Cloning and Sequence Analysis of Adiponectin Receptor 1 and Receptor 2 cDNA from Guangxi Bama Mini-pig

[ Objective] To clone and analyze the sequence of Adiponectin receptor 1 ( AdipoR1 ) and receptor 2 (AdipoR2) cDNA of Guangxi Bama mini-pig. [Method] The Adiponectin receptors cDNAs were amplified by RT-PCR using skeletal muscle total RNA as template and then ligated into pMD18-T vector after purification. The recombinant pMD18-T vector was transformed into the E. coil DH5α for identification and sequencing. And the results were compared with the cDNA sequence from other species. [Result] The fragments, 1 128 bp and 1 161 bp in size, were amplified by RT-PCR and respectively consistent with the coding sequence of AdipoR1 gene and AdipoR2 gene. The homology analysis showed that the sequences of AdipoR1 gene and AdipoR2 gene were respectively 99.8% and 99.7% homologous to the sequence of domestic pig reported in GenBank with one base and three base missense mutations correspondingly. [ Conclusion] The AdipoR1 gene and AdipoR2. gene were successfully amplified from Guangxi Bama mini-pig, laying the foundation for the further study of the biological function of AdipoR genes and the design of novel drugs with AdipoR as target. 展开更多

Dietary proline supplementation alters colonic luminal microbiota and bacterial metabolite composition between days 45 and 70 of pregnancy in Huanjiang mini-pigs 被引量:8

Among amino acids, ... Background: Pregnancy is associated with important changes in gut microbiota composition. Dietary factors may affect the diversity, composition, and metabolic activity of the intestinal microbiota. Among amino acids, proline is known to play important roles in protein metabolism and structure, cell differentiation, conceptus growth and development, and gut microbiota re-equilibration in case of dysbiosis.Results: Dietary supplementation with 1% proline decreased(P < 0.05) the amounts of Klebsiella pneumoniae,Peptostreptococcus productus, Pseudomonas, and Veillonella spp. in distal colonic contents than that in the control group. The colonic contents of Butyrivibrio fibrisolvens, Bifidobacterium sp., Clostridium coccoides, Clostridium coccoides-Eubacterium rectale, Clostridium leptum subgroup, Escherichia coli, Faecalibacterium prausnitzii,Fusobacterium prausnitzii, and Prevotella increased(P < 0.05) on d 70 of pregnancy as compared with those on d 45 of pregnancy. The colonic concentrations of acetate, total straight-chain fatty acid, and total short-chain fatty acids(SCFA) in the proline-supplemented group were lower(P < 0.05), and butyrate level(P = 0.06) decreased as compared with the control group. Almost all of the SCFA displayed higher(P < 0.05) concentrations in proximal colonic contents on d 70 of pregnancy than those on d 45 of pregnancy. The concentrations of 1,7-heptyl diamine(P = 0.09) and phenylethylamine(P < 0.05) in proximal colonic contents were higher, while those of spermidine(P = 0.05) and total bioamine(P = 0.06) tended to be lower in the proline-supplemented group than those in the control group. The concentrations of spermidine, spermine, and total bioamine in colonic contents were higher(P < 0.05) on d 70 of pregnancy than those measured on d 45 of pregnancy. In contrast, the concentration of phenylethylamine was lower(P < 0.05) on d 70 than on d 45 of pregnancy.(Continued on next page)(Continued from previous page)Conclusion: These findings indicate that L-proline supplementation modifies both the colonic microbiota composition and the luminal concentrations of several bacterial metabolites. Furthermore, our data show that both the microbiota composition and the concentrations of bacterial metabolites are evolving in the course of pregnancy. These results are discussed in terms of possible implication in terms of luminal environment and consequences for gut physiology and health. 展开更多

