Six diets supplemented with 0, 2.5, 5.0, 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln were fed to fish for 12 weeks. Supplementation with 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln significantly increased weight gain rate(WGR), protein efficiency ratio(PER), but feed conservation rate(FCR) and survival were not affected(P〉0.05). The intestinal fold height and number, digestive enzyme, Na+, K+-ATPase activities was found to be significantly high(P〈0.05) with increasing dietary Aln-Gln supplementation up to 7.5 g · kg-1, but there were no significant differences for Aln-Gln supplementation from 7.5 to 15.0 g · kg-1. The glutathione peroxidase(GPX) activity, glutathione(GSH), superoxide dismutase(SOD) activity increased and malondialdehyde(MDA) levels decreased significantly(P〈0.05) in the intestine, hepatopancreas, plasma and muscles. The plasma complement-3(C3) and complement-4(C4) levels were significantly(P〈0.05) improved at 5.0 g · kg-1 level and decreased when over 7.5 g · kg-1. The plasma lysozyme(LSZ) activity increased significantly(P〈0.05) at 7.5, 10.0, or 15.0 g · kg-1 level. In summary, the results showed that Aln-Gln improved growth performance, development and function of the intestine, the activity of the antioxidant defense system and the plasma non-specific immunity of the carps. The optimal Aln-Gln level was 8.24 g · kg-1 diet for WGR based on broken-line regression model analysis.展开更多

The experiment was designed as a single-factorial arrangement(n=8 cages/...Here, we investigated the effect of dietary arginine(Arg) supplementation on innate immunity and the antioxidant ability of broiler chickens. The experiment was designed as a single-factorial arrangement(n=8 cages/treatment, six birds/cage), and we used four dietary Arg concentrations(10.0, 15.0, 20.0 or 25.0 g kg–1). On day 21, the birds were killed to obtain spleen, cecal tonsil and liver samples to determine the gene expression and antioxidant characteristics. Increasing the Arg concentration linearly decreased(P0.05) the m RNA expression of splenic interleukin-18(IL-18) and tumor necrosis factor-α(TNF-α). Dietary Arg supplementation quadratically decreased(P0.05) the expression of interleukin-1b(IL-1b) and interferon-γ(IFN-γ) m RNA in the spleen. Increasing Arg concentrations linearly and quadratically reduced the expression of IL-18 m RNA in the spleen. Meanwhile, increasing dietary Arg supplementation linearly and quadratically increased the lymphotactin m RNA(P0.05) expression, and linearly increased the macrophage inflammatory protein-1β(MIP-1β) and toll-like receptor 15(TLR15) m RNA expression in the cecal tonsils. Dietary Arg supplementation linearly(P0.05) increased the glutathione peroxidase(GSH-Px), catalase(CAT), and lysozyme(LZM) activities in the liver. However, the malondialdehyde(MDA) activity in the liver was not influenced by the dietary Arg concentration(P0.05). No significant(P0.05) effect was found on the activity of superoxide dismutase(SOD) in the liver. Thus high levels of Arg supplementation(20.0 g kg^(–1)) may potentially suppress the innate immunity of broiler chickens, and dietary Arg supplementation enhances the antioxidant activity in broiler chickens.展开更多

Three size groups of shrimp, i.e., the small size group [Gs, with an average body length (BL) of (3.04 ± 0.36) cm and body weight (BW) (0.412± 0.35) g], the large group [GL, with a BL of (4.29±0.55) cm and BW of (1.098 ±0.42) g], and the ungraded group [Gm, with a BL of (3.47±0.81) cm and BW of (0.611 ±0.79) g], were reared under the same conditions for 8 wk. Growth performance and non- specific immunity factors were measured. The results showed that BW gain, biomass gain and the specific growth rate of body length (SGRL) were significantly influenced by size grading (one-way ANOVA, P 〈 0.05). The peroxidase (POD) and antibacterial (Ua) activities of GL were lower than those of G. Superoxide dismutase (SOD) and lysozyme (U1) activities of Gm were lower than those of G. No significant difference (P = 0.121 〉 0.05) was found on phenoloxidase (PO) activity among the three size groups. Synthetically, size grading could enhance growth and rearing efficiency, and did not have a significant influence on the immunity of L. vannamei Boone. Therefore, graded fanning in L. vannamei Boone was feasible in the culture practice.展开更多

