不同微塑料胁迫下水稻(Oryza sativa)的生理生态响应以及转录组学分析 被引量:1

为缓解传统塑料使用废弃后产生的微塑料污染问题,可降解塑料日益受到关注.可降解塑料在环境中易老化,而有关可降解塑料对植物的影响尚较鲜见报道,可降解塑料老化后给环境带来的生态风险也并不明确.为了揭示不同微塑料对植物的生态效应,... 为缓解传统塑料使用废弃后产生的微塑料污染问题,可降解塑料日益受到关注.可降解塑料在环境中易老化,而有关可降解塑料对植物的影响尚较鲜见报道,可降解塑料老化后给环境带来的生态风险也并不明确.为了揭示不同微塑料对植物的生态效应,以水稻(Oryza sativa)为研究对象,选择新鲜可降解微塑料(PLA-MPs)、老化降解后微塑料(aged PLA-MPs)和传统微塑料(PE-MPs),研究不同微塑料胁迫下水稻的生理生态响应特征和转录组学变化特征.结果表明,水稻应对不同微塑料胁迫产生的生态效应不同,PLA-MPs和APLA-MPs相比于PE-MPs更易诱导水稻产生严重的氧化应激.与对照CK组相比,PE组和APLA组水稻的SOD值分别显著提高17.41%和36.48%,PE组和PLA组的POD分别显著提高21.91%和48.65%;PLA组和APLA组胁迫下的CAT值分别显著提高29.34%和24.91%;PLA组和APLA组胁迫下的MDA含量分别显著提高70.52%和135.94%.微塑料暴露下水稻叶绿素以及叶绿素荧光参数产生显著变化,PLA组和APLA组水稻叶绿素含量分别显著降低21.28%和12.77%;最大光学量子产量(F_(v)/F_(m))分别显著降低13.95%和44.19%;非光化学荧光淬灭(NPQ_Lss)分别显著提升222.64%和143.40%.转录组测序分析表明,微塑料暴露导致水稻四吡咯结合、血红素结合、氧化还原酶、铁离子结合和苯丙烷生物合成通路富集,APLA组的水稻更多集中于水解酶类通路和氨基酸类代谢与生物合成通路富集,研究结果对于综合评价环境中可降解微塑料的潜在生态风险及其对植物的生态效应具有一定的实践与理论价值. 展开更多

格点预报数据与ORYZA模型耦合的吉林水稻花期障碍型冷害评估 被引量:1

基于格点预报数据和作物模型耦合建立水稻花期冷害产量损失评估模型,以提升水稻花期冷害评估能力。利用1993-2000年吉林省通化农业气象试验站水稻观测资料,确定ORYZA.v3模型中水稻生长发育关键参数;基于校准后模型,利用1961-2021年气象... 基于格点预报数据和作物模型耦合建立水稻花期冷害产量损失评估模型,以提升水稻花期冷害评估能力。利用1993-2000年吉林省通化农业气象试验站水稻观测资料,确定ORYZA.v3模型中水稻生长发育关键参数;基于校准后模型,利用1961-2021年气象数据资料和冷害灾情统计数据分析水稻障碍型冷害的敏感性;结合未来10d的0.125°×0.125°欧洲中期天气预报中心(ECMWF)的格点预报数据和天气要素实况,建立水稻花期冷害评估方法,定量评估2022-2023年冷害对水稻产量的影响。结果表明:(1)基于ORYZA.v3模型水稻开花期、成熟期和产量模拟值与实测值的MAE分别为1.2d、2.6d和345.7kg·hm^(-2),模型可较好地模拟水稻关键生育期和产量。(2)水稻花期障碍型冷害强度(降温幅度、持续日数)与水稻产量减损呈显著相关,冷害发生时日平均气温每降低1.0℃,水稻平均减产2.13%;低温持续日数每增加1d,水稻平均减产2.66%。典型障碍型冷害发生年份内模型模拟误差在±10.0个百分点内,模拟精度较高,可定量模拟障碍型冷害对水稻产量减损的影响。(3)2022-2023年轻、中和重度冷害导致的水稻减产率分别为3.0%、3.7%和5.1%;轻、中和重度障碍型冷害评估TS评分为0.76,0.50和0.63。评估与实际发生的冷害等级空间范围基本一致,TS综合评分为0.63,效果较好。 展开更多

