[Objective] The aim of this study was to extract RNA,DNA and protein from plants by using modified one step method.[Method]CTAB was used for the modifying of Biozol-based one step extraction method,which was then used...[Objective] The aim of this study was to extract RNA,DNA and protein from plants by using modified one step method.[Method]CTAB was used for the modifying of Biozol-based one step extraction method,which was then used for the isolation of RNA,DNA and protein from the roots of maize(Zea mays L.),soybean(Glycine max L.),alfalfa(Medicago sativa L.)and cucumber(Cucumis sativus L.).And the heme oxygenase-1 gene(HO-1)and calcium-dependent protein kinase-1 gene(CDPK1),as well as HO-1 protein were identified.[Result]The result of UV spectrophotometer determination and agarose gel electrophoresis analysis showed that the purity of obtained RNA and DNA sample was relatively high;HO-1 and CDPK1 gene exhibited the positive signals checked by RT-PCR;the extracted protein could be used for western blotting analysis;the whole isolation procedure cost 3 h.[Conclusion]This method can be used as a good reference for researcher and will offer a better value with a wide range of applications.展开更多

Thermalgravimetric analysis (TGA) and X-ray diffraction were used to determine the reaction paths of vanadium carbide, namely the following sequential reaction: V2O3→V8C7 in higher temperature stage, the rule of vanadium nitride synthesized was established, and defined conditions of temperature for the production of the carbides and nitrides were determined. Vanadium oxycarbide may consist in the front process of carbothermal reduction of vanadium trioxide. In one step method for vanadium nitride by carbothermal reduction and nitridation of vanadium trioxide, the nitridation process is simultaneous with the carbothermal reduction. A one-step mechanism of the carbothermal reduction with simultaneous nitridation leaded to a lower terminal temperature in nitridation process for vanadium nitride produced, compared with that of carbothermal reduction process without nitridation. The grain size and shape of vanadium nitride were uniform, and had the shape of a cube. The one step method combined vacuum carborization and nitridation (namely two step method) into one process. It simplified the technological process and decreased the costs.展开更多

This paper focuses on its application in cryogenic boiling heat t...Inverse heat conduction method (IHCM) is one of the most effective approaches to obtaining the boiling heat transfer coefficient from measured results. This paper focuses on its application in cryogenic boiling heat transfer. Experiments were conducted on the heat transfer of a stainless steel block in a liquid nitrogen bath, with the assumption of a 1D conduction condition to realize fast acquisition of the temperature of the test points inside the block. With the inverse-heat conduction theory and the explicit finite difference model, a solving program was developed to calculate the heat flux and the boiling heat transfer coefficient of a stainless steel block in liquid nitrogen bath based on the temperature acquisition data. Considering the oscillating data and some unsmooth transition points in the inverse-heat-conduction calculation result of the heat-transfer coefficient, a two-step data-fitting procedure was proposed to obtain the expression for the boiling heat transfer coefficients. The coefficient was then verified for accuracy by a comparison between the simulation results using this expression and the verifying experimental results of a stainless steel block. The maximum error with a revised segment fitting is around 6%, which verifies the feasibility of using IHCM to measure the boiling heat transfer coefficient in liquid nitrogen bath.展开更多
Fe-based carbon materials are widely considered promising to replace Pt/C as next-generation electrocatalysts towards oxygen reduction reaction (ORR). However, the preparation of Fe-based carbon materials is still car...Fe-based carbon materials are widely considered promising to replace Pt/C as next-generation electrocatalysts towards oxygen reduction reaction (ORR). However, the preparation of Fe-based carbon materials is still carried out by conventional heating method (CHM). Herein, a novel microwave-assisted carbon bath method (MW-CBM) was proposed, which only took 35 min to synthesize Fe/Fe3C nanoparticles encapsulated in N-doped carbon layers derived from Prussian blue (PB). The catalyst contained large specific surface area and mesoporous structure, abundant Fe-Nx and C–N active sites, unique core-shell structure. Due to the synergistic effects of these features, the as-prepared Fe/Fe3C@NC-2 displayed outstanding ORR activity with onset potential of 0.98 VRHE and halfwave potential of 0.87 VRHE, which were more positive than 20 wt.% Pt/C (0.93 VRHE and 0.82 VRHE). Besides, Fe/Fe3C@NC-2 gave a better stability and methanol tolerance than Pt/C towards ORR in alkaline media, too.展开更多