Cyrtomiumfortunei, prescription I and prescription II were orally given to 15 fishes for each experimental group once a day, with the concentration of 9,175 and 36 g/S0 kg body weight, respectively. The administration was lasted for 14 days. Meanwhile, distilled water was orally given as the control. After the experiment, some of the experimental fishes were stimulated with high temperature (30 ℃ ) for two hours. Sampling was performed from fishes treated at 22 ℃ and 30 ℃ for measuring the protein content in the serum, phagocytic activity of leucocytes and the activity of the lysozyme in six tissues. The results indicated that Cyrtomiumfortunei and prescription II could help to enhance the content of various proteins and the phagocytic activity of leucocytes. There was no significant change in the effects between these two additives at either the normal temperature or high temperature. Prescription I did not show an obvious effect on the immunity of fishes at the normal temperature, but it did in promo- ting high-efficiency response and improving immunoregulation of fish shocked by some stimuli.展开更多

Results show that the activity of serum lysozyme decreased signifi- candy with extension of stress time (P 〈 0.05). The total antioxidant capacity (T-AOC) and activity of antioxidase in liver were significantly affected. The activi- ty of T-AOC and total superoxide dismutase (T-SOD) of fish exposed to ammonia-N were initially decreasing then increasing ( P 〈 0.05 ). Activities of catalase (CAT) and glutathione peroxidase (GSH-Px) were correlated with concentrations of ammonia-N. Fish exposed to lower concentrations (2.5 mg/L or 5 mg/L) showed decreased CAT activity within 24 h (P 〈 0.05 ), while those exposed to higher concentrations (10 mg/L or 20 mg/L) initially showed increased then decreased activity of CAT. Except for the highest concentration groups, fish exposed to ammonia-N showed inductive activity of GSH-Px ( P 〈 0.05 ). Under the ex- perimental conditions, non-specific immunity of tilapia was affected by ammonia-N stress, and the impact was increased with increased concentration and extension of time.展开更多

In this study, the influences of immersion bathing in different concentrations of Bacillus velezensis DY-6 on the body weight gain rate and non-specific immune enzyme activities of the coelom fluid of sea cucumber (Ap...In this study, the influences of immersion bathing in different concentrations of Bacillus velezensis DY-6 on the body weight gain rate and non-specific immune enzyme activities of the coelom fluid of sea cucumber (Apostichopus japonicus) were determined in order to obtain the optimum bacterial concentration. The gut microbiota change in A. japonicus was then analyzed through high-throughput sequencing during the immersion bathing in B. velezensis DY-6 at the optimum concentration for 49 d. The results illustrate that the body weight growth rate of all bathing groups was higher than that of the control. The highest growth rate (25.3%) was achieved when the bacterial concentration was 1×10^3 CFU/mL. The activities of non-specific immune enzymes (ACP, AKP, SOD and LZM) of all bathing groups increased, and the activities of the enzymes of groups bathed with the bacterium at 1×10^3 and 1×10^4 CFU/mL reached the highest on day 21 and day 28. Taking the growth rate and economic cost into consideration, the optimum concentration of B. velezensis DY-6 was 1×10^3 CFU/mL. The influences of immersion bathing in B. velezensis DY-6 at 1×10^3 CFU/mL on the gut microbiota of A. japonicus were then evaluated through 16S rDNA sequencing analysis. Results showed that the gut microbiota changed with the addition of B. velezensis DY-6, and the richness and diversity of the gut microbiota peaked twice on day 14 and day 21, respectively. In association with the non-specific immune enzyme activities and if day 28 was selected as the dividing point, the community structure of the gut microbiota could be obviously divided into two types. The correlation analysis revealed that the non-specific immune enzyme activities were correlated significantly to some gut bacteria (in the phyla Firmicutes, Proteobacteria, and Bacteroidetes) after immersion bathing in B. velezensis DY-6. Our findings will provide the theoretical foundation for probiotic application in sea cucumber farming.展开更多

In order to guide the antibody generation effectively, a decision hyper plane heuristic based artificial immune network classification algorithm (DHPA1NC) is proposed. DHPAINC taboos the inner regions of the class domain, thus, the antibody generation is limited near the class domain boundary. Then, the antibodies are evaluated by their recognition abilities, and the antibodies of low recognition abilities are removed to avoid over-fitting. Finally, the high quality antibodies tend to be stable in the immune network. The algorithm was applied to two simulated datasets classification, and the results show that the decision hyper planes determined by the antibodies fit the class domain boundaries well. Moreover, the algorithm was applied to UCI datasets classification and emotional speech recognition, and the results show that the algorithm has good performance, which means that DHPAINC is a promising classifier.展开更多
Neomycin belongs to the family of 2-deoxystreptamine-containing aminoglycoside antibiotics.It is widely used for bacterial infections,targeting most gram-negative and some gram-positive bacteria.Neomycin has also been...Neomycin belongs to the family of 2-deoxystreptamine-containing aminoglycoside antibiotics.It is widely used for bacterial infections,targeting most gram-negative and some gram-positive bacteria.Neomycin has also been reported to show antiviral activity.Here,we evaluated the toxicity of neomycin sulfate,and investigated its effect on non-specific immunity and viral infection in crucian carp(Carassius auratus gibelio).The safe concentration of neomycin sulfate for crucian carp was determined to be 102.9 mg/kg in vivo.In oxidative damage assays,neomycin sulfate increased superoxide dismutase and catalase activity and decreased malondialdehyde in the liver of crucian carp.In non-specific blood immune assays,the white blood cell count and complement 3 content significantly increased after neomycin sulfate treatment,while no significant changes were observed in antibacterial or lysozyme activity.In a challenge test,neomycin sulfate protected the crucian carp from cyprinid herpesvirus 2(CyHV-2)infection and inhibited CyHV-2 replication.In cytotoxicity assays,low concentrations of neomycin sulfate had no cytotoxicity on cells from the fins of crucian carp.The results of the present study indicate that oral administration of neomycin sulfate reduced oxidative damage,enhanced immunity and provided protection against CyHV-2 in crucian carp.展开更多

