Identification of QTL for kernel number-related traits in a rice chromosome segment substitution line and fine mapping of qSP1 被引量:4

A chromosome segment substitution line (CSSL) is a powerful tool for combining quantitative trait locus (QTL) mapping with the pyramiding of desirable alleles. The rice CSSL Z1364 with increased kernel number was identified in a BC3F8 population derived from a cross of Nipponbare as the recipient with Xihui 18 as the donor parent. Z1364 carried three substitution segments distributed on chromosomes 1, 6, and 8. The mean substitution length was 1.19 Mb. Of 17 QTL identified on the substitution segments, qSP1 for spikelets per panicle, qSSD1 for seed-set density, and qNSB1 for number of secondary branches explained respectively 57.34%, 87.7%, and 49.44% of the corresponding phenotypic variance and were all linked to RM6777. Chi-square analysis showed that the increased kernel number in Z1364 was inherited recessively by a single gene. By fine mapping, qSP1 was delimited to a 50-kb region on the short arm of chromosome 1. Based on DNA sequence, a previously uncharacterized rice homolog of Arabidopsis thaliana AT4G32551 was identified as a candidate gene for qSP1 in which mutation increases the number of spikelets and kernels in Z1364. qSP1 was expressed in all tissues, but particularly in 1-cm panicles. The expression levels of OsMADS22, GN1A, and DST were upregulated and those of LAX2, GNP1, and GHD7 were downregulated in Nipponbare. Hexaploid triticale(×Triticosecale,AABBRR)is an important forage crop and a promising energy plant.Transferring D-genome chromosomes or segments from common wheat(Triticum aestivum)into hexaploid triticale is attractive in improving its economically important traits.Here,a hexaploid triticale 6D(6A)substitution line Lin 456 derived from the cross between the octoploid triticale line H400 and the hexaploid wheat Lin 56 was identified and analyzed by genomic in situ hybridization(GISH),fluorescence in situ hybridization(FISH),and molecular markers.The GISH analysis showed that Lin 456 is a hexaploid triticalewith 14 rye(Secale cereale)chromosomes and 28 wheat chromosomes,whereas non-denaturing fluorescence in situ hybridization(ND-FISH)and molecular marker analysis revealed that it is a 6D(6A)substitution line.In contrast to previous studies,the signal of Oligo-pSc119.2 was observed at the distal end of 6DL in Lin 456.The wheat chromosome 6D was associatedwith increased grain weight and decreased spikelet number using the genotypic data combined with the phenotypes of the F2 population in the three environments.The thousand-grain weight and grain width in the substitution individuals were significantly higher than those in the non-substitution individuals in the F2 population across the three environments.We propose that the hexaploid triticale 6D(6A)substitution line Lin 456 can be a valuable and promising donor stock for genetic improvement during triticale breeding. 展开更多

Following NCI design, the developmental genetic behavior of tiller number (TN) in three-line indica hybrid rice was studied using additive-dominance developmental genetic models and the corresponding statistical methods. The results showed that dominance effects were predominant for TN. The expression of those additive effects were affected by environment and genotype interaction, but the expression of dominance effects were not affected. Heterosis was the strongest in the middle developmental periods of TN. Additive effects and dominance effects were selectively expressed throughout in the entire tillering developmental stage. Analysis of genetic correlation between TN at different stages and the productive panicles indicated that a close correlation appeared earlier in the populations with higher heterosis than in those with less heterosis. Utilization of heterosis at the middle tillering stage might enhance the final biomass but reduce the percentage of productive panicles. 展开更多

Tiller is one of the most important agronomic traits which influences quantity and quality of effective panicles and finally influences yield in rice. It is important to understand "static" and "dynamic" information of the QTLs for tillers in rice. This work was the first time to simultaneously map unconditional and conditional QTLs for tiller numbers at various stages by using single segment substitution lines in rice. Fourteen QTLs for tiller number, distributing on the corresponding substitution segments of chromosomes 1, 2, 3, 4, 6, 7 and 8 were detected. Both the number and the effect of the QTLs for tiller number were various at different stages, from 6 to 9 in the number and from 1.49 to 3.49 in the effect, respectively. Tiller number QTLs expressed in a time order, mainly detected at three stages of 0-7 d, 14-21 d and 35-42 d after transplanting with 6 positive, 9 random and 6 negative expressing QTLs, respectively. Each of the QTLs expressed one time at least during the whole duration of rice. The tiller number at a specific stage was determined by sum of QTL effects estimated by the unconditional method, while the increasing or decreasing number in a given time interval was controlled by the total of QTL effects estimated by the conditional method. These results demonstrated that it is highly effective and accurate for mapping of the QTLs by using single segment substitution lines and the conditional analysis methodology. 展开更多

