches through sequencing and genomic analysis of 161 peach accessions collected from Northwest China, including 43 from the Hexi Corridor and 104 from the Tarim Basin. The results indicate that peach landraces in the Hexi Corridor and the Tarim Basin are derivatives of peaches from the east and south of China (ESC). Notably, the genetic diversity of accessions from both the Hexi Corridor and the Tarim Basin was lower than that of ESC accessions. Reduction of diversity (ROD) and linkage disequilibrium (LD) analyses detected a genetic bottleneck in peaches from these regions. Additionally, these peaches have undergone varying degrees of selection from natural environment. Moreover, genes responsive to biotic and abiotic stresses were under selection, which could be the result of the climate change of Northwest China after the Last Glacial Maximum (LGM). Our findings provide a better understanding of the genetic basis of peach migration in Northwest China. Furthermore, this study expands the available genomic data for peaches and provides critical information for future peach breeding programs. 展开更多

Residue patterns of buprofezin and teflubenzuron in treated peaches

The biological half-life and final residue levels of buprofezin and teflubenzuron were examined in peaches over a 14-day cultivation period. The residue levels of buprofezin and teflubenzuron were analyzed by chromatographic method with recovery ranging from 84.0% to 96.6%. The biological half-lives of buprofezin andteflubenzuron were 4.88 and 11.49 days at the standard dose, and 4.40 and 10.86 days at a triple dose, respectively. The initial concentration of buprofezin exceeded the maximum residue limit (MRL) set in Korea, but the concentration decreased to below the MRL within 6 days after application. The initial and persisting concentrations of teflubenzuron were all below the prescribed MRL. The final residue levels of buprofezin and teflubenzuron were 0.17 and 0.10?mg·kg﹣1 following a standard single dose, and 0.20 and 0.23 mg·kg﹣1 following a triple dose, respectively. The final re-sidue levels of buprofezin and teflubenzuron were also compared with the good agricultural practices standards of the United States and Italy. 展开更多

Softening of fleshy fruits during ripening and postharvest is a programmed physiological process that substantially impacts fruit quality and shelf life.However,the molecular mechanism underlying peach softening remains largely unknown.Lateral organ boundary(LOB)domain(LBD)proteins are pivotal regulators of plant growth and development.To date,certain LOB/LBD transcription factors are seemingly implicated in fruit softening.In this study,we identified 42 LOB/LBD genes in the peach genome.Expression analysis showed a significant upregulation of PpLOB1 transcripts toward peach fruit ripening.PpLOB1 was classified into Class II subgroup,and showed high sequence similarity to several softening-related LOB/LBD transcription factors.Transient transformation assays showed that PpLOB1 positively modulates peach softening.Further experiments demonstrated that PpLOB1 directly targets and activates the promoters of pectate lyase 1(PpPL1)and PpPL15,thereby contributing to the regulation of fruit softening.Additionally,PpNAP4 up-regulates PpLOB1 expression by binding to its promoter.Meanwhile,our findings revealed that PpNAP4 and PpNAP6 cooperatively modulate the expression of Pp LOB1.Taken together,our findings revealed a novel regulatory module involving PpNAP4 and PpLOB1 that modulates peach fruit softening. 展开更多

Integrated transcriptome and metabolome analysis reveals light induction of anthocyanin rapid accumulation in red-fleshed peach after pre-harvest bagging

Red-fleshed fruits are valued for their vibrant color and high anthocyanin content.Pre-harvest fruit bagging enhances fruit peel pigmentation,but its effect on flesh coloration remains poorly characterized.This study revealed that removing bags from‘Gengcunyangtao’red-fleshed peach fruits triggers the rapid and uniform accumulation of anthocyanins in the flesh,resulting in anthocyanin levels that exceed those in unbagged fruits.The exposure to light after bag removal triggered significant increases in anthocyanin levels within 24 h.This was accompanied by the rapid upregulation of light-responsive and flavonoid biosynthetic gene expression levels within 6 h.A metabolomic analysis indicated that anthocyanin precursors,especially p-coumaric acid,accumulated before bag removal,thereby increasing substrate availability for rapid anthocyanin synthesis.On the basis of a weighted gene co-expression network analysis,MYB transcription factors,anthocyanin transporters,glutathione S-transferase,and multidrug and toxic compound extrusion(MATE)were identified as key regulators that coordinate precursor storage along with light-induced transcriptional activation.Notably,PpMYB4 binds to the promoter of PpGSTF14 and activates its expression,thereby promoting anthocyanin accumulation.The study findings elucidated the temporal coordination of metabolic priming and light-responsive transcriptional regulation driving rapid anthocyanin biosynthesis,with possible implications for improving peach fruit flesh coloration. 展开更多

[Objectives]To analyze the microbial community structure and diversity in the rhizosphere soil of peach trees in the Tangshan area of Hebei Province,identify the dominant microbial groups,and explore their potential ecological functions.[Methods]Amplification sequencing analysis of bacterial and fungal communities in the rhizosphere soil of a peach orchard in Qian'an County,Tangshan City,Hebei Province,was performed using Illumina MiSeq high-throughput sequencing technology.[Results]The indices of Sobs,Chao,ACE,and Shannon for soil bacteria in the rhizosphere soil of peach trees were all higher than those for fungi,indicating a more uniform and diverse bacterial community structure.At the phylum level,the bacteria with relatively high abundance included Pseudomonadota(28.29%),Acidobacteriota(18.10%),Bacillota(12.17%),and Actinomycetota(11.73%).In contrast,the fungi with relatively high abundance were Ascomycota(64.64%),Basidiomycota(14.22%),and Mortierellomycota(14.09%).At the genus level,the bacteria with relatively high abundance comprised Sphingomonas(5.00%),Priestia(3.38%),Nitrospira(2.05%),etc.The fungi with relatively high abundance included Fusarium(13.13%),Mortierella(12.86%),Tausonia(6.97%),Neocosmospora(4.77%),etc.[Conclusions]This study offers a foundational dataset and theoretical reference for the regulation of rhizosphere microecology and the management of soil health in peach orchards in Tangshan. 展开更多