[Method] Totally 500 individuals of 1-day-old Guangxi Partridge chickens were randomly divided into five groups, five replicates each group and 20 chicks each replicate. Groups A, B and C were treatment groups supplemented with 0.70%, 0.35% and0.175% P. felterrae extracts in the basal diet, respectively; group D was a medical control group supplemented with 0.01% colistin sulfate premix in the basal diet; and group E was a control group with basal diet. Serum samples were collected from 10 chickens in each group at 21, 35 and 49 days of age, to analyze the biochemical and immune indexes. [Result] It had no significant impact on serum TP and GLU contents of chickens by adding different levels of P. felterrae extracts in the basal diet(P 〉0.05). However, at 21 and 35 days of age, the TBILI content of chickens in groups A, B and C were extremely lower or significantly lower than that in group E( P〈0.01; P〈0.05). At 35 days of age, the serum AKP activities in groups A, B and C were extremely higher than that in group E(P〈0.01), and the serum GOT activities at 21 and 35 days of age in group C were significantly higher than that in group E(P〈0.05). At 49 days of age, the serum Ig G content in groups A, B and C and the IL-2 content in group B and group C were significantly lower or extremely lower than that in group E( P〈0.05; P〈0.01); the T-AOC content in group B was significantly higher than that in group E(P〈0.05); the GSH content in groups A, B and C were significantly higher than that in group E( P〈0.05). Adding different levels of P. felterrae extracts in the diet significantly decreased the TBILI content, reduced the Ig G, IL-2 and IL-6 contents, and improved the AKP and GOT activities and the T-AOC and GSH contents of Guangxi Partridge chicken. [Conclusion] Adding 0.35% P. felterrae extracts in the basal diet received the best effort.展开更多
This study aimed to evaluate the effects of dietary geniposide supplementation on growth performance,lipid metabolism,health status,and ammonia stress resistance in turbot(Scophthalmus maximus).Four hundred fifty fish...This study aimed to evaluate the effects of dietary geniposide supplementation on growth performance,lipid metabolism,health status,and ammonia stress resistance in turbot(Scophthalmus maximus).Four hundred fifty fish were randomly allocated into 5 treatments with triplicate tanks(30 fish per tank).They were hand-fed to apparent satiety for 56 d with a basal diet(GP0)or diets containing 100,200,400,and 800 mg/kg geniposide(termed as GP100,GP200,GP400,GP800,respectively).After the conclusion of the feeding trial,the fish were exposed to ammonia stress for 96 h.The results showed that the growth performance were not affected by geniposide(P>0.05).Dietary supplementation with geniposide decreased crude lipid in viscera without liver,subcutaneous adipose tissue(SAT),and the liver,as well as triglyceride concentrations in plasma,the liver and SAT(P<0.05).Dietary supplementation with 400 and 800 mg/kg geniposide significantly down-regulated lipogenesis-related gene expression,as well as fatty acid uptake-related gene expression,while significantly up-regulated triglyceride secretion-related gene expression in the liver compared with the control group(P<0.05).The GP800 group exhibited a significant reduction in plasma malondialdehyde contents compared with the control group,while both the GP200 and GP800 groups showed a significant increase in plasma complement C3 activities(P<0.05).Furthermore,there was a notable enhancement in plasma lysozyme and total superoxide dismutase levels in the geniposide supplemented groups compared to the control group(P<0.05).Additionally,a significant decrease in the mRNA level of pro-inflammatory cytokine and a remarkable increase in the mRNA expression of anti-inflammatory cytokines were discovered in geniposide supplemented groups relative to the control group(P<0.05).Cumulative survival rates after ammonia stress in the GP400 and GP800 groups were statistically higher than that in the control group(P<0.05).In conclusion,dietary geniposide supplementation could reduce lipid deposition in turbot by regulating lipid metabolism and transportation,and remarkably enhance immunity,antioxidant ability,and resistance to ammonia stress in turbot.Based on the quadratic regression analyses,the optimal concentrations of geniposide were estimated to be 545.21 to 668.41 mg/kg feed.展开更多

