目的将全长γ9链mRNA和肿瘤反应性CDR3移植的δ2链mRNA共转染到抗CD3抗体刺激的PBMC,制备CDR3δ2基因修饰型γδT淋巴细胞,研究其抗肿瘤作用。方法通过PCR扩增含有肿瘤反应性特异CDR3δ2序列(OT3和OT10)的TCRδ2链及γ9链,构建重组质粒p... 目的将全长γ9链mRNA和肿瘤反应性CDR3移植的δ2链mRNA共转染到抗CD3抗体刺激的PBMC,制备CDR3δ2基因修饰型γδT淋巴细胞,研究其抗肿瘤作用。方法通过PCR扩增含有肿瘤反应性特异CDR3δ2序列(OT3和OT10)的TCRδ2链及γ9链,构建重组质粒p GEM4Z/δ2(OT3)/A64、p GEM4Z/δ2(OT10)/A64和p GEM4Z/γ9/A64;以线性化的重组质粒作为模板,体外合成δ2(OT3)、δ2(OT10)和γ9 mRNA;将δ2(OT3)mRNA和δ2(OT10)mRNA分别与γ9 mRNA共电转染入抗CD3抗体刺激的正常人PBMC;经流式细胞术检测和分选γ9δ2(OT3)T细胞和γ9δ2(T10)T细胞。ELISA及MTT法分别检测上述转染细胞的抗肿瘤作用。结果 CDR3δ2基因修饰的TCRγδ细胞γ9δ2(OT3)T细胞和γ9δ2(T10)T细胞能够分泌较高的IFN-γ及TNF-α(P<0.05),对多种肿瘤细胞具有显著的细胞毒作用(P<0.05)。结论基因修饰型γ9δ2 T细胞能够明显增强对肿瘤的杀伤活性,为肿瘤过继免疫细胞的制备提供了新的策略。 展开更多

Rice is the most important crop plant and serves as a model for cereal genomics. Rice epigenomic landscape is emerging and the function of chromatin modification regulators in gene expression, transposon repression and plant development is being characterized. Epigenomic variation that gives rise to stable or transgenerational heritable epialleles related to variation of important agronomical traits or stress responses is being characterized in rice. 目的实现腺苷酸基琥珀酸(盐)(S-AMP)的大规模合成,为开展药物学等研究提供原料。方法以pET-28-a为表达载体,利用大肠杆菌表达古细菌Pyrococcus horikoshii OT3来源的腺苷酸基琥珀酸合成酶(PhAdSS),利用Ni-NTA层析柱纯化后作为催化剂在... 目的实现腺苷酸基琥珀酸(盐)(S-AMP)的大规模合成,为开展药物学等研究提供原料。方法以pET-28-a为表达载体,利用大肠杆菌表达古细菌Pyrococcus horikoshii OT3来源的腺苷酸基琥珀酸合成酶(PhAdSS),利用Ni-NTA层析柱纯化后作为催化剂在实验室内开展这种微生物体内合成S-AMP的反应。用Bradford法测定纯化后PhAdSS的浓度。利用硅胶薄层层析检测反应进度。用硅胶柱层析法和重结晶法纯化S-AMP,利用质谱法测定合成品中S-AMP的分子量,利用紫外分光光度法测定合成品内S-AMP的含量及回收率。结果经纯化后从1 L的自动诱导培养基中至少获得20 mg的His-tagged-PhAdSS。将含有10 mmol/L肌苷酸、11 mmol/L L-天冬氨酸、20 mmol/L鸟苷三磷酸、4 mmol/L MgCl_2、2.9μmol/L的His-tagged-PhAdSS溶液于常压环境中,70℃恒温6 h以上可以实现IMP完全转化为S-AMP。纯化后可得到S-AMP的单晶体,利用紫外分光光度法测定的纯度为94%,收率为17%。结论实现了PhAdSS为催化剂的S-AMP的大量合成。 展开更多