In order to recognize the key flavor compounds influencing the consumers' choice for honey peach, nectarine, and flat peach and provide important information for orientation breeding, sugars, organic acids, and aroma volatiles in 3 types of peaches were examined by high performance liquid chromatography(HPLC) and gas chromatography–mass spectrometry(GC–MS) and their key individual characteristic flavor compounds were identified by multivariate analysis integrated with consumers' evaluation. No significant differences in sugar and organic acid were observed between 3 types of peaches. In hierarchical clustering analysis(HCA) and principal component analysis(PCA)models, 3 types of peaches could not be differentiated by sugars or organic acids, but could be identified completely by aroma volatiles or by flavor compounds. A partial least squares regression(PLSR) model revealed that the key individual characteristic volatiles in nectarine, honey peach, and flat peach are C9 compounds and terpenic compounds, norisoprenoids and C6 compounds, and benzaldehyde, γ-decalactone, andδ-dodecalactone, respectively. These results suggest that sugars and organic acids form the background flavor of peach fruit and lactones shape the typical peach aroma. However, the individual aroma volatiles play the decisive role in unique flavor determination of different types of peaches. 展开更多

The objectives of this research were to compare the performance of each individual nondestructive sensor with the destructive sensor,and to apply sensor fusion technique to explore whether a combination of sensors would give better results than a single sensor for classification of peach firmness.Tests were carried out with four peach varieties namely Royal Glory,Caterina,Tirrenia and Suidring.In this research,the three nondestructive firmness sensors acoustic firmness,low-mass impact and micro-deformation impact were used to measure firmness.A Bayesian classifier was chosen to provide a classification into three categories,namely soft,intermediate and hard.High level fusion technique was performed by using identity declaration provided by each sensor.The data fusion system processed the information of the sensors to output the fused data.The result of the high level fusion was compared with the classification provided by an unsupervised algorithm based on destructive reference measurement.The fusion process of the nondestructive sensors provided some improvements in the firmness classification;the error rate varied from 25%to 19%for individual sensor.Furthermore,the results of fusion process by using three sensors decreased the error rate from 19%to 13%.This research demonstrated that the fused systems provided more complete and complementary information and,thus,were more effective than individual sensors in the firmness classification of peaches. 展开更多

Bud endodormancy represents an ecological adaptation mechanism in perennial deciduous fruit trees to endure winter cold conditions.Sucrose serves a crucial role in bud endodormancy as both an energy metabolizer and signaling molecule.Sugars Will Eventually be Exported Transporters(SWEETs)function as sugar-efflux transporters that respond to environmental stimuli and contribute to plant growth and development.While SWEET gene families have been identified in various plant species for sugar transport regulation,their mechanism in regulating peach bud endodormancy remains undefined.In this study,we identified 15 SWEET genes in peach.The nomenclature was established through homologous alignment with the Arabidopsis SWEET gene family,resulting in four distinct clades through phylogenetic analysis.Covariance correlation analysis revealed 6 and 12 collinear SWEET genes in peach and Arabidopsis,respectively,forming 13 collinear gene pairs.Real-time quantitative polymerase chain reaction(RT-qPCR)analysis demonstrated significantly elevated expression of PpSWEET6 during peach bud endodormancy release,correlating positively with sucrose content.Transient overexpression of PpSWEET6 enhanced peach bud endodormancy release,while overexpressing PpSWEET6 in Arabidopsis enhanced seed germination and flowering.Y2H and luciferase complementation imaging(LCI)assays confirmed PpSWEET6 interacted with PpABF2.Additionally,dual luciferase reporter(DLR)assays showed that PpSWEET6 significantly decreased the activation of PpDAM6(key dormancy-inducing gene)through PpABF2,thereby modulating peach bud endodormancy release.These findings advance our understanding of SWEET genes in peach bud endodormancy regulation. 展开更多

Facile Preparation of Robust Peach Gum Polysaccharide with Remarkably Enhanced Antibacterial and Antioxidant Performance

Peach gum polysaccharide(PGP),a readily available natural polysaccharide,boasts substantial potential across diverse applications,yet its practical utility is severely limited by its vulnerability to bacterial growth and limited antioxidant activity.Herein,we introduced a simple and effective method to enhance the antibacterial and antioxidant properties of PGP by conjugating it with salicylic acid(SA).Cytotoxicity evaluation results confirmed that the resulting PGP-SA retains the excellent biocompatibility of PGP.Notably,PGP-SA demonstrates outstanding antibacterial activity against both Gram-positive(S.aureus)and Gram-negative(E.coli)bacteria,outperforming non-modified PGP.Its antibacterial mechanism is hypothesized to stem from disrupting bacterial cell membranes and proteins,targeting structures vital to microbial survival.Beyond fighting bacteria,PGP-SA also delivers robust antioxidant activity,efficiently scavenging ABTS radicals.Harnessing these dual enhancements,PGP-SA proves highly effective as a fruit preservation coating.Most impressively,it extends the post-harvest shelf life of mangoes:under identical storage conditions,PGP-SA-coatedmangoes stay fresh for seven extra days compared to uncoated fruit.With its simple synthesis process and standout performance,this work not only overcomes PGP’s key limitations but also opens new avenues for its application in food preservation and related industries. 展开更多
