局部晚期直肠癌患者血清NOP53 mRNA,FNDC1 mRNA表达与临床病理特征及其对新辅助放化疗疗效评估价值研究

目的探讨局部晚期直肠癌患者血清核糖体生物发生因子核仁蛋白53(NOP53)mRNA和Ⅲ型纤维连接蛋白域蛋白1(FNDC1)mRNA对新辅助放化疗(NACRT)疗效的评估价值。方法选取2020年1月~2023年1月西安交通大学第一附属医院榆林医院诊治的140例接受N... 目的探讨局部晚期直肠癌患者血清核糖体生物发生因子核仁蛋白53(NOP53)mRNA和Ⅲ型纤维连接蛋白域蛋白1(FNDC1)mRNA对新辅助放化疗(NACRT)疗效的评估价值。方法选取2020年1月~2023年1月西安交通大学第一附属医院榆林医院诊治的140例接受NACRT治疗的局部晚期直肠癌患者为研究组,根据NACRT治疗疗效,分为反应良好组(n=99)与反应不良组(n=41)。选取同期70例健康人为对照组。采用实时荧光定量PCR检测两组血清NOP53 mRNA,FNDC1 mRNA水平。Logistic回归分析影响NACRT治疗疗效的因素。受试者工作特征(ROC)曲线分析血清NOP53 mRNA,FNDC1 mRNA对直肠癌NACRT疗效的评估价值。结果研究组血清NOP53 mRNA(3.21±0.36),FNDC1 mRNA(2.73±0.34)表达高于对照组(0.61±0.17,0.72±0.18),差异具有统计学意义(t=57.267,46.287,均P<0.001)。T分期T4期、N分期N1+N2直肠癌患者血清NOP53 mRNA(4.08±0.43,4.10±0.40),FNDC1 mRNA(3.62±0.39,3.40±0.39)表达高于T分期T3期(2.52±0.30,2.02±0.29)、N分期N0患者(2.21±0.31,1.02±0.30),差异具有统计学意义(t=25.241~40.106,P均<0.001)。反应不良组T分期T4占比(73.17%)、N分期N1+N2占比(75.61%)、血清NOP53 mRNA(5.56±0.39)、FNDC1 mRNA(4.42±0.38)高于反应良好组(32.32%,43.43%,2.24±0.31,2.03±0.29),差异具有统计学意义(t/χ^(2)=12.045~53.337,均P<0.001)。T分期T4期、N分期N1+N2,血清NOP53 mRNA高表达和血清FNDC1 mRNA高表达是影响直肠癌NACRT疗效的危险因素(Waldχ^(2)=9.463~15.589,均P<0.001)。血清NOP53 mRNA,FNDC1 mRNA联合对直肠癌NACRT疗效评估的曲线下面积高于血清NOP53 mRNA,FNDC1 mRNA单项预测,差异具有统计学意义(Z=4.645,4.321,均P<0.001)。结论局部晚期直肠癌患者血清NOP53 mRNA,FNDC1 mRNA水平升高,两者与患者不良临床病理特征有关,联合血清NOP53 mRNA,FNDC1 mRNA能够有效预测直肠癌患者NACRT治疗的临床疗效。 展开更多

Cel proliferation is a fundamental event essential for plant organogenesis and contributes greatly to the final organ size. Although the control of cel proliferation in plants has been extensively studied, how the plant sets the cel number required for a single organ is largely elusive. Here, we describe the Arabidopsis SMALL ORGAN 4 (SMO4) that functions in the regulation of cell proliferation rate and thus final organ size. The smo4 mutant exhibits a reduced size of organs due to the decreased cell number, and further analysis reveals that such phenotype results from a retardation of the cell cycle progression during organ development. SMO4 encodes a homolog of NUCLEOLAR PROTEIN 53 (NOP53) in Saccharomy-ces cerevisiae and is expressed primarily in tissues undergoing cel proliferation. Nevertheless, further complementation tests&nbsp;show that SMO4 could not rescue the lethal defect of NOP53 mutant of S. cerevisiae. These results define SMO4 as an important regulator of cell proliferation during organ growth and suggest that SMO4 might have been evolutionarily divergent from NOP53. 展开更多