Graphitic carbon nitride(g-C_(3)N_(4))-based materials are regarded as one of the most potential photocatalysts for utilizing solar energy.In this work,we reported a facile one step in-situ hydrothermal-roasting metho...Graphitic carbon nitride(g-C_(3)N_(4))-based materials are regarded as one of the most potential photocatalysts for utilizing solar energy.In this work,we reported a facile one step in-situ hydrothermal-roasting method for preparing honeycomb-like g-C_(3)N_(4)/CeO_(2) nanosheets with abundant oxygen vacancies(g-C_(3)N_(4)/CeO_(2)-x).The hydrothermal-roasting and incomplete-sealed state can(i)generate an in-situ reducing atmosphere(CO,N2,NH3) to tune the concentration of oxygen vacancies in CeO_(2);(ii) beneficial to prevent continuous growth of g-C_(3)N_(4) and results in honeycomb-like g-C_(3)N_(4)/CeO_(2)-x hybrid nanosheets.What is more,the g-C_(3)N_(4)/CeO_(2)-x photocatalyst exhibited extended photoresponse range,increased specific surface area and obviously enhanced separation efficiency of photogenerated electron-hole pairs.As a proof-of-concept application,the optimized g-C_(3)N_(4)/CeO_(2)-xnanosheets could achieve 98% removal efficiency for Cr(Ⅵ) under visible light irradiation(λ≥420 nm)within 2.5 h,which is significantly better than those of pure g-C_(3)N_(4) and CeO_(2).This work provides a new idea for more rationally designing and constructing g-C_(3)N_(4)-based catalysts for efficient extended photochemical application.展开更多

Surface morphology and ab...Temperature-dependent photoluminescence characteristics of organic-inorganic halide perovskite CH3NH3Pb I3-xClx films prepared using a two-step method on ZnO/FTO substrates were investigated. Surface morphology and absorption characteristics of the films were also studied. Scanning electron microscopy revealed large crystals and substrate coverage. The orthorhombic-to-tetragonal phase transition temperature was-140 K. The films' exciton binding energy was 77.6 ± 10.9 meV and the energy of optical phonons was 38.8 ± 2.5 meV. These results suggest that perovskite CH3NH3Pb I(3-x)Clx films have excellent optoelectronic characteristics which further suggests their potential usage in perovskitebased optoelectronic devices.展开更多

The remaining oil distributed in the whole reservoir is calculated according to the characteristics of the space distribution of the saturation of remaining oil. Logging data are required to accomplish this. However, many such projects cannot be completed. Since the old study of remaining oil distribution could not be quantified efficiently, the "dynamic two-step method" is presented. Firstly, the water cut of every flow unit in one well at one time is calculated according to the comprehensive water cut of a single well at one time. Secondly, the remaining oil saturation of the flow unit of the well at one time is calculated based on the water cut of the flow unit at a given time. The results show that "dynamic two-step method" has characteristics of simplicity and convenience, and is especially suitable for the study of remaining oil distribution at high water-cut stage. The distribution of remaining oil presented banding and potato form, remaining oil was relatively concentrated in faultage neighborhood and imperfect well netting position, and the net thickness of the place was great. This proposal can provide an effective way to forecast remaining oil distribution and enhance oil recovery, especially applied at the high water-cut stage.展开更多

