P48-Triggered transmembrane signaling transduction of human monocytes:mobilization of calcium ion and activation of protein kinase C(PKC)

P48 is a cytokine which induces monocyte differentia-tion and the induction of cytotoxic activity. In this study,the signal transduction events involved in the stimulation of monocytes with the membrane form of P48 (mP48) were investigated. Monocyte stimulation with mP48 was found to involve the mobilization of intracellular calcium (Ca2+)and the activation and translocation of PKC from the cy-tosol to the membrane. Membane P48 induced a rapid rise of intracellular Ca2+ in a dose dependent maner. Simi-larly the stimulation of monocytes with P48 was found to involve the activation and translocation of PKC. The translocation of PKC was rapid (within 0-5 min) yet tran-sient with PKC activity returning to control levels by 8 min. The functional role of protein kineses in P48 induced TNF secretion was studied using various kinese inhibitors. The PKC inhibitors, H-7 and sphingosine, were found to inhibit P48 induced TNF secretion with 50% inhibition at 5μM HA1004, which inhibts cyclic nucleotide-dependent kinase (PKA, Ki 1.2μM), did not inhibit TNF secretion. H-8 (PKA inhibitor) was found to be an effective inhibitor of TNF secretion only at high concentrations(30μp. The Calmodulin-dependent kinase inhibitor, W7 (Ki 12μM)was found to be effective at concentration above 5μM.These findings suggest that P48-triggered TNF secretion involves transmembrane Ca2+ signaling and the subse-quent activation of at least two protein kineses, PKC and CaMK. 展开更多

In this study,37 derivatives of phorbol esters were synthesized and their anti-HIV-1 activities evaluated,building upon our previous synthesis of 51 phorbol derivatives.12-Para-electron-acceptor-trans-cinnamoyl-13-decanoyl phorbol derivatives stood out,demonstrating remarkable anti-HIV-1 activities and inhibitory effects on syncytia formation.These derivatives exhibited a higher safety index compared with the positive control drug.Among them,12-(trans-4-fluorocinnamoyl)-13-decanoyl phorbol,designated as compound 3c,exhibited the most potent anti-HIV-1 activity(EC_(50)2.9 nmol·L^(−1),CC50/EC_(50)11117.24)and significantly inhibited the formation of syncytium(EC_(50)7.0 nmol·L^(−1),CC50/EC_(50)4891.43).Moreover,compound 3c is hypothesized to act both as an HIV-1 entry inhibitor and as an HIV-1 reverse transcriptase inhibitor.Isothermal titration calorimetry and molecular docking studies indicated that compound 3c may also function as a natural activator of protein kinase C(PKC).Therefore,compound 3c emerges as a potential candidate for developing new anti-HIV drugs. 展开更多

Objective To investigate the biology function of novel protein related to bovine seminal plasma protein in human testis. Methods Recombination pcDNA3/HBRP was constructed and transfected to HEK293 cell and permanently expression cell line was established. The activity of protein kinase C (PKC) of the cell line was detected by autoradiography method. Results The stable expression cell line of HBRP was obtained. The HBRP inhibited the activity of PKC significantly. Conclusion One of the new functions of novel sperm binding protein in human is the inhibitor action on activity of PKC. It may be involved in the sperm capacitation and acrosome reaction. 展开更多
