Aim To investigate the chemical composition of Ranunculus sieboldii Miq..Methods Repeated column chromatography over silica gel, polyamide and RP-18 followed by gelfiltration on sephadex LH-20 were used to isolate chemical constituents, and their structures wereelucidated by extensive spectroscopic methods (UV, IR, MS, ~1H NMR, ^(13)C NMR) including 2D NMR(COSY, HMQC, HMBC, NOESY) techniques and by direct comparing spectral data with those reported inliterature. Results Five flavonoid glycosides named apigenin-4'-O-α-L-rhamnopyranoside (1),apigenin-7-O-β-D-glucopyranosyl-4'-O-α-L-rhamnopyranoside (2), apigenin-8-C-α-L-arabinopyranoside(3), apigenin-8-C-β-D-ga-lactopyranoside (4) , tricin-7-O-β-D-glucopyranoside (5), together withtricin (6), luteolin (7), scopoletin (8), esculetin (9), scoparone (10), ferulic acid (11),protocatechuic acid (12) , and tematolide (13) were isolated from the 95% etha-nolic extract of itswhole plant, and their cytotoxic activities were preliminarily tested. Conclusion Compounds 1-12were obtained from this genus and compound 13 from this species for the first time. Furthermore,compound 1 was for the first time isolated from nature while the ^(13)C NMR data of compounds 2 and3 are reported for the first time. The bioassay revealed that compound 1 was active against BEL-7407and A549 cell lines (IC_(45) 43, 77 μg·mL^(-1)), 8 and 10 showed inhibitory activities on KB celllines (IC_(50) 78, 44 μg·mL^(-1)) and HL-60 cell lines (Ic_(50) 85, 85 μg·mL^(-1)), while 7exerted moderate cytotoxic activities on KB, BFL-7407, A549 and HL-60 cell lines with their IC_(50)being 51, 55, 44 and 10 μg·mL^(-1) , respectively. 展开更多

Comparison between Sulfuric Acid-phenol and Sulfuric Acid-anthrone Methods Used for Determination of Polysaccharides in Shoots of Aralia elata (Miq.) Seem. 被引量:3

Sulfuric acid-phenol and sulfuric acid-anthrone methods were used to detect polysaccharide content in shoots of Aralia elata( Miq.) Seem.,and the conversion factor to glucose was measured with refined polysaccharides. Comprehensive evaluation was carried out by linear relationship,precision,reproducibility,stability and recovery rate. The results showed that the linear relationship between glucose concentration and absorbance was good when glucose concentration was0-40 μg/ml,and the average recovery rate was equal to or higher than 97. 00% with good reproducibility( RSD 〈 1. 60%,n = 5). It revealed that the two methods were accurate and reliable,and suitable for the determination of polysaccharide content in the shoots of A. elata. Polysaccharide content detected by sulfuric acid-phenol and sulfuric acid-anthrone methods was 19. 31% and 20. 40% respectively. 展开更多

Objective: To evaluate the antioxidant activity of extracts and fractions from Stachys sieboldii Miq., and to examine its effect on the cellular reactive oxygen species(ROS) and glutathione(GSH) production and genomic DNA oxidation in HT-1080 cells. Methods: The ROS generation induced by H2 O2 was measured by the dichlorofluorescein-diacetate assay. GSH levels were measured using a fluorescent method with mBBr. Genomic DNA oxidative damage was measured with levels of oxidative DNA induced by the reaction of ferritin with H2 O2. Results: The n-hexane, 85% aqueous methanol and n-butanol fractions(0.05 mg/mL concentrations) inhibited H2 O2-induced ROS generation by 63%, 35% and 45%, respectively. GSH levels were significantly increased in both acetone+methylene chloride and methanol extracts(P<0.05). Supplementation of cells with n-hexane significantly increased GSH levels at concentrations of 0.05 mg/mL(P<0.05). Both the acetone+methylene chloride and methanol extracts, as well as all fractions significantly inhibited oxidative DNA damage(P<0.05). Conclusions: These results indicate that cellular oxidation was inhibited by the n-hexane fraction and this fraction may contain valuable active compounds. 展开更多

