Stability analysis of cotton hybrids for yield and fiber quality using GGE biplot,WAASB,and MTSI approaches

Background Hybrid cotton enjoys overwhelming patronage among cotton farmers because of its superior yield capacity and fiber quality.However,various environmental factors affect its yield and fiber quality.This study aimed to assess 30 cotton hybrids for the stability of four traits(single-plant seed cotton yield,fiber upper half mean length(UHML),fiber strength,and micronaire)across three environments.Recent techniques,including genotype and genotype×environment(GGE)biplot,which provides a visual representation of performance and adaptability;weighted average absolute scores of the best linear unbiased predictions(WAASB),which balances the performance of the trait with stability;and multi-trait stability index(MTSI),which integrates multi-trait performance and stability,were used to analyze the stability of the four traits.Results Analysis of variance revealed significant genotype and environment interactions for all the traits studied,highlighting the need for comprehensive stability analysis.The environment E2 was the most suitable for the evaluation of seed cotton yield,whereas E3 was suitable for the evaluation of UHML and fiber strength.A stable hybrid,H05(TVH002×MCU5),with superior performance for seed cotton yield and UHML,was identified based on the overall results from GGE and WAASB.The which-won-where bioplot showed that H25(SVPR3×MCU5)performed the best for seed cotton yield in E3,and H27(Suraj×Sunantha)in E2 and E1.The hybrid H04(TVH002×CO14)in E1 and H30(Suraj×MCU5)in E2 and E3 performed well for UHML.Similarly,H28(Suraj×Suraksha)for E2 and E3 and H26(Suraj×Subiksha)for E1 were the best performing in the case of fiber strength.Based on the MTSI,four promising hybrids,namely,H24(SVPR3×CO14),H09(TVH2010×CO14),H18(MCU7×Suraksha),and H29(Suraj×CO14),were identified as stable with average performance for all four traits.Conclusions The study identified a stable hybrid,H05(TVH002×MCU5),with superior performance for yield and UHML.The identified hybrids in this study hold significant potential for cultivation across Tamil Nadu,with a scope for further evaluation in diverse environments. 展开更多

天麻素通过调控GSK-3β/P38 MAPK通路对过表达APOE4的SH-SY5Y细胞的保护作用及机制

为探究天麻素(GAS)对载脂蛋白E(APOE4)引发阿尔茨海默病(AD)的调控作用机制,进而为AD的干预策略提供新方案,该文采用网络药理学方法分析GAS在AD中的潜在作用靶点及相关信号通路;通过慢病毒转染技术构建稳定过表达APOE4基因的SH-SY5Y细... 为探究天麻素(GAS)对载脂蛋白E(APOE4)引发阿尔茨海默病(AD)的调控作用机制,进而为AD的干预策略提供新方案,该文采用网络药理学方法分析GAS在AD中的潜在作用靶点及相关信号通路;通过慢病毒转染技术构建稳定过表达APOE4基因的SH-SY5Y细胞模型,探究GAS对细胞活力和氧化应激状态的影响;利用RT-qPCR和Western blotting技术检测Bax、Bcl-2、p-Tau(Ser396)、p-Tau(Thr181)以及糖原合成酶激酶-3β/P38丝裂原活化蛋白激酶(GSK-3β、P38 MAPK)等关键蛋白的mRNA表达水平和蛋白表达水平变化。结果表明:GAS可显著改善因APOE4过表达所导致的SH-SY5Y细胞活力降低和氧化应激损伤,抑制细胞凋亡;同时显著抑制GSK-3β蛋白的活化,下调p-P38 MAPK蛋白的表达水平;并且有效缓解p-Tau(Ser396)与p-Tau(Thr181)的异常磷酸化及Aβ蛋白的积聚。该文研究证实,GAS可显著减轻APOE4过表达引发的氧化应激反应、抑制细胞凋亡,通过调控GSK-3β/P38 MAPK信号通路抑制Tau蛋白的过度磷酸化,发挥对神经细胞的保护作用,这为APOE4相关的AD防治提供了新的干预策略。 展开更多