文摘In order to verify the synthesis pathway of linoleic acid(LA)to generate arachidonic acid(ARA),the functions ofΔ6 FAD and Elovl 5 in Apostichopus japonicus were tested by heterologous expression in Pichia pastoris.A 60-day feeding experiment was conducted to evaluate the effects of dietary LA and ARA on growth,polyunsaturated fatty acids(PUFA)biosynthesis and im-mune function of A.japonicus.Seven diets containing graded levels of LA or ARA were formulated,and one diet without PUFA was applied as a control.The results confirmed thatΔ6 FAD from A.japonicus has a double desaturation ability ofΔ6 andΔ5 for PUFA,and it works together with Elovl 5 enzyme play important roles in biosynthesis of ARA from LA.With increasing dietary LA from 4.9 to 12.1 g/kg,the expressions ofΔ6 FAD and Elovl 5,levels of LA,ARA and EPA in tissues increased,and the activities of ACP,AKP and CAT enzymes in intestine initially increased and then decreased.Additionally,by increasing dietary ARA from 0.7 to 3.8 g/kg,the contents of ALA,EPA and DHA decreased,while theΔ6 FAD expression and the activities of ACP,AKP and CAT in intes-tine increased significantly.Therefore,appropriate dietary levels of LA(12.1 g/kg)or ARA(3.8 g/kg)build a reasonable composi-tion of n-3/n-6 PUFA in A.japonicus,and improved its growth,antioxidant capacity and immunological defenses.

文摘Exogenous alanyl-glutamine(Aln-Gln) was evaluated for its effects on growth performance, intestinal structure and function, antioxidant status and non-specific immunity of young carp(Cyprinus carpio L.). Six diets supplemented with 0, 2.5, 5.0, 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln were fed to fish for 12 weeks. Supplementation with 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln significantly increased weight gain rate(WGR), protein efficiency ratio(PER), but feed conservation rate(FCR) and survival were not affected(P〉0.05). The intestinal fold height and number, digestive enzyme, Na+, K+-ATPase activities was found to be significantly high(P〈0.05) with increasing dietary Aln-Gln supplementation up to 7.5 g · kg-1, but there were no significant differences for Aln-Gln supplementation from 7.5 to 15.0 g · kg-1. The glutathione peroxidase(GPX) activity, glutathione(GSH), superoxide dismutase(SOD) activity increased and malondialdehyde(MDA) levels decreased significantly(P〈0.05) in the intestine, hepatopancreas, plasma and muscles. The plasma complement-3(C3) and complement-4(C4) levels were significantly(P〈0.05) improved at 5.0 g · kg-1 level and decreased when over 7.5 g · kg-1. The plasma lysozyme(LSZ) activity increased significantly(P〈0.05) at 7.5, 10.0, or 15.0 g · kg-1 level. In summary, the results showed that Aln-Gln improved growth performance, development and function of the intestine, the activity of the antioxidant defense system and the plasma non-specific immunity of the carps. The optimal Aln-Gln level was 8.24 g · kg-1 diet for WGR based on broken-line regression model analysis.

文摘Here, we investigated the effect of dietary arginine(Arg) supplementation on innate immunity and the antioxidant ability of broiler chickens. The experiment was designed as a single-factorial arrangement(n=8 cages/treatment, six birds/cage), and we used four dietary Arg concentrations(10.0, 15.0, 20.0 or 25.0 g kg–1). On day 21, the birds were killed to obtain spleen, cecal tonsil and liver samples to determine the gene expression and antioxidant characteristics. Increasing the Arg concentration linearly decreased(P0.05) the m RNA expression of splenic interleukin-18(IL-18) and tumor necrosis factor-α(TNF-α). Dietary Arg supplementation quadratically decreased(P0.05) the expression of interleukin-1b(IL-1b) and interferon-γ(IFN-γ) m RNA in the spleen. Increasing Arg concentrations linearly and quadratically reduced the expression of IL-18 m RNA in the spleen. Meanwhile, increasing dietary Arg supplementation linearly and quadratically increased the lymphotactin m RNA(P0.05) expression, and linearly increased the macrophage inflammatory protein-1β(MIP-1β) and toll-like receptor 15(TLR15) m RNA expression in the cecal tonsils. Dietary Arg supplementation linearly(P0.05) increased the glutathione peroxidase(GSH-Px), catalase(CAT), and lysozyme(LZM) activities in the liver. However, the malondialdehyde(MDA) activity in the liver was not influenced by the dietary Arg concentration(P0.05). No significant(P0.05) effect was found on the activity of superoxide dismutase(SOD) in the liver. Thus high levels of Arg supplementation(20.0 g kg^(–1)) may potentially suppress the innate immunity of broiler chickens, and dietary Arg supplementation enhances the antioxidant activity in broiler chickens.

