Acne is a highly common skin condition,estimated to affect 9.4%of the global population at some point in their lives.Today,there is a growing demand for environmentally conscious products that treat skin diseases with...Acne is a highly common skin condition,estimated to affect 9.4%of the global population at some point in their lives.Today,there is a growing demand for environmentally conscious products that treat skin diseases without harming the environment,known as Clean Beauty.For this reason,the objective is to develop a delivery system based on bacterial cellulose loaded with natural actives nisin and mandelic acid for acne treatment.The system,using bacterial cellulose(BC)from Komagataebacter xylinus fermentation,was tested for antibacterial and antioxidant activity,active release,cytotoxicity,Sscanning Eelectron microscopy,and product stability.The results of this study are promising:the BC system exhibited potent antibacterial activity against both standard and skinisolated strains,along with high antioxidant activity.The controlled release of active ingredients,peaking at 300 min,and the stability of antibacterial and antioxidant properties over 180 days underscore the system's potential as a shelf-stable product.Additionally,the minimal cytotoxicity observed affirms the safety of the formulation.This innovative approach not only aligns with the principles of clean beauty by avoiding petroleumbased components but also provides a sustainable and effective solution for managing acne.The integration of natural actives within a biodegradable matrix positions this product as a frontrunner in the future of dermocosmetics,meeting consumer demands for products that are both skin-friendly and environmentally responsible.展开更多
The objective of this study is to propose a more accurate and faster MTT 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide colorimetric assay (MCA) for quantitative measurement of polypeptide bacteriocins...The objective of this study is to propose a more accurate and faster MTT 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide colorimetric assay (MCA) for quantitative measurement of polypeptide bacteriocins in solutions with nisin as an example. After an initial incubation of nisin and indicator bacterium Micrococcus luteus NCIB 8166 in tubes, MTT was added for another incubation period. After that, nisin was quantified by estimating the number of viable bacteria based on measuring the amount of purple formazan produced by cleavage of yellow tetrazolium salt MTT. Then MCA was compared to a standard agar diffusion assay (ADA). The results suggested a high correlation coefficient (r2=0.975±0.004) between optical density (OD) and the inhibitory effect of nisin on a bacterial strain Micrococcus luteus NCIB 8166 at a range of 0.125~32 IU/ml. The MCA described in this study was very quick. Quantification of nisin took only 7~8 h and the detection limit was at the level of 0.125 IU/ml when compared to 12 IU/ml and 24~28 h for ADA. The MCA provides an accurate and rapid method for quantifi-cation of nisin in solutions and is expected to be used for quantification of other antimicrobial substances.展开更多

For that purpose, both antimicrobials were loaded into Oil-in-/Water nanoemulsions, and their antimicrobial activity against Bacillus subtilis was investigated. The experimental results indicated that the O/W nanoemulsions loaded with both, nisin and carvacrol, having Sauter mean diameter (d3,2) of around 120 nm, were considerably stable under the conditions investigated (up to 1 week storage at room temperature). The interfacial tension between nisin aqueous solutions and soybean oil could be reduced up to 12 mN/m, as compared to that of pure water and soybean oil (21 mN/m), so that demonstrating the potential of nisin to be used as emulsifier. The results obtained for O/W nanoemulsions loaded with both antimicrobial compounds indicated a reduction of nearly two log cycles (around 100 times) on B. subtilis population upon incubation for 24 h, compared to the blank (10 mM phosphate buffer).展开更多
This study aimed to assess the therapeutic potential of nisin,5-fluorouracil(5FU)and selenium encapsulated in folate-conjugated thiolated chitosan nanoparticles(N/5FU/Se@FTCsNPs),combined with a probiotic cocktail of ...This study aimed to assess the therapeutic potential of nisin,5-fluorouracil(5FU)and selenium encapsulated in folate-conjugated thiolated chitosan nanoparticles(N/5FU/Se@FTCsNPs),combined with a probiotic cocktail of Lactobacillus acidophilus and Bifidobacterium bifidum,against colorectal cancer(CRC).The nanoparticles(277 nm,+9.2 mV)exhibited high drug loading efficiencies(5FU:89.11%,nisin:70.68%)and pH-responsive release,with minimal drug release under gastric conditions and∼60.7%release at colonic pH,facilitating targeted delivery.The formulation remained stable for over 40 d at−20°C and 4°C,demonstrating excellent biocompatibility(<2%hemolysis)and exhibiting strong mucoadhesive and mucus-penetrating abilities.In vitro,N/5FU/Se@FTCsNPs selectively targeted CT26 colon cancer cells(IC5n:1.57μg/ml)with minimal effects on healthy cells,enhanced cellular uptake,and induced ROS-mediated apoptosis.In vivo,oral administration-especially with probiotics-significantly reduced tumor volume,improved survival rates and alleviated chemotherapy-related side effects such as diarrhea and weight loss.Biodistribution studies confirmed increased tumor targeting and decreased off-target exposure.Mechanistically,the treatment downregulated oncogenes and inflammatory markers(2-to 12.5-fold),includingβ-catenin,mTOR,COX-2 and VEGF-α,while upregulating tumor suppressors and protective genes(4 to 14.8 fold),such as PTEN,CASP9 and Mucin 2(P<0.0001).This indicates inhibition of proliferation,metastasis,inflammation,and angiogenesis,along with improved gut barrier function.Cytokine profiling and histological analysis further confirmed reduced systemic inflammation and maintained hematological safety.These findings highlight N/5FU/Se@FTCsNPs combined with probiotics as a promising,safe and effective oral therapy for CRC,leveraging microbiota modulation and targeted delivery.展开更多