These problems arise in many fields of science and engineering including those dealing with real applications. The physical problem is modeled by partial differential equations (PDEs) and involve optimization of some quantity. The PDEs are in most cases nonlinear and solved using numerical methods. Since such numerical solutions are being used routinely, the recent trend has been to develop numerical methods and algorithms so that the optimization problems can be solved numerically as well using the same PDE-solver. We present here one such numerical method which is based on simultaneous pseudo-time stepping. The efficiency of the method is increased with the help of a multigrid strategy. Application example is included for an aerodynamic shape optimization problem.展开更多

The total length of the synthesized gene is 571 bp; 18 oligonucleotides, DNA fragments were designed and synthesized by the phosphoramidite four-step chemical method. The whole DNA sequence was synthesized by a one-step total gene synthesis method, and then inserted in pUC18 vector. Five positive clones identified by blue-white colony screening were sent to Shanghai Sangon Biological Engineering Technology and Service Co., Ltd. for sequencing. The sequencing result shows that one clone contained the complete correct gene in all the five positive clones.展开更多

The fractional steps method consists of splitting the multi-dimensional matrix inversion problem into an equivalent one dimensional problem which is successively integrated in every direction along the characteristics using the Riemann invariant associated with the cubic spline interpolation. The height and the velocity field of the shallow water equations over irregular bottom are discretized on a fixed Eulerian grid and time-stepped using the fractional steps method. Effects of the Coriolis force and the bottom topography for particular initial flows on the velocity components and the free surface elevation have been studied and the results are plotted.展开更多

文摘Graphitic carbon nitride(g-C_(3)N_(4))-based materials are regarded as one of the most potential photocatalysts for utilizing solar energy.In this work,we reported a facile one step in-situ hydrothermal-roasting method for preparing honeycomb-like g-C_(3)N_(4)/CeO_(2) nanosheets with abundant oxygen vacancies(g-C_(3)N_(4)/CeO_(2)-x).The hydrothermal-roasting and incomplete-sealed state can(i)generate an in-situ reducing atmosphere(CO,N2,NH3) to tune the concentration of oxygen vacancies in CeO_(2);(ii) beneficial to prevent continuous growth of g-C_(3)N_(4) and results in honeycomb-like g-C_(3)N_(4)/CeO_(2)-x hybrid nanosheets.What is more,the g-C_(3)N_(4)/CeO_(2)-x photocatalyst exhibited extended photoresponse range,increased specific surface area and obviously enhanced separation efficiency of photogenerated electron-hole pairs.As a proof-of-concept application,the optimized g-C_(3)N_(4)/CeO_(2)-xnanosheets could achieve 98% removal efficiency for Cr(Ⅵ) under visible light irradiation(λ≥420 nm)within 2.5 h,which is significantly better than those of pure g-C_(3)N_(4) and CeO_(2).This work provides a new idea for more rationally designing and constructing g-C_(3)N_(4)-based catalysts for efficient extended photochemical application.

文摘Temperature-dependent photoluminescence characteristics of organic-inorganic halide perovskite CH3NH3Pb I3-xClx films prepared using a two-step method on ZnO/FTO substrates were investigated. Surface morphology and absorption characteristics of the films were also studied. Scanning electron microscopy revealed large crystals and substrate coverage. The orthorhombic-to-tetragonal phase transition temperature was-140 K. The films' exciton binding energy was 77.6 ± 10.9 meV and the energy of optical phonons was 38.8 ± 2.5 meV. These results suggest that perovskite CH3NH3Pb I(3-x)Clx films have excellent optoelectronic characteristics which further suggests their potential usage in perovskitebased optoelectronic devices.
文摘The distribution of remaining oil is often described qualitatively. The remaining oil distributed in the whole reservoir is calculated according to the characteristics of the space distribution of the saturation of remaining oil. Logging data are required to accomplish this. However, many such projects cannot be completed. Since the old study of remaining oil distribution could not be quantified efficiently, the "dynamic two-step method" is presented. Firstly, the water cut of every flow unit in one well at one time is calculated according to the comprehensive water cut of a single well at one time. Secondly, the remaining oil saturation of the flow unit of the well at one time is calculated based on the water cut of the flow unit at a given time. The results show that "dynamic two-step method" has characteristics of simplicity and convenience, and is especially suitable for the study of remaining oil distribution at high water-cut stage. The distribution of remaining oil presented banding and potato form, remaining oil was relatively concentrated in faultage neighborhood and imperfect well netting position, and the net thickness of the place was great. This proposal can provide an effective way to forecast remaining oil distribution and enhance oil recovery, especially applied at the high water-cut stage.
文摘As a new method, the Level Set method had been developed to compute the interface of two-phase flow. The basic mathematical theory and the detailed method to solve the free surface hydrodynamic problem had been investigated. By using the Level Set method, the transformation of a solitary wave over a front step was simulated. The results were in good agreement with laboratory experiments.