目的探究超声微泡(UM)介导微小RNA-545-3p(miR-545-3p)调节E2F转录因子3(E2F3)信号通路对甲状腺癌(TC)细胞增殖、迁移、侵袭的影响。方法用RT-qPCR法检测甲状腺乳头状癌(PTC)细胞系(TPC-1、K1、TTA1细胞)和TC组织中miR-545-3p、E2F3的m... 目的探究超声微泡(UM)介导微小RNA-545-3p(miR-545-3p)调节E2F转录因子3(E2F3)信号通路对甲状腺癌(TC)细胞增殖、迁移、侵袭的影响。方法用RT-qPCR法检测甲状腺乳头状癌(PTC)细胞系(TPC-1、K1、TTA1细胞)和TC组织中miR-545-3p、E2F3的mRNA水平。双荧光素酶实验检测miR-545-3p与E2F3的关系。用超声技术制备微泡,将TPC-1细胞分为Control组、miR-NC组、miR-545-3p-mimics组、UM+miR-545-3p-mimics组、UM+miR-545-3p-mimics+pc DNA组、UM+miR-545-3p-mimics+E2F3组;分别检测超声微泡介导的miR-545-3p靶向E2F3对PTC细胞增殖、迁移、侵袭和上皮-间质转化(EMT)的影响。免疫印迹检测相关蛋白表达。移植瘤实验检测miR-545-3p对肿瘤生长和E2F3表达的影响。结果TC组织及TPC-1细胞中E2F3 mRNA表达水平升高,miR-545-3p表达水平降低(P<0.05)。Starbase数据库在线预测发现,miR-545-3p与E2F3间存在靶向结合位点。与miR-NC组相比,miR-545-3p-mimics组细胞克隆形成数、划痕愈合率、侵袭细胞数目减少,E2F3、CDK1、MMP-2、MMP-9、N-Cadherin、Vimentin表达水平降低,miR-545-3p、ECadherin表达水平升高(P<0.05);与miR-545-3p-mimics组相比,UM+miR-545-3p-mimics组克隆形成数、划痕愈合率、侵袭细胞数目下降,E2F3、CDK1、MMP-2、MMP-9、N-Cadherin、Vimentin表达水平降低,miR-545-3p、ECadherin表达水平升高(P<0.05);与UM+miR-545-3p-mimics+pc DNA组相比,UM+miR-545-3p-mimics+E2F3组克隆形成数、划痕愈合率、侵袭细胞数目升高,E2F3、CDK1、MMP-2、MMP-9、N-Cadherin、Vimentin表达水平升高,E-Cadherin表达水平降低(P<0.05)。miR-545-3p-mimics组肿瘤体积比miR-NC组小,质量比miR-NC组低,E2F3表达水平比miR-NC组低,miR-545-3p水平比miR-NC组高(P<0.05);UM+miR-545-3p-mimics组肿瘤体积比miR-545-3p-mimics组小,质量比miR-545-3p-mimics组低,E2F3表达水平比miR-545-3p-mimics组低,miR-545-3p表达水平比miR-545-3p-mimics组高(P<0.05)。结论UM介导miR-545-3p靶向调控E2F3信号通路抑制PTC细胞增殖、迁移、侵袭和EMT。 展开更多

