In this study, MS-HRM (Methylation-Sensitive High Resolution Melting Curve Analysis) was used to detect the methylation status of serum DNA. Gene of insulin-like growth factor binding protein 3 (IGFBP-3) was detected in this study. Results showed that the serum DNA methylation level of FGR fetus were lower than that of the control group;different methylation levels were also found between male and female fetus;and the methylation level was increased with the birth weight of the newborn. Our results showed that the IGFBP-3 gene methylation level of serum DNA of newborn could be semi-quantitative detected which guide the early prevention and treatment of IUGR. It also indicated that the methylation status of serum DNA can be conveniently identified and quantified by inspection of the melting curves.展开更多
建立了基于高效液相色谱-线性离子阱/静电场轨道阱高分辨质谱(HPLC-LTQ/Orbitrap MS)快速筛查确证生态纺织品中22种禁用分散染料的分析方法。样品在95℃水浴条件下经吡啶/水(1∶1,v/v)振荡提取后,以CAPCELL PAK C18色谱柱分离,以乙...建立了基于高效液相色谱-线性离子阱/静电场轨道阱高分辨质谱(HPLC-LTQ/Orbitrap MS)快速筛查确证生态纺织品中22种禁用分散染料的分析方法。样品在95℃水浴条件下经吡啶/水(1∶1,v/v)振荡提取后,以CAPCELL PAK C18色谱柱分离,以乙腈和5 mmol/L乙酸铵(含体积分数为0.01%的甲酸)作为色谱流动相进行梯度洗脱,采用正、负电喷雾(ESI)离子化模式,利用一级母离子的精确质量数和保留时间对22种分散染料进行快速筛查,利用碰撞诱导解离(CID)下得到的二级碎片离子进行确证。22种分散染料在各自浓度范围内线性关系良好(r2〉0.99),方法的定量限(LOQ)为0.125~2.5 mg/kg。除分散黄49外,绝大多数染料在涤纶布、棉涤混纺布两种纺织品基质中的加标回收率在65%~120%之间,相对标准偏差小于15%。应用本方法对涉及多种纤维类型的40余件纺织品样品进行了筛查,其中一个样品检出分散橙37/76。该方法简便、快速,其选择性高,抗干扰性能好,结果准确可靠,可用于纺织品中分散染料的检测。展开更多

文摘建立了高效液相色谱-线性离子阱/静电场轨道阱高分辨质谱(HPLC-LTQ/Orbitrap MS)筛查和确证生态纺织品中致癌染料的方法.样品在水浴(95℃)中用吡啶/水(1/1,v/v)振荡(150r/min)提取.上清液过聚四氟乙烯(PTFE)滤膜后,CAPCELL PAK C18色谱柱(100mm×2.0mm,5μm)分离,以乙腈和5mmol/L乙酸铵水溶液(含0.01%甲酸)、乙腈和5mmol/L乙酸铵水溶液分别作为正、负电喷雾离子化(ESI)模式的色谱流动相梯度洗脱。m/z200~800范围内进行一级质谱全扫描。以准分子离子峰的精确质量数和提取的色谱图峰面积进行筛查分析和定量.以保留时间和数据依赖扫描(data—dependent scan)模式获得的子离子质谱图进行定性确证。9种染料的质量准确度小于5×10^-6(5ppm),线性良好,相关系数大于0.99,方法检出限为0.125~25mg/kg。3个添加水平的回收率范围为62.13%~116.28%,相对标准偏差小于15%.应用该方法检测了棉、涤纶及混纺纤维等20余件纺织品样品中的致癌染料残留。该方法准确、可靠.