Oryza longistaminata is an African wild rice species with valuable agronomic traits and the donor parent of perennial rice.Endophytic bacteria play an important role in host health,adaptive evolution and stress tolerance.However,endophytic bacterial communities in O.longistaminata and their plant growth-promoting(PGP)effects on the perennial rice of O.longistaminata offspring are poorly understood.In this study,the endophytic bacterial diversity,composition and network structures in the root,stem,and leaf tissues of O.longistaminata were characterized using Illumina sequencing of the 16S rRNA gene.The results suggested that O.longistaminata contains a multitude of niches for different endophytic bacteria,among which the root endosphere is more complex and functionally diverse than the stem and leaf endospheres.Tissue-specific biomarkers were identified,including Paludibaculum,Pseudactinotalea and Roseimarinus and others,for roots,Blautia for stems and Lachnospiraceae NK4A136 for leaves.The endophytic bacterial network of O.longistaminata was reassembled for various functions,including degradation/utilization/assimilation,detoxification,generation of precursor metabolites and energy,glycan pathways,macromolecule modification and metabolism.A total of 163 endophytic bacterial strains with PGP traits of potassium release,phosphate solubilization,nitrogen fixation,siderophore activity,indole-3-acetic acid(IAA)production,and 1-aminocyclopropane-1-carboxylate(ACC)deaminase activity were isolated from O.longistaminata.Eleven strains identified as Enterobacter cloacae,Enterobacter ludwigii,Stenotrophomonas maltophilia,Serratia fonticola,and Bacillus velezensis showed stable colonization abilities and PGP effects on perennial rice seedlings.Inoculated plants generally exhibited an enhanced root system and greater photosynthesis,biomass accumulation and nutrient uptake.Interestingly,two strains of E.cloacae have host genotype-dependent effects on perennial rice growth.The results of this study provide insights into the endophytic bacterial ecosystems of O.longistaminata,which can potentially be used as biofertilizers for sustainable perennial rice productivity. 展开更多

ORYZA(V3)模型对珠三角地区双季稻参数的模拟效果及验证分析

以珠三角地区为研究区,基于2018—2020年不同播期双季稻的生长发育观测数据以及当地气象和土壤数据,利用2018—2019年的数据对ORYZA(V3)模型进行校准,以调整和确定作物的基本参数,使用2020年双季稻的生育期、叶面积指数和生物量等数据... 以珠三角地区为研究区,基于2018—2020年不同播期双季稻的生长发育观测数据以及当地气象和土壤数据,利用2018—2019年的数据对ORYZA(V3)模型进行校准,以调整和确定作物的基本参数,使用2020年双季稻的生育期、叶面积指数和生物量等数据对模型进行验证。结果表明,ORYZA(V3)模型对珠三角双季稻的生育期具有较高的模拟精度,与实测值相比误差范围为0~3 d。ORYZA(V3)模型对2020年早稻和晚稻叶面积指数动态变化的模拟准确率高,模拟值的平均线性回归系数(α值)接近于1,相关系数(R^(2))分别为0.670 4和0.766 0,t检验结果显示,模拟值与实测值之间没有显著差异(P>0.05)。ORYZA(V3)模型对水稻地上部各器官生物量的模拟误差较大,但从统计学角度来看但仍在合理范围内,且对晚稻的模拟效果优于早稻。由此可知,通过校准作物参数,ORYZA(V3)模型能准确地模拟水稻的生长情况,可以应用于珠三角地区的水稻生产。 展开更多

The N-mannosyltransferase MoAlg9 plays important roles in the development and pathogenicity of Magnaporthe oryzae

Glycosylation plays key roles in vegetative growth,development, and infection of M. oryzae. However, several glycosylation-related genes have not been characterized.In this study, we identified a Glyco_transf_22 domain-containing protein, MoAlg9, and found that MoAlg9 islocalized to the endoplasmic reticulum(ER). Deletion of MoALG9 significantly affected conidial production, normalappressorium formation, responses to stressors, and pathogenicity of M. oryzae. We also found that the ΔMoalg9mutant was defective in glycogen utilization, appressorial penetration, and invasive growth in host cells. Moreover,we further demonstrated that MoALG9 regulates the transcription of several target genes involved in conidiation,appressorium formation, and cell wall integrity. In addition, we found that the Glyco_transf_22 domain is essentialfor normal MoAlg9 function and localization. We also provide evidence that MoAlg9 is involved in N-glycosylationpathway in M. oryzae. Taken together, these results show that MoAlg9 is important for conidiation, appressoriumformation, maintenance of cell wall integrity, and the pathogenesis of M. oryzae. 展开更多