为精准评价黄淮地区大豆品系对胞囊线虫(Soybean Cyst Nematode, SCN)的抗性,本研究对211份黄淮地区育成大豆品系的SCN2抗性进行鉴定,同时利用抗SCN主效位点rhg1和Rhg4连锁的KASP标记进行基因型检测,并对含有这两个位点的抗性种质的抗... 为精准评价黄淮地区大豆品系对胞囊线虫(Soybean Cyst Nematode, SCN)的抗性,本研究对211份黄淮地区育成大豆品系的SCN2抗性进行鉴定,同时利用抗SCN主效位点rhg1和Rhg4连锁的KASP标记进行基因型检测,并对含有这两个位点的抗性种质的抗性位点拷贝数进行分析。表型鉴定结果表明:211份品系中,1份表现中抗,占品系总数的0.47%;7份表现中感,占品系总数的3.32%;203份表现感病,占品系总数的96.21%;未发现免疫和高抗品系。基因型鉴定结果表明:郑12019同时含有rhg1-a和Rhg4抗性位点,和Peking基因型一致;冀Y2305只含有rhg1-a抗性位点;中作J20287只含有rhg1-b抗性位点,和PI 88788基因型一致;科豆78只含有Rhg4抗性位点;其余207份品系均是感病基因型,占品系总数的98.10%。此外,本研究还对表现中抗的郑12019进行基因组重测序,使用CNVpytor和FREEC程序分析其抗SCN两个主效位点rhg1和Rhg4的拷贝数,两程序获得的结果较相似,CNVpytor分析得出郑12019的rhg1和Rhg4位点拷贝数分别为2.52和2.55,而FREEC分析得出郑12019的rhg1和Rhg4位点的拷贝数分别为3和2。以SKIP16为内参基因,数字PCR检测结果表明郑12019的rhg1位点拷贝数为2.66,和Peking的rhg1位点拷贝数相似。本研究为大豆新品种的审定提供依据,同时为抗SCN育种提供理论参考与材料支撑。 展开更多

By introducing the strength density and the topological current of many disclination lines, it is pointed out that the disclination lines are determined by the singulaities of the director field, and topologically quantized by the Hopf indices and Brouwer degrees. Due to the equivalence in physics of the director fields n (x) and n (x) , the Hopf indices can be integers or half integers, representing a generalization of our previous studies of integer Hopf indices. 展开更多

The electromagnetic wave growth or damping depends basically on the number density and anisotropy of energetic particles as the resonant interaction takes place between the particles and waves in the magnetosphere. The variance of both the number density and anisotropy along the magnetic field line is evaluated systematically by modeling four typically prescribed distribution functions. It is shown that in the case of "the positive anisotropy" (namely, the perpendicular temperature T⊥ exceeds the parallel temperature T||), the number density of energetic electrons always decreases with the magnetic latitude for a regular increasing magnetic field and the maximum wave growth is therefore generally confined to the equator where the resonant energy is minimum, and the number density is the largest. However, the "loss-cone" anisotropy of the electrons with a "pancake" distribution or kappa distribution keeps invariant or nearly invariant, whereas the "temperature" anisotropy with a pure bi-Maxwellian distribution or Ashour-Abdalla and Kennel's distributions decreases with the magnetic latitude. The results may provide a useful approach to evaluating the number density and anisotropy of the energetic electrons at latitudes where the observation information is not available. 展开更多

[目的]探究2,2',4,4',5,5'-六氯联苯(PCB153)对人肾上皮细胞系(293T)细胞中长散在重复序列-1(LINE-1)基因表达和转座的影响。[方法]293T细胞每2 d传代一次,传代后立即使用5μmol/L PCB153进行染毒。染毒后的细胞置于37℃、体积分数5%CO2培养箱中继续培养,连续染毒10 d。同时设置二甲基亚砜(DMSO)对照组。采用实时荧光定量PCR技术检测LINE-1开放阅读框1(ORF-1)和开放阅读框2(ORF-2)基因的m RNA表达水平及拷贝数,LINE-1拷贝数增加反映其发生转座。Western blot检测LINE-1 ORF-1和ORF-2蛋白表达水平。[结果]与对照组比较,从第4天开始PCB153染毒组细胞中LINE-1 ORF-1 m RNA表达水平增加(P<0.05),从第6天开始PCB153染毒组细胞中LINE-1 ORF-2 m RNA表达水平增加(P<0.05)。染毒第10天时,与对照组(LINE-1 ORF-1:2.74±0.51;LINE-1 ORF-2:0.57±0.10)相比,PCB153染毒组的LINE-1 ORF-1蛋白表达水平(4.39±0.85)和LINE-1 ORF-2蛋白表达水平(0.78±0.10)均明显增加(P<0.05)。染毒第10天时,PCB153染毒组与对照组的LINE-1 ORF-1拷贝数分别为1.43±0.64和0.83±0.37,LINE-1 ORF-2拷贝数分别为1.11±0.52和0.81±0.12,差异均具有统计学意义(P<0.05)。[结论]PCB153暴露可增加LINE-1的表达和拷贝数,诱导其转座。 展开更多

