Aim: To review the accumulated 30 patients with different area of Y chromosome microdeletions, focusing on their correlation with the clinical and pathological findings. Methods: A total of 334 consecutive infertile men with azoospermia (218 patients) and severe oligoasthenospermia (116 patients) were screened. Complete physical and endocrinological examinations, general chromosome study and multiplex polymerase chain reaction assay to evaluate the Y chromosome microdeletion were performed. Ten patients received testicular biopsy. Then the clinical and pathological findings were analyzed with reference to the areas of Y chromosome microdeletion. Results: There is a decline of the percentage of sperm appearing in semen in the group that the gene deletion region from AZFc to AZFb. The clinical evidence of the impairment (decreased testicular size and elevated serum FSH) is also relevantly aggravated in this group. However, the pathology of testicular biopsy specimen was poorly correlated with the different deletion areas of the Y chromosome, which may be due to the limited number of specimens. Conclusion: The clinical correlation of spermatogenic impairment to the different AZF deletion regions may provide the information for the infertile couples in pre-treatment counseling. 展开更多

Background:Oligoasthenospermia is one of the main causes of male infertility.Researchers usually use chemical drugs to directly damage germ cells to prepare oligoasthenospermia models,which disregards the adhesion and migration between spermatogenic cells and Sertoli cells.TAp73 is a critical regulator of the adhesin of germ cell;thus,we sought to explore a novel oligoasthenospermia model based on TAp73 gene suppression.Methods:Mice in the Pifithrin-αgroup were injected intraperitoneally with 2.5 mg/kg Pifithrin-α(TAp73 inhibitor)daily for 30 consecutive days.Reproductive hormone levels and epididymal sperm quality,as well as the network morphology of Sertoli cells were tested.Results:Sperm density,motility,and the relative protein and mRNA expression of TAp73 and Nectin 2 were obviously decreased in the Pifithrin-αgroup compared with the normal control group.No significant distinction was observed in the relative mRNA and protein expression of ZO-1.Furthermore,the tight junctions(TJs)and api-cal ectoplasmic specialization(ES)were destroyed in the Pifithrin-αgroup.Conclusion:The above results indicate that we successfully established a new oli-goasthenospermia mouse model.This study provides a foundation for further explo-ration of the roles of TAp73 genes during spermatogenesis and provides new research objects for further oligospermia research and future drug discovery. 展开更多

Methods The relevant seminal parameters of 32 normal donors and 35 patients with olioasthenospermia were measured by seminal routine analysis, sperm morphology and accessory, gentital glands markers.Results The percentage of normal spermatozoa, the function of sperm penetration through cervical mucus and the enzyme activity of neutral α-1,4-glucosidase significantly declined as compared with the control. There was a significant negative relationship between the semen volume and the sperm motility for the oligoasthenospermia (r= -0.415, P〈0.05), and a significant positive relationship between the sperm count and the percentage of normal sperm (r=0.393, P〈0.05).Conclusion The abnormal function of epididymis exists in patients with oligoasthenospermia. Sperm function,for the oligoasthenospermia declines with an increase of the percentage of the abnormal sperm. The poorer the spermatogenises function of the testis is, the higher the percentage of the abnormal sperm appears. 展开更多