目的探讨右美托咪定联合艾司氯胺酮在胸腔镜下肺癌根治术后镇痛效果以及对血清白细胞介素-1(IL-1)、P物质(SP)、前列腺素E2(PGE2)和细胞间黏附分子1(ICAM1)的影响。方法选择2021年1月至2023年8月南通市第六人民医院接受胸腔镜手术治疗... 目的探讨右美托咪定联合艾司氯胺酮在胸腔镜下肺癌根治术后镇痛效果以及对血清白细胞介素-1(IL-1)、P物质(SP)、前列腺素E2(PGE2)和细胞间黏附分子1(ICAM1)的影响。方法选择2021年1月至2023年8月南通市第六人民医院接受胸腔镜手术治疗的非小细胞肺癌(NSCLC)病人98例作为研究对象,采用随机数字表法分为观察组和对照组,每组49例。比较两组视觉模拟评分法(VAS)评分、Ramsay镇静评分、平均动脉压(MAP)和心率(HR)、血氧饱和度(SpO2)、血清IL-1β、SP、PGE2、ICAM1水平。结果观察组术后24 h、术后48 h静息[(2.24±0.47)分比(2.75±0.48)分、[(1.87±0.41)分比(2.08±0.40)分]]及活动[(2.76±0.53)分比(3.35±0.56)分]、[(2.06±0.43)分比(2.35±0.48)分]时VAS评分均低于对照组(P<0.05);术后6 h[(2.32±0.69)分比(3.15±0.73)分]、12 h[(2.45±0.65)分比(3.24±0.75)分]、24 h[(2.51±0.70)分比(3.12±0.59)分],观察组Ramsay镇静评分均低于对照组(P<0.05),观察组MAP与对照组比较,差异无统计学意义(P>0.05),HR明显低于对照组[(63.89±11.32)次/分比(71.64±11.86)次/分,P<0.05];术后1 d,观察组的血清IL-1β、SP、PGE2、ICAM1水平均明显低于对照组(P<0.05),此外术后血清SP、PGE2水平比术前升高(P<0.05),ICAM1水平比术前下降(P<0.05)。结论艾司氯胺酮联合右美托咪定对胸腔镜下肺癌根治术病人具有很好的术后镇痛、镇静作用,能有效稳定血流动力学指标,降低血清IL-1β、SP、PGE2、ICAM1水平,抑制炎症反应,且无明显不良反应,值得临床推广应用。 展开更多

目的探讨长链非编码RNA SOX2重叠转录本(LncRNA SOX2-OT)靶向微小RNA-186-5p(miR-186-5p)/锌指E盒结合同源框1(ZEB1)轴对黑色素瘤(MM)细胞的影响。方法体外常规培养正常人皮肤黑色素细胞系PIG1以及人MM细胞系A375、M14和A2058,检测各细... 目的探讨长链非编码RNA SOX2重叠转录本(LncRNA SOX2-OT)靶向微小RNA-186-5p(miR-186-5p)/锌指E盒结合同源框1(ZEB1)轴对黑色素瘤(MM)细胞的影响。方法体外常规培养正常人皮肤黑色素细胞系PIG1以及人MM细胞系A375、M14和A2058,检测各细胞LncRNA SOX2-OT、miR-186-5p、ZEB1 mRNA表达水平。将A2058细胞分为对照组、si-NC组、si-SOX2-OT组、si-SOX2-OT+inhibitor-NC组、si-SOX2-OT+miR-186-5p inhibitor组。检测LncRNA SOX2-OT、miR-186-5p、ZEB1 mRNA表达水平(RT-qPCR法);细胞活力(CCK-8试剂盒法)、增殖活性(克隆形成实验)、迁移(划痕试验)、侵袭能力(Transwell小室法);ZEB1、E-cadherin、TGF-β1、α-SMA蛋白表达量(Western blot法)。验证miR-186-5p与LncRNA SOX2-OT、ZEB1之间的靶向关系。结果与对照组、si-NC组比较,si-SOX2-OT组LncRNA SOX2-OT、ZEB1 mRNA表达水平、OD_(450)值、细胞克隆数、划痕愈合率、细胞侵袭数以及ZEB1、TGF-β1和α-SMA蛋白表达量降低,miR-186-5p表达水平以及E-cadherin蛋白表达量升高(P<0.05);与si-SOX2-OT+inhibitor-NC组比较,siSOX2-OT+miR-186-5p inhibitor组ZEB1 mRNA水平、OD_(450)值、细胞克隆数、划痕愈合率、细胞侵袭数以及ZEB1、TGF-β1和α-SMA蛋白表达量升高,miR-186-5p表达水平及E-cadherin蛋白表达量降低(P<0.05)。转染miR-186-5p mimic后,LncRNA SOX2-OT-WT、ZEB1-WT的荧光素酶活性降低(P<0.05)。裸鼠移植瘤实验显示,敲低SOX2-OT可提高移植瘤miR-186-5p和E-cadherin蛋白表达,降低ZEB1蛋白表达、抑制移植瘤的生长;抑制miR-186-5p的表达可减轻敲低SOX2-OT对移植瘤生长的抑制作用(P<0.05)。结论在MM细胞中LncRNA SOX2-OT异常高表达,敲低LncRNA SOX2-OT可靶向上调miR-186-5p的表达,抑制ZEB1的表达,进而抑制MM细胞的恶性生物学行为。 展开更多