W...Multi-walled carbon nanotube(MWCNT) sheet was fabricated from a drawable MWCNT forest and then deposited on poly(methyl methacrylate) film. The film was further coated with a natural antimicrobial peptide nisin. We studied the effects of nisin coating on the attachment of Bacillus anthracis spores, the germination of attached spores, and the subsequent biofilm formation from attached spores. It was found that the strong adsorptivity and the super hydrophobicity of MWCNTs provided an ideal platform for nisin coating. Nisin coating on MWCNT sheets decreased surface hydrophobicity, reduced spore attachment, and reduced the germination of attached spores by 3.5 fold, and further inhibited the subsequent biofilm formation by 94.6% compared to that on uncoated MWCNT sheet. Nisin also changed the morphology of vegetative cells in the formed biofilm.The results of this study demonstrated that the anti-adhesion and antimicrobial effect of nisin in combination with the physical properties of carbon nanotubes had the potential in producing effective anti-biofilm formation surfaces.展开更多
Objective:To examine the individual and synergistic anti-listcrial effect of nisin and leaf essential oil of Metasequoia glyptostroboides(M.glyptostroboides) against one of the leading foodhorne pathogens Listeria mon...Objective:To examine the individual and synergistic anti-listcrial effect of nisin and leaf essential oil of Metasequoia glyptostroboides(M.glyptostroboides) against one of the leading foodhorne pathogens Listeria monocytogenes(L.monocytogenes) ATCC 19116 in milk samples.Methods:The whole(8%),low(1%) and skim(no fat content) milk samples were inoculated with L.monocytogenes ATCC 19116 along with leaf essential oil of M.glyptostroboides or nisin alone as well in eomhinations.Results:In this study,the leaf essential oil at the concentrations of 2% and 5% revealed strong anti-listerial effect against L.monocytogenes ATCC 19116 in all categories of milk samples.Nisin at the concentrations of 250 and 500 IU/mL displayed a strong inhibitory effect against ATCC 19116 as compared to the control group.Additionally,synergistic combinations of leaf essential oil(1%) and nisin(62.5,125.250 and 500 IU/mL) also had a remarkable anti-listerial synergism in all the tested milk samples including whole,low and skim milk after 14 days.Conclusions:As a major finding,the leaf essential oil of M.glyptostroboides might he a useful candidate for using in food industry to control the growth of foodhorne pathogenic bacteria as confirmed by its potent anti-listerial Synergistic effect with nisin against L.monocytogenes ATCC 19116 in different milk samples.展开更多

The mutant strain secreted fully modified nisin with the N-terminal leader still attached.The presence of the leader was confirmed by N-terminal sequencing of the purified precursor. The dehydration and lanthionine formation of the precursor were already completed as active nisin could be formed by cleaving the leader from the inactive precursor by a trypsin treatment or by incubation with wild type cells. Nisin immunity of the NisP mutant strain was lowered to about 10% of the wild type immunity. The results show that NisP is needed for precursor processing and for development of high immunity of nisin.展开更多

Theretbre, here, the effect of nisin was searched in traditional fermented sausage Gombasek experimentally contaminated with Listeria innocua Lil strata Ihe antimicrobial effect of nisin was demonstrated by the Lil strain decrease at day 2 (after contamination) with the difference 1.79 logarithmic (log) cycles comparing with the control sausage (CLi). At week l, the difference 4.14 log cycles was noted in the experimental sausages comparing with CLi sausages with prolonging inhibitory effect up to week 2; although the difference was diminished (1.82 log cycles); but higher than at day 2, The pH values as well as lactic acid values were not influenced by the nisin treatment (pH value in the reference sausage was 5.0, in CLi 5.7 and in the experimental sausage E, 5.1; amount of lactic acid in t: was 0.099 mmol L^-1: and in CLi 0.107 mmol L^-1).展开更多