Rice blast,caused by Magnaporthe oryzae,is a fungal disease that causes devastating damage to rice production worldwide.During infection,pathogens secrete effector proteins that modulate plant immunity.Disulfide bond formation catalyzed by protein disulfide isomerases(PDI)is essential for protein folding and maturation.However,the biological function of Pdi1 in M.oryzae has not yet been characterized.In this study,we identified the endoplasmic reticulum(ER)-located protein,MoPdi1,in M.oryzae.MoPdi1 regulates conidiation,cell wall stress,and pathogenicity of M.oryzae.Furthermore,the CGHC active sites in the a and a'redox domain of MoPdi1 were essential for the biological function of MoPDI1.Further tests demonstrated that MoPdi1 was involved in the regulation of ER stress and positively regulated ER phagy.We also found that MoPdi1 interacted with MoHut1.Deletion of MoPDI1 led to the bereft of MoHut1 dimerization,which depends on the formation of disulfide bonds.In addition,MoPdi1 affected the normal secretion of the cytoplasmic effector AVR-Pia.We provided evidence that MoHut1 is important for the vegetative growth,conidiation,and pathogenicity in M.oryzae.Therefore,our findings could provide a suitable target point for designing antifungal agrochemicals against rice blast fungus. 展开更多

Xanthomonas oryzae pv.oryzae(Xoo)causes bacterial blight in rice,which reduces crop yield and leads to significant economic losses.Bacterial sigma(σ)factors are highly specialized proteins that allow RNA polymerase to recognize and bind to specific promoters.σ^(70) factors also regulate the expression of genes involved in stress response and virulence.However,the role of RpoD in Xoo is still unclear.In this study,we found thatσ^(70) factor RpoD is quite conservative among phytopathogenic bacteria,especially in Xanthomonas sp.In Xoo,PXO_RpoD plays an important role in oxidative stress tolerance and cell motility,as well as being essential for full virulence.Cleavage under targets and tagmentation(CUT&Tag)analyses indicated that RpoD mediates the type three secretion system(T3SS)by regulating the regulation of hrpG and hrpX.By performing bacterial one-hybrid and electrophoretic mobility assay(EMSA),we observed that RpoD directly bound to the promoters of hrpG and hrpX.Collectively,these results demonstrate the transcriptional mechanism and pathogenic functions of RpoD in regulating cell motility and oxidative stress response,providing novel insights into potential targets for disease control. 展开更多

PDIL2-3 encoding a protein disulfide isomerase-like enzyme is essential for grain yield and appearance quality in rice(Oryza sativa L.)

Rice grain size and chalkiness are important traits that influence grain yield and quality,respectively.Mining of genes for grain yield and appearance quality and clarification of their action modes are of great importance in rice breeding.In this study,a rice protein disulfide isomerase-like enzyme PDIL2-3 was characterized.Expression analysis revealed that PDIL2-3 was highly expressed in endosperm and spikelet hulls.The PDIL2-3-cri lines generated by CRISPR/Cas9 technology exhibited a chalky grain phenotype with altered storage substance accumulation and increased grain size and weight,whereas exactly opposite results were obtained for PDIL2-3 overexpression lines.Cytological experiments revealed that PDIL2-3-cri increased rice seed length mainly by increasing the cell number and rice seed width mainly by increasing the cell size in grains,implying that PDIL2-3 regulates the grain size by influencing both cell division and expansion of spikelet hulls.Further flow cytometric analysis validated that PDIL2-3 has a negative effect on cell proliferation,preventing DNA duplication and cell division in spikelet hulls.Moreover,q RT-PCR results showed that the expression levels of genes related to cell cycle and storage substance synthesis were significantly changed in PDIL2-3-cri transgenic lines.Thus,our results indicated that PDIL2-3 plays a pivotal role in influencing grain size and quality of rice by affecting cell division/expansion and storage substance accumulation,providing new insights into the function of PDIL family members in rice and enriching the genetic resources for rice breeding. 展开更多

Oxidation of self-stored carbohydrates and lipids provides the energy for the rapid morphogenetic transformation during asexual and infection-related development in Pyricularia oryzae,which results in intracellular accumulation of reducing equivalents NADH and FADH_(2),requiring a cytosolic shuttling machinery towards mitochondria.Our previous studies identified the mitochondrial D-lactate dehydrogenase MoDld1 as a regulator to channel the metabolite flow in conjunction with redox homeostasis.However,the regulator(s)facilitating the cytosolic redox balance and the importance in propelling nutrient metabolite flow remain unknown.The G-3-P shuttle is a conserved machinery transporting the cytosolic reducing power to mitochondria.In P.oryzae,the mitochondrial G-3-P dehydrogenase Gpd2 was required for cellular NAD^(+)/NADH balance and fungal virulence.In this study,we relocate the mitochondrial G-3-P dehydrogenase Gpd2 to the cytosol for disturbing cytosolic redox status.Our results showed overexpression of cytosolic gpd2^(Δmts)without the mitochondrial targeted signal(MTS)driven by Ribosomal protein 27 promoter(PR27)exerted conflicting regulation of cellular oxidoreductase activities compared to theΔModld1 deletion mutant by RNA-seq and prevented the conidiation and pathogenicity of P.oryzae.Moreover,overexpression of gpd2^(Δmts)caused defects in glycogen and lipid mobilization underlying asexual and infectious structural development associated with decreased cellular NADH production and weakened anti-oxidation activities.RNA-seq and non-targeted metabolic profiling revealed down-regulated transcriptional activities of carbohydrate metabolism and lower abundance of fatty acids and secondary metabolites in RP27:gpd2^(Δmts).Thus,our studies indicate the essential role of cytosolic redox control in nutrient metabolism fueling the asexual and infection-related development in P.oryzae. 展开更多