文摘The study aims to determine the effects of graded farming on growth performance and non-specific immunity factors of shrimp Litopenaeus vannamei Boone. Three size groups of shrimp, i.e., the small size group [Gs, with an average body length (BL) of (3.04 ± 0.36) cm and body weight (BW) (0.412± 0.35) g], the large group [GL, with a BL of (4.29±0.55) cm and BW of (1.098 ±0.42) g], and the ungraded group [Gm, with a BL of (3.47±0.81) cm and BW of (0.611 ±0.79) g], were reared under the same conditions for 8 wk. Growth performance and non- specific immunity factors were measured. The results showed that BW gain, biomass gain and the specific growth rate of body length (SGRL) were significantly influenced by size grading (one-way ANOVA, P 〈 0.05). The peroxidase (POD) and antibacterial (Ua) activities of GL were lower than those of G. Superoxide dismutase (SOD) and lysozyme (U1) activities of Gm were lower than those of G. No significant difference (P = 0.121 〉 0.05) was found on phenoloxidase (PO) activity among the three size groups. Synthetically, size grading could enhance growth and rearing efficiency, and did not have a significant influence on the immunity of L. vannamei Boone. Therefore, graded fanning in L. vannamei Boone was feasible in the culture practice.

文摘This study was to investigate the effects of aqueous decotion from three Chinese medicinal herb additives ( Cyrtomium fortunei, prescription I and prescription II) on non-specific immunity of 1 + age old Amur sturgeon (Acipenser schrencki Brandt) by oral perfusion. Cyrtomiumfortunei, prescription I and prescription II were orally given to 15 fishes for each experimental group once a day, with the concentration of 9,175 and 36 g/S0 kg body weight, respectively. The administration was lasted for 14 days. Meanwhile, distilled water was orally given as the control. After the experiment, some of the experimental fishes were stimulated with high temperature (30 ℃ ) for two hours. Sampling was performed from fishes treated at 22 ℃ and 30 ℃ for measuring the protein content in the serum, phagocytic activity of leucocytes and the activity of the lysozyme in six tissues. The results indicated that Cyrtomiumfortunei and prescription II could help to enhance the content of various proteins and the phagocytic activity of leucocytes. There was no significant change in the effects between these two additives at either the normal temperature or high temperature. Prescription I did not show an obvious effect on the immunity of fishes at the normal temperature, but it did in promo- ting high-efficiency response and improving immunoregulation of fish shocked by some stimuli.

文摘The juveniles of hybrid tilapia ( Oreochromis niloticus x Oreochromis areus) were exposed to ananonia-nitrogen (N) (0, 2.5, 5, 10 and 20 mg/L) for 0, 12, 24, 48 and 72 h to evaluate the effect of ammonia-N stress on their non-specific immunity. Results show that the activity of serum lysozyme decreased signifi- candy with extension of stress time (P 〈 0.05). The total antioxidant capacity (T-AOC) and activity of antioxidase in liver were significantly affected. The activi- ty of T-AOC and total superoxide dismutase (T-SOD) of fish exposed to ammonia-N were initially decreasing then increasing ( P 〈 0.05 ). Activities of catalase (CAT) and glutathione peroxidase (GSH-Px) were correlated with concentrations of ammonia-N. Fish exposed to lower concentrations (2.5 mg/L or 5 mg/L) showed decreased CAT activity within 24 h (P 〈 0.05 ), while those exposed to higher concentrations (10 mg/L or 20 mg/L) initially showed increased then decreased activity of CAT. Except for the highest concentration groups, fish exposed to ammonia-N showed inductive activity of GSH-Px ( P 〈 0.05 ). Under the ex- perimental conditions, non-specific immunity of tilapia was affected by ammonia-N stress, and the impact was increased with increased concentration and extension of time.

