利用CRISPR/Cas9技术靶向编辑水稻(Oryza sativa)类囊体腔蛋白基因OsTLP16.5(Os06g0705100),使该基因编码的大部分氨基酸序列发生改变,经筛选鉴定获得突变体ostlp16.5。对水稻突变体ostlp16.5及野生型ZH11进行代谢组检测,结果表明,有机... 利用CRISPR/Cas9技术靶向编辑水稻(Oryza sativa)类囊体腔蛋白基因OsTLP16.5(Os06g0705100),使该基因编码的大部分氨基酸序列发生改变,经筛选鉴定获得突变体ostlp16.5。对水稻突变体ostlp16.5及野生型ZH11进行代谢组检测,结果表明,有机酸、氨基酸、维生素及其衍生物等物质的含量在突变体ostlp16.5和野生型ZH11中表现出显著差异。进一步分析发现,突变体ostlp16.5与野生型ZH11差异较大的代谢产物(3,4二羟基苯基丙酸、多巴胺、L-花生酸和L-酪氨酸、L-2-羟基戊二酸、2-酮基戊二酸、泛酸等)及差异显著的代谢通路(异喹啉生物碱生物合成、酪氨酸代谢等)大部分直接或间接与光合反应相关,表明OsTLP16.5蛋白可能通过参与光合调控来影响水稻体内有机酸、氨基酸和维生素等的代谢。研究结果对揭示水稻类囊体腔蛋白OsTLP16.5的功能及提高水稻的生产力具有重要意义,并可为研究OsTLP16.5蛋白调控水稻体内次生代谢及生长发育提供参考。 展开更多

To screen for interacting proteins of the rice thylakoid lumen protein OsTLP16.5(encoded by Os06g0705100),which will help to elucidate the molecular regulatory mechanisms of OsTLP16.5 in rice.Total RNA was extracted from rice to construct a cDNA library.A yeast two-hybrid(Y2H)bait vector,pGBKT7-OsTLP16.5,was constructed to screen candidate target proteins using the yeast two-hybrid system,followed by interaction validation.Large-scale screening was performed using next-generation sequencing(NGS)technology.The bait vector pGBKT7-OsTLP16.5 was successfully constructed and verified to be free of autoactivation.A library screen identified 32 potential interacting proteins,of which 3 showed positive interactions after validation.NGS technology revealed 977 potential interacting proteins,including 51 chloroplast-associated proteins.A rice cDNA library was successfully constructed,and interacting proteins of OsTLP16.5 were identified,providing a foundation for studying how OsTLP16.5 functions in rice. 展开更多
