Mechanism and Efficacy of the Combination of Spherical PDRN and Spherical Collagen in Cellular Re-energization From Molecular Actions to Cellular Functional Remodeling

With the acceleration of life pace and the increase of environmental stress,skin aging has become a major concern.Traditional anti-aging methods,such as laser therapy and injections,are effective but have limitations including side effects,complex procedures,high costs,and long recovery periods.In contrast,a topical cellular re-energizing technology using a combination system of 1%Spherical PDRN(Polydeoxyribonucleotide,a registered product name with the following composition:55%mannitol,30%trehalose,10%sodium DNA,and 5%sodium hyaluronate)and 1%Spherical Collagen(a registered product name with the following composition:55%mannitol,30%trehalose,10%collagen,and 5%sodium hyaluronate)stands out with its natural,convenient,non-invasive,and long-lasting advantages.It fundamentally addresses skin aging and provides consumers with a safer,more comfortable,and comprehensive anti-aging experience.This study prepared a combination system of Spherical PDRN and Collagen via a matrix incorporation method.It investigated mitochondrial function and morphology changes in H_(2)O_(2)-stimulated fibroblasts by measuring mitochondrial membrane potential(a key function indicator),ATP content(reflecting energy production),and mitochondrial morphology(related to cell state and stress response).These analyses provided insights into fibroblast energy metabolism and cellular mechanisms.Additionally,a precise method assessed UVA-induced NAD⁺concentration changes in fibroblasts,reflecting metabolic and antioxidant states under stress.In H_(2)O_(2)-stimulated fibroblasts,the combination system of Spherical PDRN and Spherical Collagen at a concentration of 2.5%significantly increased the average value of mitochondrial membrane potential(polymer/monomer ratio)by 448.28%,and the content of ATP by 108.06%.The mitochondrial morphology in cells appeared as elongated rods,with normal fusion and fission occurring.In UVA-irradiated fibroblasts,the combination system of Spherical PDRN and Spherical Collagen at a concentration of 2.5%significantly increased the concentration of NAD^(+) per unit protein by 35.93%.This study investigated how the combination system of Spherical PDRN and Collagen affects mitochondrial function in fibroblasts.It was found that this system significantly boosts mitochondrial membrane potential,implying a marked rise in mitochondrial activity and energy conversion efficiency.This suggests optimized respiratory chain function and energy metabolism,which are vital for normal cellular functions and stress responses.Moreover,the system significantly raises intracellular ATP levels,directly reflecting enhanced mitochondrial energy production.This not only promotes cellular energy metabolism but also boosts cell vitality and stress resistance.Additionally,the system improves mitochondrial morphology,which is crucial for maintaining mitochondrial dynamics and function. 展开更多

Efficacy Study of PDRN Essence after Non-ablative Fractional Laser on Facial Rejuvenation

Polydeoxyribonucleotide(PDRN)has been utilized in clinical practice for an extended period;however,its application in the cosmetics is relatively recent.This study aims to explore the research progress of PDRN regarding its skin repair and anti-aging properties.To evaluate the potential skin repair,soothing,anti-wrinkle and firming effects of PDRN essence via both in vitro and in vivo test method.This study employed in vitro and in vivo test to assess the skin repair and anti-wrinkle effects.The 3D skin model(FulKutis®)was utilized to verify the effects of the PDRN essence on anti-inflammation,barrier repair,and anti-aging.In human test,a randomized,half-face comparison test with PDRN essence and the placebo was conducted with 33 Chinese subjects who met the test criteria.During the test,the skin conditions in terms of the a*value,facial red area ratio,and Transepidermal Water Loss(TEWL)were evaluated at 30 min,4 h,1 day,and 3 days;the skin conditions in terms of skin elasticity and firmness,skin wrinkles were evaluated at 42 days.Subjects’self-assessments were collected at 3 days and 42 days.The FulKutis■experiments demonstrated that the PDRN essence significantly decreased the levels of IL-1α,TNF-α,and PGE2,and improved the protein levels of Filaggrin,Loricrin,Collagen I,Collagen IV,and the thickness of the epidermal living cell layer.In the human study,the a*value,red area ratio,TEWL,and skin-related parameters(R2,R5,R7,F4,and skin wrinkles)on the test face side exhibited greater improvement compared to the control face side with statistically significant differences.Additionally,subject selfassessments revealed high satisfaction with the PDRN essence.The PDRN essence applied after non-ablative fractional laser treatment promotes barrier repair and soothing effects,shortens the recovery period,and enhances facial firmness and wrinkle improvement at the crow’s feet and undereye regions.No adverse events related to the tested cosmetics were observed. 展开更多

Quantitation of Polydeoxyribonucletides (PDRNs) in Human Placental Extract by Fluorescence Spectroscopy Using Ethidium Bromide

Characterization of an aqueous extract of human placenta, used as a licensed drug for wound healing, leads to the identification of several bioactive components including polydeoxyribonu-cleotides (PDRNs). PDRNs are mixture of DNA fragments of different molecular weight. A spectro-fluorimetric method of quantitation of PDRNs in the aqueous extract of human placenta by using ethidium bromide (EtBr) has been described here. It has been demonstrated by thin layer chromatography (TLC) followed by reversed phase HPLC that EtBr binds specifically with the PDRN fraction of the multi-component extract. The binding specificity of EtBr has been verified by the analysis of emission spectra of the extract. A concentration of 0.29 μg/ml EtBr exhibits a linear range of standard CT-DNA from 0.5 - 5 μg/ml of buffer (R2 = 0.992). The same concentration of EtBr shows a linear range of measurements of placenta extract from 5 - 35 μl/ml of buffer (R2 = 0.976). The points of the curve were the average of three sets where maximum variation observed was ±3%. PDRN content of the extract has been estimated based on the resultant fluorescence emission (after background correction) with respect to the standard calibration curve of calf thymus DNA (CT-DNA). Estimation of PDRN in a large number of batches of placenta extract (n = 100) has been done. The statistical analysis of the estimation was found to be significant and the lower and upper levels of PDRN were 158.30 and 239.03 μg/ml of the extract respectively. This easy-to-use method of estimation of PDRN in multi-component biological extract is reported for the first time. This will help in quantitation of PDRNs for other biological extracts. 展开更多