Objective:To investigate the effect of Xuduan Zhongzi prescription on the expression of HSP60 and HSP90 in testicular spermatogenic cells and semen quality of oligozoospermia and asthenospermia model rats.Methods:A total of 40 SPF male rats were randomly divided into 10 blank control group and 30 model group.After successful modeling,the model group was divided into model control group,levocarnitine group and Xuduan Zhongzi prescription group with 10 rats in each group.The rats in levocarnitine group and Xuduan Zhongzi prescription group were given corresponding drugs by gavage according to the equivalent measurement of humanbody,the blank control group and model control group were given the same amount of normal saline by gavage,and the rats in each group were killed after 8 weeks of continuous administration.Take the left epididymis of each group for sperm quality detection,take the left testis of each group,observe the histomorphological changes by HE staining,and detect the expression of HSP60 and HSP90 in testicular tissue by immunohistochemistry and Western blot.Results:HE staining showed that compared with the blank control group,the convoluted tubules in the model group were significantly atrophic,degenerated and irregularly arranged,the spermatogenic epithelium showed a large number of vacuoles,Sertoli cell degeneration,obvious inflammatory infiltration in stromal cells,stromal cell proliferation,scattered rows of epithelial cells,a large number of exfoliation and degeneration,and sperm were rare.After the intervention of Xuduan Zhongzi prescription,the above lesions were significantly improved,the structure of seminiferous tubules was relatively regular and orderly,denser than that of the model group,sertoli cells and stromal cells increased significantly,the seminiferous epithelium was thicker than that of the model group,with rich layers,and a large number of sperm could be seen in the lumen.Immunohistochemistry and Western Blot showed that the expression of HSP60 and HSP90 increased significantly after the intervention of Xuduan Zhongzi prescription in model rats(P<0.01).The sperm quality test results showed that compared with the blank control group,the sperm concentration and activity rate in the model group decreased significantly,(P<0.01).After continuous seed cutting,the sperm concentration and activity rate of model rats were significantly increased,(P<0.01).Conclusion:Xuduan Zhongzi prescription may inhibit the apoptosis of testicular spermatogenic cells in oligoasthenospermia model rats by increasing the expression of HSP60 and HSP90,so as to improve sperm quality. 展开更多

Objective:To investigate the therapeutic effects and pharmacological mechanisms of Yishen Xingyang capsule(YXC)in oligoasthenospermia(OA)rats.Methods:Forty-eight male SpragueeDawley rats were randomly divided into eight groups of six rats each:normal control(NC);model control(MC);three different positive drug(PD);and low-,medium-,and high-dose YXC groups.A rat model of OA was established by administering glucosides of Tripterygium wilfordii Hook.F(GTW).After YXC administration,penile erectile function was observed.The epididymis,blood,and testes of the rats were harvested for analysis of sperm quality,sex hormone levels,mitochondrial membrane potential,and the transforming growth factor(TGF)-b1/Smad signaling pathway.Results:Compared with that in the MC group,penile erectile function in the YXC groups and three PD groups increased(all P<.01).Moreover,sperm quality in the YXC groups and three PD groups improved(all P<.001).The levels of testosterone,follicle stimulating hormone,and luteinizing hormone in the three PD and YXC groups increased(all P<.05).The mitochondrial membrane potential in the three PD and YXC groups significantly improved(all P<.001).Furthermore,the YXC and three PD groups showed decreased TGF-b1 expression(all P<.05)compared with the MC group.The high-dose YXC group and three PD groups improved Smad2 and Smad4 expression(all P<.05).Conclusion:YXC improved penile erectile function and sperm quality in OA rats,and the underlying mechanism included increase in sex hormones,inhibition of sperm apoptosis,and regulation of the TGFb1/Smad signaling pathway.Meanwhile,this study provides a new effective drug option for the treatment of OA,which is beneficial to male reproductive health and social harmony. 展开更多