文摘Cinnamon essential oil(CEO)and its combination with bamboo leaf flavonoid(BLF)have been shown to exhibit an additive antibacterial effect in vitro,but their functions in broilers were not clear.An experiment was conducted to investigate the effects of dietary graded levels of CEO and its combination with BLF on the growth performance,immune responses,antioxidative ability,and intestinal morphology and microbiota of broilers fed a corn-soybean meal basal diet.A total of 576 1-d-old Arbor Acres commercial male broilers were randomly allotted to 9 treatments with 8 replicates of 8 birds each in a completely randomized design.Birds were fed on a basal corn-soybean meal diet(control,without plant extracts and antibiotics),or the basal diet supplemented with 50 mg of aureomycin kg^-1,50,100,200,400,or 800 mg of CEO kg^-1,a combination of 100 mg of CEO and 16.7 mg of BLF kg^-1,or a combination of 200 mg of CEO kg^-1 and 33.3 mg of BLF kg^-1 for 42 d.Dietary treatment affected(P<0.05)the serum immune globulin M(IgM)contents on d 42,liver malondialdehyde(MDA)contents on d 21,duodenal crypt depth on d 42,relative abundance of Lactobacillus in the cecal contents on d 21,and relative abundances of Escherichia coli and Bifidobacterium in the cecal contents on d 42,but had no effect(P>0.16)on all other measured indices.The addition of 400 mg of CEO kg^-1 or a combination of 200 mg CEO kg^-1 and 33.3 mg BLF kg^-1 increased(P<0.02)serum IgM contents on d 42.Dietary supplementation with 100 or 200 mg CEO kg^-1,or 50 mg aureomycin kg^-1 decreased(P<0.003)liver MDA contents on d 21.In addition,the supplement of 100 mg CEO kg^-1 increased(P<0.002)the Lactobacillus relative abundance in caecum on d 21 and Bifidobacterium relative abundance in caecum on d 42,and decreased(P<0.0001)E.coli relative abundance in caecum on d 42.The results indicated that dietary supplementation with CEO,an alternative to aureomycin,improved the immune status,antioxidantative ability and cecal microbiota of broilers,and dietary supplementation with the combinations of CEO and BLF did not exhibit further effects.Dietary supplementation with 100 mg CEO kg^-1 is beneficial for broilers fed a corn-soybean meal basal diet.

文摘In this study, the influences of immersion bathing in different concentrations of Bacillus velezensis DY-6 on the body weight gain rate and non-specific immune enzyme activities of the coelom fluid of sea cucumber (Apostichopus japonicus) were determined in order to obtain the optimum bacterial concentration. The gut microbiota change in A. japonicus was then analyzed through high-throughput sequencing during the immersion bathing in B. velezensis DY-6 at the optimum concentration for 49 d. The results illustrate that the body weight growth rate of all bathing groups was higher than that of the control. The highest growth rate (25.3%) was achieved when the bacterial concentration was 1×10^3 CFU/mL. The activities of non-specific immune enzymes (ACP, AKP, SOD and LZM) of all bathing groups increased, and the activities of the enzymes of groups bathed with the bacterium at 1×10^3 and 1×10^4 CFU/mL reached the highest on day 21 and day 28. Taking the growth rate and economic cost into consideration, the optimum concentration of B. velezensis DY-6 was 1×10^3 CFU/mL. The influences of immersion bathing in B. velezensis DY-6 at 1×10^3 CFU/mL on the gut microbiota of A. japonicus were then evaluated through 16S rDNA sequencing analysis. Results showed that the gut microbiota changed with the addition of B. velezensis DY-6, and the richness and diversity of the gut microbiota peaked twice on day 14 and day 21, respectively. In association with the non-specific immune enzyme activities and if day 28 was selected as the dividing point, the community structure of the gut microbiota could be obviously divided into two types. The correlation analysis revealed that the non-specific immune enzyme activities were correlated significantly to some gut bacteria (in the phyla Firmicutes, Proteobacteria, and Bacteroidetes) after immersion bathing in B. velezensis DY-6. Our findings will provide the theoretical foundation for probiotic application in sea cucumber farming.

文摘Non-specific immune response of bullfrog Rana catesbeiana to pathogenic Aeromonas hydrophila was studied to 60 individuals in two groups.Each bullfrog in bacterium-injected group was injected intraperitoneally(i.p.) with 0.2 ml bacterial suspension at a density of 5.2 × 106 CFU/ml,while each one in control group injected i.p.with 0.2 ml sterile saline solution(0.85%,w/v).Three bullfrogs in both groups were sampled at 0,1,3,7,11,15 and 20 days post-injection(dpi) for the evaluation of non-specific immune parameters.It was observed that intraperitoneal injection of A.hydrophila significantly increased the number of leucocytes and that of NBT-positive cells in peripheral blood.Significant increases in serum bactericidal activity and serum acid phosphatase activity were also observed in the bacterium-injected frogs when compared with those in the control group.However,a significant reduction was detected in vitro in phagocytosis activity of peripheral blood phagocytes.No significant difference in changes in the number of peripheral erythrocytes,serum superoxide dismutase(SOD) activity,and lysozyme activity was detected between the two groups.It is suggested that bullfrogs may produce a series of non-specific immune reactions in response to the A.hydrophila infection.

