Moringa oleifera seed essential oil was extracted by solvent-free microwave and hydrodistillation. This study assessed its chemical constituents. Cytotoxicity of the oils was investigated using hatchability and lethality tests on brine shrimps. A total of 16 and 26 compounds were isolated from the hydrodistillation extraction (HDE) and solvent-free microwave extraction (SME) oils, respectively, which accounted for 97.515% and 97.816% of total identifiable constituents, respectively. At 24 h when the most eggs had hatched, values of the SME (56.7%) and HDIE (60.0%) oils were significantly different (P〈0.05) from those of sea water (63.3%) and chloramphenicol (15.0%). Larva lethality was different significantly (P〈0.05) between HDE and SME oils at different concentrations and incubation periods. The median lethal concentration (LC50) of the oils was 〉 1000 mg/ml recommended as an index for non-toxicity which gives the oil advantage over some antioxidant, antimicrobial, therapeutic, and preservative chemicals.展开更多

At the end of treatment, body weight was determined, and the atherogenic index, coronary artery index, glucose level, insulin resistance status, liver and kidney functions were assessed. Also, the m RNA of leptin, adiponectin and resistin in visceral adipose tissue was determined by quantitative real time-PCR.Results: The results showed that M. oleifera extract down-regulated m RNA expression of leptin and resistin, while it up-regulated adiponectin gene expression in obese rats relative to untreated obese control counterparts. This amelioration of genes expression was paralleled by a reduction in body weight and improvement of the atherogenic index and coronary artery index, as well as glucose level and insulin resistance value without adverse effects on liver or kidney functions, versus the untreated obese control ones. Conclusions: It is reasonable to assume that the anti-obesity, anti-atherogenic and antidiabetic properties of M. oleifera are mechanistically achieved via working directly on the adipokines of the visceral adipose tissue. Therefore, M. oleifera may be a good therapeutic candidate for the symptoms of metabolic syndrome.展开更多

Due to the effects of oxidative stress and dopamine on male sexual function, we hypothesized that Monnga oleifera leaves might improve male sexual dysfunction induced by stress. Therefore, the effects on various factors playing important roles in male sexual behavior, such as antioxidant effects, the suppression of monoamine and phosphodiesterase type 5 (PDE-5) activities, serum testosterone and corticosterone levels, and histomorphological changes in the testes, of a hy- droethanolic extract of M. oleifera leaves were investigated. Various doses of extract including 10, 50, and 250 mg/kg body weight (BW) were given orally to male Wistar rats before exposure to 12 h-immobilization stress for 7 d. The results demonstrated that the extract showed both antioxidant and monoamine oxidase type B (MAO-B) suppression activities. At 7 d of treatment, the low dose of extract improved sexual performance in stress-exposed rats by de- creasing intromission latency and increasing intromission frequency. It also suppressed PDE-5 activity, decreased serum corticosterone level, but increased serum testosterone, numbers of interstitial cells of Leydig and spermatozoa. The increased numbers of interstitial cells of Leydig and spermatozoa might have been due to the antioxidant effect of the extract. The increased sexual performance during the intromission phase might have been due to the suppression of MAO-B and PDE-5 activities and increased testosterone. Therefore, M. oleifera is a potential aphrodisiac, but further research concerning the precise underlying mechanisms is still needed.展开更多

Methods: These fractions were prepared from the M. oleifera leaves methanol extract by solubilization in water and partitioning in n-hexane, EtOAc, DCM and n-butanol. Their phyto-components were identified by GC-MS analysis. The in vitro antioxidant effect of these fractions was carried out by assessment of 1,1-diphenyl-2-picrylhydrazyl scavenging activity. A total of 40 Sprague Dawley rats were allocated into 8 equal groups: group 1 given olive oil(1 m L/kg b.wt.), group 2 injected with CCl_4, group 3 to 7 administered with n-hexane, DCM, EtOAc, n-butanol and aqueous fractions, respectively after CCl_4, group 8 administered with silymarin after CCl_4. The activities of aspartate aminotransferase, alanine aminotransferase, and the levels of total cholesterol, triglycerides, glucose, total proteins and albumin in serum were determined spectrophotometrically. Glutathione reduced, lipid peroxide by-products levels, glutathione-s-transferase and catalase enzyme activities in the liver homogenate were determined by spectrophotometer. Liver specimens were also examined for histopathological alterations under light microscope. Results: The GCMS analysis of different fractions of the M. oleifera leaves methanol extract revealed that n-hexane, DCM, EtOAc, n-butanol, and aqueous fractions contained 17, 22, 23, 19 and 32 compounds, respectively. The percent and the molecular structure of each component in each fraction were identified. The n-butanol and EtOAc fractions exhibited the strongest in vitro antioxidant activity against 1,1-diphenyl-2-picrylhydrazyl. CCl_4 significantly decreased glutathione reduced and total proteins concentration and glutathione-s-transferase and catalase activities but increased lipid peroxide by-products and total cholesterol levels. The n-hexane followed by aqueous and DCM fractions were the most potent to regulate serum enzyme activities and lipid peroxide by-products levels in the liver homogenate. Conclusions: n-hexane, DCM, and aqueous fractions have the highest effectiveness against CCl_4-induced hepatotoxicity. Isolation and purification of the active constituents require further experiments.展开更多

