健脾清热活血方对miR-222基因沉默转染结肠癌HCT-116细胞的生物学作用及MAPK10、PANK3表达的影响

目的:研究健脾清热活血方含药血清对稳定沉默miR-222结肠癌HCT-116细胞株生物学作用及丝裂原活化蛋白激酶10(MAPK10)、泛酸激酶3(PANK3)表达变化。方法:采用慢病毒介导的RNA干扰技术沉默miR-222基因,培养细胞分为:空白对照组(HCT-116)... 目的:研究健脾清热活血方含药血清对稳定沉默miR-222结肠癌HCT-116细胞株生物学作用及丝裂原活化蛋白激酶10(MAPK10)、泛酸激酶3(PANK3)表达变化。方法:采用慢病毒介导的RNA干扰技术沉默miR-222基因,培养细胞分为:空白对照组(HCT-116)、阴性对照组、阳性对照组(sh-miR-222)和各剂量治疗组(sh-miR-222+低、中、高剂量中药)。细胞增殖检测试剂比色法检测细胞增殖,流式细胞术检测细胞凋亡及周期,Transwell检测细胞迁移;透射电镜观察细胞自噬。RT-PCR及Western Blot检测MAPK10、PANK3 mRNA及蛋白表达。结果:与空白对照组和阳性对照组比较,治疗组抑制HCT-116细胞增殖和生长活力的效应明显加强(P<0.05),细胞总凋亡率显著升高,细胞迁移数显著下降(P<0.01)。治疗组的细胞胞浆内可见明显增多的自噬体,与阳性对照组比较,高剂量治疗组MAPK10、PANK3 mRNA表达量和低剂量治疗组MAPK10、PANK3蛋白表达量显著升高(P<0.01)。结论:健脾清热活血方联合miR-222基因沉默可增强诱导结肠癌HCT-116细胞凋亡和自噬,减少细胞增殖和迁移,MAPK10、PANK3表达显著升高。 展开更多

丽江猪MAPK10基因编码区分离、序列分子特征及组织差异表达分析

【目的】分离丽江猪丝裂原活化蛋白激酶10(mitogen-activated protein kinase,MAPK10)基因序列,并分析其组织mRNA和蛋白表达情况。【方法】采用PCR技术扩增丽江猪MAPK10基因,并利用生物信息学软件对获得的序列进行分析,实时荧光定量PCR... 【目的】分离丽江猪丝裂原活化蛋白激酶10(mitogen-activated protein kinase,MAPK10)基因序列,并分析其组织mRNA和蛋白表达情况。【方法】采用PCR技术扩增丽江猪MAPK10基因,并利用生物信息学软件对获得的序列进行分析,实时荧光定量PCR检测丽江猪脑、肝脏、心脏、脾脏、肺脏、肾脏、胰脏、背脂和背最长肌等9种组织中MAPK10基因的相对表达量;以大白猪为对照,实时荧光定量PCR和Western-blotting检测丽江猪与大白猪背脂中mRNA和蛋白表达差异。【结果】MAPK10基因序列总长1281 bp,编码426个氨基酸;MAPK10蛋白属亲水蛋白,无跨膜结构和信号肽,有44个磷酸化位点和2个糖基化位点;二级结构以α-螺旋和无规则卷曲为主;丽江猪MAPK10基因序列与人、牛、山羊、绵羊、鼠、狗、猴和鸡的同源性分别为95.3%、93.5%、93.2%、93.0%、92.6%、91.2%、90.5%和85.6%;MAPK10基因在丽江猪9个组织中差异表达,以脑中最高;丽江猪背脂MAPK10基因的mRNA表达量极显著低于大白猪(P<0.01),蛋白表达量也有低于大白猪的趋势(P>0.05)。【结论】MAPK10基因可作为研究丽江猪脂肪沉积性状的候选基因,结果可为研究猪脂肪沉积提供参考。 展开更多

目的 探讨促凋亡基因MAPK10启动子区CpG岛异常甲基化引起的表达沉默在B细胞淋巴瘤中的意义.方法 提取7个B细胞淋巴瘤细胞株的总RNA和基因组DNA.分别用半定量反转录PCR(RT-PCR)和甲基化特异性PCR(MSP)检测细胞株中MAPK10基因的mRNA... 目的 探讨促凋亡基因MAPK10启动子区CpG岛异常甲基化引起的表达沉默在B细胞淋巴瘤中的意义.方法 提取7个B细胞淋巴瘤细胞株的总RNA和基因组DNA.分别用半定量反转录PCR(RT-PCR)和甲基化特异性PCR(MSP)检测细胞株中MAPK10基因的mRNA表达水平和启动子甲基化状态.用MSP检测了24例弥漫大B细胞淋巴瘤(DLBCL)、15例滤泡性淋巴瘤(FL)和12例淋巴结反应性增生(LRH)中MAPK10甲基化状态.并进一步用亚硫酸氢盐-基因组测序法(BGS)验证了部分细胞株和组织标本中MAPK10的甲基化状态.结果 MAPK10在大部分B细胞淋巴瘤细胞株中存在表达沉默现象,并与其启动子区高甲基化密切相关.MAPK10基因高甲基化在DLBCL和FL中的频率分别为71%(17/24)及100%(15/15),而未在LRH标本中检出(0/12).BGS检测结果 证实了上述结果 .结论 MAPK10基因在B细胞淋巴瘤中存在高频甲基化,由其所致的基因沉默可能在B细胞淋巴瘤发生发展中起重要作用.MAPK10基因的肿瘤特异性甲基化可作为一个良好的肿瘤分子诊断标志物. 展开更多