Objective:To investigate the protective mechanism of Xuduan Zhongzi prescription against epididymal oxidative damage in oligoasthenospermia model rats.Methods:Forty SD rats were randomly divided into blank group,model group,Xuduan Zhongzi prescription group(10g/kg)and L-carnitine group(0.1g/kg).Except blank group,all induced oligoasmospermia.The blank group and model group were given normal saline intragastric administration,the Xuduan Zhongzi prescription group was given Xuduan Zhongzi prescription solution intragastric administration,and the L-carnitine group was given L-carnitine intragastric administration.HE staining was used to observe the epididymis structure after 8 weeks.The concentration and activity rate of epididymis sperm were measured by sperm quality.MRNA and protein expression levels of Nrf2,NQO1 andγ-GCs in epididymis were detected by RT-qPCR and immunohistochemistry.Results:①HE staining:in the blank group,the epididymis tubes were arranged tightly and regularly,the tissue structure was complete,the epithelial cells were arranged orderly,and the lumen sperm were numerous and evenly distributed.The epididymis of model group showed structural atrophy,loose arrangement,enlarged mesenchyme,increased cell debris and significantly reduced sperm cells.Compared with the model group,the lumen lesions of epididymis in Xuduan Zhongzi prescription group and L-carnitine group were significantly improved,and the amount of normal sperm in lumen was increased and the distribution was uniform.②Results of sperm quality comparison among each group:sperm density and sperm motility rate:compared with blank group,sperm density and sperm motility rate in other groups were significantly decreased(P<0.05),and sperm density and sperm motility rate in model group were significantly decreased(P<0.05);Compared with model group,the sperm density and motility rate in Xuduan Zhongzi prescription group and L-carnitine group were significantly increased(P<0.05).③RT-qPCR and immunohistochemistry:Compared with the blank group,the mRNA and protein levels of Nrf2,NQO1 andγ-GCs in epididymal rats in model group were significantly decreased(P<0.05),while the mRNA and protein levels of Nrf2,NQO1 andγ-GCs were significantly increased in L-carnitine group and Continua seed formula group(P<0.05).Conclusion:Xuduan Zhongzi prescription can reduce oxidative stress damage and improve sperm quality of oligoasthenospermia.The mechanism may related to promoting the activation of Nrf2-NQO1/γ-GCS pathway in epididymis of oligoasthenospermia rats,and up-regulate the expressions of Nrf2,NQO1 andγ-GCS proteins. 展开更多

加味六味地黄丸改善肉碱代谢治疗少弱精子症模型大鼠的作用研究

目的研究加味六味地黄丸对奥硝唑(Ornidazole,ORN)所致少弱精子症模型大鼠的改善作用。方法按随机数表法将36只雄性SD大鼠分为对照组、模型组、左卡尼汀组、加味六味地黄丸组高中低三个剂量组,每组各6只。上午800 mg/(kg·d)ORN灌胃... 目的研究加味六味地黄丸对奥硝唑(Ornidazole,ORN)所致少弱精子症模型大鼠的改善作用。方法按随机数表法将36只雄性SD大鼠分为对照组、模型组、左卡尼汀组、加味六味地黄丸组高中低三个剂量组,每组各6只。上午800 mg/(kg·d)ORN灌胃,下午干预药物灌胃,连续灌胃28 d。建立少弱精子症模型,计算各组大鼠睾丸和附睾脏器系数的变化;通过苏木精-伊红(hematoxylin and eosin staining,HE)观察睾丸组织病理形态并进行Johnsen评分;酶联免疫吸附测定(ELISA)检测大鼠血浆中超氧化物歧化酶(superoxide dismutase,SOD)活性;LC-MS法检测附睾中肉碱类化合物水平,Western blot检测附睾LCAT蛋白的表达。结果与模型组大鼠睾丸脏器系数(8.00±2.00)‰比较,加味六味地黄丸低、中、高剂量组大鼠睾丸脏器系数明显升高,分别为(10.76±0.70)‰、(10.80±0.68)‰和(10.88±1.18)‰,差异均有显著统计学意义(P<0.01)。加味六味地黄丸中、高剂量组大鼠附睾脏器系数分别为(2.74±0.30)‰和(2.83±0.37)‰,明显高于模型组的(2.44±0.14)‰,且差异有统计学意义(P<0.05)。加味六味地黄高剂量组可改善大鼠睾丸曲细精管形态和管腔内各级生精细胞排列,增加精子数量,Johnsen评分与模型组比较差异有显著统计学意义(P<0.001)。酶联免疫吸附实验结果显示,与模型组SOD活性(109.14±5.58)比较,加味六味地黄丸高剂量组大鼠血浆中SOD活性明显升高,差异有显著统计学意义(P<0.001)。LC-MS法检测结果显示,加味六味地黄丸高剂量组可提高附睾中各肉碱类化合物水平,且与模型组比较差异均有统计学意义(P<0.05)。Western blot检测结果显示,与模型组(0.66±0.07)和左卡尼汀组(0.90±0.08)的LCAT蛋白表达比较,加味六味地黄丸高剂量组(1.17±0.14)显著升高,差异均有统计学意义(P<0.001)。结论加味六味地黄丸可有效改善少弱精子症模型大鼠睾丸病理组织结构,提高少弱精子症大鼠血浆中SOD活性和附睾中LCAT蛋白表达水平,通过增强机体脂质过氧化能力,改善附睾中肉碱类代谢,来达到治疗少弱精子症的目的。 展开更多