M. oleifera seeds were ground to fine powders and oil was extracted by microwave-assisted and soxhlet extraction techniques using petroleum ether. Quality attributes including yield percent, moisture content, iodine, saponification, specific gravity, viscosity, p H, thiobarbituric acid, acid and peroxide values were measured. Mineral and vitamin contents, chemical/functional groups, fatty acid(FA) composition, and reducing power of the oil were evaluated. Metabolomics of protein extracted from the defatted powders were analyzed by nuclear magnetic resonance(NMR). M. oleifera oil from MAE and SE methods had good yield(34.25 ± 0.0%,28.75 ± 0.0%), low moisture content(0.008 ± 0.0%, 0.011 ± 0.0%), non-drying and unsaturated, moderately saponified, less dense(0.91 ± 0.01, 0.92 ± 0.02 g m L^(-1)), had Newtonian flow, were weakly acidic, showed good content of FAs, recorded strong potential for long shelf-life, showed stability against oxidative rancidity and enzymatic hydrolysis, had very rich deposits of micro-and macro-nutrients as well as water-soluble and lipidsoluble vitamins, and functional groups in the oil were reflective of its content of long-and medium-chain triglycerides(LCT and MCT). Monounsaturated and saturated fatty acids(MUFA and SFA) were detected and the oil has excellent ferric ion reducing power. NMR metabolomic assay revealed the presence of nine essential amino acids(EAAs) in the protein extract. MAE technique is a feasible and acceptable alternative for high throughput extraction of M. oleifera oil with high yield and excellent quality attributes. The study revealed that MAE did not impart any remarkable advantage(s) on the physicochemical properties of M. oleifera seed oil and protein compared to SE technique.展开更多

The birds were assigned randomly into four dietary treatments containing MOLM at 0, 6%, 8%, and 10% (treatments 1, 2, 3 and 4) inclusion levels, respectively, in a complete randomized design experiment. The effect of the dietary treatments on the growth performance and blood parameters of the broiler finisher was determined. MOLM at a rate of 6% (T2), 8% (T3), and 10% (T4) of the diets (as fed basis) to replace 3.2%, 5.6% and 8.6% of the crude protein (CP) of the control diet. The daily feed, dry matter and CP intake of the chicks fed MOLM diets were higher (p〈0.05) than those fed the control diet. Average weight gain (AWG) of birds fed MOLM diets reduced as inclusion levels increases 09〈0.05) than those fed the control diet. Chicks fed T2 showed higher (p〈0.05) AWG than those on T3 and T4. Feed conversion ratios (FCR, weight gain/g and feed intake) were higher for chicks fed MOLM. MOLM showed minimal deleterious effects in birds. However, birds fed T2 (6%) diets recorded significantly (p〈0.05) the highest body weight gain. There were significant differences (p〈0.05) among groups in packed cell volume (PVC) of birds. The haemoglobin (Hb), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC) counts showed no significant difference (p〉0.05) among treatments. The results indicated that at 10% in the diets of broiler finisher chick, MOLM could be substituted with expensive conventional protein sources without any deleterious effects on performance and blood parameters of broiler finisher chicken.展开更多

Considering the current recommended approach for the use of functional foods and their bioactive components in the prevention and management of diabetes, the aim of this study was to determine the antihyperglycemic effect of Moringa oleifera functional tea in rat models and in normoglycemic human volunteers using the oral glucose tolerance test (OGTT). Moringa tea prepared by extracting Moringa leaf powder in distilled water (1:20 mg/ml) at 97°C for 30 min was administered at different doses to male Wistar rats and human volunteers prior to glucose loading. Blood glucose was measured at intervals of 30 min for 150 min. Consumption of Moringa tea prior to glucose loading suppressed the elevation in blood glucose in all cases compared to controls that did not receive the tea initially. The degree and pattern of decrease however, were dose dependent. In rats, intermediate doses of 20 ml/kg BW were more effective in reducing blood glucose overall (18.2%) vs 13.3% and 6% at doses of 10 and 30 ml/kg BW respectively. In humans, the final decrease in blood glucose was not significantly different for high 400 ml (19%) and low 200 ml (17%) doses. Of interest here was the pattern of decrease, being significantly higher (p < 0.05) at 30 min with 200 ml (22.8%) than with 400 ml (17.9%). It is suggested that low doses exert their antihyperglycemic effect more at intestinal level by inhibiting glucose absorption, whereas high doses exert their effect more in circulation. We conclude that Moringa oleifera tea has potential as a functional food in the management of hyperglycemia.展开更多