长链非编码RNA对HepG2肝癌细胞增殖及凋亡的影响

目的:探讨长链非编码RNA RP13-514E23.1在肝癌细胞系中的表达,并观察上调该基因后其下游基因MAPK10的变化及对肝癌细胞Hep G2增殖、凋亡的影响。方法:通过荧光定量PCR(q RT-PCR)检测RP13-514E23.1在正常肝细胞与肝癌细胞系中的表达差异... 目的:探讨长链非编码RNA RP13-514E23.1在肝癌细胞系中的表达,并观察上调该基因后其下游基因MAPK10的变化及对肝癌细胞Hep G2增殖、凋亡的影响。方法:通过荧光定量PCR(q RT-PCR)检测RP13-514E23.1在正常肝细胞与肝癌细胞系中的表达差异,进一步构建质粒转染肝癌Hep G2细胞上调RP13-514E23.1的表达,以Mock组(只加转染试剂)和NC组(转染空载体pc DNA3.1-NC)作为对照评估转染效率及对MAPK10表达的影响。用CCK-8实验和流式细胞术检测转染前后Hep G2细胞的增殖、凋亡的变化。结果:Lnc RNA RP13-514E23.1在大部分肝癌细胞系(Hep G2,SMMC,Huh7,Hep3B)中的表达明显低于正常肝细胞(0.58±0.05 vs 1.00,P<0.05);转染pc DNA-RP13-514E23.1后,q RT-PCR检测Hep G2细胞的RP13-514E23.1和MAPK10m RNA表达量显著升高(分别为29.90±1.40、2.42±0.25,P<0.05),western blot检测MAPK10蛋白表达量较对照组也升高(2.10±0.16,P<0.05);CCK-8结果显示Hep G2细胞在各个时间段增殖均受到抑制(P<0.01),Mock组、NC组和实验组的凋亡率分别为(5.53±1.17)%、(6.40±2.84)%和(46.87±3.45)%(P<0.01)。结论:Lnc RNA RP13-514E23.1在肝癌细胞系中表达异常降低,上调其表达后MAPK10的表达升高,且Hep G2细胞增殖受到抑制、凋亡增加。 展开更多

Gastrodin, the major component isolated from the rhizome of the Chinese traditional medicinal herb Gastrodia elata(‘‘Tianma''), has a long history in the treatment of epilepsy and other neurological disorders.However, the molecular mechanisms are not clear. Here,we found that gastrodin ameliorated pentylenetetrazole(PTZ)-induced epileptic seizures with improvement of the electroencephalographic pattern in mice. Further studies demonstrated that gastrodin decreased the levels of the pro-inflammatory cytokines interleukin-1b and tumor necrosis factor-a while increasing interleukin-10, an antiinflammatory cytokine in the brain. Furthermore, gastrodin attenuated the PTZ-induced microglial activation along with inhibition of mitogen-activated protein kinases,c AMP response element binding protein, and NF-j B. Our data suggest that gastrodin attenuates seizures by modulating the mitogen-activated protein kinase-associated inflammatory responses. 展开更多

Objective:To investigate the expressions of MAPK10,c-Jun and Itga6 in laryngeal carcinoma and its influence on the sensitivity to docetaxel,cisplatin and 5-fluorouracil(TPF)chemotherapy.Methods:Fifty-seven patients with supraglottic squamous cell carcinoma,who were treated by two cycles of TPF induction chemotherapy in our hospital,were enrolled in this study and divided into groups by chemotherapy resistance or chemotherapy sensitivity.The expressions of mRNA and protein of MAPK10,c-Jun and Itga6 in tumor tissues were evaluated by immunohistochemistry.The consistency of mRNA and protein expressions was tested,and the relation with the clinicopathological features was analyzed.Results:The positive rates of MAPK10 andc-Jun in the tumor tissues of the sensitive group were significantly higher than those of there assistant group,which was 90.48%and 100.00%,respectively.The expression rate of Itga6 was significantly higher in the resistant group,which was 83.33%(P<0.05).The mRNA levels of MAPK10 and c-Jun were significantly lower in the resistant group than in the sensitive group,whilethemRNA levelof Itga6was significantly higher in the resistant group(P<0.05).The protein expressions of MAPK10,c-Jun and Itga6 were consistent with their mRNA expressions(P<0.05).The expressions of MAPK10,c-Jun and Itga6 were not correlatedwithage,gender and tumor diameter(P>0.05).However,the expressions of MAPK10 and c-Jun were negatively correlated withclinical stage and pathological grading(P<0.05).Negative correlations between MAPK 10 and Itga6,and between c-Jun and Itga6in tumor tissues were found by Spearman’srank correlation coefficient(P<0.05).The correlation was also negative in the resistant tumor tissues(P<0.05).Conclusion:The MAPK10 and c-Jun expressions were down-regulated,while the Itga6 expression was up-regulated in the chemo-resistant laryngeal carcinoma,and the expression levels of different factors were correlated witheach other.These factorsmight be important biomarkers for predicting outcomes of TPF chemotherapy in laryngeal carcinoma in the future. 展开更多