背景:电针能够通过刺激下丘脑、垂体、睾丸中的激素合成与释放治疗少弱精子症,但其作用机制复杂,并且电针疗效与穴位配伍思路关系密切。目的:探索电针疗法能否通过调节血清相关性激素水平改善下丘脑-垂体-睾丸轴功能状态,进而治疗腺嘌... 背景:电针能够通过刺激下丘脑、垂体、睾丸中的激素合成与释放治疗少弱精子症,但其作用机制复杂,并且电针疗效与穴位配伍思路关系密切。目的:探索电针疗法能否通过调节血清相关性激素水平改善下丘脑-垂体-睾丸轴功能状态,进而治疗腺嘌呤诱导的大鼠少弱精子症。方法:采用随机数字表法将50只成年雄性SD大鼠随机分为空白组、模型组、电针组、左卡尼汀组、非经非穴组,每组10只。除空白组外,其他4组大鼠给予200 mg/(kg·d)腺嘌呤灌胃,连续灌胃28 d,建立少弱精子症模型。造模结束后,电针组选取中极、关元、足三里、三阴交四穴给予电针治疗,非经非穴组选取非穴点3处进行电针治疗,左卡尼汀组给予左卡尼汀灌胃治疗,1次/d,持续治疗28 d。治疗结束后,检测各组大鼠肾脏和睾丸脏器系数,精子数量、存活率和活力率,肾脏与睾丸组织病理形态学变化,血清中睾酮、促性腺激素释放激素、卵泡刺激素、黄体生成素、雌二醇、抑制素B、泌乳素水平。结果与结论:①模型组大鼠肾脏脏器系数高于空白组、电针组、左卡尼汀组(P<0.05),非经非穴组大鼠肾脏脏器系数高于电针组(P<0.05),各组大鼠睾丸脏器系数比较差异无显著性意义(P>0.05);②与空白组比较,模型组大鼠精子数量、精子存活率与活力率均降低(P<0.05);与模型组比较,电针组、左卡尼汀组大鼠精子数量、精子存活率与活力率均升高(P<0.05);非经非穴组大鼠精子数量、精子存活率与活力率均低于电针组(P<0.05);③苏木精-伊红染色结果显示,模型组大鼠肾脏与睾丸组织损伤严重,电针组和左卡尼汀组大鼠肾脏与睾丸组织损伤程度轻于模型组,非经非穴组大鼠肾脏与睾丸组织损伤程度重于电针组;④与空白组比较,模型组大鼠血清中睾酮、促性腺激素释放激素、抑制素B水平均降低(P<0.05),卵泡刺激素、黄体生成素、雌二醇、泌乳素水平均升高(P<0.05);与模型组比较,电针组、左卡尼汀组大鼠血清中睾酮、促性腺激素释放激素、抑制素B水平均升高(P<0.05),卵泡刺激素、黄体生成素、雌二醇、泌乳素水平均降低(P<0.05);与非经非穴组比较,电针组大鼠血清中睾酮、促性腺激素释放激素、抑制素B水平均升高(P<0.05),卵泡刺激素、黄体生成素、雌二醇、泌乳素水平均降低(P<0.05);⑤结果表明,电针“中极、关元、足三里、三阴交”4穴可通过调控“下丘脑-垂体-睾丸轴”功能状态有效治疗大鼠少弱精子症。 展开更多

