Objective This study aimed to study the effects of different crystalline states of Sheng Shigao(raw gypsum,RG)and its inorganic elements on the antipyretic efficacy of Baihu Decoction(BHT).Methods RG samples calcined at different temperatures were prepared.The phase composition of RG and Duan Shigao(calcination of gypsum,CG)as well as the changes in phase composition before and after adding water to RG calcined at specific temperatures,were determined using X-ray diffraction(XRD).A fever model was established by subcutaneously injecting 20%yeast suspension(10 mL·kg~(-1))into the backs of rats.The effects of BHT containing RG in different crystalline states on rat body temperature were measured.Serum levels of IL-1β,IL-6,and hypothalamic prostaglandin E2(PGE_2)were detected using ELISA.Serum Ca~(2+)levels were measured using a microplate method.The content of trace elements in RG and CG and the corresponding freeze-dried BHT powder was determined using inductively coupled plasma mass spectrometry(ICP-MS).The complexation of representative inorganic elements with mangiferin,a major active component in BHT,was investigated using UV-Vis spectroscopy and fluorescence spectroscopy.A validation model was established using RAW264.7 mouse macrophages.Drug-containing serum of BHT with different inorganic elements was prepared,and the nitric oxide(NO)levels in the cell supernatant of different treatment groups were measured using the Griess method.The mRNA levels of IL-6,TNF-α,and PGE2in each group were detected using qPCR(real-time fluorescent quantitative PCR).Results After calcination,the phase composition of RG changed,and the content of inorganic elements in RG,CG170(RG calcined at 170°C),and CG350(RG calcined at 350°C)showed similar trends.Compared with RG,the content of Ca,Sr,Al,and Na in CG changed significantly.Compared with BHT,the content of Ca,Sr,Si,and Na in CG changed significantly when incorporated into the formula.Intermolecular interactions confirmed strong binding between mangiferin and Cu~(2+)and Al~(3+).Cu~(2+)and Fe~(3+)exhibited fluorescence quenching effects on mangiferin solution,while Al~(3+)and Zn~(2+)showed strong fluorescence enhancement,with fluorescence intensity increasing by 120-fold and 30-fold,respectively.In vitro evaluation of synergistic anti-inflammatory effects confirmed that Ca,Fe,Cr,Al,and Si exhibited synergistic anti-inflammatory effects.Conclusion The crystalline state of RG has little effect on its antipyretic properties,while Ca,Sr,Na,Fe,and Al are likely the key material bases influencing its efficacy. 展开更多

Objective:To explore the complex prescription compatibility law of the cold and hot nature of Mahuang Decoction(麻黄汤,MHD) and Maxing Shigan Decoction(麻杏石甘汤,MXSGD),both categorized formulas but with different hot/cold natures.Methods:Oxygen consumption of mice was determined among three groups:MHD,MXSGD and the control;a cold-hot pad differentiating assay was used to observe the variability of temperature tropism among the groups of mice which was treated with MHD,MXSGD,and their compositions. Meanwhile,the total anti-oxidant capability(T-AOC) activity were detected.Results:After administration of MHD, the mice showed increased oxygen consumption(P0.01).Compared with MHD group,the remaining rate of MXSGD mice on the hot pad was found to be significantly increased with the cold-hot pad differentiating assay (P0.05).There was no significant difference(P0.05) among the remaining rates of MXSGD,MXSGD with high dose Gypsum Fibrosum(MXHGF) group,and MXSGD with low dose Gypsum Fibrosum(MXLGF) group mice.Compared with the MHD group,T-AOC activity of the mice in the Consensus Compositons group was significantly decreased(P=0.0494).Compared with the MXSGD group,T-AOC activity of Gypsum Fibrosum (GF) group was increased significantly(P=0.0013).Conclusions:The differences in cold and hot nature could be represented objectively between MHD with a hot nature and MXSGD with a cold nature.The reason may be the Gypsum Fibrosum which decreased the efficacy of the consensus compositions.However,increasing or decreasing the dose of Gypsum Fibrosum will not change the cold and hot nature of MXSGD. 展开更多

Objective To decompose and reformulate Banxia Baizhu Tianma Decoction(BBT) in order to optimize its ratio for vasodilatory effect.The fingerprints of these six component herbs were also studied via Tri-step FTIR analysis.Methods The herbal components of BBT were extracted separately with different solvents,such as 95%ethanol,50%ethanol,and distilled water.The vasodilatory effect of the extracts was studied using the in vitro pre-contracted aortic ring model.Among these extracts,the water extracts of Pinellia ternata[PT(W)]and Citri reticulatae[CR(W)],50%ethanol extracts of Castrodia elata[GE(50)]and Clycyrrhiza uralensis[GU(50)],and 95%ethanol extracts of Atractylodes macrocephala[AM(E)],and Poria cocos[PC(E)]were selected for the orthogonal stimulus-response compatibility group study due to their high effectiveness in vasodilation.Results The best combination ratio(G2) was GE(50),GU(50),AM(E),PC(E),CR(W),and PT(W) at EC0,EC5,EC5,EC5,EC5,and EC5,with EC50 of(0.06 ± 0.01) mg/mL.Comparison between the vasodilatory effects of extracts derived from G2 and BBT were also studied.From the results,the 50%ethanol extract following G2 ratio was found to be most potent extract with the highest vasodilatory effect compared with other extracts derived from G2 or BBT,with EC50 of(0.05 ±0.005) mg/mL.Conclusion The optimum ratio and appropriate extraction method for the new BBT formulae are obtained.G2 is determined as the best formulae with the highest potency in producing the vasodilatory effect on isolated rat aortic rings.The results obtained suggest that 50%ethanol is found to be the best solvent to extract the vasoactive compounds of G2 in order to exhibit the highest efficacy of vasodilatory effect. 展开更多

Comparative analysis of four active compounds of Baikal skullcap and its classical TCM prescriptions according to different clinical curative effects

Objective: A sensitive HPLC-DAD detection method was established for the comparative analysis of the four active compounds(including baicalin, baicalein, wogonoside and wogonin) of Baikal Skullcap and its classical TCM prescriptions according to different clinical curative effects. And analyze the relationship between compatibility of medicines, content and clinical curative effect.Methods: Water extracts were analyzed by high-performance liquid chromatography-diode array detection(HPLC-DAD), using an Agilent extend-C_(18)(4.6 mm×250 mm,5μm) column by using solvent A(0.2% phosphoric acid water) and solvent B(methanol) in the ratio of 52:48(v/v) at a flow rate of 1 ml/min. The column temperature was maintained at 25°C and the detection wavelength was set at 280 nm.Results: In quantitative analysis, the four selected markers showed good regression(R^2>0.999) within test ranges. The average recoveries were between 99.75~100.60% and their RSD values were between 0.97%~1.71%. The proportions of the four chemical compounds for different efficacy were 33.51~35.76: 6.78~6.87: 3.45~3.64: 1; 18.6~23.64: 3.63~4.27: 1.36~1.53: 1; 29.94~31.78: 5.14~5.23: 3.28~3.56: 1; 33.99~41.91:5.83~7.42: 2.83~3.39: 1.Conclusion: Clinical practice of thousands of years have proved that the curative effect of Traditional Chinese Medicine depends on the compatibility of different kinds of medicine material crude slices. According to this study we found that different compatibility of medicines leads to different proportions of the chemical compounds. We assume that there is a certain correlation between therapeutical effect of TCM and compatibility of medicines. 展开更多