Several trier- pene saponins extracted from the leaves of Aralia elata (Miq.) Seem possess potent cytotoxic activity, but the mechanisms remain unclear. Methods In the present study, we examined the anti-tumor activity of total saponins of Aralia elata (Miq.) Seem leaves (TSAESL) using both in vitro and in vivo experiments, and investigated the underlying molecular mechanisms. Results Briefly, TSAESL significantly inhibited the proliferation of five human cancer cell lines and induced apoptosis of MCF-7 cells in a concentration-dependent manner. TSAESL also in- creased the expression level of active caspase-3 and cleaved PARP. In addition, TSAESL dramatically activated the phosphorylation of ERK1/2, p38 and JNK in a concentration-dependent manner. Treatment with the ERK1/2 in- hibitor U0126, p38 inhibitor SB203580, or JNK inhibitor SP600125 prior to TSAESL exposure markedly attenuated TSAESL-induced phosphorylation of ERK1/2 and p38. In mice bearing MCF-7 xenografl, TSAESL markedly inhib- ited tumor growth without significantly affecting spleen coefficient and hematological parameters. Conclusion These results strongly suggest that TSAESL has anti-tumor effect via activating the MAPK-mediated signaling path- ways. 展开更多

[Objectives]This study was conducted to screen suitable seed stems of Fritillaria thunbergii Miq.from different provenances and to provide a theoretical basis for the high-yielding and high-efficiency cultivation of F.thunbergii Miq.introduced to different places.[Methods]F.thunbergii Miq.from four different provenances including Zhejiang,Nantong and Chongqing were introduced and cultivated in Wanzhou of Chongqing.The contents of available Zn,Fe,Mn,Cu,Mo,N,P,K,Ca and Mg in rhizosphere soil of F.thunbergii Miq.during five growing stages were determined after selecting different stem sizes for field cultivation.[Results]Small stems of Pan an and Ningbo provenances(SSG3,121-160/kg)and middle stems of Nantong and Fengjie provenances(SSG2,81-120/kg)showed higher soil availability.[Conclusions]In the process of introduction and cultivation of F.thunbergii Miq.,high yield and high efficiency can be achieved by selecting smaller seed stems of F.thunbergii Miq. 展开更多

[Results] In the 16 S rDNA clone library for endophytic bacteria of P. dindygulensis Miq.,95 clones separately belonged to 46 OTUs,Phylogenetic analysis indicated that these cloning sequences separately belonged to phylum Proteobacteria,phylum Bacteroidetes,phylum Actinobacteria,phylum Nitrospirae,and phylum Gemmatimonadetes. In these,phylum Proteobacteria accounted for 70. 53% of total number of clones,so it was the most dominant group in this library. [Conclusions]The endophytic bacteria diversity in the root of P.dindygulensis Miq. is rich and this study is expected to provide scientific basis for protection,development and use of resources of P. dindygulensis Miq. 展开更多

Effects of Different Harvest Periods on Seed Quality of Alpinia oxyphylla Miq.

[Objectives] To clarify the appropriate harvest period of Alpinia oxyphylla Miq. seed and improve the quality of seeds. [Methods] In this experiment,the basic parameters and germination characteristics of A. oxyphylla Miq. seeds during different harvest periods were analyzed and compared. [Results] The results showed that the A. oxyphylla Miq. fruit picking period was closely related to the seed quality,and the seed maturity was highest 100 d after the flowering of A. oxyphylla Miq.,that is,the A. oxyphylla Miq. fruit skin turned from green to yellow( yellow-green),the peel had a small number of brown spots,it was spicy enough,the fruit dry/fresh weight ratio was greater than0. 34,the seed thousand kernel weight reached 11 g or more,and the dried seed moisture content was 13. 01%. Using the TTC method,the seed viability index was determined to be 85%,and in this case,it was the best time of harvesting seed. The water permeability of A. oxyphylla Miq. seed was poor,and the seed imbibition and water absorption time was long,about 72 h. The maximum water absorption rate was about23. 71% at the imbibition and water absorption stage,and it was inferred that the A. oxyphylla Miq. seeds were hard seeds. The germination time of A. oxyphylla Miq. seeds was long,the mature A. oxyphylla Miq. seeds began to germinate on the 12 th d,and the germination rate reached 75. 56% on the 50 th d. [Conclusions] This study determined the optimum harvest period of A. oxyphylla Miq. seeds,clarified the biological characteristics of slow seed water absorption and long germination cycle,and concluded that the seed coat was the main factor leading to slow seed water absorption and long seed germination cycle. 展开更多

The analysis of chloroplast gene characteristics in Alpinia japonica(Thunb.)Miq.is of great significance for developing relevant genetic resources.The high-throughput sequencing and bioinformatic research were performed to analyze the chloroplast genome characteristics of A.japonica.The total chloroplast genome length of A.japonica was 161,906 bp,with a typical circular tetrameric structure.And 133 genes were annotated,comprising 87 protein-coding,38 tRNA,and 8 rRNA genes.Furthermore,22 genes contained two copies,and 18 genes owned introns.Repeat sequence analysis showed that it contains 321 simple sequence repeats(SSRs)and 37 long segment repeats.Compared with the chloroplast genomes of eight representative plants in the genus Alpinia,the gene structure,type,and quantity were relatively conservative.Rps12 was the highest variation site in the entire chloroplast gene.A phylogenetic tree showed that the genus Alpinia was the most closely related to the genus Amomum.Meanwhile,A.japonica is the most closely related to Alpinia chinensis belonging to the genus Alpinia.Overall,the chloroplast genome of a new species was reported in the genus Alpinia,and a basis was provided for the utilization of Alpinia plants as a medical resource. 展开更多