【目的】研究改良“益精方”改善种公牛精液品质差,少精、弱精等肾阳虚症的作用机制。【方法】采用代谢组学技术对SD大鼠血清样本(空白对照组、改良“益精方”组、模型组和左卡尼汀组)进行代谢物高通量测序。经过离子等鉴定与多元统计... 【目的】研究改良“益精方”改善种公牛精液品质差,少精、弱精等肾阳虚症的作用机制。【方法】采用代谢组学技术对SD大鼠血清样本(空白对照组、改良“益精方”组、模型组和左卡尼汀组)进行代谢物高通量测序。经过离子等鉴定与多元统计的方法鉴定出差异离子,再利用生物信息学的技术将差异离子进行富集分析。【结果】血清代谢物主要以氨基酸、肽及其衍生物、碳水化合物类、类固醇及其衍生物等为主;模型组与空白组存在有433个显著性的差异离子(FC>1.5,P<0.05);改良“益精方”组与模型组有397个显著性差异离子(FC>1.5,P<0.05);注释结果表明这些差异代谢物主要参与到鞘脂代谢、核黄素和组氨酸等代谢途径中,并显著富集到脂1-磷酸鞘氨醇(Sphingosine 1-phosphate,S1P)的表达中(P<0.05)。【结论】改良“益精方”对肾阳虚大鼠有明显改善作用,这可能与改良“益精方”抑制睾丸内细胞线粒体凋亡有关,进而发挥了治疗少弱精的作用。 展开更多

目的基于AMPK介导的线粒体自噬方面探究国医大师王琦院士研发的治疗男性不育的国家中药三类新药黄精赞育胶囊治疗少弱精子症细胞层面的分子机制。方法选择丙烯醛(acrolein,ACR)作用于GC-2spd(ts)小鼠精母细胞制作少弱精子症细胞模型,通... 目的基于AMPK介导的线粒体自噬方面探究国医大师王琦院士研发的治疗男性不育的国家中药三类新药黄精赞育胶囊治疗少弱精子症细胞层面的分子机制。方法选择丙烯醛(acrolein,ACR)作用于GC-2spd(ts)小鼠精母细胞制作少弱精子症细胞模型,通过CCK8细胞活性检测确定后续造模用ACR浓度和造模时间。造模成功后加入不同浓度含黄精赞育胶囊的完全培养基进行细胞培养,于24 h后CCK8细胞活性检测,根据细胞活力确定后续给药浓度。GC-2spd细胞贴壁后分为NC组、造模组和ACR+HJZY给药组,通过共聚焦荧光显微镜观测黄精赞育胶囊对线粒体自噬的影响;将上述3组细胞分别转染siRNA-NC和siRNA-AMPK,分为siRNA-NC+control、siRNA-NC+ACR、siRNANC+ACR+HJZY、siRNA-AMPK+control、siRNA-AMPK+ACR、siRNA-AMPK+ACR+HJZY 6组,通过Western blot验证黄精赞育胶囊对AMPK介导的p-AMPK、LC3B、P62、PINK1、Parkin、TBK1、ULK1等线粒体自噬相关蛋白的调控作用。结果通过细胞活性检测实验确定ACR的造模浓度为34μmol/L,造模时间为20 min,HJZY给药浓度为160μmol/L。共聚焦荧光显微镜显示黄精赞育胶囊对受损生精细胞线粒体膜电位具有一定正向调节作用,造模组线粒体膜电位较NC组大幅降低,ACR+HJZY给药组给药后细胞膜电位较造模组有所升高,差异有统计学意义(P<0.05);Western blot检测结果显示siRNA-NC+ACR组p-AMPK/AMPK和PINK1蛋白表达水平较siRNA-NC+control组下降(P<0.001),siRNA-NC+ACR组Parkin蛋白水平较siRNA-NC+control组有所下降,但差异无统计学意义,HJZY给药后这3种蛋白表达水平均较siRNANC+ACR组上升(P<0.001);siRNA-NC+ACR组LC3B、P62、TBK1和ULK1蛋白表达水平均较siRNA-NC+control组上升(P<0.01),siRNA-NC+ACR+HJZY组上述蛋白表达水平均较siRNA-NC+ACR组有所下降(P<0.05)。转染敲减基因siRNAAMPK后,siRNA-AMPK+ACR组p-AMPK/AMPK、PINK1和Parkin蛋白表达水平较siRNA-AMPK+control组有所下降(P<0.01);H J Z Y给药后上述3种蛋白表达水平与s i R N A-A M P K+A C R组比较差异均无统计学意义;s i R N A-AMPK+ACR+HJZY组与siRNA-AMPK+ACR组相比,LC3B蛋白表达水平仍降低(P<0.01),但P62、TBK1和ULK1蛋白表达水平差异均无统计学意义;siRNA-AMPK+control组与siRNA-NC+control组相比,p-AMPK/AMPK、ULK1和TBK1蛋白表达水平下降(P<0.001),PINK1蛋白表达水平亦下降(P<0.05),P62蛋白表达水平上升(P<0.001);siRNA-AMPK+ACR组与siRNA-NC+ACR组相比,TBK1蛋白表达水平下降(P<0.001),LC3B蛋白表达水平下降(P<0.01),ULK1蛋白表达水平亦下降(P<0.05),PINK1和Parkin蛋白表达水平有所下降但差异无统计学意义;siRNA-AMPK+ACR+HJZY组与siRNANC+ACR+HJZY组相比,p-AMPK/AMPK、PINK1和Parkin蛋白表达水平下降(P<0.05),LC3B蛋白表达水平亦下降(P<0.01),P62蛋白表达水平上升(P<0.001),TBK1和ULK1蛋白表达水平差异无统计学意义。结论黄精赞育胶囊可能通过调节AMPK介导的线粒体自噬达到治疗男性少弱精子症的效果。 展开更多