AIM: to establish a new animal model for the research of human rotavirus(HRV) infection, its pathogenesis and immunity and evaluation of potential vaccines.METHODS: 5-d, 30-d and 60-d-old Chinese mini-pigs, Guizhou and bamma, were inoculated with a single oral dose of attenuated strain Wa, G1, G3 of HRV, and PbS(control), respectively, and fecal samples of pigs from 0 to 7 d post infection(DPI) were collected individually. Enzyme linked immunosorbent assay was used to detect HRV antigen in feces. the HRV was tested by real-time PCR(Rt-PCR). the sections of the intestinal tissue were stained with hematoxylin and eosin to observe the morphologic variation by microscopy. Immunofluorescence was used to determine the HRV in intestinal tissue. HRV particles in cells of the ileum were observed by electron micrography.RESULTS: When inoculated with HRV, mini-pigs younger than 30 d developed diarrhea in an agedependent manner and shed HRV antigen of the sameinoculum, as demonstrated by Rt- PCR.Histopathological changes were observed in HRV inoculated mini-pigs including small intestinal cell tumefaction and necrosis. HRV that was distributed in the small intestine was restricted to the top part of the villi on the internal wall of the ileum, which was observed by immunofluorescence and transmission electron microscopy. Virus particles were observed in Golgi like follicles in HRV-infected neonatal minipigs. Guizhou mini-pigs were more sensitive to HRV than bamma with respect to RV antigen shedding and clinical diarrhea.CONCLUSION: these results indicate that we have established a mini-pig model of HRV induced diarrhea. Our findings are useful for the understanding of the pathogenic mechanisms of HRV infection. 展开更多

Background:Currently,increasing attention is being paid to the important role of intestinal microbiome in diabetes.However,few studies have evaluated the characteristics of gut microbiome in diabetic miniature pigs,despite it being a good model animal for assessing diabetes.Methods:In this study,a mini-pig diabetes model(DM)was established by 9-month high-fat diet(HFD)combined with lowdose streptozotocin,while the animals fed standard chow diet constituted the control group.16S ribosomal RNA(rRNA)gene sequencing was performed to assess the characteristics of the intestinal microbiome in diabetic mini-pigs.Results:The results showed that microbial structure in diabetic mini-pigs was altered,reflected by increases in levels of Coprococcus_3 and Clostridium_sensu_stricto_1,which were positively correlated with diabetes,and decreases in levels of the bac-teria Rikenellaceae,Clostridiales_vadinBB60_group,and Bacteroidales_RF16_group,which were inversely correlated with blood glucose and insulin resistance.Moreover,PICRUSt-predicted pathways related to the glycolysis and Entner-Doudoroff superpathway,enterobactin biosynthesis,and the L-tryptophan biosynthesis were significantly elevated in the DM group.Conclusion:These results reveal the composition and predictive functions of the intestinal microbiome in the mini-pig diabetes model,further verifying the relationship between HFD,gut microbiome,and diabetes,and providing novel insights into the application of the mini-pig diabetes model in gut microbiome research. 展开更多

Post-traumatic spinal cord remodeling includes both degenerating and regenerating processes,which affect the potency of the functional recovery after spinal cord injury(SCI).Gene therapy for spinal cord injury is proposed as a promising therapeutic strategy to induce positive changes in remodeling of the affected neural tissue.In our previous studies for delivering the therapeutic genes at the site of spinal cord injury,we developed a new approach using an autologous leucoconcentrate transduced ex vivo with chimeric adenoviruses(Ad5/35)carrying recombinant cDNA.In the present study,the efficacy of the intravenous infusion of an autologous genetically-enriched leucoconcentrate simultaneously producing recombinant vascular endothelial growth factor(VEGF),glial cell line-derived neurotrophic factor(GDNF),and neural cell adhesion molecule(NCAM)was evaluated with regard to the molecular and cellular changes in remodeling of the spinal cord tissue at the site of damage in a model of mini-pigs with moderate spinal cord injury.Experimental animals were randomly divided into two groups of 4 pigs each:the therapeutic(infused with the leucoconcentrate simultaneously transduced with a combination of the three chimeric adenoviral vectors Ad5/35‐VEGF165,Ad5/35‐GDNF,and Ad5/35‐NCAM1)and control groups(infused with intact leucoconcentrate).The morphometric and immunofluorescence analysis of the spinal cord regeneration in the rostral and caudal segments according to the epicenter of the injury in the treated animals compared to the control mini-pigs showed:(1)higher sparing of the grey matter and increased survivability of the spinal cord cells(lower number of Caspase-3-positive cells and decreased expression of Hsp27);(2)recovery of synaptophysin expression;(3)prevention of astrogliosis(lower area of glial fibrillary acidic protein-positive astrocytes and ionized calcium binding adaptor molecule 1-positive microglial cells);(4)higher growth rates of regeneratingβIII-tubulin-positive axons accompanied by a higher number of oligodendrocyte transcription factor 2-positive oligodendroglial cells in the lateral corticospinal tract region.These results revealed the efficacy of intravenous infusion of the autologous genetically-enriched leucoconcentrate producing recombinant VEGF,GDNF,and NCAM in the acute phase of spinal cord injury on the positive changes in the post-traumatic remodeling nervous tissue at the site of direct injury.Our data provide a solid platform for a new ex vivo gene therapy for spinal cord injury and will facilitate further translation of regenerative therapies in clinical neurology. 展开更多

