目的研究MicroRNA-137(miR-137)在膀胱癌中的表达及对膀胱癌细胞生物学特性的影响。方法收集病理诊断确诊为膀胱癌46例患者的膀胱癌组织及癌旁组织,通过Real time PCR检测miR-137在膀胱癌及癌旁组织中的表达水平,研究miR-137表达水平与...目的研究MicroRNA-137(miR-137)在膀胱癌中的表达及对膀胱癌细胞生物学特性的影响。方法收集病理诊断确诊为膀胱癌46例患者的膀胱癌组织及癌旁组织,通过Real time PCR检测miR-137在膀胱癌及癌旁组织中的表达水平,研究miR-137表达水平与膀胱癌临床分型及病理分级的关系。应用合成质粒转染T24细胞株,采用transwell实验计算miR-137过表达组、空白质粒组、抑制子质粒组的癌细胞过膜数量,研究miR-137与膀胱癌细胞的侵袭迁移作用的关系。结果 miR-137在浸润型膀胱癌中的表达(2.33±0.11)显著高于非浸润型膀胱癌组织(1.41±0.05,P<0.05)。膀胱癌组织中miR-137的表达随着病理分级的恶性程度的增高而增加(H=6.528,P<0.05)。Transwell实验显示过表达miR-137质粒组的细胞过膜数量最高[(392±21/视野)],抑制子质粒组最低[(119±9/视野)],且差异有统计学意义(P<0.05)。结论 miR-137的表达水平与膀胱癌细胞的侵袭迁移能力相关,miR-137的表达增多可以显著增强膀胱癌细胞的侵袭迁移能力,提示miR-137的高表达与膀胱癌的转移以及浸润相关,miR-137在膀胱癌的侵袭和转移过程中发挥重要作用,其可作为膀胱癌的潜在临床诊断标志物,为早期诊断和治疗膀胱癌提供了新思路。展开更多