目的探究菟枸助育汤对男性不育少弱精子症患者的临床疗效。方法选取2017年9月至2019年6月北京中医药大学东直门医院诊治的76例20~50岁男性不育少弱精子症患者作为研究对象。比较治疗前后精液常规、中医证候评分,统计研究期间及治疗结束... 目的探究菟枸助育汤对男性不育少弱精子症患者的临床疗效。方法选取2017年9月至2019年6月北京中医药大学东直门医院诊治的76例20~50岁男性不育少弱精子症患者作为研究对象。比较治疗前后精液常规、中医证候评分,统计研究期间及治疗结束后6个月内配偶受孕情况,评价疗效及安全性。结果治疗后,患者精液液化情况、前向运动精子百分率(PR)、PR+非前向运动精子百分率(NP)、精子浓度均较治疗前显著改善(P<0.05)。配偶受孕率为14.47%(11例)。治疗后,中医证候评分总分较治疗前显著降低(P<0.05),且有18项较治疗前显著改善(P<0.05)。治疗总有效率为94.74%;无不良反应发生。结论菟枸助育汤治疗男性不育少弱精子症患者的临床疗效显著,可有效改善精液常规指标,改善中医证候评分,且安全性好。 展开更多

目的:观察不同频率电针对少弱精子症(OAT)模型大鼠生精功能、睾丸形态及氧化应激的影响,探索电针干预OAT的作用机制及最优参数。方法:将60只SPF级雄性SD大鼠随机分为溶剂对照组、模型组、2 Hz电针组、100 Hz电针组和2 Hz/100 Hz电针组,... 目的:观察不同频率电针对少弱精子症(OAT)模型大鼠生精功能、睾丸形态及氧化应激的影响,探索电针干预OAT的作用机制及最优参数。方法:将60只SPF级雄性SD大鼠随机分为溶剂对照组、模型组、2 Hz电针组、100 Hz电针组和2 Hz/100 Hz电针组,每组12只。除溶剂对照组外,其余4组大鼠以奥硝唑混悬液(800 mg·kg^(-1)·d^(-1))灌胃28 d的方法制备OAT模型。从造模第1天开始,3个电针组均于“关元”“气海”及双侧“三阴交”“足三里”行电针干预,2 Hz电针组、100 Hz电针组、2 Hz/100 Hz电针组分别选择2 Hz连续波、100 Hz连续波和2 Hz/100 Hz疏密波,电流1~3 mA,每次30 min,隔日1次,连续28 d。干预后,计算各组大鼠睾丸指数,检测附睾精子质量,观察大鼠生育能力;HE染色法观察大鼠睾丸组织形态并计算Johnson评分;免疫荧光法检测大鼠睾丸组织活性氧(ROS)阳性表达;ELISA法检测大鼠睾丸组织氧化应激指标超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量;Western blot法检测大鼠睾丸组织核因子红细胞2相关因子2(Nrf2)及血红素加氧酶-1(HO-1)蛋白表达。结果:与溶剂对照组比较,模型组大鼠睾丸指数、精子浓度、精子活力及产仔数降低(P<0.01);生精小管萎缩,Johnson评分降低(P<0.01);睾丸组织SOD及CAT活性、Nrf2及HO-1蛋白表达降低(P<0.01);精子畸形率及睾丸组织ROS阳性表达、MDA含量升高(P<0.01)。