文摘A membrane element model with bending modification based on element moment equilibrium is proposed for the first time by the authors, who apply the element model in one step inverse method and simulate the forming process of a flower-shaped box using the membrane element model with and without this modification. The numerical results are compared with those of the incremental method to verify the validity of the element model developed in this paper.
文摘This paper presents a numerical method for PDE-constrained optimization problems. These problems arise in many fields of science and engineering including those dealing with real applications. The physical problem is modeled by partial differential equations (PDEs) and involve optimization of some quantity. The PDEs are in most cases nonlinear and solved using numerical methods. Since such numerical solutions are being used routinely, the recent trend has been to develop numerical methods and algorithms so that the optimization problems can be solved numerically as well using the same PDE-solver. We present here one such numerical method which is based on simultaneous pseudo-time stepping. The efficiency of the method is increased with the help of a multigrid strategy. Application example is included for an aerodynamic shape optimization problem.
文摘According to the amino acid sequence and codon preference of E. coli, the human interleukin-18(IL-18) gene was optimized to avoid the rare codons. The total length of the synthesized gene is 571 bp; 18 oligonucleotides, DNA fragments were designed and synthesized by the phosphoramidite four-step chemical method. The whole DNA sequence was synthesized by a one-step total gene synthesis method, and then inserted in pUC18 vector. Five positive clones identified by blue-white colony screening were sent to Shanghai Sangon Biological Engineering Technology and Service Co., Ltd. for sequencing. The sequencing result shows that one clone contained the complete correct gene in all the five positive clones.
文摘The two-dimensional nonlinear shallow water equations in the presence of Coriolis force and bottom topography are solved numerically using the fractional steps method. The fractional steps method consists of splitting the multi-dimensional matrix inversion problem into an equivalent one dimensional problem which is successively integrated in every direction along the characteristics using the Riemann invariant associated with the cubic spline interpolation. The height and the velocity field of the shallow water equations over irregular bottom are discretized on a fixed Eulerian grid and time-stepped using the fractional steps method. Effects of the Coriolis force and the bottom topography for particular initial flows on the velocity components and the free surface elevation have been studied and the results are plotted.
文摘Recently, the incidence of<span> </span><i><span>Candida</span></i><span> infections has substantially increased. Conventional identification methods for </span><i><span>Candida</span></i><span> species are technically difficult to conduct and cannot accurately distinguish each species. The purpose of the present study was to design primers to identify and detect simultaneously</span><span> </span><span>eight medically important </span><i><span>Candida</span></i><span> species using one-step multiplex PCR. PCR primers were designed based on partial sequences of intergenic spacer (IGS) and internal transcribed spacer (ITS) genes of eight medically important </span><i><span>Candida</span></i><span> species. These primers were able to distinguish each </span><i><span>Candida</span></i><span> species and did not display cross-reactivity with representative </span><i><span>Candida </span></i><span>species other than the eight</span><i><span> Candida</span></i><span> species. Moreover, our developed one-step multiplex PCR method is accurate, specific, cost-effective, time-saving, and worked without requiring DNA extraction.</span>
