Background:Natural astaxanthin(ASTA)has strong antioxidant properties and has been widely used as a health product to improve human health.However,the effects of ASTA on the reproductive performance of aging roosters ...Background:Natural astaxanthin(ASTA)has strong antioxidant properties and has been widely used as a health product to improve human health.However,the effects of ASTA on the reproductive performance of aging roosters have been poorly studied.We aimed to investigate the effects of dietary ASTA on semen quality and antioxidant capacity in aging roosters and to explore the potential mechanism of semen quality change via antioxidation defense system.Methods:In the present study,9653-week-old Jinghong No.1 layer breeder roosters were fed a corn-soybean meal basal diet containing 0,25,50,or 100 mg/kg ASTA for 6 weeks.Results:Semen quality in the ASTA groups remarkably improved than that in the control group,and antioxidant activities,the abilities to scavenge hydroxyl radicals and superoxide anions,increased gradually with ASTA addition(P<0.05).In addition,the mRNA levels of antioxidant enzymes as well as the mRNA and protein levels of the mitogen-activated protein kinase(MAPK)and nuclear factor-erythroid 2-related factor 2(Nrf2)were markedly increased in the 50-100 mg/kg ASTA group(P<0.05).Conclusions:Collectively,these results demonstrate that dietary ASTA may improve semen quality by increasing antioxidant enzyme activities and the ability to scavenge hydroxyl radicals,which may be related to upregulation of the MAPK/Nrf2 pathway.展开更多

Vahl, Lianqiao in Chinese, is one of the most fundamental herbs in traditional Chinese medicine (TCM) with heat-clearing and detoxicating properties. In this study, we aimed to study the antitumor activity of Lianqiao aqueous extract against melanoma using cancer cell line-based in vitro and mouse allografl tumor in vivo models. Furthermore, we also investigated the underlying molecular mechanisms, par- ticularly the involvement of anti-inflammation and anti-oxidation properties in its antitumor activity. Methods The proliferation of cancer cells was measured by MTT assay. The transplanted B16-F10 melanoma in C57BL/6 mice were established and used for the evaluation of in vivo antitumor effect of LQ. Tumor growth was monitored twice a week. Ki67 and CD31 were used to detect cancer cell proliferation and angiogenesis in tumor, respectively. The anti-oxidative property of LQ was determined by measuring the levels of ROS, MDA and GSH. The anti-inflamma- tory effect of LQ was evaluated by measuring TNF-α and IL-6 using ELISA kits. Other protein expression was deter- mined by Western Blot. Results LQ strongly inhibited the growth of B16-F10 cells in vitro and the tumor growth in vivo. The survival time of tumor-bearing mice was significantly prolonged by LQ. LQ inhibited cancer cell prolif- eration and angiogenesis in tumor as evidenced by decreased expressions of Ki67 and CD31. Levels of ROS, MDA TNF-α and IL-6 decreased, while GSH increased in LQ treatment group, indicating a strong anti-oxidative and an- ti-inflammatory activity of LQ. The expression of antioxidant proteins Nff-2 and HO-1, tumor suppressors P53 and p-PTEN, and the MAPK pathways in tumor tissues were upregulated by LQ treatment. Conclusions LQ exhibited strong antitumor activity against B16-F10 murine melanoma both in vitro and in vivo. However, the effect and mechanism of CO_2 stunning on meat color and lipid peroxidation during long-term storage remain poorly studied. We aimed to study the effects of GS methods, especial y CO_2 concentration, on meat color and meat lipid peroxidation in broilers during long-term storage at 4 °C and to explore the potential mechanism of meat color change via lipid peroxidation and the inner lipid peroxide scavenging system.Methods: Eighteen broilers were sacrificed after exposure to one of the following gas mixtures for 90 s: 40% CO_2+21% O_2+ 39% N2(G40%), 79% CO_2+ 21% O_2(G79%), or no stunning(0% CO_2, control). Meat color, serum variables,enzyme activities, and the gene expression of mitogen-activated protein kinase(MAPK), nuclear factor-erythroid2-related factor 2(Nrf2), glutathione S-transferase(GST) and superoxide dismutase(SOD) were determined.(Continued on next page)Results: The concentrations of serum triiodothyronine(T3, P = 0.03) and the ratio of serum free triiodothyronine/free thyroxine(FT3/FT4, P < 0.01) were decreased, whereas levels of serum cortisol(P < 0.01) were increased in the 40%CO_2 group compared with the control group. Additionally, the thiobarbituric acid-reactive substances(TBARS)3 d(P < 0.01) and TBARS6 d(P = 0.01) in breast meat and the TBARS3 din thigh meat(P < 0.01) were increased in the40% CO_2 group compared with the control group. Serum T3 was negatively correlated with TBARS6 dboth in the breast and thigh meat(r =-0.63, P < 0.01 and r =-0.47, P = 0.05 respectively). T3/T4 was negatively correlated with TBARS6 din the breast meat and in the thigh meat(r =-0.57, P = 0.01; and r =-0.53, P = 0.03 respectively). Compared with the control group, Lightness(L*)1 d(P = 0.03) and L*9 d(P < 0.01) were increased, whereas total chromatic aberration(E*)1 d(P = 0.05) and E*3 d(P < 0.01) were decreased in the breast meat of both the G40% and G79% groups. The values of yel owness(b*)3 d(P = 0.01), b*6 d(P < 0.01) and E*6 d(P < 0.01) in the thigh meat were lower in both the G40% and G79% groups than in the control group. In the breast muscle, the m RNA levels of c-Jun N-terminal kinase 2(JNK2, P = 0.03),GSTT1(P = 0.04), and SOD1(P = 0.05) were decreased, and the m RNA levels of JNK1(P = 0.07), Nrf2(P = 0.09), and GSTA3(P = 0.06) were slightly lower in both the G40% and G79% groups compared with the control group. However, among these genes, only the m RNA level of JNK1 was decreased in the G40% group compared with the control group and the G79% group(P = 0.03) in the thigh muscle.Conclusions: Compared with the control group, meat color quality in the breast meat was decreased, and the expression of genes in the MAPK/Nrf2/ARE(antioxidant responsive element) antioxidant pathway in breast muscle was partly suppressed by GS of both 40% and 79% CO_2. However, oxidative stress and meat lipid peroxidation during storage were aggravated by GS with 40% CO_2 compared to GS with 79% CO_2 and no GS.展开更多

文摘目的探讨槲皮素诱导Ⅰ型血红素氧化酶(HO-1)对肝细胞酒精性氧化损伤的保护效应及相关信号通路。方法乙醇(100 mmol/L)孵育的人原代肝细胞经槲皮素(100μmol/L)处理24h后,测定细胞HO-1活性、Nrf2转位表达及细胞相应的氧化损伤程度;在此基础上,联合应用HO-1及各(MAPK)通路抑制剂,观察上述指标的变化。结果槲皮素处理使HO-1进一步升高,并明显减轻乙醇孵育所导致的细胞GSH耗竭、MDA升高及胞内门冬氨酸转氨酶(AST)与乳酸脱氢酶(LDH)的释放;HO-1诱导剂血红素也具有类似的效应,而HO-1抑制剂锌原卟啉Ⅸ及p38与细胞外信号调节激酶(extracellular regulated protein kinase,ERK)抑制剂(SB203580与PD98059)则明显抑制了Nrf2的转位活化及槲皮素的保护效应。结论槲皮素通过诱导HO-1保护肝细胞免受酒精性氧化损伤,其信号通路与p38及ERK活化促使Nrf2转位入核启动HO-1表达有关。