The Eu3+-doped La2Zr207 phosphor with rod-like morphology was successfully synthesized by conventional solid state reaction and hydrothermal method. X-ray diffraction patterns, transmission electron microscopy, and photoluminescence spectra were employed to charac- terize its structure and morphology as well as luminescent properties. The results indicated that the red-emitting phosphor La2Zr207:Eu3+ had well crystallized and belonged to the cubic structure with space group of Fd3m. The as-obtained product mainly appeared as straight nanorods with an average diameter of 47 nm and length of 50-700 nm. The pos- sible growth mechanism was also discussed. It was found that under blue excitation with a wavelength of 466 nm, the La2Zr2OT:Eu3+ phosphor exhibited a characteristic red emission at 616 nm that was attributed to the hypersensitive 5D0--*TF2 electric dipole transition of Eu3+ ions. Meanwhile, it was more interesting to note that the emission of 5D1--*TFj (J=0, 1, 2) transitions and the splitting patterns of 5D0---+TFJ (J--l, 2, 4) transitions of Eu3+ ions can be observed in the luminescent spectra of La2Zr207:Eu3+. It was demonstrated that Eu3+ preferred to occupy a low symmetry site. 展开更多

In present study, the effect of the solvent annealing temperature on the crystal modifications and the phase transition behavior of the subsequently dried poly(3-octylthiophene) (P3OT) film has been investigated by the combination of DSC, WAXD and FTIR techniques. When chloroform is employed as the solvent, it is unexpectedly found that form I and form II crystal modifications of P3OT could be respectively obtained by room temperature and low temperature annealing. Comparing to the mostly used solvent reported for preparing form II, i.e. carbon disulfide (CS2) which is toxic and corrosive, chloroform is less toxic and corrosive and more suitable for solution processing of P3OT. Therefore, this finding provides an alternative way to obtain form II. By temperature dependent IR spectroscopy, the structural evolution of P3OT during the form II to form I phase transition process has also been studied in detail. 展开更多

Some high T_c superconductors REBa_2Cu_3O_(7-x)(RE=Sm,Eu,Gd,Dy,Ho,Er,Tm) with zero resist- ance temperatures were about 90K have been prepared.The effect of magnetic rare earth ions on the transi- tion temperatures is not very great,but their magnetic behaviours are different from each other.The magnetization loops of these compounds have been measured at 77K.The coexistence of superconductivity with magnetic ordering (COSM) found in the samples with RE=Gd,Dy,Ho,Er,Tm,indicates that there are two possible ways of coexistence in the samples.These expertments show that the relation of paramagnetism to the atomtc number of rare earth elements for these superconductors is similar to that of rare earth ions in some paramagnetic salt crystals,which indicates that the magnetism of the samples origi- nates from the local magnetic moments of rare earth ions.In some samples there is antiferromagnetie ordering at low temperatures,and the effective Bohr magneton numbers are consistent with that of free rare earth ions. Some superconducting parameters including the critical current density are calculated according to Ginzburg-Landau theory and the critical state model. 展开更多

Effect of annealing treatment on the performance of organic photovoltaic devices using SPFGraphene as electron-accepter material

We have researched the performances of organic photovoltaic devices with the bulk heterojunction (BHJ) structure using the organic solution-processable functionalized graphene (SPFGraphene) material as the electron-accepter material and P3OT as the donor material. The structural configuration of the device is ITO/PEDOT:PSS/P3OT:PCBM-SPFGraphene/LiF/A1. Given the P3OT/PCBM (1:1) mixture with 8wt% of SPFGraphene, the open-circuit voltage (Voc) of the device reaches 0.64 V, a short-circuit current density (J^c) reaches 5.7 mA/cm2, a fill factor (FF) reaches 0.42, and the power conversion efficiency (7?) reaches 1.53% at illumination at 100 mW/cm2 AM1.5. We further studied the reason for the device performances improvement In the P3OT:PCBM-SPFGraphene composite, the SPFGraphene material acts as exciton dissociation sites and provides the transport pathways of the lowest unoccupied molecular orbital (LUMO)-SPFGraphene-A1. Furthermore, adding SPFGraphene to P3OT results in appropriate energetic distance between the highest occupied molecular orbital (HOMO) and LUMO of the donoffacceptor and provides higher exciton dissociation volume mobility of carrier transport. We have researched the effect of annealing treatment for the devices and found that the devices with annealing treatment at 180℃ show better performances compared with devices without annealed treatment. The devices with annealed treatment show the best performance, with an enhancement of the power conversion efficiency from 1.53% to 1.75%. 展开更多
