Mammalian scent glands mediate species-specific chemical communication,yet the mechanistic basis for convergent musk production remain incompletely understood.Forest musk deer and muskrat have independently evolved sp...Mammalian scent glands mediate species-specific chemical communication,yet the mechanistic basis for convergent musk production remain incompletely understood.Forest musk deer and muskrat have independently evolved specialized musk-secreting glands,representing a striking case of convergent evolution.Through an integrated multi-omics approach,this study identified cyclopentadecanone as a shared key metabolic precursor in musk from both forest musk deer and muskrat,although downstream metabolite profiles diverged between the two lineages.Single-cell RNA sequencing revealed that these specialized apocrine glands possessed unique secretory architecture and exhibited transcriptional profiles associated with periodic musk production,distinct from those in conventional apocrine glands.Convergent features were evident at the cellular level,where acinar,ductal,and basal epithelial subtypes showed parallel molecular signatures across both taxa.Notably,acinar cells in both species expressed common genes involved in fatty acid and glycerolipid metabolism(e.g.,ACSBG1,HSD17B12,HACD2,and HADHA),suggesting a conserved molecular framework for musk precursor biosynthesis.Metagenomic analysis of musk samples further revealed parallel microbial community structures dominated by Corynebacterium and enriched in lipid metabolic pathways.These findings suggest multi-level convergence in musk biosynthesis,from molecular pathways to microbial communities,providing novel insights into mammalian chemical signaling and artificial musk production.展开更多
Background:The forest musk deer,a rare fauna species found in China,is famous for its musk secretion which is used in selected Traditional Chinese medicines.However,over-hunting has led to musk deer becoming an endang...Background:The forest musk deer,a rare fauna species found in China,is famous for its musk secretion which is used in selected Traditional Chinese medicines.However,over-hunting has led to musk deer becoming an endangered species,and their survival is also greatly challenged by various high incidence and high mortality respiratory and intestinal diseases such as septic pneumonia and enteritis.Accumulating evidence has demonstrated that Akkermannia muciniphila(AKK)is a promising probiotic,and we wondered whether AKK could be used as a food additive in animal breeding pro-grammes to help prevent intestinal diseases.Methods:We isolated one AKK strain from musk deer feces(AKK-D)using an im-proved enrichment medium combined with real-time PCR.After confirmation by 16S rRNA gene sequencing,a series of in vitro tests was conducted to evaluate the probiotic effects of AKK-D by assessing its reproductive capability,simulated gas-trointestinal fluid tolerance,acid and bile salt resistance,self-aggregation ability,hy-drophobicity,antibiotic sensitivity,hemolysis,harmful metabolite production,biofilm formation ability,and bacterial adhesion to gastrointestinal mucosa.Results:The AKK-D strain has a probiotic function similar to that of the standard strain in humans(AKK-H).An in vivo study found that AKK-D significantly amelio-rated symptoms in the enterotoxigenic Escherichia coli(ETEC)-induced murine diar-rhea model.AKK-D improved organ damage,inhibited inflammatory responses,and improved intestinal barrier permeability.Additionally,AKK-D promoted the reconsti-tution and maintenance of the homeostasis of gut microflora,as indicated by the fact that AKK-D-treated mice showed a decrease in Bacteroidetes and an increase in the proportion of other beneficial bacteria like Muribaculaceae,Muribaculum,and unclas-sified f_Lachnospiaceae compared with the diarrhea model mice.Conclusion:Taken together,our data show that this novel AKK-D strain might be a potential probiotic for use in musk deer breeding,although further extensive system-atic research is still needed.展开更多