[ Method] 24 Guangxi Bama mini-pigs were fed with high-fat and high-sucrose diet, and partial sequences of exon 2 of PPARy-2 gene were amplified by using PCR method. In addition, the contents of fasting blood glucose and insulin (INS) in Guangxi Bama mini-pigs were determined, and the glucose tolerance test (GTT) was also carried out. [ Result] There was one SNP site (19813A/G) Jn partial sequence of exon 2 of the cloned PPAFly-2 gene, and AA (7 pigs) and AG (17 pigs) genotype were detected. The contents of fasting insulin and 60-min blood glucose in GTT in AG-genotype Guangxi Bama mini-pigs were significantly higher than those of AA genotype ( P 〈0.05), while the incidence of T2DM in AG-genotype Guangxi Bama mini-pigs (71.4%) was obviously higher than that of AA gen- otype (5.9%). [ Conclusion] The polymorphism of 19813A/G in exon 2 of PPARy-2 gene was related with the susceptibility of T2DM in Guangxi Bama mini-pigs. 展开更多

本文旨在研究母-子猪饲粮中添加益生菌和合生元对子代巴马香猪肌肉营养成分及肌肉发育和脂质代谢相关基因表达的影响。选用3~5胎次妊娠巴马香猪64头,随机分为4组,分别饲喂基础饲粮(对照组)、添加维吉尼亚霉素纯品饲粮(抗生素组)、添加... 本文旨在研究母-子猪饲粮中添加益生菌和合生元对子代巴马香猪肌肉营养成分及肌肉发育和脂质代谢相关基因表达的影响。选用3~5胎次妊娠巴马香猪64头,随机分为4组,分别饲喂基础饲粮(对照组)、添加维吉尼亚霉素纯品饲粮(抗生素组)、添加复合益生菌发酵液饲粮(益生菌组)和添加复合益生菌发酵液+低聚木糖饲粮(合生元组),每组16头猪。分娩后仔猪于28日龄断奶,每窝选取2头仔猪,子代饲喂与其母猪相同的饲粮。分别于65、95和125日龄,每组选取8头仔猪采集股二头肌样品,测定其营养成分和相关基因的表达。结果表明:与对照组相比,益生菌组和合生元组的65日龄肌内脂肪、非必需氨基酸、鲜味氨基酸、总氨基酸、丙氨酸、天冬氨酸、谷氨酸、甘氨酸、丝氨酸和十六碳一烯酸含量以及95日龄脯氨酸含量显著增加(P<0.05);抗生素组的65日异亮氨酸和缬氨酸含量显著增加(P<0.05),65日龄脂肪甘油三酯脂酶(ATGL)的mRNA相对表达量显著上调(P<0.05),95日龄脂蛋白脂酶(LPL)、硬脂酰辅酶A去饱和酶(SCD)和脂肪酸结合蛋白4(FABP4)的mRNA相对表达量显著下调(P<0.05);益生菌组的125日龄油酸(C18∶1n9c)含量显著增加(P<0.05),65日龄生肌因子5(Myf5)、LPL和过氧化物酶体增殖物激活受体α(PPARα),95日龄肌球蛋白重链Ⅰ(MyHCⅠ)和生肌因子6(Myf6)及125日龄Myf5、Myf6、乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FASN)和过氧化物酶体增殖物激活受体γ(PPARγ)的mRNA相对表达量显著上调(P<0.05);合生元组65日龄十四碳酸、十六碳酸、C18∶1n9c、单不饱和脂肪酸(MUFA)含量及n-3多不饱和脂肪酸/n-6多不饱和脂肪酸显著增加(P<0.05),95日龄肌内脂肪含量显著增加(P<0.05),95日龄PPARγ及125日龄Myf6、FASN、SCD、LPL、PPARα和PPARγ的mRNA相对表达量显著上调(P<0.05),95日龄肌肉生长抑制素(MSTN)的mRNA相对表达量显著下调(P<0.05)。综上所述,母-子猪饲粮中添加益生菌和合生元可改变子代股二头肌氨基酸和脂肪酸组成,调控生肌因子和脂质代谢相关基因的表达,有利于肉品质的改善。 展开更多