旨在提升苏梅猪配套系中梅山猪高产专门化品系(M系)的产仔数并对配合力测定工作的部分研究进展进行报告,为苏梅猪配套系培育提供基础。本试验采用rs332069453突变位点结合母系指数选育方法,对M系1~3世代群体进行持续选育,跟踪统计初产... 旨在提升苏梅猪配套系中梅山猪高产专门化品系(M系)的产仔数并对配合力测定工作的部分研究进展进行报告,为苏梅猪配套系培育提供基础。本试验采用rs332069453突变位点结合母系指数选育方法,对M系1~3世代群体进行持续选育,跟踪统计初产总产仔数(TNB);进一步在2个试验场(场一、场二)开展配合力测定工作,2个试验场采用不同饲喂方式,以M系3世代母猪为母本,分别与皮特兰和大白公猪杂交,产生杂交后代皮梅猪(PM)和大梅猪(LWM)并进行体重与体尺测定,比较不同杂交组合的生长性能及不同饲喂方式下的表现差异。结果:经3个世代选育,M系猪后备猪候选群体rs332069453位点优势等位基因C频率从1世代的0.37升至3世代的0.68,3世代梅山猪后备群体平均总产仔数为13.7头,相较于1世代梅山猪后备群体平均总产仔数12.0头,提升了1.7头;通过配合力测定工作发现,场一6月龄测定中,LWM体长极显著大于PM(P<0.01),PM管围显著大于LWM(P<0.05);场二6月龄测定中,PM体重和管围均显著高于LWM(P<0.05),其余体尺性状无显著差异,但相较于梅山猪的体重与体尺性状差异显著。综上,基于分子标记的选育可有效提高梅山猪繁殖性能,杂交对梅山猪的体型劣势很大程度上进行了弥补。结合体型性状和当前优质肉市场对黑毛色的偏爱,选择体型好、毛色黑、肢蹄强的PM作为苏梅猪配套系的优质二元母本,相关试验结果为配套系选育奠定了扎实的基础。 展开更多

The expressions of nondimensional sound power are formulated andthe asymptotic forms of sound power in the low frequency regions are derived. The radiatedsound power is shown as a function of the stiffness of elastic foundation, in terms of stiffness fac-torψ, the moving speed of line force, in terms of Math number M, and the frequency, in termsof wavenumber ratio γ . Unconditional and conditional QTL mapping were conducted for growth duration (GD), plant height (PH) and effective panicle number per plant (PN) using a recombinant inbred line (RIL) population derived from a cross between two japonica rice varieties Xiushui 79 and C Bao. The RIL population consisted of 254 lines was planted in two environments, Nanjing and Sihong, Jiangsu Province, China. Results showed that additive effects were major in all of QTLs for GD, PH and PN detected by the two methods, and the epistatic effects explained a small proportion of phenotypic variation. No interactions were detected between additive QTL and environment, and between epistatic QTL pairs and environment. After growth duration was adjusted to an identical level, RM80-160bp was detected as an applicable elite allele for PN, with an additive effect of 0.71. When effective panicle number per plant was adjusted to an identical level, RM448-240bp was detected as an applicable elite allele for GD, with an additive effect of 4.64. After plant height was adjusted to an identical level, RM80-160bp was detected as an applicable elite allele for PN, with an additive effect of 0.62, and RM448-240bp was detected as an applicable elite allele for GD, with an additive effect of 3.89. These applicable elite alleles could be used to improve target traits without influencing the other two traits. 展开更多

大豆是重要的粮饲兼用作物。分枝数是影响大豆产量的重要农艺性状之一。本研究以少分枝大豆品种中黄35与多分枝品种中黄13杂交衍生的RIL F_(2:7-8)群体为材料,结合重测序基因型数据构建的高密度遗传连锁图谱,采用QTL IciMapping完备复... 大豆是重要的粮饲兼用作物。分枝数是影响大豆产量的重要农艺性状之一。本研究以少分枝大豆品种中黄35与多分枝品种中黄13杂交衍生的RIL F_(2:7-8)群体为材料,结合重测序基因型数据构建的高密度遗传连锁图谱,采用QTL IciMapping完备复合区间定位方法,在5个种植环境下定位与分枝数相关的QTL。结果显示,在2号、6号、18号和19号染色体上共定位到6个与分枝数相关的QTL。其中位于2号染色体的qVBN02-1在2个环境中均被检测到,平均解释16.07%的表型变异,为新发掘的分枝数稳定主效QTL。该位点区间遗传距离为0.3 cM,物理距离为261.37 kb,包含29个注释基因。经QTL区间内双亲错义SNP筛选,发掘出22个分枝数潜在候选基因。GO注释分析显示,这些基因编码的蛋白涉及多个影响植物生长发育的重要过程。本研究不仅为大豆株型改良提供了分子标记靶位点,也为精细定位大豆分枝数关键基因奠定了基础。 展开更多
