Magnusiomyces capitatus in Immune-Competent Patients with Pulmonary Haemorrhage and Systemic Lupus Erythematosus

Invasive fungal infections have grown significantly over the last two decades, owing to an increase in immunocompromised hosts and geriatric patients. When the host’s defenses are compromised, such infections are associated with severe morbidity and mortality. Here, a rare case of fungal infection in a 61-year-old immunocompetent male patient from Saudi Arabia was reported, who suffered from pulmonary hemorrhage and Systemic Lupus Erythematous. Bronchoalveolar Lavage was used as a diagnostic tool to identify the fungus reported in the case. The pathogenic fungal specie identified as Magnusiomyces capitatus, in macroscopic and microscopic morphological characteristics of the colonies. Based on clinical evidence, liposomal amphotericin formulation was recommended for initial therapy against fungal infection. Also, liposomal amphotericin B induced mycological eradication up to 70 percent in patients with proven Magnusiomyces capitatus infection. In addition to addressing suspected Systemic lupus erythematosus, the patient’s health has improved with no evidence of pulmonary bleeding and hemoptysis. 展开更多

Alcohol dehydrogenases(ADHs)are key enzymes in microbial ethanol metabolism and ethanol detection with significant relevance in industrial bioprocessing and synthetic biology.The study focuses on enhancing the activation of Magnusiomyces capitatus ADHs specific to ethanol for ethanol conversion and detection.This was achieved by evaluating its specific alcohol dehydrogenase(ADH)activity under varying growth conditions by following a systematic one-factor-at-a-time(OFAT)approach and a central composite rotatable design(CCRD).Using the OFAT method,the most critical factor for improving specific ADH activity were identified as glucose,ammonium sulphate,zinc sulphate,and pH,which was further optimized using the CCRD.The specific ADH activity of M.capitatus in the developed medium was 489.28±0.31 mU mg^(−1) of protein,which was greater than that of cells cultured in basal ethanol medium.Furthermore,the volatile compounds(VOCs)generated during ethanol oxidation under aerobic conditions were analyzed by GC-MS,validating the metabolic flexibility of M.capitatus under optimal circumstances.These findings offer new insights into the systems-level metabolic behavior of M.capitatus under ethanol stress and highlight its potential as a microbial platform for future biomanufacturing and enzymatic conversion processes with potential applications in biosensing and industrial bioprocessing. 展开更多

Urban life has led to the creation of human-made environments that,from a microbiological perspective,provide extreme life conditions.Certain non-ubiquitous microorganisms such as thermophilic members of the black yeast genus Exophiala are enriched within these habitats for which no counterpart is known in nature.Dishwashers consistently accumulate a number of specific black,white and red yeasts on the rubber seals of doors and in stagnant water at the interior.Several of these yeasts are primarily known as agents of human opportunistic infections.In this review,the literature data are supported by a screening study involving 937 households in 15 cities in Turkey.Fungi were detected in 230 samples(24.5%).Using rDNA sequencing,the prevalent species were identified as Exophiala dermatitidis(n=116),Candida parapsilosis(n=44),E.phaeomuriformis(n=35),Magnusiomyces capitatus(n=22),Rhodotorula mucilaginosa(n=15),and C.lusitaniae(n=14).The possible role of dishwashers in transmitting disease is discussed. 展开更多