与模型组比较,2 Hz电针组、100 Hz电针组及2 Hz/100 Hz电针组大鼠睾丸指数、精子浓度、精子活力及产仔数升高(P<0.05,P<0.01);睾丸组织病理形态改善,Johnson评分升高(P<0.01);睾丸组织SOD及CAT活性、Nrf2及HO-1蛋白表达升高(P<0.05,P<0.01);精子畸形率及睾丸组织ROS阳性表达、MDA含量降低(P<0.05,P<0.01)。与2 Hz电针组比较,2 Hz/100 Hz电针组大鼠睾丸指数、精子浓度、精子活力及睾丸组织CAT活性、HO-1蛋白表达升高(P<0.01,P<0.05),睾丸组织ROS阳性表达降低(P<0.01);与100 Hz电针组比较,2 Hz/100 Hz电针组大鼠睾丸指数升高(P<0.01),睾丸组织ROS阳性表达降低(P<0.01)。结论:2 Hz低频连续波、100 Hz高频连续波、2 Hz/100 Hz疏密波电针均可改善OAT模型大鼠生精功能障碍,降低睾丸组织氧化应激水平,其机制可能与上调Nrf2、HO-1蛋白表达,改善氧化应激有关,其中2 Hz/100 Hz疏密波电针效果最优。 展开更多

目的:通过观察俞募脑肾配穴电针疗法对少弱精子症模型大鼠的影响,探究俞募脑肾配穴电针疗法对少弱精子症大鼠生殖功能的改善作用及对睾丸TGF-β/smads信号通路的影响。方法:40只雄性SD大鼠随机分为空白组、模型组、电针组、左卡尼汀组,... 目的:通过观察俞募脑肾配穴电针疗法对少弱精子症模型大鼠的影响,探究俞募脑肾配穴电针疗法对少弱精子症大鼠生殖功能的改善作用及对睾丸TGF-β/smads信号通路的影响。方法:40只雄性SD大鼠随机分为空白组、模型组、电针组、左卡尼汀组,每组10只。除空白组外,其余组采用腺嘌呤灌胃法制备少弱精子症大鼠模型,灌胃连续28 d,造模成功后,分别对电针组(电针额旁3线、肾俞、京门、太溪,30 min/次/d,连续28 d)、左卡尼汀组(以10 mL/kg左卡尼汀口服液灌胃,连续28 d)进行干预。治疗后取各组大鼠附睾精子检测其密度和活力率;H&E染色观察睾丸病理形态学变化;ELISA检测血清性激素睾酮、卵泡刺激素、黄体生成素、雌二醇的变化;Western blot检测TGF-β、P-smad2、P-smad3在睾丸组织中的表达水平。结果:与空白组比较,模型组大鼠睾丸组织中生精细胞变性,数量减少,排列紊乱;精子密度、精子活力率、血清睾酮水平显著降低(P<0.05),黄体生成素水平显著升高(P<0.05);与模型组比较,左卡尼汀组、电针组大鼠睾丸病理损伤显著减轻,精子密度和活力率、血清睾酮水平显著增高(P<0.05),黄体生成素水平显著降低(P<0.05)。结论:俞募脑肾配穴电针疗法可提高少弱精子症大鼠精子密度和活率,调节性激素产生,改善大鼠的生精环境,其机制可能与血清性激素水平的调控和通过TGF-β/smads信号通路,影响睾丸生精过程有关。 展开更多

