Combined treatment of ischemic stroke with Chinese medicine and exogenous bone marrow mesenchymal stem cell(BMSC) transplantation may improve the removal of blood stasis and stimulation of neogenesis.Chinese medicines that remove blood stasis not only promote blood circulation but also calm the endopathic wind,remove heat,resolve phlegm,remove toxic substances and strengthen body resistance.The medicinal targeting effect of Chinese medicine can promote the homing of BMSCs,and the synergistic therapeutic effects of drugs can contribute to BMSC differentiation.As such,exogenous BMSC transplantation has potential advantages for neogenesis.Chinese medicines and exogenous BMSCs provide complementary functions for the removal of blood stasis and stimulation of neogenesis.Therefore,a combination of Chinese medicine and transplantation of exogenous BMSCs may be particularly suited to ischemic stroke treatment. 展开更多

Tubular duplication of the colon is very rare especially in adulthood,because it is frequently symptomatic earlier in newborn life,so only few cases are reported in literature.Several theories are proposed to explain the onset and the evolution of gut malformations as the aberrant lumen recanalization or the diverticular theory,the alteration of the lateral closure of the embryonal disk or finally the dorsal protrusion of the yolksac for herniation or adhesion to the ectoderm for an abnormality of the longitudinal line,but none clarifies the exact genesis of duplication.We present a case of "Y-shaped" tubular duplication of the transverse colon in a 21-year-old adult,with a history of chronic pain and constipation,referred to our department for abdominal pain with retrosternal irradiation,treated with the resection of the aberrant bowel. 展开更多

Therapeutic strategies for cerebral ischemic stroke face critical challenges in mitigating secondary cerebrovascular injury following recanalization.In this study,we engineered interleukin-4-functionalized vasculogenic-nanovesicles(IL4@SDNVs)that exhibit tripartite therapeutic efficacy:preserving blood-brain barrier integrity,suppressing neuroinflammatory cascades,specifically microglial pro-inflammatory polarization and peripheral immune cell infiltration,and inducing EMCN^(+)/CD31^(+)vascular neogenesis and achieving 144.1%increase in cerebral blood perfusion within the ischemic hemisphere in a mouse middle cerebral artery occlusion(MCAO)model.Mechanistically,post-recanalization administration of IL4@SDNV selectively promoted arginase-1^(+)(Arg1^(+))microglial polarization in the ischemic penumbra via the IL4-STAT6 signaling pathway,thereby exerting neuroprotection and cerebrovascular remodeling.Collectively,our findings demonstrate that IL4@SDNV as a promising adjuvant therapy for ischemic stroke post-recanalization through addressing cerebrovascular pathology and neuroinflammation simultaneously,thus highlighting its translational potential for clinical application. 展开更多

Fibrin-based hydrogels have been widely used in various tissue engineering because of their biocompatibility,biodegradability,tunable mechanical characteristics and nanofibrous structural properties.However,their ability to support stem cells for hair follicle neogenesis is unclear.In this study,we investigated the effect of fibrin hydrogels in supporting skin-derived precursors(SKPs)in hair follicle neogenesis.Our results showed that SKPs in fibrin hydrogels with high cell viability and proliferation,the stemness of SKPs could be maintained,and the expression of hair induction signature genes such as akp2 and nestin was enhanced.Moreover,hair follicle reconstruction experiments showed de novo hair genesis in mice and the hairs persisted for a long time without teratoma formation.More importantly,the blood vessels and sebaceous glands were also regenerated.Our study demonstrated that fibrin hydrogels are promising in hair follicle regeneration and have potential application in clinical settings for alopecia and wound healing. 展开更多