目的评价浩欧博磁微粒化学发光法(CLIA)与罗氏电化学发光法(ECLIA)检测血清免疫球蛋白E(IgE)结果的可比性,评估磁微粒CLIA检测IgE是否能够满足临床需求。方法基于美国临床和实验室标准协会(CLSI)新指南EP9-A3文件要求,收集2024年11月至2... 目的评价浩欧博磁微粒化学发光法(CLIA)与罗氏电化学发光法(ECLIA)检测血清免疫球蛋白E(IgE)结果的可比性,评估磁微粒CLIA检测IgE是否能够满足临床需求。方法基于美国临床和实验室标准协会(CLSI)新指南EP9-A3文件要求,收集2024年11月至2025年6月我院皮肤科患者新鲜血清标本510例,用罗氏ECLIA和浩欧博磁微粒CLIA平行检测IgE水平,以罗氏ECLIA为参考方法。①定性检验,设临界值为2IU/mL,计算磁微粒CLIA的灵敏度、特异性和符合率。采用成组χ2检验进行关联性分析,并用配对χ2检验比对两种方法对比定性结果的差异。②定量检验,对于结果在2~2500IU/mL的样本,采用普通线性回归和Bland-Altman一致性分析两种统计学方法进行拟合分析;并结合国家卫生健康委临床检验中心室间质量评价允许总误差要求作覆盖线性范围的浓度梯度比较分析。结果①磁微粒CLIA检测IgE的灵敏度是99.18%、特异性是90.48%、阳性预测值是99.59%、阴性预测值是82.60%和总符合率是98.82%,成组χ2检验χ2=375.856,P<0.001,配对McNemarχ2检验P=0.687。②直线线性回归分析方程是Y=0.9898 X-12.8967,R 2=0.997,r=0.998≈1;Bland-Altman分析显示有26个点超出95%置信区间,符合率是94.90%;5组IgE浓度梯度的均值偏倚均小于1/2TEa。结论浩欧博磁微粒CLIA与罗氏ECLIA检测的IgE结果可比性显著,能够满足临床需求。 展开更多

As a result, students'enthusiasm in improving their spoken English is diminished. Actually, students need some knowledge about English culture background before they can communicate effectively. It is significant for teachers to find out the students'wants and lacks during their spoken English study. Besides, attentions should be paid to English culture background in spoken English classes by both teachers and students. This paper mainly aimed at finding out the needs of students for English culture background in spoken English classes based on the theory of needs analysis. 展开更多

Objective:MicroRNAs(miRNAs)are small,non-coding RNAs that play a key role in the development of chemoresistance in various cancer types,including colorectal cancer(CRC).In this study,we aimed to study the underlying mechanisms of miRNA in chemotherapy-resistant CRC.Methods:LoVo CRC cell line was exposed to oxaliplatin at an increased dose,and cells were cultured in the presence of oxaliplatin to develop LoVo^(OXR) cells.Microarray and Quantitative Reverse Transcription Polymerase Chain Reaction(qRT-PCR),western blot,and transwell assay were used to evaluate the chemoresistance in LoVo^(OXR) CRC cells.Results:Microarray and qRT-PCR analysis showed an increased expression of miR-100-5p in LoVo^(OXR) cells.MTT assay and flow cytometry analysis revealed less apoptosis and higher cell viability in LoVo^(OXR) cells.mRNA prediction target gene analysis showed C-terminal domain small phosphatase-like(CTDSPL),a phosphatase-like tumor suppressor,as a key target of miR-100-5p.CTDSPL expression was low in LoVo^(OXR) cells compared to LoVoWT cells.miR-100-5p regulates G1/S and S-phase transitions and inhibits differentiation by targeting the CTDSPL/pRB/E2F1 signaling pathway,which involves the modulation of cell cycle effectors in LoVo^(OXR) cells.Further,we found that forkhead box P3(FOXP3),as the upstream target of miR-100-5p,is highly expressed in LoVo^(OXR) cells.Inhibiting miR-100-5p and FOXP3 down-regulates miR-100-5p expression,while increased CTDSPL expression contributed to reduced cell proliferation and promoted cell apoptosis in LoVo^(OXR) CRC cells.Conclusions:miR-100-5p plays an oncogenic role in inducing chemoresistance through modulation of the CTDSPL/retinoblastoma protein(pRB)/E2F transcription factor 1(E2F1)axis in CRC cells. 展开更多

