Decorin(DCN)is primarily found in the connective tissues of various parts of the body, including the lungs, kidneys, bone tissue, aorta, and tendons. It is an important component of the extracellular matrix(ECM)and belongs to the class I small leucine-rich proteoglycans family.DCN is increasingly attracting attention due to its significant role in tumors, fibrotic diseases, and the regulation of vascular formation. Moreover, its anti-tumor properties have positioned it as a promising biomarker in the fight against cancer.Numerous studies have confirmed that DCN can exert inhibitory effects in various solid tumors, particularly in oral squamous cell carcinoma(OSCC),by activating its downstream pathways through binding with the epidermal growth factor receptor(EGFR)and mesenchymal-epithelial transition(MET)receptor,or by stabilizing and enhancing the expression of the tumor suppressor gene p53 to mediate apoptosis in cancer cells that have undergone mutation. The occurrence of OSCC is a continuous and dynamic process, encompassing the transition from normal mucosa to oral potentially malignant disorders(OPMDs),and further progressing from OPMDs to the malignant transformation into OSCC.We have found that DCN may exhibit a bidirectional effect in the progression of oral mucosal carcinogenesis, showing a trend of initial elevation followed by a decline, which decreases with the differentiation of OSCC.In OPMDs,DCN exhibits high expression and may be associated with malignant transformation, possibly linked to the increased expression of P53 in OPMDs.In OSCC,the expression of DCN is reduced, which can impact OSCC angiogenesis, and inhibit tumor cell proliferation, migration, and invasion capabilities, serving as a potential marker for predicting adverse prognosis in OSCC patients. This article reviews the current research status of DCN,covering its molecular structure, properties, and involvement in the onset and progression of oral mucosal carcinogenesis. It elucidates DCN’s role in this process and aims to offer insights for future investigations into its mechanism of action in oral mucosal carcinogenesis and its potential application in the early diagnosis and treatment of OSCC. 展开更多

Oral squamous cell carcinoma(OSCC)is a prevalent malignancy with high morbidity and mortality.Globally, about 400000 people are affected, often with a poor quality of life. Its high mortality is mainly due to its aggressive growth and tendency to spread.Epithelial-mesenchymal transition(EMT)is a central regulatory hub driving tumor cell migration and invasion by enabling changes in cell characteristics.During EMT,epithelial cells gradually take on mesenchymal traits, gaining mobility and spreading mo re easily.Recent multi-omics studies show that many cancer cells exist in a hybrid or partial-EMT state, which lies between the full epithelial and mesenchymal forms.Cells in this state are especially invasive and metastatic, with high plasticity that promotes tumor progression. This review summarizes the role of partial-EMT in OSCC,with a focus on how it alters the tumor microenvironment(TME),promotes invasion and metastasis, and influences cancer stem cells(CSCs).We also highlight the link between partial-EMT and treatment resistance in OSCC.Based on these insights,we discuss therapeutic strategies targeting partial-EMT to improve outcomes.Targeting partial-EMT may offer promising strategies to enhance treatment effectiveness and improve patient survival and quality of life. 展开更多

Oral immunization is an alternative or supplementary approach that can significantly improve dog vaccination coverage, especially for free-roaming dogs.Safe and effective oral rabies vaccines for dogs are still being sought. In our previous studies,we generated a genetically modified rabies virus(RABV) ERA strain,rERAG_(333E),containing a mutation from arginine(Arg,R) to glutamic acid(Glu,E) at residue 333 of the G protein(G_(333E)).Our previous results demonstrated that rERAG_(333E) was safe for adult mice and dogs, and oral vaccination with rERAG_(333E) induced a strong and long-lasting protective immune response in dogs. Here,we further investigated the safety and immunogenicity of rERAG_(333E) in nontarget species, including suckling mice, rhesus monkeys, foxes, raccoon dogs, piglets, goats, and sheep.Suckling mice studies demonstrated that the G_(333E) mutation significantly reduced the virulence of the ERA strain. All of the suckling mice aged 10 days and above survived and showed no apparent signs of disease after intracerebral inoculation with rERAG_(333E).Animal studies demonstrated that rERAG_(333E) was safe in rhesus monkeys, foxes, raccoon dogs, piglets, goats, and sheep.None of those animals inoculated orally with 10 times the intended field dose of rERAG_(333E) showed abnormal clinical signs before and after the booster immunization with Rabvac 3,an inactivated rabies vaccine.Meanwhile, oral inoculation with rERAG_(333E) induced strong neutralizing antibody(NA) responses to RABV in rhesus monkeys, foxes, raccoon dogs, and piglets. These results demonstrated that rERAG_(333E) has the potential to serve as a safe oral rabies vaccine for dogs. 展开更多