Objective:To investigate the separation and extraction process of the primary chemical constituents of Alpinia oxyphylla Miq.against Salmonella typhi,and its antibacterial effi cacy,providing theoretical basis for novel gastrointestinal epidemic drugs and interventions against pathogen transmission.Methods:Alpinia oxyphylla Miq.:purified water-to-material ratio 1:5,refl uxed at 100°C for 6 hours,repeated three times.Alpinia oxyphylla Miq.extract was concentrated by freeze-drying at a 30:1 solid-to-liquid ratio to obtain the fi nal extract.The effi cacy of the extraction method and antibacterial activity were assessed using the disk diffusion method.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)were determined to establish the minimum therapeutic thresholds.Gas chromatography-mass spectrometry(GCMS)was employed to identify the primary bioactive constituents and their relative concentrations in the Alpinia oxyphylla Miq.aqueous extract.Results:The water extract of Alpinia oxyphylla Miq.Miq.demonstrated potent antibacterial activity against Salmonella typhi(P<0.001),with an inhibition zone diameter of 8.43±0.11 mm,MIC of 0.25 g/ml,and MBC of 0.5 g/ml.The extraction process was established as follows:Alpinia oxyphylla Miq.:purifi ed water ratio 1:5,refl ux heating at 100°C for 6 hours,repeated three times,followed by freeze-drying at 30:1 concentration.This yielded an extract rich in,primarily comprising octadecanoic acid(44.27%),palmitic acid(26.18%),caproic acid(8.15%),10-hydroxydecanoic acid(7.83%)as the main components,supplemented by 3-hydroxy-dodecanoic acid(1.78%),cis-13-eicosenoic acid(1.58%),valeric acid(1.21%),heptanoic acid(1.13%),nonanoic acid(1.06%),oleic acid(0.87%),and butyric acid(0.47%).Conclusion:This extraction process for novel antimicrobial compounds from Alpinia oxyphylla Miq.demonstrates high stability,operational feasibility,and precise composition and effi cacy.It provides a practical theoretical foundation for developing novel natural medicines against infectious diseases caused by Salmonella typhi,while off ering further insights for antimicrobial drug development. 展开更多

Background:The dry bulb of Fritillaria thunbergii Miq.is a traditional Chinese medicine,and is the leading product of the geo-authentic crude drugs‘Zhebawei’in Zhejiang Province.Fritillaria thunbergii requires more potassium(K)than nitrogen and phosphorus,while the response of F.thunbergii to K has rarely been studied.Objective and Methods:To evaluate the K nutritional status of F.thunbergii,a 2-year field experiment was conducted under six K application rates(0,40,80,120,160,and 200 kg K_(2)O ha−1)with two F.thunbergii cultivars(ZB1,narrow-leaf;ZB2,broad-leaf).Results:The bulbus Fritillariae Thunbergii(BFT)yield increased to a plateau with more than~120 kg K_(2)O ha-1.The BFT quality met the standard of Pharmacopoeia of P.R.China with more than 40.0 kg K_(2)O ha^(-1).Leaf K concentration was significantly correlated with BFT yield,and was more sensitive to K application rate as compared to those of stems and flowers.Potassium application extended the duration of rapid leaf-biomass accumulation and increased the accumulation rate.Two cultivars had similar response patterns to K application,but ZB2 had higher K partial productivity,leaf-biomass,and leaf K concentration than ZB1.Conclusion:108.4-128.0 kg K_(2)O ha^(-1) was the optimal K application range for BFT yield.The potassium nutrition index(KNI)model established based on leaf K concentration has biological significance and can be used for dynamic diagnosis of K nutritional status of F.thunbergii with both accuracy and simplicity. 展开更多