Tolerance analysis of a planetary reducer is conducted. By focusing the serially-arranged parts in the central line and setting an appropriate dimension as a “gap”, worst-case tolerance analysis method and statistical (RSS) tolerance analysis are used, and then a tolerance-analyzing tool CE/TOL is introduced. The acceptable value range of the “gap” is from 1.5 mm to 3.5 mm. While worst-case analysis rejects the design, RSS accepts it by a more realistic result with the law of probability accounted. With the help of CE/TOL and CAD software Pro/E, all components' dimensions and tolerances are assigned and modified, and more detailed and more comprehensible results are gives out. 展开更多

The ultrastructure of porcine kidney(PK)-15 cells was examined after lipofectamine-aided transfection of the molecular clone of the P1 agent.PK-15 cells transfected with the tandem dimer of the P1molecular DNA clone had numbers of intracytoplasmic inclusions,and a few cells had intranuclear inclusions.Intracytoplasmic inclusions were round to oval and 0.1-0.3μm in diameter,and intranuclear inclusions,which were more electron dense,were of two general types:the first were round and small(0.1μm approximately)and the second were hexagonal and larger(0.4-0.8μm in diameter).Cells transfected with the tandem dimer of the P1 molecular DNA clone tested positive for P1 DNA at passage 5.This is the first report that the P1 molecular clone has infectivity in vitro and it will provide fundamental materials for further study of the biological characterization of P1. 展开更多

Hand.-Mazz.is a rhizomatous,herbaceous,perennial plant that has been used for more than a thousand years in traditional Chinese medicine.It is facing extinction due to overharvesting.Steroids are the major therapeutic components in Paris roots,the commercial value of which increases with age.To date,no genomic data on the species have been available.In this study,transcriptome analysis of an 8-year-old root and a 4-year-old root provided insight into the metabolic pathways that generate the steroids.Using Illumina sequencing technology,we generated a high-quality sequence and demonstrated de novo assembly and annotation of genes in the absence of prior genome information.Approximately 87,577 unique sequences,with an average length of 614 bases,were obtained from the root cells.Using bioinformatics methods,we annotated approximately 65.51% of the unique sequences by conducting a similarity search with known genes in the National Center for Biotechnology Information's non-redundant database.The unique transcripts were functionally classified using the Gene Ontology hierarchy and the Kyoto Encyclopedia of Genes and Genomes database.Of 3082 genes that were identified as significantly differentially expressed between roots of different ages,1518(49.25%) were upregulated and 1564(50.75%) were downregulated in the older root.Metabolic pathway analysis predicted that 25 unigenes were responsible for the biosynthesis of the saponins steroids.These data represent a valuable resource for future genomic studies on this endangered species and will be valuable for efforts to genetically engineer P.polyphylla and facilitate saponin-rich plant development. 展开更多