The objective of the study is to evaluate and compare the effects of betaine or glycine on carcass trait,meat quality and lipid metabolism of finishing Huan Jiang mini-pigs.Betaine called trimethylglycine is a methyl derivative of glycine,but few researches were conducted to compare the impact of dietary betaine and glycine on pigs.One hundred and forty-four Huan Jiang mini-pigs(body weight=10.55±0.15 kg;70 d)were randomly divided to 3 treatment groups(basal diet,glycine or betaine).Results indicated that dietary betaine increased the average daily gain(ADG)and final weight(P<0.05).Dietary glycine or betaine markedly reduced average backfat thickness(P<0.05)and heightened lean percentage(P<0.01)compared to the control group.Moreover,in comparison with the control group,betaine significantly improved the redness(a*)and tenderness(shear force)of the longissimus dorsi(LD)muscle(P<0.05),whereas glycine only raised the value of a*of the LD muscle(P<0.05).These results showed that diet supplemented with 0.25%betaine and equimolar amounts of glycine could regulate cascass trait and meat quality of finishing Huan Jiang mini-pigs,and the effect of betaine was superior to that of glycine. 展开更多

【目的】自前期选育获得的无猪内源性反转录病毒(Porcine endogenous retrovirus,PERV)传染性“中畜”五指山小型猪近交系(Zhong Xu Wuzhishan Mini-pig Inbred line,ZXWIPIG)新品系中,选取1头雌性猪(ZXWIPIG505)进行全基因组重测序分析... 【目的】自前期选育获得的无猪内源性反转录病毒(Porcine endogenous retrovirus,PERV)传染性“中畜”五指山小型猪近交系(Zhong Xu Wuzhishan Mini-pig Inbred line,ZXWIPIG)新品系中,选取1头雌性猪(ZXWIPIG505)进行全基因组重测序分析,以进一步从分子水平上阐释无PERV传染性猪新品系的序列特征。【方法】分离ZXWIPIG505外周血单个核细胞并提取其基因组DNA,利用Illumina HiSeq-4000测序平台进行全基因组重测序。分别使用BLAST和SAMtools 0.1.19软件,提取ZXWIPIG505基因组中整合的PERV长片段和PERV-pol基因序列片段,并使用DNAStar软件对整合的PERV结构基因序列进行分析。将ZXWIPIG505中整合的PERV-pol基因序列特征与前期鉴定的PERV-pol缺陷型ZXWIPIG452中PERV-pol基因序列进行比较研究。【结果】ZXWIPIG505全基因组重测序平均测序深度为14.32×,测序数据有效率为97.21%,Q20为94.37%,Q30为87.03%,GC含量为43.38%,表明本次测序数据质量较高。ZXWIPIG505基因组中共整合5条PERV长片段,其中仅scaffold5028含有PERV全部结构基因,但均为缺陷型。ZXWIPIG505基因组中共整合85条PERV-pol基因片段序列,比对发现所有PERV-pol基因片段均不完整,为缺陷型。ZXWIPIG505和ZXWIPIG452基因组中整合的PERV-pol基因序列存在高度相似性,60条序列长度相同,75条序列在猪基因组中的整合位点相同,42条序列的PERV-pol基因提前终止位点相同。【结论】前期选育的ZXWIPIG505为PERV-pol基因缺陷型猪,并且ZXWIPIG505和ZXWIPIG452中整合的PERV-pol基因序列具有高度相似性。 展开更多