Oligoasthenospermia is the primary cause of infertility.However,there are still enormous challenges in the screening of critical candidates and targets of oligoasthenospermia owing to its complex mechanism.In this study,stem cell factor(SCF),c-kit,and transient receptor potential vanilloid 1(TRPV1)biosensors were successfully established and applied to studying apoptosis and autophagy mechanisms.Interestingly,the detection limit reached 2.787×10^(-15)g/L,and the quantitative limit reached 1.0×10^(-13)g/L.Furthermore,biosensors were used to investigate the interplay between autophagy and apoptosis.Schisandrin A is an excellent candidate to form a system with c-kit similar to SCF/c-kit with a detection constant(K_(D))of 5.701×10^(-11)mol/L,whereas it had no affinity for SCF.In addition,it also inhibited autophagy in oligoasthenospermia through antagonizing TRPV1 with a K_(D) of up to 4.181×10^(-10)mol/L.In addition,in vivo and in vitro experiments were highly consistent with the biosensor.In summary,high-potency schisandrin A and two potential targets were identified,through which schisandrin A could reverse the apoptosis caused by excessive autophagy during oligoasthenospermia.Our study provides promising insights into the discovery of effective compounds and potential targets via a well-established in vitro-in vivo strategy. 展开更多

目的通过数据挖掘,探索卞廷松教授临床治疗男性不育少弱精子症的用药规律,为临床实践提供理论依据。方法通过收集和筛选2023年6月至12月卞廷松教授在南京中医药大学附属常州市中医医院诊治的男性不育少弱精子症的有效病例处方,录入中药... 目的通过数据挖掘,探索卞廷松教授临床治疗男性不育少弱精子症的用药规律,为临床实践提供理论依据。方法通过收集和筛选2023年6月至12月卞廷松教授在南京中医药大学附属常州市中医医院诊治的男性不育少弱精子症的有效病例处方,录入中药数据,利用关联分析、聚类分析和网络可视化等工具探索处方中用药的规律。结果共纳入136首处方,涉及中药75味,使用频次前5味的中药依次为丹参、盐菟丝子、生地黄、荔枝核、白茅根。关联规则分析及聚类分析得到核心药物组成和配伍。聚类分析中将30味高频中药聚为6类时效果最佳;关联规则分析出5组最常用药对。结论卞廷松教授临床治疗男性不育症以“阴平阳秘”为理论基础,善用菟丝子、生地黄等药物平调阴阳,同时辅以宁心、健脾、补肾、活血之法,使机体趋于阴阳平衡的状态,则疾病自除。 展开更多

目的利用证据图系统梳理中医药治疗少弱精子症的随机对照试验(RCT),了解该领域证据分布及质量,发现现有治疗少弱精子症相关研究存在的问题。方法检索中国知网、万方、维普、中国生物医学文献服务系统、PubMed、EMbase、Cochrane Librar... 目的利用证据图系统梳理中医药治疗少弱精子症的随机对照试验(RCT),了解该领域证据分布及质量,发现现有治疗少弱精子症相关研究存在的问题。方法检索中国知网、万方、维普、中国生物医学文献服务系统、PubMed、EMbase、Cochrane Library等数据库中收录的中医药治疗少弱精子症的相关文献,检索时间为建库至2024年6月。提取文献样本量、研究药物、疗程、结局指标等;依据Cochrane协助网偏倚风险评价工具进行质量评价;采用图、表展示结果。结果共收集RCT文献223篇;其中药物使用占比较大的是复方玄驹胶囊、五子衍宗丸、生精胶囊、麒麟丸;结局指标包括精子活力、精子存活率、临床有效率、不良反应等;文献质量方面,103篇的随机方法正确,10篇执行了分配隐藏及盲法。结论中医药治疗少弱精子症的RCT的中医病证信息不全,结局指标较少分析中医证候积分、配偶妊娠情况、不良反应,未分析心理健康状态;整体研究质量一般。建议后续研究按照中医诊疗标准统一中医证型,针对证型实施诊疗方案;建议研究人员构建中医药治疗少弱精子症的核心指标数据集,完善疗效评价体系,在开展RCT前,参照CONSORT制定试验方案;建议引入相关量表评估患者的心理健康状况,为后续进一步开展男性不育症与心理健康状况相关性研究建立一定的基础。 展开更多