Background:The regulatory mechanisms governing vasculogenic mimicry(VM)in oral squamous cell carcinoma(OSCC)remain largely undefined. This study aimed to identify critical factors and elucidate the epigenetic mechanisms underlying VM in OSCC.Methods:Bioinformatics analysis was performed utilizing single-cell RNA-seq,bulk RNA-seq,and histone H3 lysine 27 acetylation(H3K27ac)Chromatin Immunoprecipitation(ChIP)-seq data obtained from The Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)databases.ChIP-qPCR was used to validate the binding of ETS transcription factor ELK4(ELK4)to the dihydrofolate reductase(DHFR)enhancer. In vitro VM formation and invasion of OSCC cells were assessed using Matrigel-based tube formation and Transwell assays, respectively. Results:Elevated expression of VM-related genes predicts unfavorable prognosis in OSCC patients.High-dimensional weighted gene co-expression network analysis(hdWGCNA)identified epithelial subcluster C4 as most strongly associated with VM and metastasis.Three co-expression modules within this subcluster exhibited significant positive correlations with both phenotypic traits.Among the 30 eigengenes from the three modules,DHFR emerged as a key regulator of VM and metastasis.Knockdown or inhibition of DHFR significantly suppressed VM formation and invasion in OSCC cells.Mechanistically,ELK4 activated DHFR transcription through direct binding to its enhancer.DHFR overexpression rescued VM and invasion impairment induced by ELK4 knockdown. Conclusion:DHFR was a pivotal enhancer-regulated gene driving VM and metastasis in OSCC.ELK4 directly binds to DHFR enhancer regions to activate its transcription, thereby promoting these malignant phenotypes. These findings identified the ELK4/DHFR axis as a promising therapeutic target for anti-angiogenic intervention in OSCC. 展开更多

Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials. Here, nanocarriers loaded with conventional antimicrobials and responding to environmental changes upon entry into oral biofilms are reviewed.Supra-gingival biofilms are characterized by acidic pH,the presence of bacterial enzymes, and the development of hypoxia in deeper layers.Sub-gingival biofilms are slightly alkaline, with hypoxia occurring over their entire depth.Upon entering biofilms, negatively charged,pH-and/or hypoxia-responsive nanocarriers become positively charged. This charge reversal leads to electrostatic double-layer attraction between positively charged nanocarriers towards negatively charged, waterfilled channel walls in biofilms, enhancing their accumulation in a biofilm.Degradation of bacterial enzyme-responsive nanocarriers causes in-biofilm release of antimicrobial cargo, yielding higher local antimicrobial concentrations than can be achieved through their direct, oral administration without harming soft tissues.Enhanced antibiofilm activity after in-biofilm antimicrobial release from biofilm-responsive micelles and liposomes has been demonstrated in vitro towards single-species Streptococcus mutans and Staphylococcus aureus biofilms or in vivo using specific-pathogen-free rodents inoculated with selected pathogens. This preferential antibacterial activity regulated the microbial composition of ex vivo human oral biofilm towards a more healthy microbiome composition.Although clinical confirmation is limited, the potential benefits of stimuli-responsive,antimicrobial-loaded nanocarriers for oral biofilm control and microbiome restoration are worth further investigation towards clinical translation. 展开更多

Osmanthus fragrans is most famous for its strong aroma, and different varieties have different degrees of fragrance and color.Fragrance and color are important factors affecting the ornamental quality of O.fragrans.Terpenoids are important secondary metabolites in plants, with β-carotene(C40)being the major pigment substance and linalool(C10)being the key aromatic component in O.fragrans.The geranylgeranyl pyrophosphate synthase genes(GGPPSs)play important roles in secondary metabolism in plants. However, the functions of the GGPPS family in floral color and fragrance formation has rarely been reported in O.fragrans.In this study,24 Of GGPPS genes were identified and classified into two subfamilies. The Of GGPPSs showed tissue-specific expression and Of GGPPS13 had highest expression in flowers. The Of GGPPS13 protein was localized to chloroplasts. The transcriptome data of Of GGPPS13 was verified by q RT-PCR and the expression level in‘Wanyingui'with strong aroma was higher than that in‘Zhuangyuanhong'with deep color at different flower development stages.Transient overexpression of Of GGPPS13 in O.fragrans petals showed that Of GGPPS13 increased the β-carotene content, the main color substance of O.fragrans, but decreased the linalool content, the main volatile organic compound(VOC)in the floral aroma of O.fragrans.Of GGPPS13 was indicated as the critical gene related to terpenoid synthesis in the floral aroma and color formation in O.fragrans.Our findings provide gene resources on the GGPPS gene family for further revealing the molecular regulation mechanism of the floral color and aroma formation in O.fragrans. 展开更多