目的 采取不同血氧检测手段,探讨多波长视网膜血氧仪间接检测与金标准直接检测结果的相关性及准确性。方法 6只巴马小猪分别吸入浓度为10%、21%、100%的氧气,当外周脉搏氧饱和度达到稳定水平时,通过股动脉血气分析检测股动脉血氧饱和度... 目的 采取不同血氧检测手段,探讨多波长视网膜血氧仪间接检测与金标准直接检测结果的相关性及准确性。方法 6只巴马小猪分别吸入浓度为10%、21%、100%的氧气,当外周脉搏氧饱和度达到稳定水平时,通过股动脉血气分析检测股动脉血氧饱和度;通过O_(2)微传感器损伤性地穿刺巴马小猪视网膜检测玻璃体内血氧饱和度;通过Ⅰ型、Ⅱ型多波长视网膜血氧仪无创检测视网膜血氧饱和度和动静脉管径。结果 股动脉血氧饱和度和玻璃体内血氧饱和度相关(P<0.05),玻璃体内血氧饱和度与视网膜血氧饱和度相关(P<0.05),Ⅰ型与Ⅱ型多波长视网膜血氧仪精确度无差异(P> 0.05)。在吸入10%浓度氧气的情况下,Ⅰ型与Ⅱ型多波长视网膜血氧仪测得动静脉管径有差异(P<0.05);在吸入21%浓度氧气和100%浓度氧气的情况下,Ⅰ型与Ⅱ型多波长视网膜血氧仪测得动静脉管径无差异(P 0.05)。结论 多波长视网膜血氧测定法对巴马小猪全身血氧饱和度变化较敏感;Ⅰ型与Ⅱ型多波长视网膜血氧仪精确度无差异。本实验明确了两种多波长视网膜血氧仪检测视网膜血氧饱和度的可靠性,为多波长视网膜血氧仪的临床应用提供了循证科学依据。 展开更多

【目的】研究精氨酸对妊娠环江香猪胎儿生长发育的影响。【方法】试验选取体重35—40 kg的环江后备母猪10头,单栏饲养,每栏1头。配种15 d后,随机分为2组,每组5头。试验组在基础日粮中添加0.83%L-精氨酸,对照组添加1.71%L-丙氨酸(等氮对... 【目的】研究精氨酸对妊娠环江香猪胎儿生长发育的影响。【方法】试验选取体重35—40 kg的环江后备母猪10头,单栏饲养,每栏1头。配种15 d后,随机分为2组,每组5头。试验组在基础日粮中添加0.83%L-精氨酸,对照组添加1.71%L-丙氨酸(等氮对照)。配种第45天,称取母猪体重,颈动脉放血处死母猪,收集血液,肝素抗凝,离心分离血浆;解剖母猪,记录胎儿数量、重量和存活率,采集羊水、尿囊液和胎盘,-20℃保存,用于实验室分析。【结果】结果表明,精氨酸组与丙氨酸组相比,胎儿平均个体重和黄体数分别提高14.4%(P<0.05)和6.0%(P>0.05),胎盘粗脂肪含量降低14.7%(P<0.05),羊水和尿囊液中精氨酸的浓度分别提高65.0%和79.1%(P<0.05),尿囊液中赖氨酸浓度降低28.3%(P<0.05);血浆和尿囊液中NO的含量分别提高22.9%和66.3%(P<0.05),胎盘中总NO合酶、诱导型NO合酶和结构型NO合酶的活性分别提高75.5%、61.7%和84.5%(P<0.05)。【结论】提示精氨酸通过促进胎盘一氧化氮的生成促进妊娠母猪早期胎儿的生长发育。 展开更多