In this study,we searched for dispersed repeats(DRs)in the rice(Oryza sativa)genome using the iterative procedure(IP)method.The results revealed that the O.sativa genome contained 79 DR families,comprising 992739 DNA repeats,of which 496762 and 495977 were identified on the forward and reverse DNA strands,respectively.The detected DRs were,on average,374 bp in length and occupied 66.4%of the O.sativa genome.Totally 61%of DRs,identified by the IP method,overlapped with previously annotated dispersed repeats(ADRs)detected using the Extensive De Novo TE Annotator(EDTA)pipeline. 展开更多

This study explores the broad-spectrum application of OsRALF26,a small secreted peptide belonging to the rapid alkalinization factor(RALF)family in rice.We found that the rice genome carries numerous lineage-specific OsRALFs,suggesting that this evolutionary expansion could be the result of an arms race with pathogens.Among them,we focused on the Oryza-specific Os RALF26 and its closest homolog,OsRALF27,analyzing their effects across a range of plant species from monocots to dicots.The exogenous application of OsRALF26 significantly reduced bacterial populations in rice challenged with Xanthomonas oryzae pv.oryzae(Xoo)and in Arabidopsis and tomato challenged with Pseudomonas syringae pv.tomato DC3000(Pst DC3000),whereas Os RALF27 did not enhance resistance. 展开更多

The polymerase chain reaction(PCR) is particularly useful for plant pathogen detection. In the present study, multiplex PCR and SYBR Green real-time PCR were developed to facilitate the simultaneous detection of three important rice pathogens, Xanthomonas oryzae pv.oryzae, X. oryzae pv. oryzicola, and Burkholderia glumae. The unique PCR primer sets were designed from portions of a putative glycosyltransferase gene of X. oryzae pv. oryzae, an Avr Rxo gene of X. oryzae pv. oryzicola, and an internal transcribed spacer(ITS) sequence of B. glumae. Using a multiplex PCR assay, X. oryzae pv. oryzae, X. oryzae pv. oryzicola, and B. glumae were detected in one PCR reaction that contained the newly developed primer set mix. Using SYBR Green real-time PCR assays, X. oryzae pv. oryzae, X. oryzae pv. oryzicola, and B. glumae were detected at 1, 1, and 10 fg μL-1, respectively. These newly designed molecular assays are sensitive and could be reliable tools for pathogen detection and disease forecasting. 展开更多

Oryza sativa L. ssp. japonica and indica exhibit different sensitivity to photoinhibition and they show different stability of their core proteins D1 in the chloroplast photosystem Ⅱ. Using in situ hybridization, psbA, the gene encoding D1 protein of O. sativa ssp. japonica cv. 9516, and that of O. sativa ssp. indica cv. Shanyou 63 was cloned. As revealed by homology comparison of their sequences, the sequences are identical in the regions of promoter and 5′-UTR; differences are found in individual bases in the coding region all of which, being in the third position of respective codons, however, do not affect the amino acids coded finally; a difference is noted in the length of the oligo-U sequence in the region of 3′-UTR. It is thus apparent that, rather than a result of any difference in the amino acid sequences, the differences in the sensitivity to photoinhibition of D1 proteins between japonica and indica rice may be related to the upstream factors that regulate expression of psbA or to differences of photoprotective mechanisms. 展开更多