The threshold of cyclin E expression at G1/S boundary is a characteristic feature of cell cycle progressing. In this study, we tried to develop a quantitative approach to analyze cyclin E threshold by multiparameter flow cytometry. The expression of cyclin E in exponentially growing MOLT-4 cells was detected under different photomultiplier tube (PMT) voltages by cyclin E/DNA multiparameter flow cytometry. Additionally, cyclin E was detected in cells which were treated with caffeine and cycloheximide (CHX) under the same PMT voltage. Moreover, the expression of cyclin E in MOLT-4 cells was compared with that in JURKAT cells. Cyclin E threshold was quantified by formula B2/A×C (A, B, C indicates the minimum, threshold, and maximum of cyclin E fluorescence intensity, respectively). Results showed that in MOLT-4 cells, cyclin E threshold calculated by formula B2/A×C was invariable under different PMT settings. It was decreased in cells treated with caffeine and remained changeless in cells treated with cycloheximide. Cyclin E threshold in JURKAT cells was much lower than that in MOLT-4 cells. It was suggested that Formula B2/A×C we firstly set up could be used to analyze cyclin E expression threshold quantitatively. 展开更多

P1B-type ATPase ion pumps that transport heavy metal ions across cellular membranes are essential for plant growth and development. To date, a genomic comparison overview of the family in rice, maize and sorghum is not yet available. In this study, a total of 31 heavy metal P1B-type ATPase (HMA) genes were identified, including 9 in rice, 11 each from maize and sorghum. They were classified into two distinct subfamilies based on their sequence composition and phylogenetic relationship. Four pairs of HMA genes were expanded via gene duplication with tandemly duplicated. Comprehensive analyses were performed to investigate the expression profiles of HMA genes in various tissues by using quantitative real-time PCR. Some HMA members exhibited abundant and tissue-specific expression patterns. Moreover, most of the genes were found to be differentially expressed under the Cu/Cd treatment. This study will facilitate further studies on P1B-type ATPase family and provide valuable hints for the functional validation in rice, maize and sorghum. 展开更多

Therefore it is critical to understand the landscapes and their dynamics. Patterns of landscape elements strongly influence the ecological characteristics. This study was designed to document and map the current status of the tropi-cal dry deciduous forest of the Tadoba-Andhari Tiger Reserve (TATR), Central India, (using IRS P6 LISS IV data) and to describe its landscape structure at three levels of organization viz. landscape, class, and patch. The study area was classified into 10 land cover classes that include 6 vegetation classes. The landscape structure was analyzed using FRAG-STATS using 12 set of indices. The TATR landscapes have a total of 2,307 patches with a mean patch size of 25.67 ha and patch density of 1.7 patches per km2. Amongst all land cover classes, mixed bamboo forest is dominant-it occupied maximum area (77.99%)-while riparian forest is least represented (0.32%). Mixed forest has maximum number of patches among all vegetation classes. Results have shown that despite being dominant in the area, mixed bamboo forest has low patch density (0.25/100 ha). Dominance of mixed bamboo forest is attributed to large patch sizes and not to the number of patches. This study has focussed on the approach of integrating satellite forest classification and forest inven-tory data for studying forest landscape patterns. 展开更多

Different strains from Gansu were cloned and the P1 protein was sequenced and analysed. Results indicate that there are three kinds of EV71 infections prevalent in Gansu. The VP 1 protein from one of these strains, 55F, was expressed. The recombinant protein was expressed with high level and reacted specifically with the EV71 patient antibody, the recombinant protein was also applied to raise antiserum in rabbits and after the fourth injection a high titer of antiserum was detected by ELISA assay. These data are useful for further clarification of prevalent EV71 strains in the north of China at the molecular level and provide a basis for EV71 diagnosis. 展开更多

AIM To analyze the clinicopathological characteristics of patients with both node-negative gastric carcinoma and diagnosis of recurrence during follow-up.METHODS We enrolled 41 patients treated with curative gastrectomy for p T2-4 a N0 gastric carcinoma between 1992 and 2010,who developed recurrence(Group 1).We retrospectively selected this group from the prospectively collected database of 4 centers belonging to the Italian Research Group for Gastric Cancer,and compared them with 437 p T2-4 a N0 patients without recurrence(Group 2).We analyzed lymphatic embolization,microvascular infiltration,perineural infiltration,and immunohistochemical determination of p53,Ki67,and HER2 in Group 1 and in a subgroup of Group 2(Group 2 bis)of 41 cases matched with Group 1 according to demographic and pathological characteristics.RESULTS T4 a stage and diffuse histotype were associated with recurrence in the group of p N0 patients.In-depth pathological analysis of two homogenous groups of p N0 patients,with and without recurrence during longterm follow-up(groups 1 and 2 bis),revealed two striking patterns:lymphatic embolization and perineural infiltration(two parameters that pathologists can easily report),and p53 and Ki67,represent significant factors for recurrence.CONCLUSION The reported pathological features should be considered predictive factors for recurrence and could be useful to stratify node-negative gastric cancer patients for adjuvant treatment and tailored follow-up. 展开更多