【目的】研究两种饲粮对环江香猪结肠内容物菌群结构及其代谢物的影响。【方法】选用33.5±8.1 kg的环江香猪10头,随机分为两组,每组5头猪,单栏饲养,分别饲喂高营养水平饲粮(粗蛋白质含量为13.11%,消化能为14.73 MJ/kg)和低营养水... 【目的】研究两种饲粮对环江香猪结肠内容物菌群结构及其代谢物的影响。【方法】选用33.5±8.1 kg的环江香猪10头,随机分为两组,每组5头猪,单栏饲养,分别饲喂高营养水平饲粮(粗蛋白质含量为13.11%,消化能为14.73 MJ/kg)和低营养水平饲粮(粗蛋白质含量为9.77%,消化能为12.24 MJ/kg)。饲喂75 d后屠宰,每组选3头猪采集结肠内容物提取细菌总DNA,PCR扩增获得16S r RNA基因V4标签片段,用于Mi Seq高通量测序,基于QIIME平台比较结肠内容物菌群结构多样性;每组取5头猪的结肠内容物,利用气相色谱和液相色谱分析其中短链脂肪酸、氨氮、生物胺、吲哚和粪臭素的含量。【结果】硬壁菌门、拟杆菌门、螺旋体门和软壁菌门细菌为环江香猪结肠内容物中的优势菌门;饲粮组成不影响环江香猪结肠内容物菌群结构的多样性,高营养水平饲粮组结肠内容物中广古细菌含量显著低于、瘤胃球菌属和假丁酸弧菌属细菌含量显著高于低营养水平饲粮组;高营养水平饲粮组环江香猪结肠内容物中乙酸和丙酸含量显著低于、氨氮和尸胺含量显著高于低营养水平饲粮组。【结论】硬壁菌门、拟杆菌门、螺旋体门和软壁菌门细菌为环江香猪结肠内容物中的优势菌门,短期饲喂高营养水平饲粮可改变环江香猪结肠中的部分微生物含量及其代谢特性。 展开更多

以间歇性低压低氧方法建立阻塞性睡眠呼吸暂停综合征(Obstructive sleep apnea syndrome,OSAS)小型猪模型.8只小型猪编号后随机分为A、B、C组.A为对照组,置于双室低压氧仓二室,维持海平面压力;B、C组置于双室低压氧仓一室,仓内压力约53.... 以间歇性低压低氧方法建立阻塞性睡眠呼吸暂停综合征(Obstructive sleep apnea syndrome,OSAS)小型猪模型.8只小型猪编号后随机分为A、B、C组.A为对照组,置于双室低压氧仓二室,维持海平面压力;B、C组置于双室低压氧仓一室,仓内压力约53.9 KPa、氧浓度10%~11.2%.3组每天均入仓6 h.第13 d处死B组,A、C组第23 d同时处死.各组进仓前和处死前均行咽部CT扫描、口咽部呼吸压力、血氧饱和度检查;处死后取咽部组织行病理检查.咽部CT扫描示,咽部舌骨水平咽后壁厚度和侧壁厚度,进仓第13 d(8.8±1.1 vs 6.5±0.6,8.1±0.2 vs 6.3±0.6)和第23 d(9.2±1.2 vs 6.3±0.7,8.9±0.7 vs 6.4±0.5)均较进仓前有增加(P<0.05).舌骨水平左右径和软腭水平前后径进仓第13 d(7.6±1.4 vs 9.7±1.4,3.8±1.1 vs 6.5±1.3)和第23 d(6.4±1.6 vs9.3±1.5,4.3±0.9 vs 5.9±0.8)均较进仓前减小(P<0.05).舌骨后区前后径进仓第23 d后较进仓前减小(3.7±0.9 VS 6.4±0.6,P<0.05).进仓第23d口咽呼吸压力(0.0755 Mv)较进仓前(0.0658 Mv)明显增大(P<0.05),而第13 d(0.063 Mv)较进仓前变化不明显.血氧饱和度进仓第13 d(87%)和第23 d(88.5%)均较进仓前(96.3%)明显减小(P<0.05).病理变化:A组:咽部黏膜为复层扁平上皮,黏膜下层薄,肌层横纹清、肌间脂肪少.B组:咽部上皮组织增生伴角化,黏膜下层水肿纤维组织增生,肌层厚横纹不清、肌间脂肪灶性浸润;C组较B组病理变化程度加重.提示:间歇性低压低氧,使小型猪咽腔组织结构和力学特性发生类似OSAS病人的变化,本研究建立的OSAS小型猪模型,能用于人OSAS的进一步研究. 展开更多