DNA fingerprints were divided into 112 haplotypes and 14 lineages at 73% genetic similarity level. Among the lineages, no dominant lineages were found. The population genetic structures of Magnaporthe oryzae were not distinctly different in different years. 大气中的微纳塑料(micro-and nanoplastics,MNPs)能够通过干湿沉降进入农田生态系统,这部分MNPs输入农田生态系统后对粮食作物安全生产的潜在威胁尚未明确。本研究选用聚苯乙烯纳米塑料(polystyrene nanoplastics,PS-NPs,200 nm)来模拟... 大气中的微纳塑料(micro-and nanoplastics,MNPs)能够通过干湿沉降进入农田生态系统,这部分MNPs输入农田生态系统后对粮食作物安全生产的潜在威胁尚未明确。本研究选用聚苯乙烯纳米塑料(polystyrene nanoplastics,PS-NPs,200 nm)来模拟大气中MNPs,采用室内受控土壤暴露实验,考察了大气沉降进入农田土壤中的PS-NPs对水稻(Oryza sativaL.)生长、生理生化、籽粒产量与营养品质的影响。研究发现,当土壤暴露于模拟大气沉降PS-NPs后,水稻株高(7.78%~11.09%)、根干质量(15.58%~36.27%)及籽粒产量(30.05%~59.53%)呈现剂量依赖性下降。与此同时,高剂量暴露可能延迟水稻成熟期,导致水稻籽粒含水率提升18.43%(P<0.05)。另外,土壤暴露也使水稻叶片中超氧化物歧化酶(superoxide dismutase,SOD,26.83%~38.84%),过氧化氢酶(catalase,CAT,37.19%~65.87%)和谷胱甘肽S-转移酶(glutathione S-transferase,GST,9.57%~21.81%)活性降低,丙二醛(malondialdehyde,MDA,10.43%~17.39%)含量显著上升(P<0.05);水稻籽粒中SOD(49.22%~56.49%)、CAT(55.29%)和GST(50.00%~57.14%)活性降低,MDA(12.67%~41.63%)含量显著上升(P<0.05),表明大气中的PS-NPs通过沉降输入土壤后,对水稻叶片和籽粒造成了氧化胁迫。水稻籽粒中矿质元素含量的结果表明,相比对照组,土壤中模拟大气沉降的PS-NPs显著降低籽粒中钙(calcium,Ca,20.79%~30.51%)、锰(manganese,Mn,6.60%~27.39%)、锌(zinc,Zn,23.30%~29.97%)、铁(iron,Fe,16.26%~34.32%)和铜(copper,Cu,15.13%~21.16%)的含量,但显著增加了镁(magnesium,Mg,9.31%~12.25%)的含量(P<0.05),这可能与PS-NPs对水稻根系吸收矿质元素能力、水稻体内矿质元素特异性转运蛋白表达以及水稻籽粒对矿质元素积累能力的影响有关。此外,土壤中大气高沉降PS-NPs的暴露也导致了水稻籽粒中硝态氮(13.99%)含量的显著下降,硝酸还原酶(nitrate reductase,NR,21.59%)活性、粗蛋白(11.39%)和酸水解氨基酸(4.71%~21.88%)含量的显著上升(P<0.05),说明PS-NPs可能通过激活NR活性,促使水稻籽粒中酸水解氨基酸与粗蛋白的富集。本研究系统性阐明了植物生育期土壤中大气沉降源PS-NPs的暴露能够通过氧化损伤-矿物质元素失衡-促进有机氮积累等多种途径影响水稻产量与品质,为MNPs农田生态风险评估提供理论支撑。 展开更多

校准与验证水稻生长模型ORYZA2000,为模型本地化、区域化研究应用提供依据。采用湖南双季稻区作物田间观测数据,结合栽培管理措施、土壤以及同期逐日气象数据等资料对ORYZA2000进行参数校正,调试确定了早稻、晚稻有代表性品种的作物参数... 校准与验证水稻生长模型ORYZA2000,为模型本地化、区域化研究应用提供依据。采用湖南双季稻区作物田间观测数据,结合栽培管理措施、土壤以及同期逐日气象数据等资料对ORYZA2000进行参数校正,调试确定了早稻、晚稻有代表性品种的作物参数;利用独立的数据资料,对双季稻生育期、叶面积指数、生物量、产量等指标的模拟结果进行了详细地验证与适应性评价。结果表明:模型对双季稻品种的生育期模拟较好,开花期和成熟期的相对模拟误差为1—2d;早稻和晚稻叶面积指数的归一化均方根误差(NRMSE)均为24%,地上部总生物量、绿叶生物量、茎生物量和穗生物量的NRMSE值分别为18%、22%、22%、24%和19%、24%、28%、28%,产量的NRMSE值分别为11%和16%。校验的作物参数反映了湖南早稻和晚稻品种的生物学特性,参数值合理、有效。通过校准作物参数,ORYZA2000可较为准确地模拟双季稻生长发育及其生物量的动态累积过程,适应性较强,能够应用于双季稻生产。 展开更多