文摘This experiment was to investigate ginger extract on production performance,antioxidant ability and immunity of laying hens.A total of 600 Hy-Line brown laying hens aged at 25 wk old were randomly divided into 2 treatments,4 replicates per treatment,75 layers each replicate.The control group hens were fed a basal diet;the experimental group hens were fed basal diets with 0.1%ginger extract.The results were shown as follows:1)ginger extract significantly enhanced laying rates(P<0.05)and daily egg weight(P<0.05),substantially reduced the ratio of feed to egg(P<0.05)of the hens;2)ginger extract did not change the activities of glutathione peroxidase(GSH-PX)and total antioxidant capacity(TAOC)but significantly improved plasma superoxide dismutase(SOD)activity(P<0.05),reduced malondialdehyde(MDA)content(P<0.05)of the birds;3)ginger extract did not affect the contents of serum total protein(TP),albumin(ALB),globulin(GLB),but significantly increased lysozyme(LZM)activity(P<0.05);4)ginger extract also significantly reduced plasma prostaglandin E2(PGE2)content(P<0.05).This study shows that ginger extract not only can improve the birds’antioxidant capacity,enhance immune function,but also has a potential of reducing inflammatory response.

However, the relationship of the methionine and immunity has not been elucidated clearly, this review aims to clarify the effects of methionine on immune function in the aspects of growth and development immune organs, the histological structure of the immune organs, non-specific immunity, humoral immunity, cellular immunity and cytokines, and to provide foundations for further studies on the relationship between methionine and immune function.

文摘Most of the developed immune based classifiers generate antibodies randomly, which has negative effect on the classification performance. In order to guide the antibody generation effectively, a decision hyper plane heuristic based artificial immune network classification algorithm (DHPA1NC) is proposed. DHPAINC taboos the inner regions of the class domain, thus, the antibody generation is limited near the class domain boundary. Then, the antibodies are evaluated by their recognition abilities, and the antibodies of low recognition abilities are removed to avoid over-fitting. Finally, the high quality antibodies tend to be stable in the immune network. The algorithm was applied to two simulated datasets classification, and the results show that the decision hyper planes determined by the antibodies fit the class domain boundaries well. Moreover, the algorithm was applied to UCI datasets classification and emotional speech recognition, and the results show that the algorithm has good performance, which means that DHPAINC is a promising classifier.

文摘Neomycin belongs to the family of 2-deoxystreptamine-containing aminoglycoside antibiotics.It is widely used for bacterial infections,targeting most gram-negative and some gram-positive bacteria.Neomycin has also been reported to show antiviral activity.Here,we evaluated the toxicity of neomycin sulfate,and investigated its effect on non-specific immunity and viral infection in crucian carp(Carassius auratus gibelio).The safe concentration of neomycin sulfate for crucian carp was determined to be 102.9 mg/kg in vivo.In oxidative damage assays,neomycin sulfate increased superoxide dismutase and catalase activity and decreased malondialdehyde in the liver of crucian carp.In non-specific blood immune assays,the white blood cell count and complement 3 content significantly increased after neomycin sulfate treatment,while no significant changes were observed in antibacterial or lysozyme activity.In a challenge test,neomycin sulfate protected the crucian carp from cyprinid herpesvirus 2(CyHV-2)infection and inhibited CyHV-2 replication.In cytotoxicity assays,low concentrations of neomycin sulfate had no cytotoxicity on cells from the fins of crucian carp.The results of the present study indicate that oral administration of neomycin sulfate reduced oxidative damage,enhanced immunity and provided protection against CyHV-2 in crucian carp.

文摘[Objectives]This trial was conducted to investigate the effects of diet with"Lianteng"additive of Chinese herbal medicine on blood biochemical indexes,immune and antioxidant function of Guangxi Sanhuang chickens.[Methods]480 Guangxi Sanhuang chickens at the age of 1 d were divided into 4 groups randomly and each group was assigned to 6 replicates with 20 chicks in each replicate;groups A,B and C were treatment groups and fed basal diets with 0.1%,0.2%and 0.4%of"Lianteng"additive of Chinese herbal medicine respectively during a trial of 70 d.Group D was control group and fed only basal diet.The serum samples were taken from 6 chicks in each group at the age of 21,35 and 49 d respectively and analyzed for biochemical,immune and antioxidant parameters in serum.[Results]Results from the trial showed that no difference was found in serum content of GLU,TG,TP,TC,and UN in the measurement at the age of 21,35 and 49 d(P>0.05);however,the content of ALB from groups A and B at 35 d was significantly higher than that from group D(P<0.01),and the ALB content from group B at 49 d was also higher than that from group D(P<0.05);the IgM content at 49 d from group A was lower than that from group D(P<0.05),in addition to this,there was no differences found in the contents of IgG,IgM and IFN-y among groups A,B,C and D during the trial(P>0.05);but the content of IL-2 from groups A and B at 49 d was significantly greater than that from group D(P<0.05);T-AOC from groups A,B and C at 49 d was very significantly higher than that from group D(P<0.01),and the one of group B at 35 d was still obviously higher than that of group D(P<0.05);also at 49 d,the GSH content in groups A and B was much greater than that in group D(P<0.01);T-SOD activity of group B and C at 35 d was distinctly greater than that of group D(P<0.05).[Conclusions]It can be concluded that the addition of 0.2%of"Lianteng"additive of Chinese herbal medicine in diet can increase immune function in a certain extent and promote the antioxidant ability observably for Guangxi Sanhuang chickens.