本试验旨在研究高、低营养水平饲粮对妊娠母猪繁殖性能和体成分的影响,并探讨其生化机制。选用首次妊娠的环江香猪48头,根据体重随机分为2组,每组8个重复(栏),每重复3头。配种后分别饲喂NRC饲粮[消化能(DE)为14.73 MJ/kg,粗蛋白质(CP)... 本试验旨在研究高、低营养水平饲粮对妊娠母猪繁殖性能和体成分的影响,并探讨其生化机制。选用首次妊娠的环江香猪48头,根据体重随机分为2组,每组8个重复(栏),每重复3头。配种后分别饲喂NRC饲粮[消化能(DE)为14.73 MJ/kg,粗蛋白质(CP)含量为13.11%]和地方猪饲粮(DE为12.24 M J/kg,CP含量为9.77%)。分别于配种后45、75和110 d称取母猪体重,每栏选取1头母猪,前腔静脉采血用于生化参数分析,放血处死后解剖,记录胎猪头数,称取胎猪个体重,并测定母体成分。结果表明:与地方猪饲粮组相比,NRC饲粮组母猪的繁殖性能和体成分指标差异均不显著(P>0.05),妊娠45和75 d时血浆谷丙转氨酶以及妊娠75 d时谷草转氨酶活性均显著降低(P<0.05),妊娠45 d时低密度脂蛋白-胆固醇浓度,妊娠75 d时碱性磷酸酶活性、高密度脂蛋白-胆固醇和总胆固醇浓度以及整个妊娠期甘油三酯浓度均显著升高(P<0.05);在不同妊娠期,2种饲粮对各测定指标的影响有一定差异。由此可见,2种饲粮对环江香猪的繁殖性能和体成分均无显著影响;NRC饲粮在一定程度上改善了机体代谢,促进了生长发育,但同时增加了脂肪沉积。 展开更多

本试验旨在研究脯氨酸代谢对妊娠环江香猪繁殖性能的影响,并探讨其可能的生理生化机制。选取48头后备环江母猪,配种15 d后随机分成3组,每组8个重复,每个重复2头猪。各组分别饲喂添加0.77%L-丙氨酸(丙氨酸组)、1%L-脯氨酸(脯氨酸组)、0.7... 本试验旨在研究脯氨酸代谢对妊娠环江香猪繁殖性能的影响,并探讨其可能的生理生化机制。选取48头后备环江母猪,配种15 d后随机分成3组,每组8个重复,每个重复2头猪。各组分别饲喂添加0.77%L-丙氨酸(丙氨酸组)、1%L-脯氨酸(脯氨酸组)、0.77%L-丙氨酸+0.016 7%二氟甲基鸟氨酸(DFMO)(DFMO组)的试验饲粮。分别于配种第45和70天,每重复随机取1头母猪,放血处死,收集血液,用于生化参数分析;解剖母猪,记录黄体数、胎儿数和胎儿重。结果表明:妊娠第70天时,脯氨酸组窝重显著高于丙氨酸组(P<0.05);妊娠第45天时,脯氨酸组血浆谷丙转氨酶活性以及白蛋白浓度均显著低于丙氨酸组(P<0.05)。妊娠第70天与第45天相比,脯氨酸组血浆总胆固醇浓度、DFMO组血浆高密度脂蛋白和白蛋白浓度均显著降低(P<0.05)。其余指标各组间均无显著差异(P>0.05)。由此可见,饲粮添加脯氨酸可能通过改善机体氮代谢和脂肪代谢而促进妊娠环江香猪胎儿的生长发育。 展开更多