Named entity recognition(NER)in musk deer domain is the extraction of specific types of entities from unstructured texts,constituting a fundamental component of the knowledge graph,Q&A system,and text summarizatio...Named entity recognition(NER)in musk deer domain is the extraction of specific types of entities from unstructured texts,constituting a fundamental component of the knowledge graph,Q&A system,and text summarization system of musk deer domain.Due to limited annotated data,diverse entity types,and the ambiguity of Chinese word boundaries in musk deer domain NER,we present a novel NER model,CAELF-GP,which is based on cross-attention mechanism enhanced lexical features(CAELF).Specifically,we employ BERT as a character encoder and advocate the integration of external lexical information at the character representation layer.In the feature fusion module,instead of indiscriminately merging external dictionary information,we innovatively adopted a feature fusion method based on a cross-attention mechanism,which guides the model to focus on important lexical information by calculating the correlation between each character and its corresponding word sets.This module enhances the model’s semantic representation ability and entity boundary recognition capability.Ultimately,we introduce the decoding module of GlobalPointer(GP)for entity type recognition,capable of identifying both nested and non-nested entities.Since there is currently no publicly available dataset for the musk deer domain,we built a named entity recognition dataset for this domain by collecting relevant literature and working under the guidance of domain experts.The dataset facilitates the training and validation of the model and provides data foundation for subsequent related research.The model undergoes experimentation on two public datasets and the dataset of musk deer domain.The results show that it is superior to the baseline models,offering a promising technical avenue for the intelligent recognition of named entities in the musk deer domain.展开更多

Chinese forest musk deer(FMD),an endangered species,have exhibited low reproductive rates even in captivity due to stress conditions.Investigation revealed the presence of di(2-ethylhexyl)phthalate(DEHP),an environmen...Chinese forest musk deer(FMD),an endangered species,have exhibited low reproductive rates even in captivity due to stress conditions.Investigation revealed the presence of di(2-ethylhexyl)phthalate(DEHP),an environmental endocrine disruptor,in the serum and skin of captive FMDs.Feeding FMDs with maslinic acid(MA)has been observed to alleviate the stress response and improve reproductive rates,although the precise molecular mechanisms remain unclear.Therefore,this study aims to investigate the molecular mechanisms underlying the alleviation of DEHP-induced oxidative stress and cell apoptosis in primary peritubular myoid cells(PMCs)through MA intake.Primary PMCs were isolated and exposed to DEHP in vitro.The results demonstrated that DEHP significantly suppressed antioxidant levels and promoted cell apoptosis in primary PMCs.Moreover,interfering with the expression of PRDX6 was found to induce excessive reactive oxygen species(ROS)production and cell apoptosis in primary PMCs.Supplementation with MA significantly upregulated the expression of PRDX6,thereby attenuating DEHP-induced oxidative stress and cell apoptosis in primary PMCs.These findings provide a theoretical foundation for mitigating stress levels and enhancing reproductive capacity of in captive FMDs.展开更多

These solid-liquid equilibrium data and the heats of fusion are reported for the first time. Then UNIFAC model is used to correlate the sofid-liquid equilibrium data. Background The musk glands of adult male Chinese forest musk deer(Moschus berezovskii Flerov,1929)(FMD),which are considered as special skin glands,secrete a mixture of sebum,lipids,and proteins into the musk pod.Toge...Background The musk glands of adult male Chinese forest musk deer(Moschus berezovskii Flerov,1929)(FMD),which are considered as special skin glands,secrete a mixture of sebum,lipids,and proteins into the musk pod.Together,these components form musk,which plays an important role in attracting females during the breeding season.However,the relationship between the musk glands and skin of Chinese FMD remains undiscovered.Here,the musk gland and skin of Chinese FMD were examined using histological analysis and RNA sequencing(RNA-seq),and the expression of key regulatory genes was evaluated to determine whether the musk gland is derived from the skin.Methods A comparative analysis of musk gland anatomy between juvenile and adult Chinese FMD was conducted.Then,based on the anatomical structure of the musk gland,skin tissues from the abdomen and back as well as musk gland tissues were obtained from three juvenile FMD.These tissues were used for RNA-seq,hematoxylin-eosin(HE)staining,immunohistochemistry(IHC),western blot(WB),and quantitative real-time polymerase chain reaction(qRT-PCR)experiments.Results Anatomical analysis showed that only adult male FMD had a complete glandular organ and musk pod,while juvenile FMD did not have any well-developed musk pods.Transcriptomic data revealed that 88.24%of genes were co-expressed in the skin and musk gland tissues.Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway analysis found that the genes co-expressed in the abdomen skin,back skin,and musk gland were enriched in biological development,endocrine system,lipid metabolism,and other pathways.Gene Ontology(GO)enrichment analysis indicated that the genes expressed in these tissues were enriched in biological processes such as multicellular development and cell division.Moreover,the Metascape predictive analysis tool demonstrated that genes expressed in musk glands were skin tissue-specific.qRT-PCR and WB revealed that sex-determining region Y-box protein 9(Sox9),Caveolin-1(Cav-1),and androgen receptor(AR)were expressed in all three tissues,although the expression levels differed among the tissues.According to the IHC results,Sox9 and AR were expressed in the nuclei of sebaceous gland,hair follicle,and musk gland cells,whereas Cav-1 was expressed in the cell membrane.Conclusions The musk gland of Chinese FMD may be a derivative of skin tissue,and Sox9,Cav-1,and AR may play significant roles in musk gland development.展开更多

The two representative polycyclic musks, 1-(5,6,7,8-tetrahydro-3,5,5,6,8,8-hexamethyl-2-naphthenyl)-ethanone (AHTN) and 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[g]-2-benzopyrane (HHCB), were measur...The two representative polycyclic musks, 1-(5,6,7,8-tetrahydro-3,5,5,6,8,8-hexamethyl-2-naphthenyl)-ethanone (AHTN) and 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[g]-2-benzopyrane (HHCB), were measured in aqueous samples and sludge samples of three sewage treatment plants (STPs) in Beijing, China using a gas chromatograph-mass spectrometry. The HHCB and AHTN concentration ranges in the influents were 1251.4-3003.8 ng/L and 111.9-286.3 ng/L, respectively. Meanwhile, the ranges of 492.8-1285.3 ng/L for HHCB and 47.3-89.3 ng/L for AHTN were present in the effluents. The musks in the sludges were three to four orders of magnitude higher than those in aqueous sewages. The removal efficiencies.of the two musks varied in the ranges of 41.7%- 70.1% for HHCB, and 25.5%-68.8% for AHTN. Adsorption onto suspended particulate matter played an important role in removing musks from the sewages. The HHCB/AHTN ratio along the treatment processes showed that the two musks had high similarity of removal from sewage by each reactor in STP. The musks in the effluent sewages may pose a low potential risk to aquatic environment in terms of the predicted-non-effect concentration. Nevertheless, considering the possible additive and synergistic effects with other compounds emitted via STPs, their bioconcentration and bioaccumulation in aquatic organisms and so on, it is essential to monitor these compounds in various compartments and to study their environmental fate.展开更多
Synthetic musks are used as additives in many household products.After absorption into the human body,they accumulate and their concentrations in human milk reflect both the mother and her infant's exposure level.Con...Synthetic musks are used as additives in many household products.After absorption into the human body,they accumulate and their concentrations in human milk reflect both the mother and her infant's exposure level.Concentrations of four synthetic musks,musk xylene(1-tert-butyl-2,6-dimethyl-2,4,6-trinitrobenzene,MX),musk ketone(4-tert-butyl-2,6-dimethyl-3,5-dinitroacetophenone,MK),1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[γ]-2-benzopyran(HHCB) and 7-acetyl-1,1,3,4,4,6-hexamethyl-1,2,3,4tetrahydronaphthalene(AHTN),were determined in human milk samples collected from Shanghai,Wuxi,and Shaoxing in Eastern China.The four synthetic musks were found in most samples analyzed,with HHCB the dominant component followed by MX.The median(mean) values for HHCB,AHTN,MX and MK concentrations were 63(82),5(12),17(24) and 4(9) ng/g lipid weight,respectively.These data suggested the total synthetic musk contamination was low,and the distribution percentage was HHCB 〉 MX 〉 AHTN ≈ MK.The relative high ratio of nitro to polycylic musk indicated that nitro musks were still widely used.The musk concentrations in these cities were not significantly different from each other(p 〉 0.05).Principal components score plots were obtained,which showed similar exposure sources.The amount of total synthetic musks in human milk were not associated with mother's age,although HHCB was significantly correlated with AHTN(p 〈 0.05).Daily ingestion of HHCB,AHTN,MX and MK for infants from human milk were estimated as(2526 ± 2926),(370 ± 524),(7391 ± 832),and(277 ± 462) ng/day,respectively.Those doses were 1-2 orders of magnitude below the provisional tolerable daily intakes.展开更多