[Method] Tissue culture techniques and large scale micropropagation of M. oleifera were studied in this paper. By means of a series of experiments, we used the leaf of aseptic seedling from M. oleifera as explants to optimize and establish the regeneration system cultured in vitro by means of direct organogenesis. [Result] It was observed that using the fresh shelled M. oleifera seeds with 0.1% mercuric chloride for 6 minutes could reach the best disinfection effect. The seed germination rate was 85%. The leaf could produce cluster buds well using the medium with MS+2.0 mg/L 6-BA+0.05 mg/L NAA, while the best proliferation condition was under MS+6-BA 1.0 mg/L+KT 0.1 rag/L+2, 4-D 2.0 mg/L+NAA 0.05 mg/L. The best rooting induction culture medium was MS+0.5 mg/L IBA, with the rooting rate as 100%. [Cenclusien] This protocol might find use in mass production of true- to-type plants and in production of transgenic plants through Agrobacterium/biolisticmediated transformation.展开更多

文摘Obesity has brought great challenges to global human health, and how to effectively prevent and control the occurrence and development of obesity has become an urgent problem. The role and mechanism of 4-[(α-Lrhamnosyloxy)benzyl]isothiocyanate(MITC),an active ingredient of Moringa oleifera Lam.,in the regulation of lipid metabolism have not been comprehensively investigated. In the present study,we investigated the mechanism of MITC in inhibiting lipid accumulation in mice fed with a high-fat diet(HFD)in terms of both lipolysis and central appetite regulation mediated by the gut microbe-gut-brain axis.MITC enhanced the characteristic indices associated with HFD mice and also promoted adipocytolysis and brown fat thermogenesis. Moreover,MITC was observed to improve leptin resistance, modulate the composition of gut microbiota such as Ruminococcaceae,Parasutterella, and Acetatifactor, promote 5-hydroxytryptamine secretion, further enhance the secretion of glucagon-like peptide-1(GLP-1)and peptide tyrosine-tyrosine(PYY)to activate peroxisome proliferator-activated receptor(PPAR)signaling in the hypothalamus, and modulate feeding behavior to inhibit lipid accumulation in HFD mice. These data suggest that MITC supplementation can help to alleviate obesity or obesity-related diseases.
文摘Moringa oleifera have laxative effects, but their active compositions and mechanisms are not very clear thus far.To this end,we systematically explored the active components and mechanism of M.oleifera leaves in relieving constipation by using the slow transit constipation(STC)mouse model and network pharmacology. The results of animal experiments showed that M.oleifera aqueous extract(MOA)had good laxative activity, and its 70%alcohol soluble part(ASP)also showed significant laxative activity(P<0.01).Network pharmacological prediction results suggested that L-phenylalanine(Phe)was the key compound of ASP,and it might relieve constipation through tachykinin receptor 1(TACR1)and three kinds of adrenergic receptors,includingα_(1A)(ADRA1A),α_(2A)(ADRA2A),andα_(2B)(ADRA2B).Further animal experiment results showed that Phe significantly promoted gastrointestinal motility.Phe may relieve STC by enhancing the release of substance P(SP)and upregulating the m RNA expression of TACR1 in the ileum.Importantly,Phe may also promote intestinal movement by downregulating the m RNA expression of ADRA2A and ADRA2B and upregulating the m RNA expression of Calm and the m RNA and protein expression of myosin light chain 9 in the ileum, thereby activating the G protein-coupled receptor-myosin light chain signaling pathway. These results lay a foundation for the application of M.oleifera and Phe in constipation.

文摘The extensive use of polymeric materials in single-use packaging has driven the need to develop biodegradable alternatives. This study investigates the incorporation of graphene oxide(GO)and Moringa oleifera seed oil(MOSO)into a gelatin matrix to create polymer films and evaluate their potential as active packaging materials. The properties of these films were evaluated using structural, thermal, mechanical, optical, and physicochemical methods to determine their suitability for food packaging applications. The results showed that GO and MOSO were homogeneously dispersed in the gelatin matrix, forming colloidal particles(around 5μm in diameter). The addition of GO increased opacity by approximately 20 times the base value while MOSO affected light transmittance without impacting opacity.Mechanical properties were affected differently,GO acted as a crosslinking agent reducing elongation and increasing tensile strength at break,on the other hand MOSO acted as a plasticizer, making films more plastic increasing elongation a 30%.These effects counteracted each other, and similar behavior was recorded in differential scanning calorimetry. The films exhibited an improved water vapor resistance, which is crucial for food packaging. These findings indicate that the incorporation of GO and MOSO into a gelatin matrix may produce biodegradable polymer films with enhanced properties, suitable for active packaging in the food industry.

