Microsatellite instability (MSI) is used as a molecular marker for defective DNA mismatch repair (MMR) genes.We report here alterations of MSI in 15 malignant astrocytomas (WHO grade Ⅲ) and glioblastomas (GBM; WHO grade Ⅳ) of pediatric patients (2-21 years) and 12 GBM from adults (44-68 years) by comparative analysis of BAT25/BAT26 loci and 10 other microsatellite markers. Gastric cancer(GC)remains a major health challenge worldwide,with Helicobacter pylori(H.pylori)infection playing a key role in its development.H.pylori creates a mutagenic environment in the stomach by causing chronic inflammation,oxidative DNA damage,inducing DNA double-strand breaks,and disrupting DNA repair mechanisms and cell cycle checkpoints.Cytotoxin-associated gene A is the main carcinogenic component of H.pylori and interacts with signaling pathways to promote carcinogenesis.Deleted in malignant brain tumors 1(DMBT1),a potential tumor suppressor gene,shows variable expression patterns in GC.DMBT1 is frequently downregulated in well-differentiated gastric adenocarcinomas but upregulated in other GC types.The correlation between DMBT1 expression and H.pylori infection is important,as maintained DMBT1 expression in precancerous states may protect against gastric carcinogenesis,while its downregulation may facilitate tumor progression.DMBT1 functions as a pattern recognition receptor that binds to H.pylori and other pathogens and modulates inflammatory and immune responses.H.pylori colonization of gastric mucosa can induce an inflammatory microenvironment,which influences tumor suppressor gene expression,including DMBT1.Understanding the interactions between DMBT1 and H.pylori may reveal new therapeutic targets and preventive strategies for H.pylori-associated gastric disease. 展开更多

Paraffin-embedded tissue of skin biopsy specimens taken retrospectively from 24 patients with cutaneous malignant lymphomas (CML) and 8 patients with cutaneous lymphoid infiltrates (CLI) and other dennatoses were studied retrospectively with PCIO immunostaining. The results show a statistical significant difference among PCIO indices for cutaneous genuine histiocytic lymphoma (CGHL), cutaneous germinal center cell-derived lymphomas (CGCCL), cutaneous peripheral T-cell lymphomas (CPTL), non-mycosis fungoides (MF) and Sezary's syndrome (SS), and MF when compared with those for CLI. There is a linear relationship between PCIO index and square root of PCIO density, both of which seem to have a parallel relationship to the severity of malignancy in CML. The nuclear volume of the positive tumor cell or lymphocyte with PCIO immunostaining may be also useful in differentiating CML from CLI. 展开更多