文摘Chinese forest musk deer(FMD),an endangered species,have exhibited low reproductive rates even in captivity due to stress conditions.Investigation revealed the presence of di(2-ethylhexyl)phthalate(DEHP),an environmental endocrine disruptor,in the serum and skin of captive FMDs.Feeding FMDs with maslinic acid(MA)has been observed to alleviate the stress response and improve reproductive rates,although the precise molecular mechanisms remain unclear.Therefore,this study aims to investigate the molecular mechanisms underlying the alleviation of DEHP-induced oxidative stress and cell apoptosis in primary peritubular myoid cells(PMCs)through MA intake.Primary PMCs were isolated and exposed to DEHP in vitro.The results demonstrated that DEHP significantly suppressed antioxidant levels and promoted cell apoptosis in primary PMCs.Moreover,interfering with the expression of PRDX6 was found to induce excessive reactive oxygen species(ROS)production and cell apoptosis in primary PMCs.Supplementation with MA significantly upregulated the expression of PRDX6,thereby attenuating DEHP-induced oxidative stress and cell apoptosis in primary PMCs.These findings provide a theoretical foundation for mitigating stress levels and enhancing reproductive capacity of in captive FMDs.

文摘Anhui musk deer [ Moschus ( moschiferus/berezovskii ) anhuiensis ] has been a taxonomic mystery since its discovery in early 80's.In this paper,with museum samples,we sequenced the complete cytochrome b gene of five Anhui musk deer.When compared with other species in Genus Moschus ,Anhui musk deer showed a rather level of sequence divergence from all the other species in this genus.The phylogenetic trees constructed by multiple methods supported the same topology,in which the monophyly of Anhui musk deer was clearly demonstrated.Therefore,our molecular data suggest a full species status for Anhui musk deer ( Moschus anhuiensis ),rather than a subspecies of either M moschiferus or M berezovskii previously suggested by morphological studies.

文摘[Objective] The pathogenic Escherichia coli in musk deer was classified at molecular level to provide basic materials for molecular epidemiology of pathogenic Escherichia coli in musk deer. [Method] Plasmids from 24 pathogenic Escherichia coli in musk deer were extracted by the Lysis Triton method, and then identified by single enzyme digestion with three endonucleases of Hind Ⅲ, EcoR Ⅰ and BamH Ⅰ. [Result] The yield rate of plasmids was 91.6%, and 24 pathogenic Escherichia coli in musk deer had the identical or similar plasmid profiles. [Conclusion] Plasmid DNA analysis offers scientific basis for molecular epidemiology of pathogenic Escherichia coli in musk deer in Sichuan Institute of Musk Deer Breeding.

文摘A multivalent inactivated Escherichia coli vaccine for forest musk deer by using serotypes O4,O26,and O139 with Al(OH)3 adjuvant was prepared.The vaccine did not cause any adverse reactions in forest musk deer.The immunogenic effects of the vaccine were experimentally investigated in pregnant and young forest musk deer.The serum antibody titers of pregnant and young forest musk deer were determined by performing the micro-agglutination test.The serum antibody titers of pregnant forest musk deer were more stable from 35th to 68th d after the third vaccination,and the serum antibody titers of four pregnant forest musk deer were maintained 25,25,25,and 24 on 68th d after the third vaccination.Young forest musk deer showed serum antibody titers which were obtained due to nursing.Young forest musk deer were administered the first intramuscular vaccine injection at an age of approximately 60 days due to a fall in maternal antibody titers.The serum antibody titers of young forest musk deer were higher after the third vaccination and maintained at approximately the same level until they were 137 days old.The maternal antibodies and the antibodies produced by young forest musk deer could be helpful for protecting the young musk deer from the infections of pathogenic Escherichia coli strains(serotypes O4,O26,and O139)for 137 days after birth(during the nursing period and the period when the forest musk deer were susceptible to diseases).