Background:Oral health issues persistently affect the overall health and well-being of rural populations.Village health volunteers(VHVs)play a crucial role in advancing oral health literacy in their community. This study aimed to examine the factors related to nutritional literacy for oral health among VHVs.Methods:This was a mixed-methods study employing an explanatory sequential design. The quantitative data were gathered through questionnaires distributed to a cohort of 10,514 VHVs registered in Health Region 1.A stratified random sampling technique was used to ensure adequate representation of various subgroups within the VHV population, considering factors such as age, education level, and geographical distribution across the region. This approach allowed for a more representative sample that accurately reflects the diversity of the VHV population.Qualitative data were obtained through semi-structured interviews with a purposive sampling of 20 participants based on specific criteria.Quantitative data were analyzed using descriptive statistics and biserial correlation techniques, while qualitative data were analyzed using content analysis. Results:The study found that the sample group possessed a moderate level of knowledge of health literacy principles and nutrition for oral health. However, their self-assessed skills in nutritional literacy for oral health were rated as high.A statistically significant negative correlation was found between knowledge of nutrition for oral health and skills in nutritional literacy for oral health.VHVs equated health literacy with knowledge because their public health training had focused more on imparting knowledge rather than developing skills based on health literacy principles. Conclusion:There is a need to emphasize skill-based health literacy training and to use effective skill development techniques that are tailored to the specific roles and responsibilities of health volunteers. It is also recommended to continuously monitor and evaluate the outcomes of these efforts. 展开更多

Oral submucous fibrosis(OSF),characterized by excessive deposition of extracellular matrix(ECM)that causes oral mucosal tissue sclerosis, and even cancer transformation,is a chronic, progressive fibrosis disease. However, despite some advancements in recent years,no targeted antifibrotic strategies for OSF have been approved;likely because the complicated mechanisms that initiate and drive fibrosis remain to be determined. In this review,we briefly introduce the epidemiology and etiology of OSF.Then,we highlight how cell-intrinsic changes in significant structural cells can drive fibrotic response by regulating biological behaviors, secretion function, and activation of ECM-producing myofibroblasts. In addition,we also discuss the role of innate and adaptive immune cells and how they contribute to the pathogenesis of OSF.Finally,we summarize strategies to interrupt key mechanisms that cause OSF,including modulation of the ECM,inhibition of inflammation, improvement of vascular disturbance. This review will provide potential routes for developing novel anti-OSF therapeutics. 展开更多

Understanding microbial-host interactions in the oral cavity is essential for elucidating oral disease pathogenesis and its systemic implications. In vitro bacteria-host cell coculture models have enabled fundamental studies to characterize bacterial infection and host responses in a reductionist yet reproducible manner. However, existing in vitro coculture models fail to establish conditions that are suitable for the growth of both mammalian cells and anaerobes, thereby hindering a comprehensive understanding of their interactions. Here,we present an asymmetric gas coculture system that simulates the oral microenvironment by maintaining distinct normoxic and anaerobic conditions for gingival epithelial cells and anaerobic bacteria, respectively.Using a key oral pathobiont,Fusobacterium nucleatum,as the primary test bed,we demonstrate that the system preserves bacterial viability and supports the integrity of telomerase-immortalized gingival keratinocytes.Compared to conventional models, this system enhanced bacterial invasion, elevated intracellular bacterial loads, and elicited more robust host pro-inflammatory responses, including increased secretion of CXCL10,IL-6,and IL-8.In addition, the model enabled precise evaluation of antibiotic efficacy against intracellular pathogens. Finally,we validate the ability of the asymmetric system to support the proliferation of a more oxygen-sensitive oral pathobiont,Porphyromonas gingivalis. These results underscore the utility of this coculture platform for studying oral microbial pathogenesis and screening therapeutics, offering a physiologically relevant approach to advance oral and systemic health research. 展开更多

BACKGROUND Oral squamous cell carcinoma(OSCC)is a frequent cancer affecting the oral cavity.OSCC is usually preceded by occurrence of precursor lesions, which demonstrate a varying degree of malignant transformation potential.Epithelialmesenchymal transition is a significant biological phenomenon that facilitates tumor growth and metastasis through the involvement of diverse epithelial and mesenchymal proteins.E-cadherin down regulation and over-expression of zincfinger E-box-binding homeobox 1(ZEB1)has been reported in several cancers.AIM To evaluate the role of E-cadherin and ZEB1 in oral leukoplakia and OSCC.METHODS A total of 60 cases i.e.,oral leukoplakia/oral epithelial dysplasia(OED)(n=30)and OSCC(n=30)were included.Immunohistochemistry was performed utilizing two markers:E-cadherin and ZEB1.The Mann-Whitney U test was employed to compare individual markers across the study groups.Spearman’s correlation was done followed by discriminant function analysis.RESULTS Reduction in the expression E-cadherin and altered localization were noted from OED to OSCC.Overexpression of ZEB1 along with cytoplasmic accumulation was noted in OED,with marked expression in OSCC.ZEB1 epithelium intensity and ZEB1 connective tissue percentage contributed to the discriminant function with an accuracy of 85%for the classification of OED from OSCC.CONCLUSION Loss of E-cadherin and up-regulation of ZEB1 from OED to OSCC,predisposes to induction of epithelial-mesenchymal transition.Discriminant formulas are developed to classify cases of OED and OSCC with 85%accuracy. 展开更多