文摘Renewed interest in natural materials as food flavors and preservatives has led to the search for suitable essential oils. The median lethal concentration (LC50) of the oils was 〉 1000 mg/ml recommended as an index for non-toxicity which gives the oil advantage over some antioxidant, antimicrobial, therapeutic, and preservative chemicals.

文摘Objective: To elucidate the molecular mechanisms of the potent anti-obesity effect of Moringa oleifera Lam.(M. oleifera) ethanolic extract and to clarify the link between these mechanisms and the associated metabolic and vascular risks in the experimental model of visceral obesity. Methods: M. oleifera ethanolic extract was orally administered at 600 mg/kg body weight in obese female rats daily for 12 weeks. Therefore, M. oleifera may be a good therapeutic candidate for the symptoms of metabolic syndrome.

文摘Aphrodisiacs are required to improve male sexual function under stressful conditions. Therefore, M. oleifera is a potential aphrodisiac, but further research concerning the precise underlying mechanisms is still needed.
文摘Objective: To evaluate the potential antioxidant and hepatoprotective effects of n-hexane, dichloromethane(DCM), ethyl acetate(EtOAc), n-butanol and aqueous fractions of Moringa oleifera(M. oleifera) leaves methanol extract against carbon tetrachloride(CCl_4)-induced liver injury in rats. Isolation and purification of the active constituents require further experiments.

文摘There is a constant search for biomaterials from natural products like plants for food and industrial applications. The work embodied in this report aimed at investigating the effects of microwave-assisted and soxhlet extraction(MAE and SE) techniques on the functional physicochemical quality characteristics of Moringa oleifera seed oil and proteins extracts. M. oleifera seeds were ground to fine powders and oil was extracted by microwave-assisted and soxhlet extraction techniques using petroleum ether. The study revealed that MAE did not impart any remarkable advantage(s) on the physicochemical properties of M. oleifera seed oil and protein compared to SE technique.

文摘To investigate the effects ofMoringa oleifera leaf meal (MOLM) on the performance and haematology of broiler finisher using 120 Cobb chicks of 4 weeks old broiler chicks. The results indicated that at 10% in the diets of broiler finisher chick, MOLM could be substituted with expensive conventional protein sources without any deleterious effects on performance and blood parameters of broiler finisher chicken.
文摘Maintenance of glycemic control is important in preventing diabetes and its associated complications. We conclude that Moringa oleifera tea has potential as a functional food in the management of hyperglycemia.

文摘[Objective] The aim was to optimized and established the regeneration system of Moringa oleifera and provided foundation for its rapid propagation and further research. [Cenclusien] This protocol might find use in mass production of true- to-type plants and in production of transgenic plants through Agrobacterium/biolisticmediated transformation.

文摘Carbon tetrachloride(CCl4)is a hepatotoxin that triggers liver damage. This study aimed to evaluate the protective effect of phytochemicals detected in Moringa oleifera Lam.leaf extract(MOLE)on CCl4-induced hepatotoxicity in mice.Phytochemicals, total phenolics, and total fl avonoids were detected in MOLE.MOLE markedly decreased the elevation of serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in consistence with the ameliorating effect on CCl4-induced histopathological abnormalities. Moreover,MOLE significantly alleviated the decrease in the antioxidant defense mechanism induced by CCl4.The suppressing effect of MOLE on the boosted inflammatory pathway triggered by CCl4 was detected by measuring the protein levels of nuclear factor kappa-light-chain-enhancer of activated B-cells(NF-κB-p65),toll-like receptor 4(TLR4),tumor necrosis factor-α(TNF-α),interleukin(IL)-6,IL-1β,and IL-8 as well as the relative expressions of nuclear factor kappa B(NF-κB),TNF-α,IL-1β,and TLR4 genes.Apoptosis and genotoxicity induced by CCl4 were signifi cantly alleviated by MOLE.MOLE co-administration modulated TLR4/NF-κB pathway as presented by the suppressed gene expression of TLR4 and NF-κB as well as by the reduced protein expression of TLR4 and NF-κB-p65.In conclusion,MOLE has a multifarious protective role against hepatotoxicity through control of oxidative stress and modulation of TLR4/NF-κB.