文摘The solid-liquid equilibria of musk ketone + musk xylene, musk xylene +l,3-dimethyl-2,4-dinitro-5-tert-butyl benzene are measured by differential scanning calorimeter (DSC), these systems are proved to be simple eutectics. Moreover the melting points and the fusion enthalpies of musk ketone, musk xylene and 1,3-dimethyl-2,4-dinitro-5-tert-butyl benzene are also measured by the DSC. These solid-liquid equilibrium data and the heats of fusion are reported for the first time. Then UNIFAC model is used to correlate the sofid-liquid equilibrium data. It is shown that the solid-liquid equilibria of musk systems can be predicted by the UNIFAC model.

文摘Background The musk glands of adult male Chinese forest musk deer(Moschus berezovskii Flerov,1929)(FMD),which are considered as special skin glands,secrete a mixture of sebum,lipids,and proteins into the musk pod.Together,these components form musk,which plays an important role in attracting females during the breeding season.However,the relationship between the musk glands and skin of Chinese FMD remains undiscovered.Here,the musk gland and skin of Chinese FMD were examined using histological analysis and RNA sequencing(RNA-seq),and the expression of key regulatory genes was evaluated to determine whether the musk gland is derived from the skin.Methods A comparative analysis of musk gland anatomy between juvenile and adult Chinese FMD was conducted.Then,based on the anatomical structure of the musk gland,skin tissues from the abdomen and back as well as musk gland tissues were obtained from three juvenile FMD.These tissues were used for RNA-seq,hematoxylin-eosin(HE)staining,immunohistochemistry(IHC),western blot(WB),and quantitative real-time polymerase chain reaction(qRT-PCR)experiments.Results Anatomical analysis showed that only adult male FMD had a complete glandular organ and musk pod,while juvenile FMD did not have any well-developed musk pods.Transcriptomic data revealed that 88.24%of genes were co-expressed in the skin and musk gland tissues.Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway analysis found that the genes co-expressed in the abdomen skin,back skin,and musk gland were enriched in biological development,endocrine system,lipid metabolism,and other pathways.Gene Ontology(GO)enrichment analysis indicated that the genes expressed in these tissues were enriched in biological processes such as multicellular development and cell division.Moreover,the Metascape predictive analysis tool demonstrated that genes expressed in musk glands were skin tissue-specific.qRT-PCR and WB revealed that sex-determining region Y-box protein 9(Sox9),Caveolin-1(Cav-1),and androgen receptor(AR)were expressed in all three tissues,although the expression levels differed among the tissues.According to the IHC results,Sox9 and AR were expressed in the nuclei of sebaceous gland,hair follicle,and musk gland cells,whereas Cav-1 was expressed in the cell membrane.Conclusions The musk gland of Chinese FMD may be a derivative of skin tissue,and Sox9,Cav-1,and AR may play significant roles in musk gland development.
文摘The ternary phase diagrams of musk ketone, musk xylene and various solvent were obtained by calcu- lations. Moreover, the double saturated lines were found to be curve for the first time. The methods to separate eutectic mixtures of musks by solution crystallization were suggested on the basis of these phase diagrams. A new process was proposed to get musk ketone and musk xylene from the eutectics by adding acetonitrile and heptane in turn, which was verified by experiments. Musk xylene with purity of 97.50% and musk ketone with purity of 98.10% were obtained.