文摘[Objective] This trial was to investigate the effect of dietary Picria felterrae Lour extracts on serum biochemical indexes and immune function of Guangxi Partridge chickens. [Method] Totally 500 individuals of 1-day-old Guangxi Partridge chickens were randomly divided into five groups, five replicates each group and 20 chicks each replicate. Groups A, B and C were treatment groups supplemented with 0.70%, 0.35% and0.175% P. felterrae extracts in the basal diet, respectively; group D was a medical control group supplemented with 0.01% colistin sulfate premix in the basal diet; and group E was a control group with basal diet. Serum samples were collected from 10 chickens in each group at 21, 35 and 49 days of age, to analyze the biochemical and immune indexes. [Result] It had no significant impact on serum TP and GLU contents of chickens by adding different levels of P. felterrae extracts in the basal diet(P 〉0.05). However, at 21 and 35 days of age, the TBILI content of chickens in groups A, B and C were extremely lower or significantly lower than that in group E( P〈0.01; P〈0.05). At 35 days of age, the serum AKP activities in groups A, B and C were extremely higher than that in group E(P〈0.01), and the serum GOT activities at 21 and 35 days of age in group C were significantly higher than that in group E(P〈0.05). At 49 days of age, the serum Ig G content in groups A, B and C and the IL-2 content in group B and group C were significantly lower or extremely lower than that in group E( P〈0.05; P〈0.01); the T-AOC content in group B was significantly higher than that in group E(P〈0.05); the GSH content in groups A, B and C were significantly higher than that in group E( P〈0.05). Adding different levels of P. felterrae extracts in the diet significantly decreased the TBILI content, reduced the Ig G, IL-2 and IL-6 contents, and improved the AKP and GOT activities and the T-AOC and GSH contents of Guangxi Partridge chicken. [Conclusion] Adding 0.35% P. felterrae extracts in the basal diet received the best effort.

文摘This study aimed to evaluate the effects of dietary geniposide supplementation on growth performance,lipid metabolism,health status,and ammonia stress resistance in turbot(Scophthalmus maximus).Four hundred fifty fish were randomly allocated into 5 treatments with triplicate tanks(30 fish per tank).They were hand-fed to apparent satiety for 56 d with a basal diet(GP0)or diets containing 100,200,400,and 800 mg/kg geniposide(termed as GP100,GP200,GP400,GP800,respectively).After the conclusion of the feeding trial,the fish were exposed to ammonia stress for 96 h.The results showed that the growth performance were not affected by geniposide(P>0.05).Dietary supplementation with geniposide decreased crude lipid in viscera without liver,subcutaneous adipose tissue(SAT),and the liver,as well as triglyceride concentrations in plasma,the liver and SAT(P<0.05).Dietary supplementation with 400 and 800 mg/kg geniposide significantly down-regulated lipogenesis-related gene expression,as well as fatty acid uptake-related gene expression,while significantly up-regulated triglyceride secretion-related gene expression in the liver compared with the control group(P<0.05).The GP800 group exhibited a significant reduction in plasma malondialdehyde contents compared with the control group,while both the GP200 and GP800 groups showed a significant increase in plasma complement C3 activities(P<0.05).Furthermore,there was a notable enhancement in plasma lysozyme and total superoxide dismutase levels in the geniposide supplemented groups compared to the control group(P<0.05).Additionally,a significant decrease in the mRNA level of pro-inflammatory cytokine and a remarkable increase in the mRNA expression of anti-inflammatory cytokines were discovered in geniposide supplemented groups relative to the control group(P<0.05).Cumulative survival rates after ammonia stress in the GP400 and GP800 groups were statistically higher than that in the control group(P<0.05).In conclusion,dietary geniposide supplementation could reduce lipid deposition in turbot by regulating lipid metabolism and transportation,and remarkably enhance immunity,antioxidant ability,and resistance to ammonia stress in turbot.Based on the quadratic regression analyses,the optimal concentrations of geniposide were estimated to be 545.21 to 668.41 mg/kg feed.
