Other members of the same family of transcription factors were found to have a role in intestinal metaplasia and gastric cancer (OCT-1) and in trophectoderm differentiation of embryonic stem cells (OCT3/4) via modulation ofCDX2. These findings prompted us to evaluate whether OCT-2 could in fact have a role in gastric oncogenesis and lymph node metastasis in human gastric cancer, designing an immunohistochemistry study of the putative expression of OCT-2 on human gastric cancer tissue of 69 surgical archival specimens of intestinal and diffuse type. We could not find expression of OCT-2 on any of our cancer tissues, metaplasia or normal gastric mucosa despite the expression of OCT-2 on tonsil lymphatic cells (tissue test) and normal lymph nodes. We concluded that, based on immunohistochemistry, OCT-2 does not have a role in gastric oncogenesis nor does have any relationship with lymph node metastasis in gastric cancer.展开更多

文摘目的探讨转录因子Oct-4与Sox-2在卵巢癌组织中的表达及临床意义。方法选取36例卵巢癌组织和19例正常卵巢上皮组织标本,采取免疫组织化学方法和Western blot方法检测两组组织标本中Oct-4和Sox-2的蛋白表达水平,采用荧光定量PCR法检测两组组织标本中Oct-4、Sox-2 mRNA的表达量,分析卵巢癌组织Oct-4、Sox-2的蛋白表达与卵巢癌临床病理参数的关系。结果卵巢癌组织中Oct-4与Sox-2蛋白表达的平均光密度值显著高于正常卵巢上皮组织(P<0.01);卵巢癌组织中Oct-4与Sox-2 m RNA的相对表达水平亦显著高于正常卵巢上皮组织(P<0.01)。Oct-4与Sox-2在卵巢癌组织中的表达与卵巢癌的肿瘤分级密切相关(P<0.01),与患者年龄、病理类型和淋巴结转移无关(P>0.05)。结论卵巢癌组织中Oct-4和Sox-2的表达上调,并与卵巢癌的肿瘤分级密切相关。