前胡属药材中24种香豆素类成分的定量表征及化学模式判别研究

目的构建前胡属药材中24种香豆素类成分的多组分同步定量分析方法,阐明前胡与紫花前胡的化学特征差异及其种属特异性标志物,为品种鉴定和道地性评价提供参考。方法基于色谱与质谱联用技术(UPLC-MS/MS)技术建立多组分定量分析方法,系统... 目的构建前胡属药材中24种香豆素类成分的多组分同步定量分析方法,阐明前胡与紫花前胡的化学特征差异及其种属特异性标志物,为品种鉴定和道地性评价提供参考。方法基于色谱与质谱联用技术(UPLC-MS/MS)技术建立多组分定量分析方法,系统验证线性范围、灵敏度、精密度、稳定性、基质效应及回收率等,结合化学计量学如主成分分析(PCA)、偏最小二乘法-判别分析(PLS-DA)、聚类分析对多批次前胡及紫花前胡样本进行化学模式解析。结果基于UPLC-MS/MS法建立的前胡属中24种香豆素类成分分析方法经方法学验证,考察项目均符合分析要求;前胡及紫花前胡成分含量分析显示,两者在香豆素类成分分布上种属特异性和化学特征差异显著,如紫花前胡苷(10~30倍)、7-羟基香豆素(2~10倍)等在前胡中含量远低于紫花前胡;PCA、PLS-DA、聚类分析表明24种香豆素类成分显示前胡及紫花前胡在化学空间中显著分离特点,其中前胡以线型呋喃香豆素中的白花前胡甲素、乙素等为核心标志物,紫花前胡富集角型吡喃香豆素中的异紫花前胡内酯及羟基化衍生物如7-羟基香豆素等。结论本研究开发的UPLC-MS/MS方法高效、精准,结合化学计量学模型,系统揭示前胡属药材香豆素成分的种属分布规律,为品种鉴定、质量标准化及中药监管提供多指标协同控制策略。 展开更多

钩藤茎秆基质栽培食药用菌试验初报

钩藤作为我国传统大宗中药材,其非药用茎秆资源长期没有得到有效利用。以钩藤采收药材后剩余的茎秆为原料,设计4种钩藤茎秆屑基质配方,并与全阔叶树木屑基质进行对比,探索栽培食药用菌的可行性及方法,分析其经济效益。试验采用袋料林内... 钩藤作为我国传统大宗中药材,其非药用茎秆资源长期没有得到有效利用。以钩藤采收药材后剩余的茎秆为原料,设计4种钩藤茎秆屑基质配方,并与全阔叶树木屑基质进行对比,探索栽培食药用菌的可行性及方法,分析其经济效益。试验采用袋料林内栽培灵芝菌和黑木耳菌,结果表明:含有钩藤茎秆基质的各个处理,其子实体产量均显著高于对照组的全阔叶树木屑基质,其中灵芝产量高出对照组36.7%~55.0%,黑木耳产量高出对照组73.0%~122.6%。经对栽培菌菌丝生长阶段观察,发现灵芝菌各个处理所接种的菌棒无一感染杂菌,而黑木耳各个处理所接种的菌棒均有一定程度杂菌感染,平均感染率为9.6%,说明灵芝菌对杂菌有很强的抗性,而黑木耳菌则对杂菌抗性较弱。经对钩藤茎秆进行产量测定,造林10年林分单位面积年产鲜茎秆为28812 kg/hm^(2),折合干茎秆为14406 kg/hm^(2),以每个袋料菌棒用干茎秆屑0.72 kg(占基质80%)计算,可制作菌棒2万个,参照试验处理D的单棒产量,可年产鲜灵芝3080 kg/hm^(2)、鲜黑木耳13860 kg/hm^(2),年新增产值分别为17.17万元/hm^(2)、11.09万元/hm^(2)。指出钩藤茎秆在栽培食药用菌方面极具优势和潜力,且可完全替代传统的阔叶树基质用于生产食药用菌,兼具资源高效利用与生态保护价值。 展开更多

为了认识湖南特有种——绒毛皂荚的果实形态特有性及其与山皂荚的区别,采用现场观测记录与收集样本测量分析的方法,进行了该项研究。结果表明:(1)绒毛皂荚果实特有特征为果实密被颜色随生长季节发生变化的绒毛,果喙细长、扭转、成绳索条... 为了认识湖南特有种——绒毛皂荚的果实形态特有性及其与山皂荚的区别,采用现场观测记录与收集样本测量分析的方法,进行了该项研究。结果表明:(1)绒毛皂荚果实特有特征为果实密被颜色随生长季节发生变化的绒毛,果喙细长、扭转、成绳索条状,果体端部呈三角状平展或微翘、顶部锐角状,背缝线方形、宽度>腹缝线等。(2)山皂荚果实生长全过程无绒毛、光滑,果喙短促、顶平、不扭转,背腹缝线汇合处紧缩成“鹰嘴”状向内收缩、并与两缝线汇合段“勺状”端部融合形成“鹰嘴勺状联合体”形态,背缝线刀脊形、宽度<腹缝线等。两种果实在以上形态上的区别表现出有与无,或特征完全相反的关系。两者在其他组织结构形态特征上也存在明显差异。结合其他研究资料综合分析,绒毛皂荚采用种的学名更为合理。 展开更多