文摘目的探讨骨骼肌特异性酪氨酸激酶(muscle-specific tyrosine kinase,MuSK)抗体阳性重症肌无力(MuSK-myasthenia gravis,MMG)临床特点和对治疗的反应。方法分析2017年7月至2018年12月就诊于我院的MG患者的临床资料,计算MMG占全部MG患者比例,并对MMG患者的起病年龄,起病方式,临床进展模式和对治疗的反应进行分析,并随访至少6个月,每3个月电话或面对面随访记录患者日常生活能力评分(myasthenia gravis-activity of daily living score,MG-ADL)。结果401例MG患者中,完成MuSK检测319例,MuSK抗体阳性率为3.13%(10/319)。10例MMG患者男女比为1:9,起病年龄17耀50岁,平均31.4岁。90%患者以眼外肌无力为首发症状。1年内进展为全身型的比例高达90%。溴吡斯的明有效率90%,重频刺激低频递减阳性率为77.78%,无胸腺异常者。经过至少6个月的随访,大多数患者ADL评分降至0耀1分。结论眼动障碍和球部肌无力为这组患者突出的临床表现,免疫治疗有效。

文摘The two representative polycyclic musks, 1-(5,6,7,8-tetrahydro-3,5,5,6,8,8-hexamethyl-2-naphthenyl)-ethanone (AHTN) and 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[g]-2-benzopyrane (HHCB), were measured in aqueous samples and sludge samples of three sewage treatment plants (STPs) in Beijing, China using a gas chromatograph-mass spectrometry. The HHCB and AHTN concentration ranges in the influents were 1251.4-3003.8 ng/L and 111.9-286.3 ng/L, respectively. Meanwhile, the ranges of 492.8-1285.3 ng/L for HHCB and 47.3-89.3 ng/L for AHTN were present in the effluents. The musks in the sludges were three to four orders of magnitude higher than those in aqueous sewages. The removal efficiencies.of the two musks varied in the ranges of 41.7%- 70.1% for HHCB, and 25.5%-68.8% for AHTN. Adsorption onto suspended particulate matter played an important role in removing musks from the sewages. The HHCB/AHTN ratio along the treatment processes showed that the two musks had high similarity of removal from sewage by each reactor in STP. The musks in the effluent sewages may pose a low potential risk to aquatic environment in terms of the predicted-non-effect concentration. Nevertheless, considering the possible additive and synergistic effects with other compounds emitted via STPs, their bioconcentration and bioaccumulation in aquatic organisms and so on, it is essential to monitor these compounds in various compartments and to study their environmental fate.

文摘Synthetic musks are used as additives in many household products.After absorption into the human body,they accumulate and their concentrations in human milk reflect both the mother and her infant's exposure level.Concentrations of four synthetic musks,musk xylene(1-tert-butyl-2,6-dimethyl-2,4,6-trinitrobenzene,MX),musk ketone(4-tert-butyl-2,6-dimethyl-3,5-dinitroacetophenone,MK),1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[γ]-2-benzopyran(HHCB) and 7-acetyl-1,1,3,4,4,6-hexamethyl-1,2,3,4tetrahydronaphthalene(AHTN),were determined in human milk samples collected from Shanghai,Wuxi,and Shaoxing in Eastern China.The four synthetic musks were found in most samples analyzed,with HHCB the dominant component followed by MX.The median(mean) values for HHCB,AHTN,MX and MK concentrations were 63(82),5(12),17(24) and 4(9) ng/g lipid weight,respectively.These data suggested the total synthetic musk contamination was low,and the distribution percentage was HHCB 〉 MX 〉 AHTN ≈ MK.The relative high ratio of nitro to polycylic musk indicated that nitro musks were still widely used.The musk concentrations in these cities were not significantly different from each other(p 〉 0.05).Principal components score plots were obtained,which showed similar exposure sources.The amount of total synthetic musks in human milk were not associated with mother's age,although HHCB was significantly correlated with AHTN(p 〈 0.05).Daily ingestion of HHCB,AHTN,MX and MK for infants from human milk were estimated as(2526 ± 2926),(370 ± 524),(7391 ± 832),and(277 ± 462) ng/day,respectively.Those doses were 1-2 orders of magnitude below the provisional tolerable daily intakes.