OBJECTIVE: To investigate the association of E-selectin $128R polymorphisms with ischemic stroke. DATA SOURCES: A computer-based online search was conducted in PubMed, Elsevier, Ovid Database, the China National Knowledge Infrastructure, and Wanfang Database between January 1998 and December 2010. STUDY SELECTION: Case-controlled studies addressing the association of the E-selectin polymorphism and ischemic stroke were included in this review. The genotype distribution complied with the Hardy-Weinberg genetic equilibrium. The included reports were evaluated by two authors for strict quality screening. Meta-analysis software, REVMAN 5.1, was used to investigate heterogeneity, pooled odds ratio (OR) and 95% confidence interval (CO in individual studies. MAIN OUTCOME MEASURES: Genotype and allele distributions at the E-selectin $128R site. RESULTS: Six case-controlled studies were included after screening and application of inclusion and exclusion criteria. There was no heterogeneity in the genotype and allele frequencies, and no publication bias was found. Meta-analysis of the pooled data showed that the OR value of the (AC+CC)/AA genotype was 1.93 (95% CI: 1.55 2.41, Z= 5.80, P 〈 0.000 01), and the ORfor the C/A allele was 1.80 (95% CI: 1.47 2.22, Z= 5.59, P 〈 0.000 01) in the ischemic stroke group, compared with control group. Results of pooled data in Chinese subjects showed that the ORvalue of (AC+CC)/AA was 2.36 (95% CI: 1.68 3.31, Z = 4.99, P 〈 0.000 01), and the OR value of the C/A allele was 2.25 (95% CI: 1.63 3.12, Z= 4.89, P 〈 0.000 01). CONCLUSION: Polymorphism of E-selectin S128R was significantly associated with susceptibility to ischemic stroke; the AC and CC genotypes as well as the C allele may be factors associated with susceptibility to ischemic stroke. 展开更多

BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human papillomavirus(HPV)E6/E7 mRNA in cervical tissue of patients with different types of epithelial cell neoplasia(CIN)and its relationship with CIN progression and diagnosis.METHODS One hundred women with HPV infection detected by cervical exfoliation cytology between January 2022 and January 2023 were retrospectively selected.These patients were graded CIN based on colposcopy and cervical pathology.The positive expression rates of HPV E6/E7 mRNA and HPV[polymerase chain reaction(PCR)-reverse dot crossing]were compared among all groups.Patients with HPV E6/E7 mRNA expression in the grade 1 CIN group were followed up for 1 yr.The relationship between atypical squamous epithelium and high malignant epithelial neoplasia was investigated by univariate and multivariate analysis.RESULTS The diagnostic sensitivity,specificity,and sensitivity of PCR-reverse point hybrid ization technology for secondary CIN were 70.41%,70.66%,and 0.714,respectively.Sensitivity and specificity for secondary CIN were 752%and 7853%,respectively,the area under the curve value was 0.789.Logistic Multifactorial model analysis revealed that the HPV positive rates and the HPV E6/E7 mRNA positive rates were independent risk factors of CIN grade I(P<0.05).In CIN grade I patients with positive for HPV E6/E7 mRNA,in its orientation to grade CIN patients,in its orientation to grade CIN patients,at 69.2%,compared with patients negative for HPV E6/E7 mRNA(30.8%),significant difference(P<0.05).CONCLUSION HPV E6/E7 mRNA and HPV(PCR-reverse dot hybrid)positive expression have a close relationship with CINgrade disease progression and is an independent risk factor for high-grade CIN lesions. 展开更多
